1Effects of Predeformation on the Reverse Martensitic Transformation of TiNi Shape Memory Alloy显示文摘DSC was used to study the effects of predeformation on the reverse martensitic transformation of near-equiatomic TiNi alloy. Both the start temperature A. and the finish temperature Af of the reverse transformation increased with increasing degree of predeformation, but the algebraic difference between A. and Af decreased with increasing predeformation until it reached a minimum value, then remained unchanged with further deformation. Transformation heat also increased with increasing predeformation until it reached a maximum value, then decreased with further predeformation. 4Toripalimab plus chemotherapy as second-line treatment in previously EGFR-TKI treated patients with EGFR-mutant-advanced NSCLC:a multicenter phase-II trial显示文摘This multicenter phase-II trial aimed to investigate the efficacy,safety,and predictive biomarkers of toripalimab plus chemotherapy as second-line treatment in patients with EGFR-mutant-advanced NSCLC.Patients who failed from first-line EGFR-TKIs and did not harbor T790M mutation were enrolled.Toripalimab plus carboplatin and pemetrexed were administrated every three weeks for up to six cycles,followed by the maintenance of toripalimab and pemetrexed.The primary endpoint was objective-response rate(ORR).Integrated biomarker analysis of PD-L1 expression,tumor mutational burden(TMB),CD8+tumor-infiltrating lymphocyte(TIL)density,whole-exome,and transcriptome sequencing on tumor biopsies were also conducted.Forty patients were enrolled with an overall ORR of 50.0%and disease-control rate(DCR)of 87.5%.The median progression free survival(PFS)and overall survival were 7.0 and 23.5 months,respectively.The most common treatment-related adverse effects were leukopenia,neutropenia,anemia,ALT/AST elevation,and nausea.Biomarker analysis showed that none of PD-L1 expression,TMB level,and CD8+TIL density could serve as a predictive biomarker.Integrated analysis of whole-exome and transcriptome sequencing data revealed that patients with DSPP mutation had a decreased M2 macrophage infiltration and associated with longer PFS than those of wild type.Toripalimab plus chemotherapy showed a promising anti-tumor activity with acceptable safety profiles as the second-line setting in patients with EGFR-mutant NSCLC.DSPP mutation might serve as a potential biomarker for this combination.A phase-III trial to compare toripalimab versus placebo in combination with chemotherapy in this setting is ongoing(NCT03924050).Tao Jiang Pingyang Wang Jie Zhang Yanqiu Zhao Jianying Zhou Yun Fan Yongqian Shu Xiaoqing Liu Helong Zhang Jianxing He Guanghui Gao Xiaoqian Mu Zhang Bao Yanjun Xu Renhua Guo Hong Wang Lin Deng Ningqiang Ma Yalei Zhang Hui Feng Sheng Yao Jiarui Wu Luonan Chen Caicun Zhou Shengxiang Ren 2021Signal Transduction and Targeted Therapy2021,6,11:8
5Seasonal variation of sedimentation in the Changjiang Estuary mud area显示文摘Seasonal distributions of suspended matter and their sedimentary effect on the Changjiang Estuary mud area of the East China Sea were discussed, based on three cruise data of total suspended matter, temperature and salinity collected from the Changjiang Estuary and its adjacent area in summer and winter. The results show that the basic pattern of distributions of suspended matter in the study area is almost the same in winter and in summer. Sediments from Changjiang (Yangtze River) to the sea are chiefly trapped to the west of 123 o 15’E due to a strong obstruction of the Taiwan Warm Current. This suggests that these sediments are mainly transported and deposited in the inner shelf. The sediment supply, Taiwan Warm Current, and Zhejiang Coastal Current show a strong seasonal variation, which results in a strong seasonal variation of the sedimentary effect on this mud area. 7Activation of anterior thalamic reticular nucleus GABAergic neurons promotes arousal from propofol anesthesia in mice显示文摘Propofol is widely used for the induction and maintenance of anesthesia,which causes a rapid loss of consciousness.However,the mechanisms underlying the hypnosis effect of propofol are still not fully understood.The thalamic reticular nucleus(TRN)is crucial for regulating wakefulness,sleep rhythm generation,and sleep stability,while the role of TRN in the process of propofol-induced anesthesia is still unknown.Here,we investigated the function of the anterior TRN in propofol general anesthesia.Our results demonstrated that the neural activity of anterior TRN is suppressed during propofol anesthesia,whereas it is robustly activated from anesthesia by recording the calcium signals using fiber photometry technology.The results showed that the activation of anterior TRN neurons by chemogenetic and optogenetic methods shortens the emergency time without changing the induction time.Conversely,chemogenetic or optogenetic inhibition of the TRN neurons leads to a delay in the recovery time.Our study showed that anterior TRN is crucial for behavioral arousal without affecting the induction time of propofol anesthesia.Yanjun Liu Bing Chen Yirong Cai Yuan Han Ying Xia Nanqi Li Bingqian Fan Tianjie Yuan Junli Jiang Po Gao Weifeng Yu Yingfu Jiao Wenxian Li 2021Acta Biochimica et Biophysica Sinica2021,53,7:3
