1PARylation regulates stress granule dynamics, phase separation, and neurotoxicity of disease-related RNA-binding proteins显示文摘Mutations in RNA-binding proteins (RBPs) localized in ribonucleoprotein (RNP) granules, such as hnRNP A1 and TDP-43, promote aberrant protein aggregation, which is a pathological hallmark of various neurodegenerative diseases, such as amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). Protein posttranslational modifications (PTMs) are known to 佗gulate RNP granules. In this study, we investigate the function of poly(ADP-ribosyl)ation (PARylation), an important PTM involved in DNA damage repair and cell death, in RNP granule-related neurodegeneration. We reveal that PARylation levels are a major regulator of the assembly-disassembly dynamics of RNP granules containing disease-related RBPs, hnRNP A1 and TDP-43. We find that hnRNP A1 can both be PARylated and bind to PARylated proteins or poly(ADP-ribose)(PAR). We further uncover that PARylation of hnRNP A1 at K298 controls its nucleocytoplasmic transport, whereas PAR-binding via the PAR-binding motif (PBM) of hnRNP Al regulates its association with stress granules. Moreover, we reveal that PAR not only dramatically enhances the liquid-liquid phase separation of hnRNP A1, but also promotes the co-phase separation of hnRNP A1 and TDP-43 in vitro and their interaction in vivo. Finally, both genetic and pharmacological inhibition of PARP mitigates hnRNP Al- and TDP-43-mediated neurotoxicity in cell and Drosophila models of ALS. 2Comprehensive analysis of NAC transcription factors in diploid Gossypium: sequence conservation and expression analysis uncover their roles during fiber development显示文摘Determining how function evolves following gene duplication is necessary for understanding gene expansion.Transcription factors(TFs)are a class of proteins that regulate gene expression by binding to specific cis-acting elements in the promoters of target genes,subsequently activating or repressing their transcription.In the present study,we systematically examined the functional diversification of the NAC transcription factor(NAC-TFs)family by analyzing their chromosomal location,structure,phylogeny,and expression pattern in Gossypium raimondii(Gr)and G.arboreum(Ga).The 145 and 141 NAC genes identified in the Gr and Ga genomes,respectively,were annotated and divided into 18 subfamilies,which showed distinct divergence in gene structure and expression patterns during fiber development.In addition,when the functional parameters were examined,clear divergence was observed within tandem clusters,which suggested that subfunctionalization had occurred among duplicate genes.The expression patterns of homologous gene pairs also changed,suggestive of the diversification of gene function during the evolution of diploid cotton.These findings provide insights into the mechanisms underlying the functional differentiation of duplicated NAC-TFs genes in two diploid cotton species.Haihong Shang Zhongna Wang Changsong Zou Zhen Zhang Weijie Li Junwen Li Yuzhen Shi Wankui Gong Tingting Chen Aiying Liu Juwu Gong Qun Ge Youlu Yuan 2016Science China(Life Sciences)2016,59,2:12
3Influence of Substrate Negative Bias on Structure and Properties of TiN Coatings Prepared by Hybrid HIPIMS Method显示文摘TiN coatings were deposited using a hybrid home-made high power impulse magnetron sputtering(HIPIMS)technique at room temperature.The effects of substrate negative bias voltage on the deposition rate,composition,crystal structure,surface morphology,microstructure and mechanical properties were investigated.The results revealed that with the increase in bias voltage from-50 to-400 V,TiN coatings exhibited a trend of densification and the crystal structure gradually evolved from(111) orientation to(200)orientation.The growth rate decreased from about 12.2 nm to 7.8 nm per minute with the coating densification.When the bias voltage was-300 V,the minimum surface roughness value of 10.1 nm was obtained,and the hardness and Young's modulus of TiN coatings reached the maximum value of 17.4 GPa and 263.8 GPa,respectively.Meanwhile,the highest adhesion of 59 N was obtained between coating and substrate.Zhenyu Wang Dong Zhang Peiling Ke Xincai Liu Aiying Wang 2015Journal of Materials Science & Technology2015,31,1:10
