2Osteoimmunomodulatory effects of biomaterial modification strategies on macrophage polarization and bone regeneration显示文摘Biomaterials as bone substitutes are always considered as foreign bodies that can trigger host immune responses.Traditional designing principles have been always aimed at minimizing the immune reactions by fabricating inert biomaterials.However,clinical evidence revealed that those methods still have limitations and many of which were only feasible in the laboratory.Currently,osteoimmunology,the very pioneering concept is drawing more and more attention-it does not simply regard the immune response as an obstacle during bone healing but emphasizes the intimate relationship of the immune and skeletal system,which includes diverse cells,cytokines,and signaling pathways.Properties of biomaterials like topography,wettability,surface charge,the release of cytokines,mediators,ions and other bioactive molecules can impose effects on immune responses to interfere with the skeletal system.Based on the bone formation mechanisms,the designing methods of the biomaterials change from immune evasive to immune reprogramming.Here,we discuss the osteoimmunomodulatory effects of the new modification strategies—adjusting properties of bone biomaterials to induce a favorable osteoimmune environment.Such strategies showed potential to benefit the development of bone materials and lay a solid foundation for the future clinical application.Yajuan Xie Cheng Hu Yi Feng Danfeng Li Tingting Ai Yulei Huang Xiaodan Chen Lijia Huang Jiali Tan 2020Regenerative Biomaterials2020,7,3:22

4Mammalian target of rapamycin pathway inhibition enhances the effects of 5-aza-dC on suppressing cell proliferation in human gastric cancer cell lines显示文摘The present study aimed to evaluate the relationship between mTOR signaling pathway and DNA methylation in cell survival,cell cycle,gene expression and protein level on human gastric cancer cells. Human gastric cancer cell lines,MKN45 and SGC7901 were treated with 5-aza-dC,rapamycin and/or LY294002.Cell viability was analyzed by MTT.Cell cycle distribution was evaluated by flow cytometry (FCM).The transcription level of PTEN and p27 Kip1 genes was detected by using real-time PCR.Protein expressions were detected by Western blotting.We found that cell viability was moderately reduced when treated with 5-aza-dC alone,but remarkably reduced when mTOR pathway was inhibited together (P<0.01).mTOR inhibition enhances the effects of 5-aza-dC on arresting cell cycle at G2 phase in human gastric cancer cell lines.The expression of PTEN and p27 Kip1 mRNA was remarkably increased in the gastric cancer cells treated with combind drugs(P<0.01).Phosphorylation of Akt,p70S6K and 4E-BP1 were significantly reduced in the cells treated with LY294002 or RAPA(P<0.01),but we failed to find that 5-aza-dC enhance these effects.We suggested that mTOR inhibition could enhance the effects of 5-aza-dC on suppressing cell proliferation and arresting cell cycle in human gastric cancer cell lines, which might be a potential target for tumor therapy.SUN DanFeng,TIAN XiaoQing,ZHANG YanJie,CHEN YingXuan,LU Rong,WANG Xia &FANG JingYuan Shanghai Jiao-Tong University Sc0hool of Medicine Renji Hospital,Shanghai Institute of Digestive Disease,Shanghai 200001, China 2008Science China(Life Sciences)2008,51,7:12
5MicroRNAs and Their Cross-Talks in Plant Development显示文摘Plant development is a complex process influenced by exogenous and endogenous elements.A series of postembryonic developmental events is involved to form the final architecture and contend with the changing environment.MicroRNA(miRNA) is one of endogenous non-coding RNAs,which plays an important role in plant developmental regulation.In this review,we summarized 34 miRNA families that are closely associated with plant development.Among these families,nine are expressed only in specific organs,whereas 20 families are expressed in at least two different organs.It is known that some miRNAs are expressed across most processes of plant growth,while some appear only at particular developmental stages or under special environmental conditions such as drought,waterlogging and short-day time.These miRNAs execute their diverse functions by regulating developmental gene expression levels,interacting with phytohormone signaling response,participating in the biogenesis of ta-siRNAs and affecting the production of miRNAs.Danfeng Jin Yue Wang Yuhua Zhao Ming Chen 2013Journal of Genetics and Genomics2013,40,4:9
