Sintilimab versus docetaxel as second-line treatment in advanced or metastatic squamous non-small-cell lung cancer:an open-label,randomized controlled phase 3 trial(ORIENT-3)显示文摘Background:Treatment options for Chinese patients with locally advanced or metastatic squamous-cell non-small-cell lung cancer(sqNSCLC)after failure of first-line chemotherapy are limited. This study(ORIENT-3)aimed to evaluate the efficacy and safety of sintilimab versus docetaxel as second-line treatment in patients with locally advanced or metastatic sqNSCLC.Methods:ORIENT-3 was an open-label,multicenter,randomized controlled phase 3 trial that recruited patients with stage IIIB/IIIC/IV sqNSCLC after failure with first-line platinum-based chemotherapy.Patients were randomized in a 1:1 ratio to receive either 200 mg of sintilimab or 75 mg/m^(2) of docetaxel intravenously every 3 weeks, stratified by the Eastern Cooperative Oncology Group performance status. The primary endpoint was overall survival(OS)in the full analysis set(FAS).Secondary endpoints included progression-free survival(PFS),objective response rate(ORR),disease control rate(DCR),duration of response(DoR)and safety. Results:Between August 25,2017,and November 7,2018,290 patients were randomized. For FAS,10 patients fromthe docetaxel armwere excluded.Themedian OS was 11.79(n=145;95%confidence interval[CI],10.28-15.57)months with sintilimab versus 8.25(n=135;95%CI,6.47-9.82)months with docetaxel(hazard ratio[HR]:0.74;95%CI,0.56-0.96;P=0.025).Sintilimab treatment significantly prolonged PFS(median 4.30 vs.2.79 months;HR:0.52;95%CI,0.39-0.68;P<0.001)and showed higher ORR(25.50%vs.2.20%,P<0.001)and DCR(65.50%vs.37.80%,P<0.001)than the docetaxel arm.The median DoRwas 12.45(95%CI,4.86-25.33)months in the sintilimab arm and 4.14(95%CI,1.41-7.23)months in the docetaxel arm(P=0.045).Treatment-related adverse events of grade≥3were reported in 26(18.1%)patients in the sintilimab arm and 47(36.2%)patients in the docetaxel arm.Exploratory biomarker analysis showed potential predictive values of expression levels of two transcription factors, including OVOL2(HR:0.35;P<0.001)and CTCF(HR:3.50;P<0.001),for sintilimab treatment. Conclusions:Compared with docetaxel, sintilimab significantly improved the OS,PFS,and ORR of Chinese patients with previously treated locally advanced or metastatic sqNSCLC.Yuankai Shi Lin Wu Xinmin Yu Puyuan Xing Yan Wang Jianying Zhou Airong Wang Jianhua Shi Yi Hu Ziping Wang Guangyu An Yong Fang Sanyuan Sun Caicun Zhou Changli Wang Feng Ye Xingya Li Junye Wang Mengzhao Wang Yunpeng Liu Yanqiu Zhao Ying Yuan Jifeng Feng Zhendong Chen Jindong Shi Tao Sun Gang Wu Yongqian Shu Qisen Guo Yi Zhang Yong Song Shucai Zhang Yuan Chen Wei Li Hongrui Niu Wenwei Hu Lijun Wang Jianan Huang Yang Zhang Ying Cheng Zhengdong Wu Bo Peng Jiya Sun Christoph Mancao Yanqi Wang Luyao Sun 2022Cancer Communications2022,42,12:8
Synthesis and Characterization of Sphere-like Pt Nanoparticles Supported on DWCNT/WO_3 Nanorods with Electrocatalytic Activity显示文摘Tungsten oxide(WO3) nanorods, which were used to load platinum(Pt) nanoparticles, were investigated. H2WO4 nanorods with diameters from 10 to 50 nm were obtained when tungsten precursor was added into homogenous double-walled carbon nanotubes(DWCNT) and ethylene glycol(EG) solution.Nanosized rod-like WO3 were achieved after calcination of the DWCNT/H2WO4 composite. Sphere-like Pt nanoparticles were loaded on the surface of the nanorods by EG in-situ reduction. Pt particles were isolated by DWCNT/WO3 nanorods and secondary accumulation could be prevented when Pt particles appeared in the DWCNT/WO3 nanorod/EG dispersion solution. Therefore, Pt nanoparticles with mean diameters of 2–6 nm could be obtained. Pt-deposited on DWCNT/WO3 nanorods exhibited high electrochemical activity, which could facilitate the low-cost mass production of Pt catalyst.Yanhong Yin Chun'an Ma Ziping Wu Man Zhao Litao Chen Youqun Chu 2015Journal of Materials Science & Technology2015,31,9:4
