| 1 | Frequent Mutation of rs13281615 and Its Association with PVT1 Expression and Cell Proliferation in Breast Cancer显示文摘The q24 band of chromosome 8(8q24)is frequently amplified in human cancers including breast cancer,and several SNPs(single nucleotide polymorphisms)at 8q24,including rs13281615,have been identified for their association with cancer risks.These SNPs are in a'gene desert'region,and their functions in cancer development remain to be illustrated,although several of the SNPs appear to influence the genes in the'desert'in a long-range manner,including the v-myc avian myelocytomatosis viral oncogene homolog(MYC)and the nonprotein coding plasmacytoma variant translocation 1(PVT1),both of which have been implicated in human cancers.In the current study,we examined rs13281615 for its potential role in breast cancer using normal and cancer tissues from 121 Chinese women with breast cancer.In addition to confirming the association of the GG genotype of rs13281615 with breast cancer risk,we found that germline GG genotype was significantly associated with estrogen receptor(ER)positivity,higher tumor grade and higher proliferation index.We also found frequent somatic mutations(22/121 or 18.2%)of this SNP in breast cancer.Interestingly,the majority of the mutations(17/22 or 77%)involved a G/A change,resulting in a decrease in the number of cancers with the GG risk genotype and subsequent loss of GG association with higher tumor grade and proliferation index in cancers.Furthermore,PVT1 expression was increased in cancers,and the increase was associated with the GG genotype of rs13281615.These results suggest that the GG genotype of SNP rs13281615 plays a role in breast cancer likely by influencing PVT1 expression,and that during oncogenesis,'protective'mutations could occur. | Zhiqian Zhang Zhengmao Zhu Baotong Zhang Weidong Li Xin Li Xiao Wu Lijuan Wang Liya Fu Li Fu Jin-Tang Dong | 2014 | Journal of Genetics and Genomics2014,41,4: | 12 |

| 4 | Genomic landscapes of Chinese sporadic autism spectrum disordersrevealed by whole-genome sequencing显示文摘Autism spectrum disorder (ASD) is a neurodevelopmental disorder with considerable clinical and genetic heterogeneity.In this study,we identified all classes of genomic variants from whole-genome sequencing (WGS) dataset of 32 Chinese trios with ASD,including de novo mutations,inherited variants,copy number variants (CNVs) and genomic structural variants.A higher mutation rate (Poisson test,P<2.2×10^(-16)) in exonic (1.37×10^(-8)) and 3'-UTR regions (1.42×10^(-8)) was revealed in comparison with that of whole genome (1.05×10^(-8)).Using an integrated model,we identified 87 potentially risk genes (P<0.01) from 4832 genes harboring various rare deleterious variants,including CHD8 and NRXN2,implying that the disorders may be in favor to multiple-hit.In particular,frequent rare inherited mutations of several microcephaly-associated genes (ASPM,WDR62,and ZNF335)were found in ASD.In chromosomal structure analyses,we found four de novo CNVs and one de novo chromosomal rearrangement event,including a de novo duplication of UBE3A-containing region at 15q11.2-q13.1,which causes Angelman syndrome and microcephaly,and a disrupted TNR due to de novo chromosomal translocation t (1;5) (q25.1;q33.2).Taken together,our results suggest that abnormalities of centrosomal function and chromatin remodeling of the microcephaly-associated genes may be implicated in pathogenesis of ASD.Adoption of WGS as a new yet efficient technique to illustrate the full genetic spectrum in complex disorders,such as ASD,could provide novel insights into pathogenesis,diagnosis and treatment. | Jinyu Wu Ping Yu Xin Jin Xiu Xu Jinchen Li Zhongshan Li Mingbang Wang Tao Wang Xueli Wu Yi Jiang Wanshi Cai Junpu Mei Qingjie Min Qiong Xu Bingrui Zhou Hui Guo Ping Wang Wenhao Zhou Zhengmao Hu Yingrui Li Tao Cai Yi Wang Kun Xia Yong-Hui Jiang Zhong Sheng Sun | 2018 | Journal of Genetics and Genomics2018,45,10: | 3 |