4Correlation between BRAF^(V600E) mutation and clinicopathological features in pediatric papillary thyroid carcinoma显示文摘In adults, the presence of the BRAF^(V600E) mutation in papillary thyroid cancer(PTC) has been demonstrated to be strongly associated with aggressive cancer-cell characteristics and poor patient prognosis. In contrast, the frequency of this mutation in pediatric PTC has undergone limited study, and the few available estimates range from 0 to 63%. Furthermore, the role of the BRAF^(V600E) mutation in pediatric PTC is controversial; thus, the present study aimed to investigate the prevalence and role of the BRAF^(V600E) mutation in48 pediatric patients with PTC, aged 3–13 years. Of these patients, 41 were diagnosed with classic PTC, five were found to have a follicular variant of PTC, and two to exhibit a diffuse sclerosing PTC variant. The BRAF^(V600E) mutation was identified to be present in 35.4% of the 48 analyzed patients, and in 41.5% of the patients diagnosed with classical PTC. Furthermore, the presence of the BRAF^(V600E) mutation was found to be associated with a patient age at diagnosis of less than ten years(P=0.011), the performance of a thyroidectomy(P=0.03), exhibited tumor multifocality(P=0.02) and/or extra-thyroidal invasion(P=0.003), and both a low MACIS(Metastases, Age, Completeness of resection, Invasion, Size)(P=0.036) and AMES(Age, Metastasis, Extent of tumor,Size)(P=0.001)score. 5Manipulation of SlMXl for enhanced carotenoids accumulation and drought resistance in tomato显示文摘Plants are the ultimate source of nutrients in the human diet. To ensure adequate availability of high quality food for an increasing world population, traits including improved tolerance of stresses and nutrient levels need to be selected in crops, both individually and in combination. Here we report the identification of Sl MX1 encoding a MIXTA-like MYB transcription factor in tomato that simultaneously modulates drought resistance and metabolic processes through regulating key structural and regulatory genes of the corresponding pathways. Over-expression of Sl MX1 results in substantially increased drought tolerance and improved fruit quality, while knocking down Sl MX1 resulted in the opposite phenotypes. 6Constructing a high-density linkage map for Gossypium hirsutum × Gossypium barbadense and identifying QTLs for lint percentage显示文摘To introgress the good fiber quality and yield from Gossypium barbadense into a commercial Upland cotton variety, a high-density simple sequence repeat(SSR) genetic linkage map was developed from a BC1F1 population of Gossypium hirsutum×Gossypium barbadense. The map comprised 2,292 loci and covered 5115.16 centi Morgan(c M) of the cotton AD genome, with an average marker interval of 2.23 c M. Of the marker order for 1,577 common loci on this new map, 90.36% agrees well with the marker order on the D genome sequence genetic map. Compared with five published high-density SSR genetic maps, 53.14% of marker loci were newly discovered in this map. Twenty-six quantitative trait loci(QTLs) for lint percentage(LP) were identified on nine chromosomes. Nine stable or common QTLs could be used for marker-assisted selection. Fifty percent of the QTLs were from G. barbadense and increased LP by 1.07%–2.41%. 73D MHD simulation of the double-gradient instability of the magnetotail current sheet显示文摘Flapping motion of the current sheet(CS) is an important physical process in the Earth's magnetotail. The magnetic doublegradient model, which includes both the instability and wave modes, offers a reasonable explanation for the exciting and propagation of the flapping wave. In this paper, we apply an advanced numerical magnetohydrodynamic(MHD) scheme(conservation element and solution element(CESE)-MHD) to simulate the magnetic double-gradient instability in an idealized current sheet that mimics the magnetotail configuration. We initialize the simulations with a numerically relaxed magnetotail equilibrium, in which the normal component of the magnetic field has a tailward gradient. It is confirmed in our simulation that the instability develops in the current layer. The growth rate of the instability yielded from the simulation is very close to the prediction of theory, with a relative deviation of only ten percent. 8Phase orientation improved the corrosion resistance and conductivity of Cr_(2)AlC coatings for metal bipolar plates显示文摘In view of the M_(n+1)AX_(n)(MAX)phase coatings benefting the adaptive passivation flm for good corrosion resistance and high electronic density of states for excellent electrical conductivity,here,we reported the Cr_(2)Al C MAX phase coatings with different preferred orientations by a homemade technique consisting of vacuum arc and magnetron sputtering.The dependence of surface and interface microstructural evolution upon the corrosion and electrochemical properties of deposited coating was focused.Results showed that all the Cr_(2)Al C coatings with different phase orientations greatly improved the performance of stainless steel(SS)316 L substrate.Specifcally,the lowest value of interface contact resistance(ICR)reached to 3.16 mΩcm^(2)and the lowest corrosion current density was 2×10^(-2)μA cm^(-2),which were much better than those of bare SS316L.The combined studies of electrochemical properties and theoretical calculations demonstrated that the Cr_(2)Al C coatings with preferred(103)orientation were easier to form oxide passivation flm on their surface to increase the corrosion resistance.Guanshui Ma Dong Zhang Peng Guo Hao Li Yang Xin Zhenyu Wang Aiying Wang 2022Journal of Materials Science & Technology2022,,10:2
