1Biofunctional magnesium coated Ti6Al4V scaffold enhances osteogenesis and angiogenesis in vitro and in vivo for orthopedic application显示文摘The insufficient osteogenesis and osseointegration of porous titanium based scaffold limit its further application.Early angiogenesis is important for scaffold survival.It is necessary to develop a multifunctional surface on titanium scaffold with both osteogenic and angiogenic properties.In this study,a biofunctional magnesium coating is deposited on porous Ti6Al4V scaffold.For osseointegration and osteogenesis analysis,in vitro studies reveal that magnesium-coated Ti6Al4V co-culture with MC3T3-E1 cells can improve cell proliferation,adhesion,extracellular matrix(ECM)mineralization and ALP activity compared with bare Ti6Al4V cocultivation.Additionally,MC3T3-E1 cells cultured with magnesium-coated Ti6Al4V show significantly higher osteogenesisrelated genes expression.In vivo studies including fluorochrome labeling,micro-computerized tomography and histological examination of magnesium-coated Ti6Al4V scaffold reveal that new bone regeneration is significantly increased in rabbits after implantation.For angiogenesis studies,magnesium-coated Ti6Al4V improve HUVECs proliferation,adhesion,tube formation,wound-healing and Transwell abilities.HUVECs cultured with magnesium-coated Ti6Al4V display significantly higher angiogenesis-related genes(HIF-1αand VEGF)expression.Microangiography analysis reveal that magnesium-coated Ti6Al4V scaffold can significantly enhance the blood vessel formation.This study enlarges the application scope of magnesium and provides an optional choice to the conventional porous Ti6Al4V scaffold with enhanced osteogenesis and angiogenesis for further orthopedic applications.Peng Gao Bo Fan Xiaoming Yu Wenwen Liu Jie Wu Lei Shi Di Yang Lili Tan Peng Wan Yulin Hao Shujun Li Wentao Hou Ke Yang Xiaokang Li Zheng Guo 2020Bioactive Materials2020,5,3:15
2Secretory/releasing proteome-based identification of plasma biomarkers in HBV-associated hepatocellular carcinoma显示文摘For successful therapy, hepatocellular carcinoma (HCC) must be detected at an early stage. Herein, we used a proteomic approach to analyze the secretory/releasing proteome of HCC tissues to identify plasma biomarkers. Serum-free conditioned media (CM) were collected from primary cultures of cancerous tissues and surrounding noncancerous tissues. Proteomic analysis of the CM proteins permitted the identification of 1365 proteins. The enriched molecular functions and biological processes of the CM proteins, such as hydrolase activity and catabolic processes, were consistent with the liver being the most important metabolic organ. Moreover, 19% of the proteins were characterized as extracellular or membrane-bound. For validation, secretory proteins involved in transforming growth factor-β signaling pathways were validated in plasma samples. Alphafetoprotein (AFP), metalloproteinase (MMP)1, osteopontin (OPN), and pregnancy-specific beta-1-glycoprotein (PSG)9 were significantly increased in HCC patients. The overall performance of MMP1 and OPN in the diagnosis of HCC remained greater than that of AFP. In addition, this study represents the first report of MMP1 as a biomarker with a higher sensitivity and specificity than AFP. Thus, this study provides a valuable resource of the HCC secretome with the potential to investigate serological biomarkers. 3Post-surgical resection prognostic value of combined OPN, MMP7, and PSG9 plasma biomarkers in hepatocellular carcinoma显示文摘Biomarkers for hepatocellular carcinoma (HCC) following curative resection are not currently sufficient for prognostic indication of overall survival (OS) and disease-free survival (DFS). The aim of this study was to investigate the prognostic performance of osteopontin (OPN), matrix metalloproteinase 7 (MMP7), and pregnancy specific glycoprotein 9 (PSG9) in patients with HCC. A total of 179 prospective patients with HCC provided plasma before hepatectomy. Plasma OPN, MMP7, and PSG9 levels were determined by enzyme-linked immunosorbent assay. Correlations between plasma levels, clinical parameters, and outcomes (OS and DFS) were overall analyzed. High OPN (≥149.97 ng/mL), MMP7 (≥2.28 ng/mL), and PSG9 (≥45.59 ng/mL) were prognostic indicators of reduced OS (P<0.001, P<0.001, and P=0.007, respectively). Plasma PSG9 protein level was an independent factor in predicting OS (P=0.008) and DFS (P=0.038). Plasma OPN+MMP7+PSG9 elevation in combination was a prognostic factor for OS (P<0.001). OPN was demonstrated to be a risk factor-associated OS in stage I patients with HCC and patients with low α-fetoprotein levels (<20 ng/mL). 4Method of moving target detection based on sub-image cancellation for single-antenna airborne synthetic aperture radar显示文摘The method of moving target detection based on subimage cancellation for single-antenna airborne SAR is presented.First the subimage is obtained through frequency processing is pointed out.The imaging difference of a stationary objects and moving object in the subimage based on the frequency division is analyzed from the fundamental principle.Then the developed method combines the shear averaging algorithm to focus on the moving target in the subimage,after the clutter suppression and the focusing position in each subimage is obtained. 