| 1 | A Resource for Inactivation of MicroRNAs Using Short Tandem Target Mimic Technology in Model and Crop Plants显示文摘microRNAs (miRNAs)are endogenous small non-coding RNAs that bind to mRNAs and target them for cleavage and/or translational repression,leading to gene silencing.We previously developed short tandem target mimic (STTM)technology to deactivate endogenous miRNAs in Arabidopsis.Here,we created hundreds of STTMs that target both conserved and species-specific miRNAs in Arabidopsis,tomato,rice,and maize,providing a resource for the functional interrogation of miRNAs.We not only revealed the functions of several miRNAs in plant development,but also demonstrated that tissue-specific inactivation of a few miRNAs in rice leads to an increase in grain size without adversely affecting overall plant growth and development.RNA-seq and small RNAseq analyses of STTM156/157 and STTM165/166 transgenic plants revealed the roles of these miRNAs in plant hormone biosynthesis and activation,secondary metabolism,and ion-channel activity-associated electrophysiology,demonstrating that STTM technology is an effective approach for studying miRNA functions.To facilitate the study and application of STTM transgenic plants and to provide a useful platform for storing and sharing of information about miRNA-regulated gene networks,we have established an online Genome Browser (http://gffzzc750bc0108fa4044swxb66oc955ov6fun.ffgz.tsg.suse.edu.cn/designindex2.php) to display the transcriptomic and miRNAomic changes in STTMinduced miRNA knockdown plants. | Ting Peng Mengmeng Qiao Haiping Liu Sachin Teotia Zhanhui Zhang Yafan Zhao Bobo Wang Dongjie Zhao Lina Shi Cui Zhang Brandon Le Kestrel Rogers Chathura Gunasekara Haitang Duan Yiyou Gu Lei Tian Jinfu Nie Jian Qi Fanrong Meng Lan Huang Qinghui Chen Zhenlin Wang Jinshan Tang Xiaoqing Tang Ting Lan Xuemei Chen Hairong Wei Quanzhi Zhao Guiliang Tang | 2018 | Molecular Plant2018,11,11: | 7 |
| 2 | A novel PIK3CD C896T mutation detected in bilateral sudden sensorineural hearing loss using next generation sequencing:An indication of primary immunodeficiency显示文摘Objective: To investigate immune-related genetic background in bilateral sudden sensorineural hearing loss(SSNHL).Case report and methods: The case is a 45-year-old man presenting with a 7-year history of bilateral profound SSNHL. Blood biochemical testing demonstrated increased levels of total cholesterol(5.88 mmol/L). Tests for hepatitis B showed a positive antibody against the hepatitis B core antigen. Complement C3 was below the normal value, and complement C4 and Ig G were in the lower range of normal values. CT images showed a normal inner ear and vestibular aqueduct but round window membranous ossification on both sides. A total number of 232 immuneassociated genes were sequenced using the next generation sequencing technique.Results: Mutations were detected in 5 genes, including the phosphoinositide 3-kinase catalytic subunit delta(PIK3CD), caspase recruitment domain-containing protein 9(CARD9), complement factor H-related(CFHR2), immunoglobulin lambda-like polypeptide 1 Protein(IGLL1),and transmembrane channel-like gene family 8(TMC8). In the PIK3 CD gene, a C896 T substitute in exon 7 was detected. | 5 | Study of pyrolyzed hemin/C as non-platinum cathodic catalyst for direct methanol fuel cells显示文摘Biological reduction of O2 to H2O justifies a serious look at heme as a potential O2 reduction reaction(ORR) catalyst for low temperature fuel cells.In this study,a novel non-platinum electrocatalyst for ORR was prepared through hemin,which is hydrochloride of heme,supported on Black Pearls 2000 carbon black(Hm-BP) pyrolyzed at 700-900℃ in Ar atmosphere.The physical and electrocatalytic properties of as-prepared catalysts were characterized by TGA,XRD,XPS,TEM,rotating disk electrode(RDE) and rotating ring disk electrode(RRDE).It has found that the catalyst treated at 750℃(Hm-BP-750) exhibits the best property among the Hm-BP catalysts prepared.The onset potential of ORR on the Hm-BP-750 at 30℃ was measured ca.0.90 V(vs.RHE) in 0.1 M H2SO4,and mass current density was reached 15.3 mA mg-1 at 0.75 V.It has revealed that O2 could be reduced directly to water in a 4e process between 0.9 and 0.83V,and the yield of H2O2 was 0-18% in the potential range of 0.83-0.63 V.This methanol-tolerant catalyst also presents excellent stability in medium-term test of direct methanol fuel cell at 80℃. | Qiang Wang ZhiYou Zhou DeJun Chen JianLong Lin FuSheng Ke GuiLiang Xu ShiGang Sun | 2010 | Science China Chemistry2010,53,9: | 0 |

