1De novo assembly of a Tibetan genome and identification of novel structural variants associated with high-altitude adaptation显示文摘Structural variants(SVs) may play important roles in human adaptation to extreme environments such as high altitude but have been under-investigated. Here, combining long-read sequencing with multiple scaffolding techniques, we assembled a high-quality Tibetan genome(ZF1), with a contig N50 length of 24.57 mega-base pairs(Mb) and a scaffold N50 length of 58.80 Mb. The ZF1 assembly filled 80 remaining N-gaps(0.25 Mb in total length) in the reference human genome(GRCh38). Markedly, we detected17 900 SVs, among which the ZF1-specific SVs are enriched in GTPase activity that is required for activation of the hypoxic pathway. Further population analysis uncovered a 163-bp intronic deletion in the MKL1 gene showing large divergence between highland Tibetans and lowland Han Chinese. This deletion is significantly associated with lower systolic pulmonary arterial pressure, one of the key adaptive physiological traits in Tibetans. Moreover, with the use of the high-quality de novo assembly, we observed a much higher rate of genome-wide archaic hominid(Altai Neanderthal and Denisovan) shared non-reference sequences in ZF1(1.32%–1.53%) compared to other East Asian genomes(0.70%–0.98%),reflecting a unique genomic composition of Tibetans. One such archaic hominid shared sequence-a662-bp intronic insertion in the SCUBE2 gene-is enriched and associated with better lung function(the FEV1/FVC ratio) in Tibetans. 2Effect of fluoride on the corrosion behavior of nanostructured Ti-24Nb-4Zr-8Sn alloy in acidulated artificial saliva显示文摘The surface of titanium dental implants is highly susceptible to aggressive fluoride ions in the oral environment. Nanotechnology has proven an effective approach to improve the stability and corrosion resistance of titanium by applying a passive film. In this study, we investigated the effects of fluoride on the corrosion behavior of nanostructured(NS) Ti-24 Nb-4 Zr-8 Sn(Ti2448) alloy in acidulated artificial saliva(AAS)at 37 ℃, and then conducted comparisons with its coarse grained(CG) counterpart. Electrochemical techniques, such as potentiodynamic polarization and electrochemical impedance spectroscopy(EIS), as well as surface analysis including X-ray photoelectron spectroscopy(XPS) with argon ion sputtering, and scanning electronic microscopy(SEM) were employed to evaluate the effects of fluoride on sensitivity to pitting and the tolerance of Ti2448 to fluoride in AAS solution. The results demonstrate that corrosion current density increased with F-concentration. In all respects, the NS Ti2448 alloy presented corrosion resistance superior to that of its coarse grained(CG) counterpart at low F-concentrations(<0.1%).Furthermore, a high content of F-(1%) was shown to promote the active dissolution of both alloys by increasing the rate of corrosion. Following immersion in the fluoridated AAS solution for 60 days, a tissuefriendly compound, Ca_3(PO_4)_2, was detected on the surface of the NS when F-= 0.01% and Na_2 TiF_6 was identified as the main component in the corrosion products of the CG as well as NS Ti2448 alloys when F-= 1%. High concentrations of F-produced pitting corrosion on the CG alloy, whereas NS Ti2448 alloy presented general corrosion in the form of lamellar separation under the same conditions. 3CsPb(I_(x)Br_(1-x))_(3) solar cells显示文摘Owing to its nice performance, low cost, and simple solution-processing, organic-inorganic hybrid perovskite solar cell(PSC) becomes a promising candidate for next-generation high-efficiency solar cells.The power conversion efficiency(PCE) has boosted from 3.8% to 25.2% over the past ten years. Despite the rapid progress in PCE, the device stability is a key issue that impedes the commercialization of PSCs. Recently, all-inorganic cesium lead halide perovskites have attracted much attention due to their better stability compared with their organic-inorganic counterpart. In this progress report, we summarize the properties of CsPb(IxBr1-x)3 and their applications in solar cells. The current challenges and corresponding solutions are discussed. 4Optimization of Technology for Increasing Total Ester Content of Jujube Wine by Adding Nutrients显示文摘Using jujube juice prepared from red jujube as raw material and Saccharomyces cerevisiae as experimental strain,the effect of nutrients on the content of total ester in jujube wine was studied. The nutrients affecting the total ester content of jujube wine were glutamine,arginine,calcium chloride and biotin by Plackett-Burman design. Through the steepest ascent test,it was determined that the central points of the optimum addition of nutrients were glutamine 40 mg/L,arginine 50 mg/L,calcium chloride 35 mg/L and biotin 0. 25 mg/L,respectively.Through Box-Behnken experiment design,taking glutamine,arginine,calcium chloride and biotin as experimental factors,and the content of total ester in jujube wine as response value,the response surface optimization experiment was carried out and the verification experiment was also carried out. It was found that the response surface model can be used to predict the total ester content of jujube wine when nutritional elements were added. When the content of total ester in jujube wine was the highest,the optimal combination of nutrients was glutamine38. 