6PSG9 promotes angiogenesis by stimulating VEGFA production and is associated with poor prognosis in hepatocellular carcinoma显示文摘Hepatocellular carcinoma(HCC) is a common malignant solid tumor characterized by rich vascularization. Pregnancy-specific glycoprotein 9(PSG9) is a member of the carcinoembryonic antigen(CEA)/PSG family and is produced at high levels during pregnancy. We previously identified PSG9 as an HCC-related protein. However, the expression of PSG9 and its regulation during HCC carcinogenesis remain poorly explored. In the present study, we first found that the levels of PSG9 protein were significantly increased in the plasma of HCC patients. PSG9 overexpression also increased the proliferation ability of an HCC cell line. High expression of PSG9 was associated with angiogenesis by accelerating VEGFA expression. In addition, Cox's proportional hazards model analysis revealed that the plasma level of PSG9 was an independent prognostic factor for overall survival. We propose that PSG9 is a novel indicator of prognosis in patients with HCC and could serve as a novel therapeutic target for HCC. 7Effect of fluoride on the corrosion behavior of nanostructured Ti-24Nb-4Zr-8Sn alloy in acidulated artificial saliva显示文摘The surface of titanium dental implants is highly susceptible to aggressive fluoride ions in the oral environment. Nanotechnology has proven an effective approach to improve the stability and corrosion resistance of titanium by applying a passive film. In this study, we investigated the effects of fluoride on the corrosion behavior of nanostructured(NS) Ti-24 Nb-4 Zr-8 Sn(Ti2448) alloy in acidulated artificial saliva(AAS)at 37 ℃, and then conducted comparisons with its coarse grained(CG) counterpart. Electrochemical techniques, such as potentiodynamic polarization and electrochemical impedance spectroscopy(EIS), as well as surface analysis including X-ray photoelectron spectroscopy(XPS) with argon ion sputtering, and scanning electronic microscopy(SEM) were employed to evaluate the effects of fluoride on sensitivity to pitting and the tolerance of Ti2448 to fluoride in AAS solution. The results demonstrate that corrosion current density increased with F-concentration. In all respects, the NS Ti2448 alloy presented corrosion resistance superior to that of its coarse grained(CG) counterpart at low F-concentrations(<0.1%).Furthermore, a high content of F-(1%) was shown to promote the active dissolution of both alloys by increasing the rate of corrosion. Following immersion in the fluoridated AAS solution for 60 days, a tissuefriendly compound, Ca_3(PO_4)_2, was detected on the surface of the NS when F-= 0.01% and Na_2 TiF_6 was identified as the main component in the corrosion products of the CG as well as NS Ti2448 alloys when F-= 1%. High concentrations of F-produced pitting corrosion on the CG alloy, whereas NS Ti2448 alloy presented general corrosion in the form of lamellar separation under the same conditions. 8CD4+T cells memorize obesity and promote weight regain显示文摘Body weight regain often causes failure of obesity therapies while the underlying mechanism remains largely unknown.In this study,we report that immune cells,especially CD4+T cells,mediate the‘memory’of previous obese status.In a weight gain-loss-regain model,we found that C57BL/6J mice with an obesity history showed a much faster rate of body weight regain.This obesity memory could last for at least 2 months after previously obese mice were kept at the same body weight as non-obese mice.Surprisingly,such obesity memory was abrogated by dexamethasone treatment,whereas immunodeficient Rag1−/−and H2A−/−mice failed to establish such memory.Rag1−/−mice repossessed the obesity memory when immune cells or CD4+T cells isolated from previously obese mice were transferred.Furthermore,depletion of CD4+T cells led to obesity memory ablation.Taken together,we conclude that CD4+T cells mediate obesity memory and promote weight regain.Jianghuan Zou Beibei Lai Mingzhu Zheng Qin Chen Shujun Jiang Anying Song Zan Huang Peiliang Shi Xin Tu Di Wang Linrong Lu Zhaoyu Lin Xiang Gao 2018Cellular & Molecular Immunology2018,15,6:2

14A joint call for actions to advance taxonomy in China显示文摘Taxonomy plays an important role in understanding the origin, evolution, and ecological functionality of biodiversity. There are large number of unknown species yet to be described by taxonomists, which together with their ecosystem services cannot be effectively protected prior to description. Despite this, taxonomy has been increasingly underrated insufficient funds and permanent positions to retain young talents. Further, the impact factordriven evaluation systems in China exacerbate this downward trend, so alternative evaluation metrics are urgently necessary. When the current generation of outstanding taxonomists retires,there will be too few remaining taxonomists left to train the next generation. 19Genome-wide developed microsatellite markers for the melon thrips Thrips palmi Karny(Thysanoptera:Thripidae)显示文摘The melon thrips, Thrips palmi Karny, is an economically important pest on many vegetables from Solanaceae and Cucurbitaceae. In this study, we developed novel microsatellite markers for Thrips palmi. First, we obtained randomly sequenced regions from the genome using next-generation sequencing and assembled 356 Mb genomic sequences. In total, 155,789 microsatellites were identified from the genomic sequences, of which, 64.02% were loci of dinucleotide repeat. Sixty primer pairs were initially validated in seven individuals. Thirty-five polymorphic markers were retained according to the amplification efficiency and polymorphism.These 35 microsatellite loci were then assessed in 96 individuals in four geographical populations collected from China. The allele numbers ranged from 2 to 19 with an average value of 6.9 per locus, while the polymorphism information content ranged from 0.2219 to 0.8490.The observed and expected heterozygosity varied from 0.5252 to 0.5367 and 0.5934 to 0.6148,respectively. Genetic structure was found between populations from southern and northern China.