6Percent free prostate-specific antigen for prostate cancer diagnosis in Chinese men with a PSA of 4.0-10.0 ng/mL:Results from the Chinese Prostate Cancer Consortium显示文摘Objective:To test the diagnostic performance of percent free prostate-specific antigen(%fPSA)in predicting any prostate cancer(PCa)and high-grade prostate cancer(HGPCa)in a retrospective multi-center biopsy cohort with a PSA level of 4.0e10.0 ng/mL in China.Methods:Consecutive patients with a PSA of 4.0-10.0 ng/mL who underwent transrectal ultrasound-guided biopsy were enrolled at 16 Chinese medical centers from January 1st,2010 to December 31st,2013.Total and free serum PSA determinations were performed using three types of electro-chemiluminescence immunoassays recalibrated to the World Health Organization(WHO)standard.The diagnostic accuracy of PSA,%fPSA,and %fPSA in combination with PSA(%fPSA t PSA)was determined using the area under the receiver operating characteristic(ROC)curve(AUC).Results:A total of 2310 consecutive men with PSA levels between 4.0 and 10.0 ng/mL were included,and the detection rate of PCa was 25.1%.The AUC of%fPSA and %fPSA t PSA in predicting any PCa was superior to PSA alone in men aged≥60 years(0.623 vs.0.534,p<0.0001)but not in men aged 40e59 years(0.517 vs.0.518,p=0.939).Similar result was yield in predicting HGPCa.Conclusion:In a clinical setting of Chinese men with 4.0e10.0 ng/mL PSA undergoing initial prostate biopsy,adding %fPSA to PSA can moderately improve the diagnostic accuracy for any PCa and HGPCa compared with PSA alone in patients≥60 but not in patients aged 40-59 years.Rui Chen Liping Xie Xiaobing Cai Yiran Huang Liqun Zhou Lulin Ma Xu Gao Chuanliang Xu Shancheng Ren Pengfei Shao Danfeng Xu Kexin Xu Zhangqun Ye Chunxiao Liu Dingwei Ye Li Lu Qiang Fu Jianquan Hou Jianlin Yuan Dalin He Tie Zhou Fubo Wang Biming He Yinghao Sun 2015Asian Journal of Urology2015,2,2:8
7Circulating tumor cells:biology and clinical significance显示文摘Circulating tumor cells(CTCs)are tumor cells that have sloughed off the primary tumor and extravasate into and circulate in the blood.Understanding of the metastatic cascade of CTCs has tremendous potential for the identification of targets against cancer metastasis.Detecting these very rare CTCs among the massive blood cells is challenging.However,emerging technologies for CTCs detection have profoundly contributed to deepening investigation into the biology of CTCs and have facilitated their clinical application.Current technologies for the detection of CTCs are summarized herein,together with their advantages and disadvantages.The detection of CTCs is usually dependent on molecular markers,with the epithelial cell adhesion molecule being the most widely used,although molecular markers vary between different types of cancer.Properties associated with epithelial-tomesenchymal transition and stemness have been identified in CTCs,indicating their increased metastatic capacity.Only a small proportion of CTCs can survive and eventually initiate metastases,suggesting that an interaction and modulation between CTCs and the hostile blood microenvironment is essential for CTC metastasis.Single-cell sequencing of CTCs has been extensively investigated,and has enabled researchers to reveal the genome and transcriptome of CTCs.Herein,we also review the clinical applications of CTCs,especially for monitoring response to cancer treatment and in evaluating prognosis.Hence,CTCs have and will continue to contribute to providing significant insights into metastatic processes and will open new avenues for useful clinical applications.Danfeng Lin Lesang Shen Meng Luo Kun Zhang Jinfan Li Qi Yang Fangfang Zhu Dan Zhou Shu Zheng Yiding Chen Jiaojiao Zhou 2021Signal Transduction and Targeted Therapy2021,6,12:8
8Determining reference conditions for TN,TP,SD and Chl-a in eastern plain ecoregion lakes,China显示文摘Establishing the nutrient reference condition(baseline environmental condition) of lakes in an ecoregion is a critical consideration in the development of scientifically defensible aquatic nutrient criteria.Three methods were applied to determine reference conditions in the Eastern plain ecoregion lakes with respect to total phosphorus(TP),total nitrogen(TN),planktonic chlorophyll a(Chl-a) and Secchi depth(SD).The reference condition value for the lakes in the Eastern plain ecoregion by the trisection method is TP of 0.029 mg/L,TN of 0.67 mg/L,Chl-a of 3.92 mg/m 3,SD of 0.85 m,and the reference condition range by the lake population distribution approach is TP of 0.014-0.043 mg/L,TN of 0.360-0.785 mg/L,Chl-a of 1.78-4.73 mg/m 3,SD of 0.68-1.21 m.Additionally,empirical models were developed for estimating the reference Chl-a concentration and SD successfully for lakes in the Eastern plain ecoregion.Overall,the data suggest that multiple methods can be used to determine reference conditions and that in Eastern plain ecoregion lakes the reference condition corresponds to a mesotrophic status.Shouliang Huo Beidou Xi Jing Su Fengyu Zan Qi Chen Danfeng Ji Chunzi Ma 2013Journal of Environmental Sciences2013,25,5:7