9Microstructure and Tribological Behavior of Self-lubricating(Si:N)-DLC/MAO Coatings on AZ80 Magnesium Substrate显示文摘The combined micro arc oxidation(MAO)and a hybrid beam deposition process was used to deposit duplex(Si:N)-DLC/MAO coatings on AZ80 magnesium alloy.The microstructure and composition of the duplex coatings were analyzed by Raman spectroscopy,X-ray photoelectron spectroscope(XPS),scanning electron microscope(SEM)and atomic force microscopy(AFM).Tribological behaviors of the coatings were studied by ball-on-disk friction test.It was found that the ID/IG ratio of the(Si:N)-DLC(diamond-like carbon)top films increases with decreasing C2H2/N2ratio.The(Si:N)-DLC top film with Si3N4was formed on the MAO coated sample as the C2H2/N2ratio was 10sccm:5sccm,which showed an increasing critical load compared with the pure DLC directly deposited on the Mg alloy substrate.As a result,the(Si:N)-DLC/MAO coating exhibited an advanced wear protection for the substrate.Wei YANG Zhennan DENG Dong ZHANG Peiling KE Aiying WANG 2013Acta Metallurgica Sinica(English Letters)2013,26,6:2

13QTL mapping for plant height and fruit branch number based on RIL population of upland cotton显示文摘Background:Plant height(PH)and fruit branch number(FBN)are important traits for improving yield and mechanical harvesting of cotton.In order to identify genes of PH and FBN in cotton germplasms to develop superior cultivars,quantitative trait loci(QTLs)for these traits were detected based on the phenotypic evaluation data in nine environments across four locations and 4 years and a previously reported genetic linkage map of an recombinant inbred line(RIL)population of upland cotton.Results:In total,53 QTLs of PH and FBN,were identified on 21 chromosomes of the cotton genome except chromosomes c02,c09-c11,and c22.For PH,27 QTLs explaining 3.81%–8.54%proportions of phenotypic variance were identified on 18 chromosomes except c02,c08-c12,c15,and c22.For FBN,26 QTLs explaining 3.23%–11.00%proportions of phenotypic variance were identified on 16 chromosomes except c02-c03,c06,c09-c11,c17,c22-c23,and c25.Eight QTLs were simultaneously identified in at least two environments.Three QTL clusters containing seven QTLs were identified on three chromosomes(c01,c18 and c21).Eleven QTLs were the same as previously reported ones,while the rest were newly identified.Conclusions:The QTLs and QTL clusters identified in the current study will be helpful to further understand the genetic mechanism of PH and FBN development of cotton and will enhance the development of excellent cultivars for mechanical managements in cotton production.LIU Ruixian XIAO Xianghui GONG Juwu LI Junwen ZHANG Zhen LIU Aiying LU Quanwei SHANG Haihong SHI Yuzhen GE Qun IQBAL Muhammad Sajid CHEN Quanjia YUAN Youlu GONG Wankui 2020Journal of Cotton Research2020,3,1:1

18Strong restrictions on the trait range of co-occurring species in the newly created riparian zone of the three Gorges Reservoir Area, china显示文摘Aims community assembly links plant traits to particular environmental conditions.Numerous studies have adopted a trait-based approach to understand both community assembly processes and changes in plant functional traits along environmental gradients.In most cases these are long-established,natural or semi-natural environments.However,increasingly human activity has created,and continues to create,a range of new environmental conditions,and under-standing community assembly in these‘novel environments’will be increasingly important.Methods Built in 2006,the three Gorges Dam,largest hydraulic project in china,created a new riparian area of 384 km^(2),with massively al-tered hydrology.this large,newly created ecosystem is an ideal platform for understanding community assembly in a novel environment.We sampled environment variables and plant communities within 103 plots located in both the reservoir riparian zone(RRZ)and adjacent non-flooded and semi-natural upland(Upland)at the three Gorges Reservoir Area.We measured six traits from 168 plant species in order to calculate community-level distribution of trait values.We expected that the altered hydrology in RRZ would have a profound effect on the