Survival difference between EGFR Del19 and L858R mutant advanced non-small cell lung cancer patients receiving gefitinib:a propensity score matching analysis显示文摘Objective: Although superior clinical benefits of epidermal growth factor receptor(EGFR) tyrosine kinase inhibitors(TKIs) in the treatment of advanced non-small-cell lung cancer(NSCLC) had been reported, the survival difference between exon 19 deletion(Del19) and exon 21 Leu858 Arg substitution(L858 R) remains controversial. The purpose of this study is to investigate the differences in progression-free survival(PFS) and overall survival(OS) between different EGFR mutant subtypes among advanced NSCLC patients receiving gefitinib. Methods: There were 204 advanced NSCLC patients with EGFR mutations treated with gefitinib were enrolled in this retrospective cohort study.Patients were divided into the EGFR Del19 group and the L858 R mutated group according to their mutant subtype.Propensity score matching(PSM) was conducted by using a nearest-neighbor algorithm(1:1) to adjust for demographical and clinical covariates.Survival curves were constructed with the Kaplan-Meier method and compared by using the log-rank test. Results: The PFS in Del19 group was similar to that in the L858 R group [before PSM 8.6 vs.7.2 months,P=0.072; after PSM 7.3 vs.7.2 months,P=0.155].No differences were detected in OS between the L858 R and the Del19 group(before PSM 17.8 vs.13.1 months,P=0.253; after PSM 16.9 vs.13.1 months,P=0.339).The Del19 group was significantly younger compared with the L858 R mutation group in age(P=0.015).Conclusions: No significant difference was found in the PFS or OS between the Del19 and L858 R mutant NSCLC patients receiving gefitinib. The age gap might contribute to the survival differences between Del19 and L858 R groups.PSM is of important value to the elimination of potential bias.Minglei Zhuo Qiwen Zheng Jun Zhao Meina Wu Tongtong An Yuyan Wang Jianjie Li Shuhang Wang Jia Zhong Xue Yang Hanxiao Chen Bo Jia Zhi Dong Emei Gao JingjingWang Ziping Wang 2017Chinese Journal of Cancer Research2017,29,6:4

Terahertz quantum cascade lasers with sampled lateral gratings for single mode operation显示文摘In this paper,we presented single mode terahertz quantum cascade lasers(THz QCLs)with sampled lateral grating emitting approximately 3.4 THz.Due to strong mode selection, the implementation of sampled lateral grating on THz QCL ridges can result in stable single longitudinal mode emission with a side-mode suppression ratio larger than 20 dB.The measured peak power of the grating laser is improved by about 11.8%compared to the power of devices with uniform distributed feedback gratings.Furthermore, the far-field pattern of the presented device is uninfluenced by grating structures.Dixiang SHAO Chen YAO Zhanglong FU Wenjian WAN Ziping LI Juncheng CAO 2021Frontiers of Optoelectronics2021,14,1:1

Paclitaxel liposome for injection (Lipusu) plus cisplatin versus gemcitabine plus cisplatin in the first-line treatment of locally advanced or metastatic lung squamous cell carcinoma: A multicenter, randomized, open-label, parallel controlled clinical study显示文摘Background:Lipusu is the first commercialized liposomal formulation of pacli-taxel and has demonstrated promising efficacy against locally advanced lung squamous cell carcinoma(LSCC)in a small-scale study. Here,we conducted a multicenter, randomized, phase 3 study to compare the efficacy and safety of cis-platin plus Lipusu(LP)versus cisplatin plus gemcitabine(GP)as first-line treat-ment in locally advanced or metastatic LSCC.Methods:Patients enrolled were aged between 18 to 75 years, had locally advanced(clinical stage IIIB,ineligible for concurrent chemoradiation or surgery)or metastatic(Stage IV)LSCC,had no previous systemic chemother-apy and at least one measurable lesion as per the Response Evaluation Criteria in Solid Tumors(version 1.1)before administration of the trial drug. The primary endpoint was progression-free survival(PFS).The secondary endpoints included objective response rate(ORR),disease control rate(DCR),overall survival(OS),and safety profiles. To explore the possible predictive value of plasma cytokines for LP treatment, plasma samples were collected from the LP group at baseline and first efficacy evaluation time and were then subjected to analysis by 45-Plex ProcartaPlex Panel 1 to detect the presence of 45 cytokines using the Luminex xMAP technology. The correlation between treatment outcomes and dynamic changes in the levels of cytokines were evaluated in preliminary analyses. Results:The median duration of follow-up was 15.4 months.237 patients in the LP group and 253 patients in the GP group were included in the per protocol