| 7 | Long-term outcomes and prognosis factors of vagus nerve stimulation in patients with refractory epilepsy显示文摘Background:Vagus nerve stimulation(VNS)is an effective treatment for patients with refractory epilepsy,yet with varied predictive factors and heterogeneous long-term outcomes.Adjustment of VNS parameters is critical for obtaining favorable efficacy.In this study,we aimed to investigate the long-term outcomes and the possible predictive factors of VNS in patients with refractory epilepsy.Methods:Eighty-six patients(59 males and 27 females)who underwent VNS implantation for treatment of refractory epilepsy between May 2016 and May 2017 at five Epilepsy Centers were enrolled.The clinical data,including sex,age at epilepsy onset,VNS implantation,epilepsy duration,seizure type,MRI findings,history of neurosurgical operations,and responder rate(responders were those with≥50%seizure reduction),were analyzed.Results:Four-year follow-up data were available for 76 patients(53 males and 23 females).The mean current intensity at the last follow-up was 1.8±0.3 mA(range:0.75–2.5 mA).The mean seizure reduction was 36.2%at 6 months,38.5%at 1 year,69.4%at 3 years,and 56.7%at 4 years.A favorable outcome of≥50%reduction in seizure frequency occurred in 40.0%of the patients at 6 months,55.9%at 1 year with 4 patients being seizure-free,63.2%at 3 years with 5 patients being seizure-free,and 68.4%at 4 years with 5 patients being seizure-free.Earlier onset age(P<0.001)and shorter duration(P=0.042)were associated with favorable prognosis.Compared with generalized tonicclonic seizures,tonic seizures had a favorable outcome(P=0.026).Twenty-three patients underwent neurosurgical operations before VNS implantation,and the responder rate was 60.9%at the last follow-up.Conclusions:VNS is an adjunctive and effective treatment for patients with refractory epilepsy who are not good candidates for surgical resection or have failed to respond to surgical treatment.The stimulation efficacy increases over time after implantation,and earlier exposure to VNS improves the prognosis. | Cuiping Xu Hua Lin Jiwen Xu Xiaohua Zhang Guiliang Hao Qiang Qiang Liu Chengyun Ding Shan Wang Quanjun Zhao Xiaojun Bai Kui Chen Duanyu Ni Yongjie Li Tao Yu Yuping Wang | 2022 | Acta Epileptologica2022,5,3: | 0 |
| 8 | Molecular Characterization and Expression Analysis of SKIV Infection of Interferon-Induced Protein with Tetratricopeptide Repeats 1(IFIT1) in Epinephelus lanceolatus显示文摘Interferon-induced protein with tetratricopeptide repeats 1(IFIT1), also known as interferon-induced protein 56(IFI56) or Interferon-stimulated protein 56(ISG56), was originally identified as a protein induced upon treatment with interferon and inhibited by viral replication and translational initiation. In this study, Epinephelus lanceolatus IFIT1(ELIFIT1) gene was cloned for the first time. The complete cDNA of El IFIT1 gene includes 2921 nucleotides, and encodes a 437-amino acid(AA) protein. The putative ELIFIT1 protein has 9 TRP domains and is highly similar with IFIT1 proteins in other teleosts. In healthy fish, ELIFIT1 gene was highly expressed in the blood, which indicate its specific function in the peripheral immune system. Its expression was also observed in various immunity-related tissues including spleen, intestine, and kidney, Inducted with spotted knifejaw iridovirus(SKIV), ELIFIT1 gene expression was upregulated in the spleen, kidney, and liver 24 h after induction and reached its peak at 72 h, indicating that ELIFIT1 may play an important role in antivirus.
