1Comparative Analysis of JmjC Domain-containing Proteins Reveals the Potential Histone Demethylases in Arabidopsis and Rice显示文摘Histone methylation homeostasis is achieved by controlling the balance between methylation and demethylation to maintain chromatin function and developmental regulation. In animals, a conserved Jumonji C (JmjC) domain was found in a large group of histone demethylases. However, it is still unclear whether plants also contain the JmjC domaincontaining active histone demethylases. Here we performed genome-wide screen and phylogenetic analysis of JmjC domain-containing proteins in the dicot plant, Arabidopsis, and monocot plant rice, and found 21 and 20 JmjC domaincontaining, respectively. We also examined the expression of JmjC domain-containing proteins and compared them to human JmjC counterparts for potential enzymatic activity. The spatial expression patterns of the Arabidopsis JmjC domaincontaining genes revealed that they are all actively transcribed genes. 2Efficacy and safety evaluation of intravenous infusion of cervus and cucumis polypeptides for treatment of avascular necrosis of the femoral head: a randomized clinical trial显示文摘OBJECTIVE: To investigate the efficacy and safety of intravenous cervus and cucumis polypeptides for treating avascular necrosis of the femoral head(ANFH) in regard to pain and hip function in a randomized clinical trial.METHODS: A total of 96 subjects with ANFH who were recruited at the Orthopaedic Hospital Affiliated with Hebei United University and Qian Hai Femoral Head Hospital of Beijing were assigned by lottery to an intervention group(n = 48) or a control group(n = 48). All subjects underwent physical therapy and rehabilitation exercises. In addition,subjects in the intervention group were given intravenous infusions of cervus and cucumis polypeptides. Visual analogue scale(VAS), Harris hip score,and radiography or magnetic resonance imaging were applied to assess all subjects at the beginning of treatment and 3, 6, and 9 months afterward. All the subjects were followed up for 2 years.RESULTS: At the beginning of treatment, there were no statistically significant differences between the two groups in terms of the general condition of patients or the VAS and Harris hip scores(all P > 0.05). At 3, 6, and 9 months after treatment,however, the VAS score decreased and the Harris hip score increased in all patients, with the improvement of intervention group significantly greater than that of the control group(P < 0.05).The total effectiveness rates for the intervention and control groups were 89.58% and 70.83%, respectively, with the difference being statistically significant(P < 0.05). 4Association of KIR Genotypes and Haplotypes with Susceptibility to Chronic Hepatitis B Virus Infection in Chinese Han Population显示文摘Killer immunoglobulin-like receptor (KIR) genes can regulate the activation of NK and T cells upon interaction with HLA class I molecules. Hepatitis B virus (HBV) infection has been regarded as a multi-factorial disorder disease. Previous studies revealed that KIRs were involved in HCV and HIV infection or clearance. The aim of this study was to explore the possibility of the inheritance of KIR genotypes and haplotypes as a candidate for susceptibility to persistent HBV infection or HBV clearance. The sequence specific primer polymerase chain reaction (SSP-PCR) was employed to identify the KIR genes and pseudogenes in 150 chronic hepatitis B (CHB) patients, 251 spontaneously recovered (SR) controls, and 412 healthy controls. The frequencies of genotype G, M, FZ1 increased in CHB patients compared with healthy control subjects. The frequency of genotype AH was higher in SR controls than that in both CHB patients and healthy controls. The carriage frequencies of genotype G and AH were higher; while, the frequencies of AF and AJ were lower in SR controls than those in healthy control subjects. The frequency of A haplotype was lower, whereas, the frequency of B haplotype was higher in CHB patients and SR controls than those in healthy controls. In healthy controls, haplotype 4 was found lower compared with that in CHB patients and SR controls and the frequency of haplotype 5 was higher in SR controls than that in other two groups. Based on these findings, it seems that the genotypes