set(PPS).In the PPS,the median PFS was 5.2 months versus 5.5 months in the LP and GP group(hazard rtio[HR]:1.03,P=0.742)respectively.The median OS was 14.6 months versus 12.5 months in the LP and GP group(HR:0.83,P=0.215).The ORR(41.8%versus 45.9%,P=0.412)and DCR(90.3%versus 88.1%,P=0.443)were also similar between the LP and GP group.A significantly lower proportion of patients in the LP group experienced adverse events(AEs)leading to treatment interruptions(10.9%versus 26.4%,P<0.001)or treatment termination(14.3%versus 23.1%,P=0.011).The analysis of cytokine levels in the LP group showed that low baseline levels of 27 cytokines were associated with an increased ORR,and 15 cytokines were associated with improved PFS,with 14 cytokines, including TNF-a,IFN-y,IL-6,and IL-8,demonstrating an overlapping trend. Conclusion:The LP regimen demonstrated similar PFS,OS,ORR and DCR as the GP regimen for patients with locally advanced or metastatic LSCC but had more favorable toxicity profiles. The study also identified a spectrum of different cytokines that could be potentially associated with the clinical benefit in patients who received the LP regimen.Jie Zhang Yueyin Pan Qin Shi Guojun Zhang Liyan Jiang Xiaorong Dong Kangsheng Gu Huijuan Wang Xiaochun Zhang Nong Yang Yuping Li Jianping Xiong Tienan Yi Min Peng Yong Song Yun Fan Jiuwei Cui Gongyan Chen Wei Tan Aimin Zang Qisen Guo Guangqiang Zhao Ziping Wang Jianxing He Wenxiu Yao Xiaohong Wu Kai Chen Xiaohua Hu Chunhong Hu Lu Yue Da Jiang Guangfa Wang Junfeng Liu Guohua Yu Junling Li Jianling Bai Wenmin Xie Weihong Zhao Lihong Wu Caicun Zhou 2022Cancer Communications2022,42,1:1

Terahertz metalens of hyper-dispersion显示文摘Chromatic optical lenses have promising applications in three-dimensional imaging, which allows fast spectral tomography without mechanical moving parts. The scanning range of current chromatic optical lenses is limited by their dispersion ability. The recent development in metasurfaces provides ideal blocks for optical wavefront manipulation and dispersion engineering of artificial materials at sub-wavelength scales. Hyper-dispersive metalenses can be realized by utilizing dispersive meta-atoms, which have enhanced dispersion compared to regular diffractive lenses. This is critical for increasing the imaging depth of fast spectral tomography. In this work, a hyper-dispersive metalens is realized with a chromatic dispersion 1.76 times greater than that of a regular diffractive metalens in the THz frequency range of 2.40–2.61 THz by simultaneously controlling the frequencydependent phase, group delay(GD), and GD dispersion of the metalens. This approach can also be extended to other optical spectra and improve the performance of spectral tomography.Fen Zhao Ziping Li Sheng Li Xuemei Dai Yi Zhou Xiaoyu Liao JCCao Gaofeng Liang Zhengguo Shang Zhihai Zhang Zhongquan Wen Hua Li Gang Chen 2022Photonics Research2022,10,4:1
In-situ polymerized carbonate induced by Li-Ga alloy as novel artificial interphase on Li metal anode显示文摘Li metal is considered an ideal anode material because of its high theoretical capacity and low electrode potential. However, the practical usage of Li metal as an anode is severely limited because of inevitable parasitic side reactions with electrolyte and dendrites formation. At present,single-component artificial solid electrolyte interphase cannot simultaneously meet the multiple functions of promoting ion conduction, guiding lithium ion deposition, inhibiting dendrite growth, and reducing interface side reactions.Therefore,multi-component design on Li metal surface is widely investigated to achieve long-term cycling.Herein,we report a Li_(2)Ga-carbonate polymer interphase layer to solve volume changes,Li dendrites formation and side-reactions.As a result, the Li symmetric cell can be stabilized at 3.0 m A/cm^(2)in carbonate electrolyte with limited volume of 20μL.Coupled with 13.6 mg/cm^(2)(loading of 2 mAh/cm^(2))LiFePO_(4)cathode,discharge capacity retains at 90%for over 150 cycles under limited electrolyte conditions. With such an alloy-polymer interphase layer, higher energy density Li metal batteries become prominent in the near future.Ziping Wang Shuyuan Xie Xuejie Gao Xinyang Chen Lina Cong Jun Liu Haiming Xie Chuang Yu Yulong Liu 2023Chinese Chemical Letters2023,34,9:0