9Discovery of the ancient Ordovician oil-bearing karst cave in Liuhuanggou,North Tarim Basin,and its significance显示文摘A typical ancient karst cave is found in the Yijianfang Formation of Ordovician at Liuhuanggou,North Tarim Basin.With an entrance of 4.8 m wide and 20 m high,the Liuhuanggou ancient karst cave extends to NW 315° and has been filled by sediments,which can be classified into three types according to their nature and origin:clastic sediments(gravels),chemical sediments(palaeotravertine),and mixed sediments.Among them,the Paleotravertine and gravels are most common.The palaeotravertine is laminated type and yellowish brown or black in color due to oil inside.When saturated with oil,the palaeotravertine is black.Under microscope,oil mainly accumulates in the cracks between calcite crystals.The filling period of the Liuhuanggou karst cave can be divided into three stages:mixed filling,chemical filling,and clastic(palaeotravertine breccia) filling.Numerous fractures are distributed on one side of the karst cave.The fractures can be as wide as 40 cm and over 40 m long.Fractures of different periods and sizes constitute a fracture network and are all filled with calcite veins.Oil can be found inside the fractures.In the chemical fillings,sulfur is also found,which can indicate the magma influence on karst caves.The existence of sulfur also confirms that the Liuhunggou karst cave was formed in ancient times rather than recently or nowadays.Different from normal sedimentary limestone,the ancient Liuhuanggou karst cave has a distinct isotope fractionation and the depletion of heavy carbon and oxygen isotope.Controlled by the northwest oriented vertical strike-slip shear fault,the karst cave is actually a dissolved vertical fault belt.ZHONG JianHua MAO Cui LI Yong LI Yang YUAN XiangChun NIU YongBin CHEN Xin HuANG ZhiJuan SHAO ZhuFu WANG PeiJun LI Jia ZHANG DanFeng 2012Science China Earth Sciences2012,55,9:4
10Analysis of the Global Burden of Disease study highlights the trends in death and disability-adjusted life years of leukemia from 1990 to 2017显示文摘Background:The patterns of leukemia burden have dramatically changed in recent years.This study aimed to estimate the global trends of leukemiarelated death and disability-adjusted life-years(DALYs)from 1990 to 2017.Methods:The data was acquired from the latest version of the Global Burden of Disease(GBD)study.Estimated annual percentage changes(EAPCs)were calculated to estimate the trend of age-standardized rate(ASR)of death and DALYs due to leukemia and its main subtypes from 1990 to 2017.Results:Globally,the numbers of death and DALYs due to leukemia were 347.58×10^(3)(95%uncertainty interval[UI]=317.26×10^(3)-364.88×10^(3))and 11975.35×10^(3)(95%UI=10749.15×10^(3)-12793.58×10^(3))in 2017,with a 31.22% and 0.03% increase in absolute numbers from 1990 to 2017,respectively.Both of their ASR showed decreasing trends from 1990 to 2017 with the EAPCs being−1.04(95%confidence interval[CI]=(−1.10-−0.99)and−1.52(95%CI=−1.59-−1.44),respectively.Globally,the most pronounced decreasing trend of death and DALYs occurred in chronic myeloid leukemia with EAPCs of−2.76(95%CI=−2.88-−2.64)and−2.84(95%CI=−2.97-−2.70),respectively,while the trend increased in acute myeloid leukemia.The death and DALYs of leukemia decreased in most areas and countries with high socio-demographic index(SDI)including Bahrain,Finland,and Australia.Conclusions:The disease burden of death and DALYs due to leukemia decreased globally,and for most regions and countries from 1990 to 2017.However,the leukemia burden is still a substantial challenge globally and required adequate and affordable medical resources to improve the survival and quality of life of leukemia patients.Zejin Ou Danfeng Yu Yuanhao Liang Wenqiao He Yongzhi Li Minyi Zhang Fangfei You Huan He Qing Chen 2020Cancer Communications2020,40,11:4