M and FZ1 are HBV susceptive genotypes; AH, on the other hand, may be protective genotypes that facilitate the clearance of HBV. 5ULK1 and JNK are involved in mitophagy incurred by LRRK2 G2019S expression显示文摘Mutations in LR RK2(Leucine rich repeat kinase 2)are a major cause of Parkinson’s disease(PD).We and others reported recently that expression of the pathogenic gain-of-function mutant form of LRRK2,LRRK2 G2019S,induc-es mitochondrial fi ssion in neurons through DLP1.Here we provide evidence that expression of LRRK2 G2019S stimulates mitochondria loss or mitophagy.We have char-acterized several LRRK2 interacting proteins and found that LRRK2 interacts with ULK1 which plays an essential role in autophagy.Knockdown of either ULK1 or DLP1 expression with shRNAs suppresses LRRK2 G2019S expression-induced mitochondrial clearance,suggesting that LRRK2 G2019S expression induces mitochondrial fi ssion through DLP1 followed by mitophagy via an ULK1 dependent pathway.In addition to ULK1,we found that LRRK2 interacts with the endogenous MKK4/7,JIP3 and coordinates with them in the activation of JNK signaling.Interestingly,LRRK2 G2019S-induced loss of mitochon-dria can also be suppressed by 3 different JNK inhibitors,implying the involvement of the JNK pathway in the path-ogenic mechanism of mutated LRRK2.Thus our fi ndings may provide an insight into the complicated pathogenesis of PD as well as some clues to the development of novel therapeutic strategies.Yuangang Zhu Chunyan Wang Mei Yu Jie Cui Liang Liu Zhiheng Xu 2013Protein & Cell2013,4,9:6
6IL-17A-dependent gut microbiota is essential for regulating diet-induced disorders in mice显示文摘The gut microbiota plays a key role in obesity and related metabolic disorders, and multiple factors including diet, host genotype, and age regulate it. Many studies have examined the contribution of extrinsic factors to the regulation of the gut microbiota, but the importance of the host genetic constitution cannot be ignored. Interleukin 17A(IL-17A), a pro-inflammatory cytokine, is important in the defense against infection and diseases. Here, we investigated the association among IL-17, a high-fat diet(HFD), and the gut microbiota. Mice deficient in IL-17A were resistant to diet-induced obesity and related diseases. Compared with the Ⅱ-17^(-/-)mice, wild-type(WT) mice challenged with HFD showed obvious weight fluctuations, such as those seen in type 2 diabetes, and hematological changes similar to those associated with metabolic syndrome. However, housing WT mice and Ⅱ-17a^(-/-)mice together significantly alleviated these symptoms in the WT mice. A metagenomic analysis of the mouse feces indicated that the microbial community compositions of these two groups differed before HFD feeding. The HFD mediated shifts in the gut microbial compositions, which were associated with the mouse phenotypes.We also identified potentially beneficial and harmful species present during this period, and drew networks of the most abundant species. A functional analysis indicated pathway changes in the WT and Ⅱ-17a^(-/-)mice when fed the HFD. 7Enhanced production of seed oil with improved fatty acid composition by overexpressing NAD^(+)-dependent glycerol-3-phosphate dehydrogenase in soybean显示文摘There is growing interest in expanding the production of soybean oils(mainly triacylglycerol, or TAG) to meet rising feed demand and address global energy concerns. We report that a plastidlocalized glycerol-3-phosphate dehydrogenase(GPDH), encoded by GmGPDHp1 gene, catalyzes the formation of glycerol-3-phosphate(G3 P), an obligate substrate required for TAG biosynthesis.Overexpression of GmGPDHp1 increases soybean seed oil content with high levels of unsaturated fatty acids(FAs), especially oleic acid(C18:1), without detectably affecting growth or seed protein content or seed weight. Based on the lipidomic analyses, we found that the increase in G3 P content led to an elevated diacylglycerol(DAG) pool, in which the Kennedy pathwayderived DAG was mostly increased, followed by PC-derived DAG, thereby promoting the synthesis of TAG containing relatively high proportion of C18:1. The increased G3 P levels induced several transcriptional alterations of genes involved in the glycerolipid pathways. In particular, genes encoding the enzymes responsible for de novo glycerolipid synthesis were largely upregulated in the transgenic lines, in-line with the identified biochemical phenotype. 