| 2 | Characteristics of distribution of Mycobacterium tuberculosis lineages in China显示文摘The genotyping methods of Mycobacterium tuberculosis would dramatically improve our understanding of the molecular epidemiology of tuberculosis. 3,929 isolates, from a National Survey of Drug-Resistant Tuberculosis in 2007 in China, were successfully genotyped by large sequence polymorphisms and 15 loci variable number tandem repeats. We found that 2,905(2,905/3,929, 73.9%) cases belonged to Lineage 2, dominated in the east and central regions, 975 cases(975/3,929, 24.8%) were Lineage 4, highly prevailed in the west regions, and 36 and 13 cases were Lineage 3 and Lineage 1, respectively. We also explored the associations between lineages(Lineage 2 vs. Lineage 4) and clinical characteristics by logistic regression. For Lineage 2, the risk factors were Han-ethnicity population and fever. However, for Lineage 4, they were occupation(farmer), and degree of education(non-literate). In this process, egg white was added as protein feeder for facilitating the formation of CLEAs. The optimal conditions of the immobilization process were determined. Michaelis constants(Km) of free NHase and NHase CLEAs were also determined. The NHase CLEAs exhibited increased stability at varied pH and temperature conditions compared to its free counterpart. | 5 | Dominance rank of adult females and mating competition in Sichuan snub-nosed monkeys(Rhinopithecus roxellana)in the Qinling Mountains,China显示文摘Dominance hierarchy reflects resource competition in male primates as well as females.In this study,we collected data on the social rank and mating competition of female Sichuan snub-nosed monkeys from September 2009 to June 2010 in the Qinling Mountains,Shaanxi,China.Displacement was used to determine social hierarchy of the studied wild females.A total of 1223 displacement bouts between adult females and 316 mating behaviors within one-male units(OMUs) were recorded.Competitive displacement among females was very low(0.22 displacements per observation hour).The dominant ranks of females in target OMUs were determined by displacement,from one OMU to another:JB unit:YL > DBC > YCM >XBC > BD;JZT unit:XHW > JG > DT;PK unit:QQ > TM > HF;LP unit:SK > TH > WM.Higher-ranking females performed significantly more sexual behavior than lower-ranking females in this rare monkey.High-ranking females were more likely to win mating contests within those OMUs.Based on these studies,we assessed the social relationships among adult females and clarified possible explanations for the strategies of female mating competition in the OMUs.Furthermore,the notion that reproductive success may also be related to female dominance hierarchies in this polygynous species should be considered in future research. | HE HaiXia ZHAO HaiTao QI XiaoGuang WANG XiaoWei GUO SongTao JI WeiHong WANG ChengLiang WEI Wei LI BaoGuo | 2013 | Chinese Science Bulletin2013,58,17: | 5 |

| 10 | Overexpression of amplified in breast cancer 1 (AIB1) gene promotes lung adenocarcinoma aggressiveness in vitro and in vivo by upregulating C-X-C motif chemokine receptor 4显示文摘Background:We previously found that overexpression of the gene known as amplified in breast cancer 1(AIB1)was associated with lymph node metastasis and poor prognosis in patients with lung adenocarcinoma.However,the role of AIB1 in that malignancy remains unknown.The present study aimed to investigate the function of AIB1 in the process of lung adenocarcinoma cell metastasis.Methods:A series of in vivo and in vitro assays were performed to elucidate the function of AIB1,while real-time PCR and Western blotting were utilized to identify the potential downstream targets of AIB1 in the process of lung adenocarcinoma metastasis.Rescue experiments and in vitro assays were performed to investigate whether the invasive-ness of AIB1-induced lung adenocarcinoma was mediated by C-X-C motif chemokine receptor 4(CXCR4).Results:The ectopic overexpression of AIB1 in lung adenocarcinoma cells substantially enhanced cell migration and invasive abilities in vitro and tumor metastasis in vivo,whereas the depletion of AIB1 expression substantially inhibited lung adenocarcinoma cell migration and invasion.CXCR4 was identified as a potential downstream target of AIB1 in lung adenocarcinoma.The knockdown of AIB1 greatly reduced CXCR4 gene expression at both the transcription and protein levels,whereas the knockdown of CXCR4 in cells with AIB1 ectopic overexpression diminished AIB1-induced migration and invasion in vitro and tumor metastasis in vivo.Furthermore,we found a significant positive association between the expression of AIB1 and CXCR4 in lung adenocarcinoma patients(183 cases),and the co-overexpression of AIB1 and CXCR4 predicted the poorest prognosis.Conclusions:These findings suggest that AIB1 promotes the aggressiveness of lung adenocarcinoma in vitro and in vivo by upregulating CXCR4 and that it might be usable as a novel prognostic marker and/or therapeutic target for this disease. | Liru He Haixia Deng Shiliang Liu Jiewei Chen Binkui Li Chenyuan Wang Xin Wang Yiguo Jiang Ningfang Ma Mengzhong Liu Dan Xie | 2018 | Cancer Communications2018,38,1: | 2 |

