1Inulin Can Alleviate Metabolism Disorders in ob/ob Mice by Partially Restoring Leptinrelated Pathways Mediated by Gut Microbiota显示文摘Inulin has been used as a prebiotic to alleviate glucose and lipid metabolism disorders in mice and humans by modulating the gut microbiota. However, the mechanism underlying the alleviation of metabolic disorders by inulin through interactions between the gut microbiota and host cells is unclear. We use ob/ob mice as a model to study the effect of inulin on the cecal microbiota by16 S rRNA gene amplicon sequencing and its interaction with host cells by transcriptomics. The inulin-supplemented diet improved glucose and lipid metabolism disorder parameters in ob/ob mice,alleviating fat accumulation and glucose intolerance. The a diversity of gut microbial community of ob/ob mice was reduced after inulin treatment, while the b diversity tended to return to the level of wild type mice. Interestingly, Prevotellaceae UCG 001(family Prevotellaceae) was obviously enriched after inulin treatment. A comparative analysis of the gene expression profile showed that the cecal transcriptome was changed in leptin gene deficiency mice, whereas the inulin-supplemented diet partially reversed the changes in leptin gene-related signaling pathways, especially AMPK signaling pathway, where the levels of gene expression became comparable to those in wild type mice.Further analysis indicated that Prevotellaceae UCG 001 was positively correlated with the AMPK signaling pathway, which was negatively correlated with markers of glycolipid metabolism disorders. 3Sequence analysis of the gene correlated with cytoplasmic male sterility (CMS) in rape-seed (Brassica napus) Polima and Shaan 2A显示文摘orf224 is a CMS-related mitochondrial gene discovered in Polima cytoplasm. Shaan 2A CMS line is the parent of the first rapeseed hybrid cultivar Qinyou No. 2 that has been grown in many regions of China. In this work, genomic DNA of Polima CMS line and Shaan 2A CMS line were used as templates, two primers of specific oligonucleo-tides at 5’ and 3’ ends were used, PCR was performed, the amplification fragments were cloned into pGEM-T Easy vectors and DNA sequences were determined. The CMS-associated gene, orf224-1 present in Shaan 2A CMS line, has a sequence highly homologous to the orf224 of the Polima CMS line, except for one nucleotidc at position +398. There were only one base (AAC→AGC) and one amino acid (Asn→Ser) differences between the two. The homologies of the two sequences in nucleotide and amino acid were 99.9% and 99.6%, respectively. 5The temporal dynamics of antimicrobial-resistant Salmonella enterica and predominant serovars in China显示文摘Salmonella enterica is one of the most common bacterial pathogens in humans and animals.Systematic studies on the trends and geographical distribution of antimicrobial-resistant Salmonella and dominant serovars have been well studied in European and American countries while not in China.Here,taking the One-Health strategy,we used>35000 Salmonella enterica isolates to explore the temporal and spatial dynamics of dominant serovars in China.We found that Salmonella Typhimurium was the dominant serovar causing human infection in China,which was consistent with Australia but inconsistent with North American and European countries.The proportion of Salmonella serovars Typhimurium,London,Rissen,Corvallis,Meleagridis,Kentucky,and Goldcoast showed an increasing trend during 2006-2019.We randomly selected 1962 isolates for comparative genomics and antimicrobial resistance studies and found that the number of antibiotic resistance genes(ARGs)per isolate increased 1.84 and 2.69 times of human and non-human origins,respectively,spanning 14 years.The proportion of antimicrobial-resistant Salmonella isolates had an increasing trend during 2006-2019,especially beta-lactam,quinolone,tetracycline,and rifampicin resistance.Moreover,we found that higher diversity of sequence types(STs)in S.Typhimurium than in other serovars,ST34 from pig and ST 19 from chicken origin,were mainly associated with isolates causing child and adult gastro-infection,respectively.Our results fill in the data gap on the trends of dominant serovars and antimicrobial resistance of Salmonella enterica in China.These data provide useful information for public health decision-makers prioritizing interventions for foodborne diseases and food safety.Yanan Wang Yue Liu Na Lyu Zhiyuan Li Sufang Ma Demin Cao Yuanlong Pan Yongfei Hu Hua Huang George F.Gao Xuebin Xu Baoli Zhu 2023National Science Review2023,10,3:4
