| 1 | Vitamin B12 modulates Parkinson’s disease LRRK2 kinase activity through allosteric regulation and confers neuroprotection显示文摘Missense mutations in Leucine-Rich Repeat Kinase 2(LRRK2)cause the majority of familial and some sporadic forms of Parkinson’s disease(PD).The hyperactivity of LRRK2 kinase induced by the pathogenic mutations underlies neurotoxicity,promoting the development of LRRK2 kinase inhibitors as therapeutics.Many potent and specific small-molecule LRRK2 inhibitors have been reported with promise.However,nearly all inhibitors are ATP competitive—some with unwanted side effects and unclear clinical outcome—alternative types of LRRK2 inhibitors are lacking.Herein we identify 5′-deoxyadenosylcobalamin(AdoCbl),a physiological form of the essential micronutrient vitamin B12 as a mixed-type allosteric inhibitor of LRRK2 kinase activity.Multiple assays show that AdoCbl directly binds LRRK2,leading to the alterations of protein conformation and ATP binding in LRRK2.STD-NMR analysis of a LRRK2 homologous kinase reveals the contact sites in AdoCbl that interface with the kinase domain.Furthermore,we provide evidence that AdoCbl modulates LRRK2 activity through disrupting LRRK2 dimerization.Treatment with AdoCbl inhibits LRRK2 kinase activity in cultured cells and brain tissue,and prevents neurotoxicity in cultured primary rodent neurons as well as in transgenic C.elegans and D.melanogaster expressing LRRK2 disease variants.Finally,AdoCbl alleviates deficits in dopamine release sustainability caused by LRRK2 disease variants in mouse models.Our study uncovers vitamin B12 as a novel class of LRRK2 kinase modulator with a distinct mechanism,which can be harnessed to develop new LRRK2-based PD therapeutics in the future. | Adam Schaffner Xianting Li Yacob Gomez-Llorente Emmanouela Leandrou Anna Memou Nicolina Clemente Chen Yao Farinaz Afsari Lianteng Zhi Nina Pan Keita Morohashi Xiaoluan Hua Ming-Ming Zhou Chunyu Wang Hui Zhang Shu G.Chen Christopher J.Elliott Hardy Rideout Iban Ubarretxena-Belandia Zhenyu Yue | 2019 | Cell Research2019,29,4: | 7 |

| 6 | Evaluating alternative hypotheses behind biodiversity and multifunctionality relationships in the forests of Northeastern China显示文摘Background:The importance of biodiversity in maintaining multiple ecosystem functions has been widely accepted.However,the specific mechanisms affecting biodiversity and ecosystem multifunctionality(BEMF)relationships in forests are largely unknown.This is particularly evident for the macroscale of a large forested landscape.Methods:Based on 412 one-tenth hectare field plots distributed over forested areas across northeastern China,we evaluated three alternative hypotheses explaining the relationships between BEMF,namely:niche complementarity,mass ratio,and vegetation quantity effect.We used Rao's quadratic entropy and community weighted mean trait values to quantify forest“biodiversity”.These two variables represent two complementary aspects of functional properties,which are in line with niche complementary and mass ratio effects,respectively.Results:Ecosystem multifunctionality was negatively associated with the community weighted mean values of acquisitive traits(a proxy of mass ratio effect).Rao's quadratic entropy(a proxy of niche complementarity)had no relationship with ecosystem multifunctionality.Higher stand biomass greatly increased ecosystem multifunctionality,which is in line with the vegetation quantity effect.Our results confirm that in the temperate forests of northeastern China,the relationship of BEMF was primarily affected by vegetation quantity,followed by mass ratio effects.Conclusions:The results of this study contribute to a better understanding of the main drivers of ecosystem multifunctionality in forest ecosystems.The results of this study provide additional evidence to support the vegetation quantity and mass ratio hypotheses in forest ecosystems. | Qingmin Yue Minhui Hao Yan Geng Xuerui Wang Klaus von Gadow Chunyu Zhang Xiuhai Zhao Lushuang Gao | 2022 | Forest Ecosystems2022,9,3: | 1 |

| 20 | Dt1 inhibits SWEET-mediated sucrose transport to regulate photoperiod-dependent seed weight in soybean显示文摘Soybean is a photoperiod-sensitive short-day crop whose reproductive period and yield are markedly affected by day-length changes.Seed weight is one of the key traits determining the soybean yield;how-ever,the prominent genes that control the final seed weight of soybean and the mechanisms underlying the photoperiod's effect on this trait remain poorly understood.In this study,we identify SwW19 as a major locus controlling soybean seed weight by QTL mapping and determine Dt1,an orthologous gene of Arabidopsis TFL1 that is known to govern the soybean growth habit,as the causal gene of the SW19 locus.We showed that Dt1 is highly expressed in developing seeds and regulates photoperiod-dependent seed weight in soybean.Further analyses revealed that the Dt1 protein physically interacts with the sucrose transporter GmSWEET10a to negatively regulate the import of sucrose from seed coat to the embryo,thus modulating seed weight under long days.However,Dt1 does not function in seed development under short days due to its very low expression.Importantly,we discovered a novel natural allelic variant of Dt1(H4 haplotype)that decouples its pleiotropic effects on seed size and growth habit;i.e.,this variant remains functional in seed development but fails to regulate the stem growth habit of soybean.Collectively,our findings provide new insights into how soybean seed development responds to photoperiod at different latitudes,offering an ideal genetic component for improving soybean's yield by manipulating its seed weightandgrowth habit. | Xiaoming Li Zhonghui Chen Haiyang Li Lin Yue Cuirong Tan Hongjie Liu Yilong Hu Yuhua Yang Xiani Yao Lingping Kong Xiang Huangi Bin Yu Chunyu Zhang Yuefeng Guan Baohui Liu Fanjiang Kong Xingliang Hou | 2024 | Molecular Plant2024,17,3: | 0 |