8Tea eggs-inspired high-strength natural polymer hydrogels显示文摘Natural polymer(NP)hydrogels are an irreplaceable class of biomaterials owing to their identified biosafety;however,the intrinsic poor mechanical strengths severely limit their applications as structural tissue engineering scaffolds.Inspired by the stiffening albumen gel of tea eggs,a traditional Chinese snack,high-strength NP hydrogels are constructed by simply soaking in aqueous solution of tea polyphenols(TP),an active ingredient extracted from tea.The TP-treated representative NP hydrogels exhibit considerably enhanced multifaceted mechanical properties with maximum 19-/30-,8.4-,6.1-,72-fold increases in tensile/compressive strengths,Young’s modulus,elongation at break and facture toughness,respectively,compared with pristine hydrogels,primarily due to the hydrogen bonding interactions between TP and NP chains.The TP-treated NP hydrogels can resist different large deformations,which cannot be achieved by their original species at all.In aqueous solution,the TP-treated NP hydrogels can still maintain robust mechanical performances,in spite of somewhat decline in strengths with release of TP,which just favorably affords increased water contents,antibacterial and antioxidant activities.GelMA-TP hydrogel is shown to facilitate wound healing in a full-thickness skin defect model.Importantly,the weak 3D printed GelMA scaffolds are significantly strengthened by TP treatment,broadening the possibility for customizing individualized bioscaffolds.Tengling Wu Chunyan Cui Chuanchuan Fan Ziyang Xu Yang Liu Wenguang Liu 2021Bioactive Materials2021,6,9:3

10Biomedical polymers: synthesis, properties, and applications显示文摘Biomedical polymers have been extensively developed for promising applications in a lot of biomedical fields, such as therapeutic medicine delivery, disease detection and diagnosis, biosensing, regenerative medicine, and disease treatment. In this review, we summarize the most recent advances in the synthesis and application of biomedical polymers, and discuss the comprehensive understanding of their property-function relationship for corresponding biomedical applications. In particular, a few burgeoning bioactive polymers, such as peptide/biomembrane/microorganism/cell-based biomedical polymers, are also introduced and highlighted as the emerging biomaterials for cancer precision therapy. Furthermore, the foreseeable challenges and outlook of the development of more efficient, healthier and safer biomedical polymers are discussed. 18A hyperbranched polymer-based water-resistant adhesive:Durable underwater adhesion and primer for anchoring anti-fouling hydrogel coating显示文摘Direct deployment of gluing and achieving durable robust adhesion in water is challenging due to difficulty in repelling interface water.This work reports a novel hyperbranched polymer-based water-resistant adhesive(HBPBA)based on Michael addition reaction of multi-vinyl monomers with dopamine and 3-aminophenylboronic acid.Upon encountering water,the HBPBA forms coacervates whose hydrophobic chains aggregate to displace interface water,and meanwhile the catechols exposing outwards contribute to underwater adhesion.The HBPBA can strongly glue diverse substrates including PTFE,PE,PET,ceramic,Ti and stainless steel.The HBPBA can maintain stable adhesion in different environments,such as tap water,simulated sea water,PBS,and a wide range of pH solutions(pH 2 to 10)for 3 months,supposedly due to the complexation of catechol with boronic acid.Intriguingly,HBPBA film can be bonded to the titanium surface as a primer,which firmly anchors the antifouling PNAGAPCBAA hydrogel coating through copolymerization of remaining double bonds in HBPBA and NAGA plus CBAA.The PNAGA-PCBAA hydrogel-modified titanium is biocompatible and shows outstanding antifouling ability both in vitro and in vivo.This work proposes a new strategy for creating underwater deployable and water-resistant adhesives that may find promising applications in engineering and biomedical fields.ZHANG YaHan CUI ChunYan SUN YaGe ZHANG XiaoPing YANG Rong YANG JianHai XIE Fei LIU WenGuang 2022Science China(Technological Sciences)2022,65,1:1