8Self-immolative micellar drug delivery:The linker matters显示文摘Redox-responsive polymer-drug conjugate micelles are excellent nanoscale vehicles for self-immolative intracellular drug delivery.To covalently connect the polymer and drug,disulfide-bearing linkers,such as 3,3'-dithiodipropionic acid (DDPA) and 4,4'-dithiodibutyric add (DDBA),are used.In this paper,we report the influence of linker length on the therapeutic outcome of redox-sensitive conjugate micelles.Curcumin was selected as the model drug and it was conjugated to a multivalent methoxy poly(ethylene glycol)-polylysine copolymer with DDPA or DDBA as the linker.The obtained two polymer-curcumin conjugates were amphiphilic and could self-assemble into micelles that have a hydrodynamic diameter less than 100 nm.The loading of curcumin in both micelles was above 20% (w/w).Irrespective of the linker type,micelle disassembly was observed due to the collapse of the disulfide bond in a reducing environment.However,the rate of curcumin release was much faster with the DDBA linker than with the DDPA linker as the side product was a 5-membered ring with a low ring strain.The linker length-induced variation of curcumin release kinetics caused a significant difference in the intracellular drug concentration and a higher cytotoxicity was witnessed in three model cell lines (HeLa,PC3,and 4T1) for the micelles with a DDBA linker compared to those containing a DDPA linker.As expected,this phenomenon was also observed in HeLa tumor-bearing nude mice in vivo.The current work highlights the significance of linker length in engineering redox-responsive on-demand delivery systems.Xuan Meng Min Gao Jian Deng Di Lu Aiping Fan Dan Ding Deling Kong Zheng Wang Yanjun Zhao 2018Nano Research2018,11,12:2

17HvWRKY2 acts as an immunity suppressor and targets HvCEBiP to regulate powdery mildew resistance in barley显示文摘Plants use a sophisticated immune system to perceive pathogen infection and activate immune responses in a tightly controlled manner.In barley,Hv WRKY2 acts as a repressor in barley disease resistance to the powdery mildew fungus,Blumeria graminis f.sp.hordei(Bgh).However,the molecular features of Hv WRKY2 in its DNA-binding and repressor functions,as well as its target genes,are uncharacterized.We show that the W-box binding of Hv WRKY2 requires an intact WRKY domain and an upstream sequence of~75 amino acids,and the Hv WRKY2 W-box binding activity is linked to its repressor function in disease resistance.Chromatin immunoprecipitation(ChIP)-seq analysis identified HvCEBiP,a putative chitin receptor gene,as a target gene of Hv WRKY2 in overexpressing transgenic barley plants.ChIP-qPCR and Electrophoretic Mobility Shift Assay(EMSA)verified the direct binding of Hv WRKY2 to a W-boxcontaining sequence in the HvCEBiP promoter.Hv CEBiP positively regulates resistance against Bgh in barley.Our findings suggest that Hv WRKY2 represses barley basal immunity by directly targeting pathogen-associated molecular pattern(PAMP)recognition receptor genes,suggesting that Hv CEBiP and likely chitin signaling function in barley PAMP-triggered immune responses to Bgh infection.Deshui Yu Renchun Fan Ling Zhang Pengya Xue Libing Liao Meizhen Hu Yanjun Cheng Jine Li Ting Qi Shaojuan Jing Qiuyun Wang Arvind Bhatt Qian-Hua Shen 2023The Crop Journal2023,11,1:1
18Triple-functional bone adhesive with enhanced internal fixation,bacteriostasis and osteoinductive properties for open fracture repair显示文摘At present,effective fixation and anti-infection implant materials represent the mainstay for the treatment of open fractures.However,external fixation can cause nail tract infections and is ineffective for fixing small fracture fragments.Moreover,closed reduction and internal fixation during the early stage of injury can lead to potential bone infection,conducive to bone nonunion and delayed healing.Herein,we designed a bone adhesive with anti-infection,osteogenic and bone adhesion fixation properties to promote reduction and fixation of open fractures and subsequent soft tissue repair.It was prepared by the reaction of gelatin(Gel)and oxidized starch(OS)with vancomycin(VAN)-loaded mesoporous bioactive glass nanoparticles(MBGNs)covalently cross-linked with Schiff bases.Characterization and adhesion experiments were conducted to validate the successful preparation of the Gel-OS/VAN@MBGNs(GOVM-gel)adhesive.Meanwhile,in vitro cell experiments demonstrated its good antibacterial effects with the ability to stimulate bone marrow mesenchymal stem cell(BMSCs)proliferation,upregulate the expression of alkaline phosphatase(ALP)and osteogenic proteins(RunX2 and OPN)and enhance the deposition of calcium nodules.Additionally,we established a rat skull fracture model and a subcutaneous infection model.The histological analysis showed that bone adhesive enhanced osteogenesis,and in vivo experiments demonstrated that the number of inflammatory cells and bacteria was significantly reduced.Overall,the adhesive could promote early reduction of fractures and antibacterial and osteogenic effects,providing the foothold for treatment of this patient population.Yusheng Yang Shenghui Su Shencai Liu Weilu Liu Qinfeng Yang Liangjie Tian Zilin Tan Lei Fan Bin Yu Jian Wang Yanjun Hu 2023Bioactive Materials2023,,7:1
19Optical Design and Stray Light Analysis of the Space Infrared Optical System显示文摘This article describes a novel configuration design for a re-imaging off-axis catadioptric space infrared optical system,and in order to satisfy the signal noise ratio requirements of the system,the stray light of the system is necessary to analyze and restrain. The optical system with a focal length of 1 200 mm,an entrance pupil diameter of 600 mm,an F-number of 2,a field of view of 3°× 0. 15°,a working wave band of 8 μm-10 μm,and the image quality of the optical system almost approach to diffraction limits in all field of view.Then the mathematical models of stray light are built,and the suppressive structure is established to eliminate the effect of stray light. Finally,TraceP ro is used to analyze and simulate stray light with and without the suppressive structure,and also get the results of the PST curves.