12Bioresponsive nanotherapy for preventing dental caries by inhibiting multispecies cariogenic biofilms显示文摘Early childhood caries(ECC)is a public healthcare concern that greatly reduces the quality of life of young children.As a leading factor of ECC,cariogenic biofilms are composed of acidogenic/aciduric pathogens and extracellular polysaccharides(EPSs),creating an acidic and protected microenvironment.Antimicrobial photodynamic therapy(aPDT)is a noninvasive,painless,and efficient therapeutic approach that is suitable for treating ECC.However,due to the hyperfine structure of cariogenic biofilms,most photosensitizers(PSs)could not access and penetrate deeply in biofilms,which dramatically hamper their efficiency in the clinic.Herein,bioresponsive nanoparticle loaded with chlorin e6(MPP-Ce6)is developed,which largely increases the penetration depth(by over 75%)and retention(by over 100%)of PS in the biofilm compared with free Ce6.Furthermore,MPP-Ce6-mediated aPDT not only kills the bacteria in preformed biofilms but also inhibits multispecies biofilm formation.A rampant caries model is established to mimic ECC in vivo,where the population of cariogenic bacteria is decreased to 10%after MPP-Ce6-mediated aPDT.Importantly,the number and severity of carious lesions are efficiently reduced via Keyes’scoring and micro-CT analysis.This simple but effective strategy can serve as a promising approach for daily oral hygiene in preventing ECC.Danfeng Liu Xianbin Ma Yaoting Ji Rourong Chen Shuhui Zhou Hantao Yao Zichen Zhang Mengjie Ye Zhigang Xu Minquan Du 2022Bioactive Materials2022,7,8:2

The architecture comprises a Single Instruction Multiple Data (SIMD) DSP core and three coprocessors that are used for synchronization, FFT and channel decoder. In this MIMO OFDM system, the Zero Correlation Zone (ZCZ) code is used as the synchronization word preamble of packet in the physical layer in order to avoid the interference from other transmitting antennas. 16Tumor extracellular vesicles mediate anti-PD-L1 therapyresistance by decoying anti-PD-L1显示文摘PD-L1+tumor-derived extracellular vesicles(TEVs)cause systemic immunosuppression and possibly resistance to anti-PD-L1 antibody(αPD-L1)blockade.However,whether and how PD-L1+TEVs mediateαPD-L1 therapy resistance is unknown.Here,we show that PD-L1+TEVs substantially decoyαPD-L1 and that TEV-boundαPD-L1 is more rapidly cleared by macrophages,causing insufficient blockade of tumor PD-L1 and subsequentαPD-L1 therapy resistance.Inhibition of endogenous production of TEVs by Rab27a or Coro1a knockout reversesαPD-L1 therapy resistance.Either an increasedαPD-L1 dose or macrophage depletion mediated by the clinical drug pexidartinib abolishesαPD-L1 therapy resistance.Moreover,in the treatment cycle with the same total treatment dose ofαPD-L1,high-dose and low-frequency treatment had better antitumor effects than low-dose and highfrequency treatment,induced stronger antitumor immune memory,and eliminatedαPD-L1 therapy resistance.Notably,in humanized immune system mice with human xenograft tumors,both increasedαPD-L1 dose and high-dose and low-frequency treatment enhanced the antitumor effects ofαPD-L1.Furthermore,increased doses ofαPD-L1 andαPD-1 had comparable antitumor effects,butαPD-L1 amplified fewer PD-1+Treg cells,which are responsible for tumor hyperprogression.Altogether,our results reveal a TEV-mediated mechanism ofαPD-L1-specific therapy resistance,thus providing promising strategies to improveαPD-L1 efficacy.Jiming Chen Jie Yang Wenhui Wang Danfeng Guo Chengyan Zhang Shibo Wang Xinliang Lu Xiaofang Huang Pingli Wang Gensheng Zhang Jing Zhang Jianli Wang Zhijian Cai 2022Cellular & Molecular Immunology2022,19,11:1

18Influence of the microbiota on epigenetics in colorectal cancer显示文摘Colorectal cancer is one of the most common malignancies and is the second leading cause of cancer death worldwide.Generally;there are three categories of colorectal cancer development mechanism-genetic,epigenetic and aberrant immunological signaling pathways-all of which may be initiated by an imbalanced gut microbiota.Epigenetic modifications enable host cells to change gene expression without modifying the gene sequence.The microbiota can interact with the host genome dynamically through the interface presented by epigenetic modifications.In particular,bacterially derived short-chain fatty acids have been identified as one clear link in the interaction of the microbiota with host epigenetic pathways.This review discusses recent findings relating to the cross talk between the microbiota and epigenetic modifications in colorectal cancer.Danfeng Sun Yingxuan Chen Jing-Yuan Fang 2019National Science Review2019,6,6:1

20Synchronous bilateral multiple chromophobe cell renal carcinoma complicated with right kidney cyst: a case report显示文摘Synchronous bilateral multiple chromophobe cell renal carcinoma is rare; here we report a case diagnosed with bilateral renal multiple tumors complicated with a cyst in the right kidney. Retroperitoneal laparoscopic bilateral nephron sparing surgery was performed and there was no serious postoperative renal dysfunction. Pathological and immunohistochemical diagnoses of both tumors were chromophobe cell renal carcinoma.
