1Analysis of DNA methylation variation in wheat genetic background after alien chromatin introduction based on methylation-sensitive amplification polymorphism显示文摘During the process of alien germplasm introduced into wheat genome by chromosome engineering, extensive genetic variations of genome structure and gene expression in recipient could be induced. In this study, we performed GISH (genome in situ hybridization) and AFLP (amplified fragment length po-lymorphism) on wheat-rye chromosome translocation lines and their parents to detect the identity in genomic structure of different translocation lines. The results showed that the genome primary struc-ture variations were not obviously detected in different translocation lines except the same 1RS chromosome translocation. Methylation sensitive amplification polymorphism (MSAP) analyses on genomic DNA showed that the ratios of fully-methylated sites were significantly increased in translo-cation lines (CN12, 20.15%; CN17, 20.91%; CN18, 22.42%), but the ratios of hemimethylated sites were significantly lowered (CN12, 21.41%; CN17, 23.43%; CN18, 22.42%), whereas 16.37% were fully-me-thylated and 25.44% were hemimethylated in case of their wheat parent. Twenty-nine classes of me-thylation patterns were identified in a comparative assay of cytosine methylation patterns between wheat-rye translocation lines and their wheat parent, including 13 hypermethylation patterns (33.74%), 9 demethylation patterns (22.76%) and 7 uncertain patterns (4.07%). 2Zanthoxylum-specific whole genome duplication and recent activity of transposable elements in the highly repetitive paleotetraploid Z.bungeanum genome显示文摘Zanthoxylum bungeanum is an important spice and medicinal plant that is unique for its accumulation of abundant secondary metabolites,which create a characteristic aroma and tingling sensation in the mouth.Owing to the high proportion of repetitive sequences,high heterozygosity,and increased chromosome number of Z.bungeanum,the assembly of its chromosomal pseudomolecules is extremely challenging.Here,we present a genome sequence for Z.bungeanum,with a dramatically expanded size of 4.23 Gb,assembled into 68 chromosomes.This genome is approximately tenfold larger than that of its close relative Citrus sinensis.After the divergence of Zanthoxylum and Citrus,the lineage-specific whole-genome duplication event q-WGD approximately 26.8 million years ago(MYA)and the recent transposable element(TE)burst~6.41 MYA account for the substantial genome expansion in Z.bungeanum.The independent Zanthoxylum-specific WGD event was followed by numerous fusion/fission events that shaped the genomic architecture.Integrative genomic and transcriptomic analyses suggested that prominent speciesspecific gene family expansions and changes in gene expression have shaped the biosynthesis of sanshools,terpenoids,and anthocyanins,which contribute to the special flavor and appearance of Z.bungeanum.In summary,the reference genome provides a valuable model for studying the impact of WGDs with recent TE activity on gene gain and loss and genome reconstruction and provides resources to accelerate Zanthoxylum improvement.Shijing Feng Zhenshan Liu Jian Cheng Zihe Li Lu Tian Min Liu Tuxi Yang Yulin Liu Yonghong Liu He Dai Zujun Yang Qing Zhang Gang Wang Jisen Zhang Huifeng Jiang Anzhi Wei 2021Horticulture Research2021,8,1:5
3Comparative FISH and molecular identification of new stripe rust resistant wheat-Thinopyrum intermedium ssp. trichophorum introgression lines显示文摘Thinopyrum intermedium(2 n = 6 x = 42, JJJ^sJ^sStSt) has been hybridized extensively with common wheat and has proven to be a valuable germplasm source for improving disease resistance, quality attributes, and yield potential in wheat. We characterized new disease resistant wheat-Th. intermedium derivatives A1082 and A5-5 using sequential multi-color fluorescence in situ hybridization(mc-FISH), genomic in situ hybridization(GISH), PCRbased landmark unique gene(PLUG) and intron targeting(IT) markers. A1082 was identified as a wheat-Th. intermedium 3J disomic addition line, and A5-5 was a T4BS·5J^sL homozygous Robertsonian translocation line. Seventy-one and 106 pairs of primers amplified Th.intermedium-specific bands allowing chromosomes 3J and 5J^s to be tracked, respectively. A new oligonucleotide probe, Oligo-6 H-2-100, was developed for FISH labeling of the subterminal region of the long arm of chromosome 5J^s. Both lines were highly resistant to stripe rust pathogen races prevalent in Chinese field screening nurseries. A5-5 also displayed a significant increase in tiller number compared to its wheat parent. 