| 1 | Ciliary parathyroid hormone signaling activates transforming growth factor-βto maintain intervertebral disc homeostasis during aging显示文摘Degenerative disc disease(DDD) is associated with intervertebral disc degeneration of spinal instability. Here, we report that the cilia of nucleus pulposus(NP) cells mediate mechanotransduction to maintain anabolic activity in the discs. We found that mechanical stress promotes transport of parathyroid hormone 1 receptor(PTH1 R) to the cilia and enhances parathyroid hormone(PTH) signaling in NP cells. PTH induces transcription of integrin α_vβ_6 to activate the transforming growth factor(TGF)-β-connective tissue growth factor(CCN2)-matrix proteins signaling cascade. Intermittent injection of PTH(iPTH) effectively attenuates disc degeneration of aged mice by direct signaling through NP cells, specifically improving intervertebral disc height and volume by increasing levels of TGF-β activity, CCN2, and aggrecan. PTH1 R is expressed in both mouse and human NP cells. Importantly,knockout PTH1 R or cilia in the NP cells results in significant disc degeneration and blunts the effect of PTH on attenuation of aged discs. | 3 | Optimization Potentials for the Waste Heat Recovery of a Gas-Steam Combined Cycle Power Plant Based on Absorption Heat Pump显示文摘A new waste heat recovery system is presented to recover exhausted steam waste heat from the steam turbine by absorption heat pump(AHP) in a gas-steam combined cycle(GSCC) power plant. The system can decrease energy consumption and further improve the energy utilization. The performance evaluation criteria are calculated, and exergy analysis for key components are implemented in terms of the energy and exergy analysis theory. Besides, the change of these criteria is also revealed before and after modification. The net power output approximately increases by 21738 kW, and equivalent coal consumption decreases by 5.58 g/kWh. A 1.81% and 1.92% increase in the thermal and exergy efficiency is respectively obtained in the new integrated system as the heating load is 401095 kJ at 100% condition. Meanwhile, the appropriate extraction parameters for heating have been also analyzed in the two systems. | 7 | Non-equilibrium Plasma Assisted Combustion of Low Heating Value Fuels显示文摘This paper describes the effects of non-equilibrium air plasma generated by a dielectric barrier discharge (DBD) on the combustion of low heating value fuels. The experimental results indicate that addition of a very small amount of energy to the air flow in the form of DBD significantly improves the flame stability. Moreover, main combustion characteristics such as flame propagation speed, combustion intensity and lean blow-off limits are also enhanced by the effect of plasma. Some active radicals such as excited O atom and excited N2 molecule are observed by spectrograph in the discharge area. | 8 | Engineering an enthesis-like graft for rotator cuff repair:An approach to fabricate highly biomimetic scaffold capable of zone-specifically releasing stem cell differentiation inducers显示文摘Rotator cuff(RC)attaches to humerus across a triphasic yet continuous tissue zones(bone-fibrocartilage-tendon),termed“enthesis”.Regrettably,rapid and functional enthesis regeneration is challenging after RC tear.The existing grafts bioengineered for RC repair are insufficient,as they were engineered by a scaffold that did not mimic normal enthesis in morphology,composition,and tensile property,meanwhile cannot simultaneously stimulate the formation of bone-fibrocartilage-tendon tissues.Herein,an optimized decellularization approach based on a vacuum aspiration device(VAD)was developed to fabricate a book-shaped decellularized enthesis matrix(O-BDEM).Then,three recombinant growth factors(CBP-GFs)capable of binding collagen were synthesized by fusing a collagen-binding peptide(CBP)into the N-terminal of BMP-2,TGF-β3,or GDF-7,and zone-specifically tethered to the collagen of O-BDEM to fabricate a novel scaffold(CBP-GFs/O-BDEM)satisfying the above-mentioned requirements.After ensuring the low immunogenicity of CBP-GFs/O-BDEM by a novel single-cell mass cytometry in a mouse model,we interleaved urine-derived stem cell-sheets into this CBP-GFs/O-BDEM to bioengineer an enthesis-like graft.Its high-performance on regenerating enthesis was determined in a canine model.These findings indicate this CBP-GFs/O-BDEM may be an excellent scaffold for constructing enthesis-like graft to patch large/massive RC tears,and provide breakthroughs in fabricating graded interfacial tissue. | Can Chen Qiang Shi Muzhi Li Yang Chen Tao Zhang Yan Xu Yunjie Liao Shulin Ding Zhanwen Wang Xing Li Chunfeng Zhao Lunquan Sun Jianzhong Hu Hongbin Lu | 2022 | Bioactive Materials2022,7,10: | 4 |
| 9 | PhaseⅡstudy of induction chemotherapy followed by concurrent chemoradiotherapy with raltitrexed and cisplatin in locally advanced nasopharyngeal carcinoma显示文摘Objective:For locally advanced nasopharyngeal carcinoma(LA-NPC)patients,high incidences of distant metastases and severe treatment related toxicities are the main obstacles needed to be overcome.Raltitrexed,a specific thymidylate synthase inhibitor with a convenient administration schedule,has an acceptable and manageable toxicity,and possesses radio-sensitizing properties.To investigate the efficacy and safety of raltitrexed and cisplatin induction chemotherapy and concurrent chemoradiotherapy(IC+CCRT)in patients with LA-NPC,a phaseⅡclinical study was conducted.Methods:Sixty eligible patients with LA-NPC were enrolled into this study.A raltitrexed-cisplatin combination was used as part of an IC+CCRT regimen.Raltitrexed-cisplatin IC was given once every 3 weeks(q3 w)for two cycles,followed by raltitrexed-cisplatin based CCRT q3 w for two cycles.Intensity-modulated radiotherapy(IMRT)was given for all enrolled patients.Results:All patients were