| 2 | cDNA cloning, expression and antibacterial activity of lysozyme C in the blue shrimp (Litopenaeus stylirostris)显示文摘The gene coding for lysozyme in blue shrimp (Litopenaeus stylirostris) was cloned, sequenced and expressed in pET-32a vector. The deduced amino acid sequence of F. merguiensi lysozyme showed 37–93% similarity with the mouse, human, chicken, and tiger prawn counterparts. The lysozyme was purified to homogeneity and observed as a band of approximately 15 kDa in 15% SDS–PAGE. Semiquantitative RT-PCR analysis demonstrated that mRNA transcripts of lysozyme could be mainly detected in the tissues of haemocytes, gill, gonad and the lymphoid organ of unchallenged shrimps, whereas the expression of lysozyme transcripts was increased in all the tested tissues after the heat-killed Vibrio alginolyticus challenge. The temporal expression of lysozyme mRNA in haemolymph challenged by Micrococcus luteus and V. alginolyticus was both up-regulated and reached the maximum level at 8 and 16 h post-stimulation, respectively, and then dropped back to the original level. Bacteriolytic activity of the lysozyme against different bacterial cultures was determined by the solid phase and turbidimetric assays. The results demonstrated that the lysozyme we obtained was not only against Grampositive and Gram-negative bacteria but also against shrimp pathogens V. alginolyticus and V. parahemolyticus. | 3 | Non-equilibrium Plasma Assisted Combustion of Low Heating Value Fuels显示文摘This paper describes the effects of non-equilibrium air plasma generated by a dielectric barrier discharge (DBD) on the combustion of low heating value fuels. The experimental results indicate that addition of a very small amount of energy to the air flow in the form of DBD significantly improves the flame stability. Moreover, main combustion characteristics such as flame propagation speed, combustion intensity and lean blow-off limits are also enhanced by the effect of plasma. Some active radicals such as excited O atom and excited N2 molecule are observed by spectrograph in the discharge area. | 4 | Post-Orogenic Tectonic Evolution of the Jiangnan-Xuefeng Orogenic Belt:Insights from Multiple Geochronometric Dating of the Mufushan Massif,South China显示文摘The Mufushan massif, as continental intra-plate magmatites located in the Jiangnan-Xuefeng orogenic belt of the South China. The Mufushan massif constitutes the largest Mesozoic intrusive complex, intruded the Mesoproterozoic Lengjiaxi Formation. Multiple geochronometric dating was used to reconstruct their evolution from emplacement to exhumation. The Mufushan granitoids were emplaced at ~150 Ma(U-Pb zircon) as post-orogenic magmatites contributing to Triassic crustal thickening. Onset of regional extension at ~128 Ma(40Ar/39Ar white mica and biotite) manifests a tectonic regime switch. Intense exhumation prior to ~55 Ma was followed by slow denudation and peneplanation for the next 37 Ma(~55–18 Ma). Accelerated cooling since ~18 Ma may have been caused by a far-field effect of the collision between IndiaAsia Plate or the Pacific-Plate subduction. | 5 | Characterization of mitochondrial genome of sea cucumber Stichopus horrens:A novel gene arrangement in Holothuroidea显示文摘The complete mitochondrial DNA sequence contains useful information for phylogenetic analyses of metazoa.In this study,the complete mitochondrial DNA sequence of sea cucumber Stichopus horrens (Holothuroidea:Stichopodidae:Stichopus) is presented.The complete sequence was determined using normal and long PCRs.The mitochondrial genome of Stichopus horrens is a circular molecule 16257 bps long,composed of 13 protein-coding genes,two ribosomal RNA genes and 22 transfer RNA genes.Most of these genes are coded on the heavy strand except for one protein-coding gene (nad6) and five tRNA genes (tRNA Ser(UCN),tRNA Gln,tRNA Ala,tRNA Val,tRNA Asp) which are coded on the light strand.The composition of the heavy strand is 30.8% A,23.7% C,16.2% G,and 29.3% T bases (AT skew=0.025;GC skew= 0.188).A non-coding region of 675 bp was identified as a putative control region because of its location and AT richness.The intergenic spacers range from 1 to 50 bp in size,totaling 227 bp.A total of 25 overlapping nucleotides,ranging from 1 to 10 bp in size,exist among 11 genes.All 13 protein-coding genes are initiated with an ATG.The TAA codon is used as the stop codon in all the protein coding genes except nad3 and nad4 that use TAG as their termination codon.The most frequently used amino acids are Leu (16.29%),Ser (10.34%) and Phe (8.37%).All of the tRNA genes have the potential to fold into typical cloverleaf secondary structures.We also compared the order of the genes in the mitochondrial DNA from the five holothurians that are now available and found a novel gene arrangement in the mitochondrial DNA of Stichopus horrens. | FAN SiGang HU ChaoQun WEN Jing ZHANG LvPing | 2011 | Science China(Life Sciences)2011,54,5: | 4 |

| 8 | The Karyotypes, C-banding Patterns and AgNORs of Epinephelus malabaricus显示文摘The chromosome specimens of Epinephelus malabaricus (Bloch & Schneider, 1801) are obtained from metaphase of kindney cell by vivi-injection of PHA and culture of colchicines, hypatoic-air drying technique, and then by studying their Giemsa stain, C-bands and AgNORs. The results are as follows: (1)E. malabaricus has a diploid chromosome number of 48 and its karyotype formula is 48t, NF=48, sex chromosome is not found. (2) There is a pair of chromosomes with secondary constriction near the centromere of chromosome t24. (3) 1~4 nucleoli appear in the nucleus of interphase, 55% nuclei has 1 nucleolus and only 2% for 4 nucleoli. (4) AgNORs appear in the chromosome t24 of 50% metaphase, sometimes in the chromosome t5, but not in other chromosomes. (5) The AgNORs polymorphisms are individually specific, 1~4 pairs of the number, and the frequency of 4 AgNORs are lowest. (6) The secondary constrictions and positive C-bands are coincident, close to the centromere of the chromosome, and mass constrictive heterochromatins appear in that region. (7) All the centromeres of chromosomes are darkly stained C-bands, and the whole arm of chromosome t24 and its centromere are same positive C-bands. | 9 | Effect of Shrimp(Litopenaeus vannamei) Farming Waste on the Growth, Digestion, Ammonium-Nitrogen Excretion of Sea Cucumber(Stichopus monotuberculatus)显示文摘In this study, specific growth rate(SGR), ingestion rate(IR), food conversion ratio(FCR), apparent digestion ratio(ADR) and ammonium-nitrogen excretion were determined for sea cucumber(Stichopus monotuberculatus) reared in plastic containers(70 L; 4 containers each diet treatment). Sea cucumbers were fed with five diets containing different amounts of farming waste from shrimp(Litopenaeus vannamei)(100%, 75%, 50%, 25% and 0) and a formulated compound(20% sea mud and 80% powdered algae). Sea cucumbers grew faster when they were fed with diet D(25% shrimp waste and 75% formulated compound) than those fed with other diets. Although IR value of sea cucumber fed with diet A(shrimp waste) was higher than those fed with other diets, both the lowest SGR and the highest FCR occurred in this diet group. The highest and the lowest ADR occurred in diet E(formulated compound) and diet A group, respectively, and the same to ammonium-nitrogen excretion. The contents of crude protein, crude lipid and total organic matter(TOM) in feces decreased in comparison with corresponding diets.