9A novel method for reconstruction in laparoscopic pancreaticoduodenectomy:an experience of 13 cases显示文摘Laparoscopic pancreaticoduodenectomy(LPD)is a challenging operation to general surgeon.Up to date,only about 135 cases have been reported,16 cases in China,119 cases outside China.The reconstruction of alimentary system is a key procedure to ensure success of the whole surgery.It is worth investigating the methods of reconstruc-tion in LPD.A retrospective study is made to investigate the methods of reconstruction in LPD.We analyze 13 cases of LPD performed in our center.Child’s or modified Child’s method was used to make the reconstruction in our practice.We tried three methods to make the anastomosis of pancre-aticojejunostomy,including end-to-end dunking binding pancreaticojejunostomy in two cases,end-to-end dunking pancreaticojejunostomy using interrupted suture in two cases,and duct-to-jejunal end-to-side embedding pancreaticojeju-nostomy in nine cases.The clinical data was collected and analyzed.Three of four patients,who underwent end-to-end pancreaticojejunostomy,had a little pancreatic leakage,especially in the first case.None of other nine patients,who underwent duct-to-jejunal end-to-side embedding pancreati-cojejunostomy,was detected to have pancreatic leakage,and the operating time of these nine cases was less than other four cases.Duct-to-jejunal end-to-side embedding pancreaticoje-junostomy is a safe and efficient method of reconstruction in LPD.LU Bangyu HUANG Yubin CAI Xiaoyong HUANG Fei LI Jie LU Wenqi XU Jing LIU Zujun YAN Yihe LI Jianjun 2007Frontiers of Medicine2007,1,4:1

13Influence of CO_(2)/HCO_(3)^(-) on Microbial Communities in Two Karst Caves with High CO_(2)显示文摘There is limited knowledge about microbial communities and their ecological functions in karst caves with high CO_(2)concentrations.Here,we studied the microbial community compositions and functions in Shuiming Cave( 'SMC',CO_(2)concentration 3 303 ppm) and Xueyu Cave( 'XYC',CO_(2)concentration 8 753 ppm) using Illumina MiSeq high-throughput sequencing in combination with BIOLOG test.The results showed that Proteobacteria,Actinobacteria and Bacteroidetes were dominant phyla in these two caves,and Thaumarchaeota was the most abundant in the rock wall samples of SMC.The microbial diversity in the water samples decreased with increasing HCO_(3)^(-)concentration,and it was higher in XYC than that in SMC.The microbial community structures in the sediment and rock wall samples were quite different between the two caves.High concentrations of CO_(2)can reduce the microbial diversity on the rock walls in karst caves,probably through changing microbial preference for different types of carbon sources and decreasing the microbial utilization rate of carbon sources.These results expanded our understanding of microbial community and its response to environments in karst caves with high CO_(2).Jiyu Chen Qiang Li Qiufang He Heinz C.Schroder Zujun Lu Daoxian Yuan 2023Journal of Earth Science2023,34,1:0

15Theoretical analysis on GaAs sub-cell doping concentration for triple-junction solar cells irradiated by proton based on TCAD simulation显示文摘The degradation on the GaInP/GaAs/Ge triple-junction solar cells was irradiated by proton, and the solar cells with various GaAs sub-cell doping concentrations are modeled by the technology computer aided design(TCAD) simulation. The degradation results of related electrical parameters and external quantum efficiency(EQE) are studied. The degradation mechanism irradiated by proton is discussed. The short-circuit current, maximum power and conversion efficiency decrease with the increasing of GaAs sub-cell doping concentration. When the base doping concentration of GaAs sub-cell is 1×1016cm-3, the degradation of short-circuit current is less than that of other base doping concentrations. Furthermore, under proton irradiation, with the increase of doping concentration of GaAs sub-cell, the open-circuit voltage first increases and then decreases. Meanwhile, when the base doping concentration of GaAs sub-cell is 2×10^(17)cm^(-3), the degradation of open-circuit voltage is less than that of other base doping concentrations. 17Genome sequencing of Sitopsis species provides insights into their contribution to the B subgenome of bread wheat显示文摘Wheat(Triticum aestivum,BBAADD)is an allohexaploid species that originated from two polyploidization events.The progenitors of the A and D subgenomes have been identified as Triticum urartu and Aegilops tauschii,respectively.Current research suggests that Aegilops speltoides is the closest but not the direct ancestor of the B subgenome.However,whether Ae.speltoides has contributed genomically to the wheat B subgenome and which chromosome regions are conserved between Ae.speltoides and the B subgenome remain unclear.Here,we assembled a high-quality reference genome for Ae.speltoides,resequenced 53 accessions from seven species(Aegilops bicornis,Aegilops longissima,Aegilops searsii,Aegilops sharonensis,Ae.speltoides,Aegilops mutica[syn.Amblyopyrum muticum],and Triticumdicoccoides)and revealed their genomic contributions to the wheat B subgenome.Our results showed that centromeric regions were particularly conserved between Aegilops and Triticum and revealed 0.17 Gb of conserved blocks between Ae.speltoides and the B subgenome.We classified five groups of conserved and non-conserved genes between Aegilops and Triticum,revealing their biological characteristics,differentiation in gene expression patterns,and collinear relationships between Ae.speltoides and the wheat B subgenome.We also identified gene families that expanded in Ae.speltoides during its evolution and 789 genes specific to Ae.speltoides.These genes can serve as genetic resources for improvement of adaptability to biotic and abiotic stress.The newly constructed reference genome and large-scale resequencing data for Sitopsis species will provide a valuable genomic resource for wheat genetic improvement and genomic studies.Yuxin Yang Licao Cui Zefu Lu Guangrong Li Zujun Yang Guangyao Zhao Chuizheng Kong Danping Li Yaoyu Chen Zhencheng Xie Zhongxu Chen Lichao Zhang Chuan Xia Xu Liu Jizeng Jia Xiuying Kong 2023Plant Communications2023,4,4:0
