| 1 | A Modified Plate Assay for Rapid Screening of Potassium-Solubilizing Bacteria显示文摘The utility of microorganisms for solubilizing the unavailable forms of potassium(K) from soil has led to renewed interest in fabrication of rapid and sensitive plate assays for their isolation and screening. The present study developed a modified plate assay and compared it with previously reported methods for the isolation and screening of K-solubilizing bacteria. The newly developed plate assay is based on improved visualization of halo zone formation around the colonies on agar plates, through inclusion of an acid-base indicator dye, bromothymol blue(BTB), to modify the previously reported Aleksandrov medium. The halo zone exhibited a significant correlation(R = 0.939) with K released in liquid medium. The visualization of potential K solubilizers was improved using this method, which would help in detection of weak/non-acid producers based on secretion of organic acids in the medium. Organic acids in plate diffuse radially and form halo zones in response to reaction with the acid-base indicator dye BTB. Furthermore, K solubilization on plates with this method can be observed within 48–72 h, against the incubation time of 4–5 d needed in the earlier method. | 2 | Microtexture and Distribution of Minerals in Hydrothermal Barite-Silica Chimney from the Franklin Seamount, SW Pacific: Constraints on Mode of Formation显示文摘An extinct hydrothermal barite-silica chimney from the Franklin Seamount of the Woodlark Basin, in the southwestern Pacific Ocean, was investigated for mineral distribution and geochemical composition. Six layers on either side of the orifice of a chimney show significant disparity in color, mineral assemblage and major element composition. Electron microscope(SEM) images reveal that the peripheral wall of the chimney is composed of colloform silica, suggesting that incipient precipitation of silica-saturated hydrothermal fluid initiated the development of the chimney wall. Intermediate layers, between the exterior wall and the inner fluid-orifice, dominate with barite and sulfides. Low Sr-to-Ba ratios(SrO/BaO = 0.015–0.017) indicate restricted fluid-seawater mixing, which causes relatively high-temperature formation of the intermediate layers. Whereas the innermost layer bordering the chimney orifice is characterized by more silica and a higher Sr-to-Ba ratio(SrO/BaO = 0.023), could have formed due to a paragenetic shift from a high-temperature active phase to a cooler waning stage of formation. | 3 | Role of angiotensin converting enzyme and angiotensinogen gene polymorphisms in angiotensin converting enzyme inhibitor-mediated antiproteinuric action in type 2 diabetic nephropathy patients显示文摘AIM To investigate the role of genetic variants of angiotensin converting enzyme(ACE) and angiotensinogen(AGT) genes in the antiproteinuric efficacy of ACE inhibitor therapy in diabetic nephropathy(DN) patients.METHODS In the present study, 270 type 2 diabetes mellitus patients with nephropathy were enrolled and treated with ACE inhibitor(ramipril) and followed at 6 mo for renal function and albumin excretion by estimating serum creatinine, end stage renal disease, and albumin/creatinine ratio(ACR) in urine. Genotyping of ACE I/D and AGT M235 T polymorphisms were performed by using primer specific polymerase chain reaction(PCR) and PCR-RFLP techniques, respectively. RESULTS Forty-eight percent of DN patients(responders) benefited with respect to proteinuria from ACE inhibitor therapy at 6 mo follow-up. A significant reduction in ACR was observed after 6 mo treatment with ACE inhibitor irrespective of whether DN patients were micro-albuminuric(≥ 30 and < 300 mg/g creatinine) or macro-albuminuric(≥ 300 mg/g creatinine) at the time of enrollment. However, macro-albuminuric patients(55%) showed better response to therapy. A reduction in urinary ACR was found independent of genotypes of ACE I/D and AGT M235 T polymorphisms although macro-albuminuric patients having TT genotype showed statistically insignificant increased response(72%). | 9 | Clinical outcomes for nasopharyngeal cancer with intracranial extension after taxanebased induction chemotherapy and concurrent chemo-radiotherapy in the modern era显示文摘Objective:To evaluate the survival outcomes for a cohort of nasopharyngeal cancer with intracranial extension(ICE)treated with induction chemotherapy(ICT)followed by chemo-intensity-modulated radiotherapy(CTRT)at a tertiary cancer center.Methods:We retrospectively analyzed 45 patients with histologically proven,non-metastatic NPC with ICE treated at our institute between October 2008 and October 2016.Patients were classified as minor ICE or major ICE,based on the extent of ICE.All the patients received 2-3 cycles of a taxane-based ICT regimen followed by CTRT.Radiotherapy was delivered with'riskadapted'intensity-modulated radiotherapy(IMRT)technique