| 17 | Quasi-4-dimension ionospheric modeling and its application in PPP显示文摘Ionospheric delay modeling is not only important for Global Navigation Satellite System(GNSS)based space weather study and monitoring,but also an efficient tool to speed up the convergence time of Precise Point Positioning(PPP).In this study,a novel model,denoted as Quasi-4-Dimension Ionospheric Modeling(Q4DIM)is proposed for wide-area high precision ionospheric delay correction.In Q4DIM,the Line Of Sight(LOS)ionospheric delays from a GNSS station network are divided into different clusters according to not only the location of latitude and longitude,but also satellite elevation and azimuth.Both Global Ionosphere Map(GIM)and Slant Ionospheric Delay(SID)models that are traditionally used for wide-area and regional ionospheric delay modeling,respectively,can be regarded as the special cases of Q4DIM by defining proper grids in latitude,longitude,elevation,and azimuth.Thus,Q4DIM presents a resilient model that is capable for both wide-area coverage and high precision.Four different sets of clusters are defined to illustrate the properties of Q4DIM based on 200 EUREF Permanent Network(EPN)stations.The results indicate that Q4DIM is compatible with the GIM products.Moreover,it is proved that by inducting the elevation and azimuth angle dependent residuals,the precision of the 2-dimensional GIM-like model,i.e.,Q4DIM 2-Dimensional(Q4DIM-2D),is improved from around 1.5 Total Electron Content Units(TECU)to better than 0.5 TECU.In addition,treating Q4DIM as a 4-dimensional matrix in latitude,longitude,elevation,and azimuth,whose sparsity is less than 5%,can result in its feasibility in a bandwidth-sensitive applications,e.g.,satellite-based Precising Point Positioning Real-Time Kinematic(PPP-RTK)service.Finally,the advantages of Q4DIM in PPP over the 2-dimensional models are demonstrated with the one month's data from 30 EPN stations in both high solar activity year 2014 and low solar activity year 2020. | Shengfeng Gu Chengkun Gan Chengpeng He Haixia Lyu Manuel Hernandez-Pajares Yidong Lou Jianghui Geng Qile Zhao | 2022 | Satellite Navigation2022,3,3: | 1 |

In recent years,C. nitidissima planting area has been increasing,resulting in the growing gap between the supply and demand of C. nitidissima seedlings. At present,C. nitidissima can be propagated through seeding,cutting,grafting,plant tissue culture and air layering. Among them,cutting and tissue culture are the most commonly used methods for rapid propagation of C. nitidissima. Optimal rooting medium,type and concentration of hormones and environmental conditions are critical factors for C. nitidissima propagation by cuttings,while detoxification and optimization of medium composition are important for C. nitidissima propagation by tissue culture. | 20 | Variant rs8400 enhances ALKBH5 expression through disrupting miR-186 binding and promotes neuroblastoma progression显示文摘Objective:AlkB homolog 5(ALKBH5)has been proven to be closely related to tumors.However,the role and molecular mechanism of ALKBH5 in neuroblastomas have rarely been reported.Methods:The potential functional single-nucleotide polymorphisms(SNPs)in ALKBH5 were identified by National Center for Biotechnology Information(NCBI)dbSNP screening and SNPinfo software.TaqMan probes were used for genotyping.A multiple logistic regression model was used to evaluate the effects of different SNP loci on the risk of neuroblastoma.The expression of ALKBH5 in neuroblastoma was evaluated by Western blotting and immunohistochemistry(IHC).Cell counting kit-8(CCK-8),plate colony formation and 5-ethynyl-2'-deoxyuridine(EdU)incorporation assays were used to evaluate cell proliferation.Wound healing and Transwell assays were used to compare cell migration and invasion.Thermodynamic modelling was performed to predict the ability of miRNAs to bind to ALKBH5 with the rs8400 G/A polymorphism.RNA sequencing,N6-methyladenosine(mA)sequencing,mA methylated RNA immunoprecipitation(MeRIP)and a luciferase assay were used to identify the targeting effect of ALKBH5 on SPP1.Results:ALKBH5 was highly expressed in neuroblastoma.Knocking down ALKBH5 inhibited the proliferation,migration and invasion of cancer cells.miR-186-3p negatively regulates the expression of ALKBH5,and this ability is affected by the rs8400 polymorphism.When the G nucleotide was mutated to A,the ability of miR-186-3p to bind to the 3'-UTR of ALKBH5 decreased,resulting in upregulation of ALKBH5.SPPI is the downstream target gene of the ALKBH5 oncogene.Knocking down SPP1 partially restored the inhibitory effect of ALKBH5 downregulation on neuroblastoma.Downregulation of ALKBH5 can improve the therapeutic efficacy of carboplatin and etoposide in neuroblastoma.Conclusions:We first found that the rs8400 G>A polymorphism in the m6A demethylase-encoding gene ALKBH5 increases neuroblastoma susceptibility and determines the related mechanisms.The aberrant regulation of ALKBH5 by miR-186-3p caused by this genetic variation in ALKBH5 promotes the occurrence and development of neuroblastoma through the ALKBH5-SPP1 axis. | Qian Guan Huiran Lin Wenfeng Hua Lei Lin Jiabin Liu Linqing Deng Jiao Zhang Jiwen Cheng Zhonghua Yang Yong Li Jun Bian Haixia Zhou Suhong Li Li Li Lei Miao Huimin Xia Jing He Zhenjian Zhuo | 2023 | Chinese Journal of Cancer Research2023,35,2: | 0 |