18New PCR based markers allowed to identify Secale chromatin in wheat-Secale africanum introgression lines显示文摘The genus of Secale has many agronomically important characters.In order to use the best of this species,markers tracking the rye chromatin incorporated into wheat must be developed.In this study,one rye genome-specific random amplified polymorphic DNA(RAPD)marker was isolated from Secale africanum(Ra genome).Two cloned markers,named OPP13_(1165) and OPP13_(662),were 1165 bp and 662 bp,respectively.Sequence analysis revealed that OPP13_(1165) was highly homologous to a part of a new class of transposon-like gene called the Revolver family,and OPP13_(662) was partially similar to LTR gypsy-like retrotransposon.Fluorescence in situ hybridization(FISH)showed only OPP13_(1165) localized within the whole arms of rye except their terminal regions and no signal was detected on wheat chromosomes,while OPP13_(662) had no hybridization signal detected on wheat and rye genomes.Based on these sequences,two pairs of sequence-characterized amplified region(SCAR)primers were designed,and the resulted SCAR markers were able to target both cultivated and wild Secale species.The FISH patterns and the two SCAR markers should be able to identify and track all wheat-rye translocation lines,especially the S.africanum chromatin.Juqing JIA Guangrong LI Cheng LIU Jianping ZHOU Zujun YANG 2010Frontiers in Biology2010,5,2:0
19Novel expressed sequence tags of an alpine-cold plant species,Gymnadenia conopsea (Orchidaceae)显示文摘Gymnadenia conopsea,an alpine Orchidaceae plant,was one of the widely used Tibetan traditional medicines.In this study,we sequenced total 105 expressed sequence tags (ESTs) from a full-length cDNA expression library constructed by the Oligo-capping technique.The further bioinformatic analyses suggested that the 65 represented unique sequences showed high homology to previously identified genes in other plants:30 sequences matched to other uncharacterized expressed sequence tags (ESTs),and 10 sequences showed no good matches to available sequences in DNA databases.Gene ontology annotation by InterProScan indicated that many of these cDNAs (7 percent) have no known molecular functions and may be unique to G.conopsea.Fifty-five ESTs with matched proteins were involved in a series of diverse functions,in which molecular function such as 'binding' (42.9 percent) and 'catalytic activity' (25.0 percent) were the most frequent functions of the cDNAs.This cDNA library provided a critical basis for further investigation of functional genes expression under cold stress in this alpine species.In addition,13 ESTs-based polymerase chain reaction (PCR) primers were designed and can also be used for genotypic identification and for the genetic diversity analysis of G.conopsea and its closely related species.JuQing Jia JianPing Zhou Tao Zhang ZhengNan Wen GuangRong Li ZuJun Yang 2010Research in Cold and Arid Regions2010,2,2:0
20Haploidentical transplantation has a superior graft-versus-leukemia effect than HLA-matched sibling transplantation for Ph- high-risk B-cell acute lymphoblastic leukemia显示文摘Background: Compared with human leukocyte antigen (HLA)-matched sibling donor (MSD) transplantation, it remains unclear whether haploidentical donor (HID) transplantation has a superior graft-versus-leukemia (GVL) effect for Philadelphia-negative (Ph-) high-risk B-cell acute lymphoblastic leukemia (B-ALL). This study aimed to compare the GVL effect between HID and MSD transplantation for Ph- high-risk B-ALL.Methods: This study population came from two prospective multicenter trials (NCT01883180, NCT02673008). Immunosuppressant withdrawal and prophylactic or pre-emptive donor lymphocyte infusion (DLI) were administered in patients without active graft-versus-host disease (GVHD) to prevent relapse. All patients with measurable residual disease (MRD) positivity posttransplantation (post-MRD+) or non-remission (NR) pre-transplantation received prophylactic/pre-emptive interventions. The primary endpoint was the incidence of post-MRD+.Results: A total of 335 patients with Ph- high-risk B-ALL were enrolled, including 145 and 190, respectively, in the HID and MSD groups. The 3-year cumulative incidence of post-MRD+ was 27.2% (95% confidence interval [CI]: 20.2%-34.7%) and 42.6% (35.5%-49.6%) in the HID and MSD groups(P = 0.003), respectively. A total of 156 patients received DLI, including 60 (41.4%) and 96 (50.5%), respectively, in the HID and MSD groups (P= 0.096). The 3-year cumulative incidence of relapse was 18.6% (95% CI: 12.7%-25.4%) and 25.9% (19.9%-32.3%;P = 0.116) in the two groups, respectively.