community assembly process for the local plants.Important Findingsconsistent with previous work on community assembly,the dis-tribution of trait values(range,variance,kurtosis and the standard deviation of the distribution neighbor distances)within all plots was significantly lower than those from random distributions,indicat-ing that both habitat filtering and limiting similarity simultaneously shaped the distributions of traits and the assembly of plant commu-nities.considering the newly created RRZ relative to nearby sites,community assembly was different in two main ways.First,there was a large shift in the mean trait values.compared to Upland commu-nities,plant communities in the RRZ had higher mean specific leaf area(SLA),higher nitrogen per unit leaf mass(Nmass),and lower max-imum height(MH).Second,in the RRZ compared to the Upland,for the percentage of individual plots whose characteristic of trait values was lower than null distributions,the reductions in the community-level range for SLA,Nmass,nitrogen per unit leaf area(Narea)and phos-phorus per unit leaf area(Parea)were much larger,suggesting that the habitat filter in this newly created riparian zone was much stronger compared to longer established semi-natural upland vegetation.this stronger filter,and the restriction to a subset of plants with very similar trait values,has implications for predicting riparian ecosystems’responses to the hydrological alterations and further understanding for human’s effect on plant diversity and plant floras.Aiying Zhang Will Cornwell Zhaojia Li Gaoming Xiong Dayong Fan Zongqiang Xie 2019Journal of Plant Ecology2019,12,5:0
19Evolution of pectin synthesis relevant galacturonosyltransferase gene family and its expression during cotton fiber development显示文摘Background:Pectin is a key substance involved in cell wall development,and the galacturonosyltransferases(GAUTs)gene family is a critical participant in the pectin synthesis pathway.Systematic and comprehensive research on GAUTs has not been performed in cotton.Analysis of the evolution and expression patterns of the GAUT gene family in different cotton species is needed to in crease kno wledge of the functi on of pectin in cotto n fiber development.Results:In this study,we have identified 131 GAUT genes in the genomes of four Gossypium species(G.raimondii,G barbadense,G.hirsutum,and G.arboreum),and classified them as GAUT-A,GAUT-B and GAUT-C,which coding probable galacturonosyltransferases.Among them,the GAUT genes encode proteins GAUT1 to GAUT15.All GAUT proteins except for GAUT7 contai n a con served glycosyl transferase family 8 domain(H-DN-A-SW-S-V-H-T-F).The conserved sequence of GAUT7 is PLN(phospholamban)02769 domain.According to c/s-elemet analysis,GAUT genes transcript levels may be regulated by horm ones such as JA,GA,SA,ABA,Me-JA,and IA A.The evoluti on and transcription patterns of the GAUT gene family in different cotton species and the transcript levels in upland cotton lines with different fiber st「ength were analyzed.Peak transcript level of GhGAUT genes have been observed before 15 DPA.In the six materials with high fiber strength,the transcription of GhGAUT genes were concentrated from 10 to 15 DPA;while the highest transcript levels in low fiber st「ength materials were detected between 5 and 10 DPA.These results lays the foundation for future research on gene function during cotton fiber development.Conclusions:The GAUT gene family may affect cotton fiber development,including fiber elongation and fiber thickening.In the low strength fiber lines,GAUTs mainly participate in fiber elongation,whereas their major effect on cotton with high strength fiber is related to both elongation and thickening.FAN Senmiao LIU Aiying ZOU Xianyan ZHANG Zhen GE Qun GONG Wankui LI Junwen GONG Juwu SHI Yuzhen DENG Xiaoying JIA Tingting YUAN Youlu SHANG Haihong 2021Journal of Cotton Research2021,4,3:0
20A Method of Peak Detecting for Nanosecond Pulse in Optical Performance Monitoring System显示文摘In optical performance monitoring system,the analog to digital converter is needed to detect the peak of nanosecond pulse and get the signal envelope.A scheme based on a designed anti-aliasing filter and analog to digital converter is proposed to broaden the nanosecond pulse and make it easier for the analog to digital converter to catch the peak of the nanosecond pulse.The experimental results demonstrate that,with the proposed scheme,the optical performance system needs less time to get the recovered eye-diagram of high speed optical data signal,and is robust to phase mismatch in the analog to digital converter circuit.FENG Lihui ZHANG Qing LIU Xiang ZUO Lin YANG Aiying 2014China Communications2014,11,2:0