included in survival analysis according to the intent-to-treat principle.The objective response rate(ORR)3 months after treatment was 98%.The 2-year overall survival(OS)rate was 92%.The median relapse-free survival(RFS)time was 30.5[95%confidence interval(95%CI),28.4-32.3]months.The 2-year RFS rate was 85%.The 2-year local failure-free survival(LFFS)rate was 97%and the 2-year distant metastasis-free survival(DMFS)rate was 88%.Acute toxicities were mostly grade 2 and 3 reactions in bone marrow suppression,gastrointestinal side effect and oropharyngeal mucositis.Only two patients occurred grade 4 acute toxicities,one was bone marrow suppression and the other was dermatitis radiation.Conclusions:The combination of raltitrexed and cisplatin has a comparable efficacy to those in standard firstline therapy. | Yuan Wu Xueyan Wei Zilong Yuan Hongbin Xu Yanping Li Ying Li Liu Hu Guang Han Yu Qian Desheng Hu | 2020 | Chinese Journal of Cancer Research2020,32,5: | 4 |

| 11 | Very Short-term Spatial and Temporal Wind Power Forecasting: A Deep Learning Approach显示文摘In power systems that experience high penetration of wind power generation,very short-term wind power forecast is an important prerequisite for look-ahead power dispatch.Conventional univariate wind power forecasting methods at presentonly utilize individual wind farm historical data.However,studies have shown that forecasting accuracy canbe improved by exploring both spatial and temporal correlations among adjacent wind farms.Current research on spatial-temporal wind power forecasting is based on relatively shallow time series models that,to date,have demonstrated unsatisfactory performance.In this paper,a convolution operation is used to capture the spatial and temporal correlations among multiple wind farms.A novel convolution-based spatial-temporal wind power predictor(CSTWPP)is developed.Due to CSTWPP’s high nonlinearity and deep architecture,wind power variation features and regularities included in the historical data can be more effectively extracted.Furthermore,the online training of CSTWPP enables incremental learning,which makes CSTWPP non-stationary and in conformity with real scenarios.Graphics processing units(GPU)is used to speed up the training process,validating the developed CSTWPP for real-time application.Case studies on 28 adjacent wind farms are conducted to show that the proposed model can achieve superior performance on 5-30 minutes ahead wind power forecasts. | Tianyu Hu Wenchuan Wu Qinglai Guo Hongbin Sun Libao Shi Xinwei Shen | 2020 | CSEE Journal of Power and Energy Systems2020,6,2: | 3 |
| 12 | Genome-wide mapping of conserved microRNAs and their host transcripts in Tribolium castaneum显示文摘MicroRNAs (miRNAs) are endogenous 22-nt RNAs,which play important regulatory roles by post-transcriptional gene silencing. A computational strategy has been developed for the identification of conserved miRNAs based on features of known metazoan miRNAs in red flour beetle (Tribolium castaneum),which is regarded as one of the major laboratory models of arthropods. Among 118 putative miRNAs,47% and 53% of the predicted miRNAs from the red flour beetle are harbored by known protein-coding genes (intronic) and genes located outside (intergenic miRNA),respectively. There are 31 intronic miRNAs in the same transcriptional orientation as the host genes,which may share RNA polymerase Ⅱ and spliceosomal machinery with their host genes for their biogenesis. | 13 | IAP's Solar-Powered Unmanned Surface Vehicle Actively Passes through the Center of typhoon Sinlaku(2020)显示文摘The solar-powered marine unmanned surface vehicle(USV) developed by the USV team of the Institute of Atmospheric Physics is a rugged, long-duration, and autonomous navigation vessel designed for the collection of longrange, continuous, real-time, meteorological and oceanographic measurements, especially under extreme sea conditions(sea state 6–7). These solar-powered USVs completed a long-term continuous navigation observation test over 26 days.During this time, they coordinated double-USV observations and actively navigated into the path of Typhoon Sinlaku(2020) before collecting data very close to its center during the 2020 USV South China Sea Typhoon Observation Experiment. Detailed high temporal resolution(1 min) real-time observations collected by the USV on the typhoon were used for operational typhoon forecasting and warning for the first time. | 19 | Simultaneous, hybrid single-molecule method by optical tweezers and fluorescence显示文摘As studies on life sciences progress toward the single-molecule level,new experiments have put forward more requirements for simultaneously displaying the mechanical properties and conformational changes of biomolecules.Optical tweezers and fluorescence microscopy have been combined to solve this problem.The combination of instruments forms a new generation of hybrid single-molecule technology that breaks through the limitations of traditional biochemical analysis.Powerfulmanipulation and fluorescence visualization have beenwidely used,and these techniques provide new possibilities for studying complex biochemical reactions at the singlemolecule level.This paper explains the features of this combined technique,including the application characteristics of single-trap and dual-traps,the anti-bleaching method,and optical tweezers combined with epifluorescence,confocal fluorescence,total internal reflection fluorescence,and other fluorescence methods.Using typical experiments,we analyze technical solutions and explain the factors and principles that instrument designers should consider.This review aims to give an introduction to this novel fusion technology process and describe important biological results. | Guoteng Ma Chunguang Hu Shuai Li Xiaoqin Gao Hongbin Li Xiaotang Hu | 2019 | Nanotechnology and Precision Engineering2019,2,4: | 1 |