| 5 | Modulation response performances of a Fabry-Perot semiconductor laser subjected to light injection from another Fabry-Perot semiconductor laser显示文摘The modulation response characteristics of a Fabry-Perot semiconductor laser(slave FP-LD) subjected to light injection from another Fabry-Perot semiconductor laser(master FP-LD) have been investigated theoretically.The results show that the modulation response performances of the slave FP-LD depend on the light injection strength,the central mode frequency detuning Δf and the mode spacing difference Δλ between the slave FP-LD and the master FP-LD.With the increase of the light injection strength from the master FP-LD,the 3 dB modulation bandwidth of the slave FP-LD will be enhanced;however,if the injection strength is increased to certain degree,the front side of the relaxation oscillation peak will be reduced to below 3 dB,which results in the rapid decrease of the 3 dB modulation bandwidth.With the increase of Δf,for a relatively small injection strength level,the 3 dB modulation bandwidth will behave a monotonous enlargement until the slave FP-LD operates in a period-one state(P1).For a relatively strong optical injection level,the slave FP-LD has a broad injection-locked frequency detuning area.Within the injection-locked area,with the increase of frequency detuning Δf,the 3dB modulation bandwidth increases at first and then decreases after experiencing a maximum value.For given light injection strength and frequency detuning,the 3 dB modulation bandwidth of the slave FP-LD has two maximums and behaves a symmetrical distribution with the change of Δλ.By choosing reasonable parameters,the 3 dB modulation bandwidth can be significantly improved.For the parameter values selected in this paper,the 3 dB modulation bandwidth can increase 5.5 times compared with the case of free running. | XIA GuangQiong WU ZhengMao YANG Qi LIN XiaoDong | 2009 | Chinese Science Bulletin2009,54,20: | 3 |
| 6 | Nanoporous membrane device for ultra high heat flux thermal management显示文摘High power density electronics are severely limited by current thermal management solutions which are unable to dissipate the necessary heat flux while maintaining safe junction temperatures for reliable operation.We designed,fabricated,and experimentally characterized a microfluidic device for ultra-high heat flux dissipation using evaporation from a nanoporous silicon membrane.With~100 nm diameter pores,the membrane can generate high capillary pressure even with low surface tension fluids such as pentane and R245fa.The suspended ultra-thin membrane structure facilitates efficient liquid transport with minimal viscous pressure losses.We fabricated the membrane in silicon using interference lithography and reactive ion etching and then bonded it to a high permeability silicon microchannel array to create a biporous wick which achieves high capillary pressure with enhanced permeability.The back side consisted of a thin film platinum heater and resistive temperature sensors to emulate the heat dissipation in transistors and measure the temperature,respectively.We experimentally characterized the devices in pure vaporambient conditions in an environmental chamber.Accordingly,we demonstrated heat fluxes of 665±74 W/cm^(2) using pentane over an area of 0.172mm×10mm with a temperature rise of 28.5±1.8 K from the heated substrate to ambient vapor.This heat flux,which is normalized by the evaporation area,is the highest reported to date in the pure evaporation regime,that is,without nucleate boiling.The experimental results are in good agreement with a high fidelity model which captures heat conduction in the suspended membrane structure as well as non-equilibrium and sub-continuum effects at the liquid–vapor interface.This work suggests that evaporative membrane-based approaches can be promising towards realizing an efficient,high flux thermal management strategy over large areas for high-performance electronics. | Daniel F.Hanks Zhengmao Lu Jay Sircar Todd R.Salamon Dion S.Antao Kevin R.Bagnall Banafsheh Barabadi Evelyn N.Wang | 2018 | Microsystems & Nanoengineering2018,4,1: | 3 |

| 8 | Zinc finger factor ZNF121 is a MYC-interacting protein functionally affecting MYC and cell proliferation in epithelial cells显示文摘MYC is a potent oncoprotein that modulates multiple cellular processes including proliferation,apoptosis, differentiation, stemness, senescence, and migration. Functioning primarily as a transcriptional factor, MYC interacts with a large number of proteins, and identification and characterization of MYC-interacting proteins are important for understanding how MYC functions. In this study, we used different systems to demonstrate that a novel zinc finger transcription factor, ZNF121, physically interacted with MYC, and the interaction involved their N-terminal regions. Overexpression of ZNF121 increased, while its knockdown decreased, the expression of MYC in multiple epithelial cell lines, and MYC had similar effects on ZNF121 expression. An expression