8 mg/L,arginine 53. 8 mg/L,calcium chloride 35. 7 mg/L and biotin 0. 3 mg/L. At this point,the content of total ester in jujube wine was 628 mg/L,which was very close to the predicted value. 7Effect of adenovirus-mediated gene transfer of Olig1 on oligodendrocyte differentiation and remyelination in a rat model of focal cerebral ischemia显示文摘BACKGROUND:The transcription factor Olig1 is required for oligodendrocyte maturation and demyelinated lesion repair,and is a key regulator of myelinogenesis following ischemia. OBJECTIVE:To examine the efficacy of intraventricular injection of a recombinant adenovirus-expressing Olig1 gene(Ad5-Olig1-eGFP) on oligodendrocyte maturation and myelin repair following focal cerebral ischemia. DESIGN,TIME AND SETTING:A randomized,controlled,animal experiment was performed at the Department of Neurology,Beijing Friendship Hospital Affiliated to Capital Medical University from January 2007 to March 2008. MATERIALS:Adenovirus and a recombinant adenovirus containing Olig1 gene(Ad5-Olig1) were provided by Vector Gene Technology,China. METHODS:All 50 rats were induced by middle cerebral artery occlusion.A total of 46 rats were successfully induced and were subsequently randomly assigned to a adenovirus(Ad5) group and a recombinant adenovirus-expression Olig1 gene(Ad5-Olig1) group,with 23 rats per group.One day after middle cerebral artery occlusion,either Ad5-Olig1-eGFP or Ad5-eGFP(10μL,2.3×10^(11)/mL) was injected into the lateral ventricle on the ischemic hemisphere. MAIN OUTCOME MEASURES:Adenovirus-mediated Olig1 gene expression in vitro and in vivo was measured by reverse transcription-polymerase chain reaction and immunofluorescence, respectively.Myelin basic protein(MBP) levels were evaluated by Western Blot,immunostaining, and electron microscopy. RESULTS:Exogenous Olig1 expression was measured at the periventricular zone of the lateral ventricle 1 day after Ad5-Olig1 injection.In the Ad5-Olig1-treated group,MBP protein levels and average intensity of MBP-immunoreactivity(-ir) increased 28 days after middle cerebral artery occlusion,compared with the control group(P<0.01,P<0.05).Furthermore,myelinated axonal numbers markedly increased following Ad5-Olig1 treatment. 9Short-term Effects of Nitrogen Deposition on Soil Physical and Chemical Properties of Tibetan Forest-Grassland Landscape Boundary显示文摘[Objective] The paper was to study the effects of nitrogen deposition on soil nutrients and soil dissolved organic carbon in forest-grass-land landscape in Linzhi, Tibet, and to provide scientific basis and basic data for understanding and assessing the effect of atmospheric nitrogen de-position on soil nutrients and soil dissolved organic carbon. [Method] From July 2014 to August 2015, in situ nitrogen deposition(CK 0 kg·hm^2/a, LN25 kg·hm^2/a, MN 50 kg·hm^2/a, HN 150 kg·hm^2/a) was simulated in the forest-grassland boundary of Zhuqudeng village, Bujiu Township, Linzhi County, Tibet. The soil samples were collected for analyzing nutrient and dissolved contents in the soil layer of 0-20 and 20-40 cm. The effects of different nitrogen deposition levels on soil nutrients and dissolved organic carbon(DOC) were studied. [Result] Nitrogen deposition had significant impacts on soil organic matter, total N, total P, total K, available N, available P, available K, exchangeable Ca, exchangeable Mg, p H, and DOC( P<0.05).(2) With the deepening of nitrogen deposition from CK, LN, MN to HN in the 0-20 cm boundary soil, the contents of organic matter, total N,total P, available P, exchangeable Ca, exchangeable Mg and DOC kept decreasing, and the content of total K and available N increased continuous-ly. The p H increased in LN treatment and decreased in HN treatment, while the available K content was decreased in LN and HN treatment, but increased in MN treatment.(3) The contents of organic matter, total N, total P, available N, available P, exchangeable Ca, exchangeable Mg and DOC all decreased at the soil layer of 20-40 cm under the same nitrogen deposition. The p H increased in LN treatment, but decreased in HN treat-ment; the content of total K decreased in LN treatment and increased in MN and HN treatments; the content of available K decreased in LN and HN treatments, but increased in MN treatment.(4) With the deepening of boundary soil layer(0-20 to 20-40 cm), the organic matter, total N, total P, available P, available K, exchangeable Ca, exchangeable Mg, DOC showed the same response to simulated nitrogen deposition, while the avail-able N and total K responded differently. 