Theanine, a tea-plant-specific non-proteinogenic amino acid, is involved in the regulation of lateral root development in response to nitrogen status显示文摘Glutamine synthetase type I(GSI)-like proteins are proposed to mediate nitrogen signaling and developmental fate by synthesizing yet unidentified metabolites.Theanine, the most abundant non-proteinogenic amino acid in tea plants,is the first identified metabolite synthesized by a GSI-like protein(CsTSI)in a living system. However, the roles of theanine in nitrogen signaling and development are little understood. In this study we found that nitrogen deficiency significantly reduced theanine accumulation and increased lateral root development in tea plant seedlings.Exogenous theanine feeding significantly repressed lateral root development of seedlings of tea plants and the model plant Arabidopsis. The transcriptomic analysis revealed that the differentially expressed genes in the roots under theanine feeding were enriched in the apoplastic pathway and H2O_(2) metabolism.Consistently, theanine feeding reduced H2O_(2) levels in the roots.Importantly, when co-treated with H2O_(2),theanine abolished the promoting effect of H2O_(2) on lateral root development in both tea plant and Arabidopsis seedlings. The results of histochemical assays confirmed that theanine inhibited reactive oxygen species accumulation in the roots.Further transcriptomic analyses suggested the expression of genes encoding enzymes involved in H2O_(2) generation and scavenging was down-and upregulated by theanine, respectively. Moreover, the expression of genes involved in auxin metabolism and signaling, cell division, and cell expansion was also regulated by theanine.Collectively, these results suggested that CsTSI-synthesized theanine is likely involved in the regulation of lateral root development, via modulating H2O_(2) accumulation,in response to nitrogen levels in tea plants. This study also implied that the module consisting of GSI-like protein and theanine-like metabolite is probably conserved in regulating development in response to nitrogen status in plant species.Tingting Chen Shijia Lin Ziping Chen Tianyuan Yang Shupei Zhang Jinsong Zhang Guohua Xu Xiaochun Wan Zhaoliang Zhang 2023Horticulture Research2023,10,2:0
Responses of Drought-Resistant Mutant vem1 to Stress and Cloning of VEM1 Gene in Arabidopsis显示文摘[Objective]This study aimed to investigate the responses of drought-resistant mutant vem1 to stress and clone VEM1 gene in Arabidopsis. [Method] A drought-resistant mutant vem1 was isolated from the Arabidopsis mutant pool. The germination rates of wild-type( WT) and mutant vem1 were detected to investigate the responses of mutant vem1 to mannitol,NaCl and ABA stress. [Result] The mutant vem1 was resistant to mannitol and NaCl stress but sensitive to ABA stress. VEM1 gene was cloned by Tail-PCR technology and sequenced. The sequencing result was submitted to NCBI for sequence alignment and gene mapping using BLAST. Database analysis suggested that VEM1 gene was a transposable element gene. [Conclusion]This study laid the foundation for functional analysis of drought-resistant gene VEM1.Junxin SHI Jiaojiao ZHANG Ziping CHEN Xiaohui DING Jie YANG Li JIANG 2013Agricultural Biotechnology2013,2,5:0
Haem Oxygenase 1 is a potential target for creating etiolated/albino tea plants (Camellia sinensis) with high theanine accumulation显示文摘Theanine content is highly correlated with sensory quality and health benefits of tea infusion. The tender shoots of etiolated and albino tea plants contain higher theanine than the normal green tea plants and are valuable materials for high quality green tea processing. However, why these etiolated or albino tea plants can highly accumulate theanine is largely unknown. In this study,we observed an Arabidopsis etiolated mutant hy1–100(mutation in Haem Oxygenase 1,HO1)that accumulated higher levels of glutamine(an analog of theanine). We therefore identified CsHO1 in tea plants and found CsHO1 is conserved in amino acid sequences and subcellular localization with its homologs in other plants.Importantly,CsHO1 expression in the new shoots was much lower in an etiolated tea plants‘Huangkui’and an albino tea plant‘Huangshan Baicha’than that in normal green tea plants. The expression levels of CsHO1 were negatively correlated with theanine contents in these green, etiolated