correlation was also detectable in a panel of epithelial cell lines and a cohort of human breast cancer tissues. Functionally, knockdown of ZNF121 in several breast epithelial cell lines attenuated the expressions of MYC and its target genes(e.g., EGR1,CDC2, and nucleolin) and slowed cell proliferation, accompanied by cell cycle arrest in the G1 phase and expression alteration of cell cycle regulators(cyclin D1, p14 and p21). Analysis of publically available databases showed that ZNF121 expression is up-regulated in human breast cancer, and the up-regulation significantly associates with worse patient survival in the luminal A subtype of breast cancer. The positive clones were examined by western-blotting and immunofluorescence,cell growth was analyzed with MTT assay. | 13 | Loss-of-function of KMT5B leads to neurodevelopmental disorder and impairs neuronal development and neurogenesis显示文摘Autism spectrum disorder(ASD)is a group of neurodevelopmental disorders that cause severe social,communication,and behavioral problems.Recent studies show that the variants of a histone methyltransferase gene KMT5B cause neurodevelopmental disorders(NDDs),including ASD,and the knockout of Kmt5b in mice is embryonic lethal.However,the detailed genotype-phenotype correlations and functional effects of KMT5B in neurodevelopment are unclear.By targeted sequencing of a large Chinese ASD cohort,analyzing published genome-wide sequencing data,and mining literature,we curated 39 KMT5B variants identified from NDD individuals.A genotype-phenotype correlation analysis for 10 individuals with KMT5B pathogenic variants reveals common symptoms,including ASD,intellectual disability,languages problem,and macrocephaly.In vitro knockdown of the expression of Kmt5b in cultured mouse primary cortical neurons leads to a decrease in neuronal dendritic complexity and an increase in dendritic spine density,which can be rescued by expression of human KMT5B but not that of pathogenic de novo missense mutants.In vivo knockdown of the Kmt5b expression in the mouse embryonic cerebral cortex by in utero electroporation results in decreased proliferation and accelerated migration of neural progenitor cells.Our findings reveal essential roles of histone methyltransferase KMT5B in neuronal development,prenatal neurogenesis,and neuronal migration. | Guodong Chen Lin Han Senwei Tan Xiangbin Jia Huidan Wu Yingting Quan Qiumeng Zhang Bin Yu Zhengmao Hu Kun Xia Hui Guo | 2022 | Journal of Genetics and Genomics2022,49,9: | 1 |

| 20 | Integrated construction technology for natural gas gravity drive and underground gas storage显示文摘Based on the mechanisms of gravity displacement,miscibility,viscosity reduction,and imbibition in natural gas flooding,an integrated reservoir construction technology of oil displacement and underground gas storage(UGS)is proposed.This paper systemically describes the technical connotation,site selection principle and optimization process of operation parameters of the gas storage,and advantages of this technology.By making full use of the gravity displacement,miscibility,viscosity reduction,and imbibition features of natural gas flooding,the natural gas can be injected into oil reservoir to enhance oil recovery and build strategic gas storage at the same time,realizing the win-win situation of oil production and natural gas peak shaving.Compared with the gas reservoir storage,the integrated construction technology of gas storage has two profit models:increasing crude oil production and gas storage transfer fee,so it has better economic benefit.At the same time,in this kind of gas storage,gas is injected at high pressure in the initial stage of its construction,gas is injected and produced in small volume in the initial operation stage,and then in large volume in the middle and late operation stage.In this way,the gas storage wouldn’t have drastic changes in stress periodically,overcoming the shortcomings of large stress variations of gas reservoir storage during injection-production cycle due to large gas injection and production volume.The keys of this technology are site selection and evaluation of oil reservoir,and optimization of gravity displacement,displacement pressure,and gas storage operation parameters,etc.The pilot test shows that the technology has achieved initial success,which is a new idea for the rapid development of UGS construction in China. | JIANG Tongwen WANG Zhengmao WANG Jinfang | 2021 | Petroleum Exploration and Development2021,48,5: | 1 |