10Optimization of Fermentation Conditions of Lepista sordida Mycelium显示文摘[Objective] This study aimed to optimize the fermentation condition of Lepista sordida mycelium. [Method] The effects of carbon sources,nitrogen sources,pH and incubation time on the dry weight of mycelium in L. sordida fermentation were investigated. [Result] The descending order of the influences on L. sordida mycelium growth was: carbon sources>incubation time>pH>nitrogen sources. Based on orthogonal experiment, the optimal fermentation condition of L. sordida mycelium was determined as: fermentation medium of 3% corn powder and 0.2% yeast extract, pH of 6.0, and incubation time of 8 d. 12Structural covariance network of the hippocampus–amygdala complex in medication-nale patients with first-episode major depressive disorder显示文摘Background The hippocampus and amygdala are densely interconnected structures that work together in multiple affective and cognitive processes that are important to the etiology of major depressive disorder(MDD).Each of these structures consists of several heterogeneous subfields.We aim to explore the topologic properties of the volume-based intrinsic network within the hippocampus–amygdala complex in medication-nale patients with first-episode MDD.Methods High-resolution T1-weighted magnetic resonance imaging scans were acquired from 123 first-episode,medication-nale,and noncomorbid MDD patients and 81 age-,sex-,and education level-matched healthy control participants(HCs).The structural covariance network(SCN)was constructed for each group using the volumes of the hippocampal subfields and amygdala subregions;the weights of the edges were defined by the partial correlation coefficients between each pair of subfields/subregions,controlled for age,sex,education level,and intracranial volume.The global and nodal graphmetrics were calculated and compared between groups.Results Compared with HCs,the SCN within the hippocampus–amygdala complex in patients with MDD showed a shortened mean characteristic path length,reduced modularity,and reduced small-worldness index.At the nodal level,the left hippocampal tail showed increased measures of centrality,segregation,and integration,while nodes in the left amygdala showed decreased measures of centrality,segregation,and integration in patients with MDD compared with HCs.Conclusion Our results provide the first evidence of atypical topologic characteristics within the hippocampus–amygdala complex in patients with MDD using structure network analysis.It provides more delineate mechanism of those two structures that underlying neuropathologic process in MDD.Lianqing Zhang Xinyue Hu Yongbo Hu Mengyue Tang Hui Qiu Ziyu Zhu Yingxue Gao Hailong Li Weihong Kuang Weidong Ji 2022Psychoradiology2022,2,4:1
13Triple oxygen isotope constraints on the origin of ocean island basalts显示文摘Understanding the origin of ocean island basalts(OIB) has important bearings on Earth's deep mantle.Although it is widely accepted that subducted oceanic crust, as a consequence of plate tectonics, contributes material to OIB's formation, its exact fraction in OIB's mantle source remains ambiguous largely due to uncertainties associated with existing geochemical proxies. Here we show, through theoretical calculation, that unlike many known proxies, triple oxygen isotope compositions(i.e.D^(17 )O) in olivine samples are not affected by crystallization and partial melting. This unique feature, therefore, allows olivine D^(17 )O values to identify subducted oceanic crusts in OIB's mantle source. Furthermore, the fractions of subducted ocean sediments and hydrothermally altered oceanic crust in OIB's mantle source can be quantified using their characteristic D^(17 )O values. 19Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND 20An Empirical Research on the Relationship of Market Structure,Efficiency and Performance of Chinese Traditional Patent Medicine Industry显示文摘Objective To explore the relationship of market structure,efficiency and performance of Chinese traditional patent medicine(CTPM)industry,and to provide suggestions for the government and related enterprises to take the corresponding strategies.Methods Econometric model were empirically analyzed about 44 ongoing sample enterprises from 2007 to 2017,based on market power hypothesis and efficiency structure hypothesis of the industrial organization theory.Results and Conclusion The CTPM industry and the unlisted enterprises only conform to the relative market power hypothesis,and there is a significant positive correlation between performance and market share.The listed enterprises do not conform to the market power hypothesis and efficiency structure hypothesis:There is no significant correlation between performance and efficiency,market structure.There is no significant correlation between market share and efficiencies,while the industrial concentration has a significant correlation with efficiencies.The CTPM industry is relatively immature,so it’s necessary for the government to establish the elimination mechanism of CTPM industry in line with market rules,to enhance the industrial concentration and resource utilization efficiency by encouraging enterprises to improve management and scale production level.The government should improve the innovation policy of CTPM industry to encourage the enterprises to pursue product innovation.The CTPM enterprises,especially the unlisted enterprises should increase the market shares and enterprise performance through production differentiation.Gao Yongbo Cao Linlin Xiao Yao Sun Lihua 2019Asian Journal of Social Pharmacy2019,14,4:0