in all patients.Results:After a median follow up of 45 months(range:8-113 months),the estimated 5-year DFS,LRFS,DMFS,and OS of the entire cohort was 58%,82%,67%and 74%respectively.On multivariate analysis,histological subtype was an independent predictor of LRFS,and age was an independent predictor of DFS.The extent of ICE showed only a trend towards worse DFS(P=0.06).None of the factors significantly predicted for DMFS or OS.Gender,N-stage,and response to ICT did not significantly affect any of the outcomes.Grade 2 or worse subcutaneous fibrosis was seen in 22%of patients and grade 2 or worse xerostomia was seen in 24%of patients at last follow up.Thirty-three percent of the patients developed clinical hypothyroidism at last follow up.None of the patients experienced any neurological or vascular complications.Conclusions:Taxane-based induction chemotherapy followed by chemo-intensity modulated radiotherapy resulted in excellent locoregional control and survival with acceptable toxicities in patients of nasopharyngeal cancer with intracranial extension.Distant metastasis continues to be the predominant problem in these patients. | Sarbani Ghosh-Laskar Avinash Pilar Carlton Johnny Kumar Prabhash Amit Joshi Jai Prakash Agarwal Tejpal Gupta Ashwini Budrukkar Vedang Murthy Monali Swain Vanita Noronha Vijay Maruthi Patil Prathamesh Pai Deepa Nair Devendra Arvind Chaukar Shivakumar Thiagarajan Gouri Pantvaidya Anuja Deshmukh Pankaj Chaturvedi Sudhir Nair Anil D’Cruz | 2020 | World Journal of Otorhinolaryngology-Head and Neck Surgery2020,6,1: | 1 |

| 14 | Antimalarial activity of a novel series of artemisinin-derived 1, 2, 3-triazole dimers显示文摘Objective: To obtain suitable artimisinin-based drug candidates with high antimalarial activity.Methods: Three different reaction schemes were used to synthesize a total of 15 artemisininbased compounds.The first synthetic scheme involved the synthesis of diazido aliphatic and aromatic compounds from commercially available dihalides and azido derivatives of artemisinin.The second scheme consisted of the reaction of dibromoaliphatic compounds with sodium azide in dimethylformamide which yielded the desired compounds.Artemisinin-based compounds on treatment with sodium azide and bromotrimethylsilane in dichloromethane produced the most potent compound GB-2.Another potent compound GB-1 was synthesized from artemisinin by treatment with alcohols in the presence of Aberlyst-15 in anhydrous dichloromethane.The third scheme involved the Huisgen 1,3-dipolar cycloaddition between the synthesized aliphatic and aromatic diazides and two alkyne derivatives of artemisinin to obtain the desired artemisinin dimers with average yields.Results: The best in vitro antiplasmodial activity was shown by the compound GB-2 registering IC_(50) value 0.066 μg/mL against chloroquine-sensitive and 0.865 μg/mL against chloroquineresistant strains of Plasmodium falciparum.It suppressed 59.0% parasitaemia in vivo of rodent malaria parasite Plasmodium berghei in Swiss albino model at 50 μg/kg body weight dosage.Molecular docking interactions of Plasmodium falciparum ATP6(PfATP6) protein revealed strong bonding of GB-2 with Thr255 residue which is likely to be the reason for excellent antimalarial activity of this compound.Conclusion: Two compounds GB-1 and GB-2 exhibited excellent in vitro antiplasmodial activity and fair in vivo antimalarial activity.Of the two, GB-2 showed better activity which could be attributed to its strong bonding interactions with Thr255 as evidenced from the molecular docking study.Study helped in identifying artemisinin analogues possessing good antimalarial properties and further research in structural alterations of the selected molecules should be carried out which may result in obtaining potent drug candidates against the malarial parasite. | Kabita Gogoi Gokul Baishya Biswajit Saikia Nabin Chandra Barua Chandrajit Dohutia Akalesh Kumar Verma Anil Prakash | 2019 | Asian Pacific Journal of Tropical Medicine2019,12,5: | 0 |

Standard procedures have been followed to evaluate the pharmacognostical properties and preliminary screening of phytochemicals of Cuscuta reflexa Roxb. Albino Wistar rats(either sex) weighing 150–200 g were taken and made to acclimatize by providing them with the appropriate room conditions in the experimental room. Excision wound model has been selected for analgesic and wound healing activity for evaluating potential of the water-extract and ethanolic extract from the stem of Cuscuta reflexa Roxb. The macroscopic and microscopic studies showed the various characteristic features of stems. Preliminary screening of phytochemicals reveled that alkaloids, glycosides, flavonoids were the major groups of phytochemicals present in the successive extracts. Water and ethanolic extracts from stem of Cuscuta reflexa Roxb at 200 mg/kg and 400 mg/kg were able to alleviate pain and wounds.