and albino shoots.Moreover,CsHO1 expression levels in various organs and different time points were also negatively correlated with theanine accumulation. The hy1–100 was hypersensitive to high levels of theanine and accumulated more theanine under theanine feeding, and these phenotypes were rescued by the expression of CsHO1 in this mutant.Transient knockdown CsHO1 expression in the new shoots of tea plant using antisense oligonucleotides(asODN)increased theanine accumulation.Collectively, these results demonstrated CsHO1 negatively regulates theanine accumulation in tea plants, and that low expression CsHO1 likely contributes to the theanine accumulation in etiolated/albino tea plants.Ziping Chen Shijia Lin Tingting Chen Mengxue Han Tianyuan Yang Yan Wang Shilai Bao Zhougao Shen Xiaochun Wan Zhaoliang Zhang 2023Horticulture Research2023,10,2:0
Study on Distribution Characteristics and Occurrence Form of Soil Fluorine in Sorghum Base of Renhuai City显示文摘In order to find out the effects of soil fluorine on ecological environment and crop safety in the sorghum cultivation base of Renhuai City, Guizhou Province, the content and the occurrence forms of soil fluorine and its influencing factors of Maoba Town were investigated and studied. The results showed that the total fluorine content in the soil of the sorghum base ranged from 668.60 to 2 596.80 mg/kg, with an average of 1 483.25 mg/kg, which was 3.10 and 1.85 times of the national soil background value and the average value of the soil in endemic areas, respectively. The spatial distribution of fluorine in the soil of the study area was uneven, and the fluorine content was in the order of Anliang Village>Yangliu Village>Houba Village>Xiongfeng Village. The soil fluorine contents of various forms varied greatly, and the distribution law was as follows: residual state>water soluble state>organically bound state>exchangeable state>Fe/Mn-F state. The residual fluorine content was the highest, accounting for more than 99% of total fluorine content, while the sum of other four forms was less than 1%, with their respective proportions ranging from 0.10% to 0.30%. The content and proportion of available fluorine in water-soluble and exchangeable states were relatively low, which had limited influence on the surrounding water environment and crop safety. The physicochemical properties of soil, such as pH, organic matter, cation exchange capacity and clay particles had a certain influence on the occurrence forms of soil fluorine, thus changing the availability and migration of soil fluorine, which might endanger the ecological environment safety, food safety and human health.Ziping PAN Maoyan SHAO Xubin HUANG Ting CHEN Luyao SUI 2023Agricultural Biotechnology2023,12,1:0
Discovery of a highly potent and selective Bruton’s tyrosine kinase inhibitor avoiding impairment of ADCC effects for B-cell non-Hodgkin lymphoma显示文摘Dear Editor,Bruton’s tyrosine kinase(BTK)plays a crucial role in the B-cell receptor(BCR)signaling which is essential for B-cell proliferation, differentiation, and cell migration.Aberrant BCR activation has been identified as a major pathogenic factor in several B-cell non-Hodgkin lymphoma(B-NHL)subtypes,including diffuse large Bcell lymphoma(DLBCL),mantle cell lymphoma(MCL),follicular lymphoma(FL),and chronic lymphocytic leukemia(CLL).1 Therefore,BTK has been recognized as a validated therapeutic target for B-cell malignancies.Ibrutinib, the first approved BTK inhibitor that binds irreversibly to cysteine residue 481,has shown potent clinical activity in the majority of CD20 positive B-cell malignancies.2 However, due to the inhibition of off-target kinases such as EGFR,ITK,and TXK,which have a cysteine residue at the identical position of Cys481 of BTK,Ibrutinib also results in some adverse events, such as the antagonizing Rituximab-dependent NK-cell-mediated antibody-dependent cell-mediated cytotoxicity(ADCC)due to its irreversible binding to ITK,which is required for FcR-stimulated NK cell function.3 Although several secondary generation inhibitors have shown improved selectivity,4,5 more pharmacologically diverse novel inhibitors are still highly demanded in the clinic.Juan Liu Qianmao Liang Aoli Wang Fengming Zou Ziping Qi Kailin Yu Qingwang Liu Cheng Chen Jing Liu Qingsong Liu 2020Signal Transduction and Targeted Therapy2020,5,1:0