6Bifunctional carbon-based cathode catalysts for zinc-air battery: A review显示文摘Efficient bifunctional OER/ORR catalysts are crucial for the further development of zinc-air battery. From a sustainable point of view, it is important that electrocatalysts are efficient, low cost, and composed of abundant resources instead of scarce metals. Due to their good conductivity, low cost, and strong durability, carbon-based materials are considered a promising alternative in the field of commercial zinc-air battery catalysts. Herein, we briefly introduce the zinc-air battery and then summarize recent progress in the development of carbon-based bifunctional catalysts by defect engineering, heteroatom doping and metal doping. 8Robust superamphiphobic coatings with gradient and hierarchical architecture and excellent anti-flashover performances显示文摘Biomimetic superhydrophobic(SH)coatings have emerged as a promising alternative to traditional room temperature vulcanizing(RTV)silicone rubber coatings for improving the flashover strength of insulators.However,organic contamination occurs in outdoor applications and thus a superamphiphobic(SAP)surface is more desirable but not yet reported for improving flashover performance.Herein,we developed a novel anti-flashover technique by fabricating robust SAP coating with unique gradient and micro-nanoscale hierarchical architecture.The SAP coating was fabricated by sequentially spray-depositing a resin-based primer and a silica-based topcoat on substrates(i.e.,glass slides and insulators).The primer not only functions as an adhesive offering strong adhesion to the substrate but also offers a micromastoid-like structure facilitating the subsequent formation of hierarchical micro-nanostructure.The appropriate spraying pressure leads to a diffusion of the fluorocarbon-modified silica nanoparticles into the primer to form a unique gradient structure,by analogy to inserting bullets into a wood.These features render the SAP coating excellent robustness with strong abrasive resistance,excellent ultraviolet(UV)resistance,and excellent chemical and thermal stability.Pollution flashover property of the SAP coating was explored and compared with that of SH and RTV specimens,from which a novel organic-contamination model to evaluate the flashover performance was proposed.The coated SAP glass insulator demonstrated 42.9%pollution flashover voltage improvement than RTV-coated insulator.These stated unique features reveal the convincing potential of the present SAP coatings to be applied for not only outdoor transmission line insulators for antiflashover but also other fields for self-cleaning,anti-fouling,and anti-icing.Yi Xie Wei Xiong Shefiu Kareem Chuxiong Qiu Yongfei Hu Ivan PParkin Shengwu Wang Huayun Wang Junwu Chen Lee Li Zhi Chen Huajun Sun Xiujian Zhao 2022Nano Research2022,15,8:2
9Two Ultraviolet Radiation Datasets that Cover China显示文摘Ultraviolet(UV) radiation has significant effects on ecosystems, environments, and human health, as well as atmospheric processes and climate change. Two ultraviolet radiation datasets are described in this paper. One contains hourly observations of UV radiation measured at 40 Chinese Ecosystem Research Network stations from 2005 to 2015. CUV3 broadband radiometers were used to observe the UV radiation, with an accuracy of 5%, which meets the World Meteorology Organization's measurement standards. The extremum method was used to control the quality of the measured datasets. The other dataset contains daily cumulative UV radiation estimates that were calculated using an all-sky estimation model combined with a hybrid model. The reconstructed daily UV radiation data span from 1961 to 2014. The mean absolute bias error and root-mean-square error are smaller than 30% at most stations, and most of the mean bias error values are negative, which indicates underestimation of the UV radiation intensity. These datasets can improve our basic knowledge of the spatial and temporal variations in UV radiation. All scale simulation of the two systems provide the basis for design and improvement of air conditioning system fans to improve the internal flow, increase air volume, and reduce the noise. The conclusion that disturbance on flow of nuzzle outlet is the main sources of measurement error about 5% can be proved by the comparative analysis of the results between computation and experiment. 17A thermally crosslinked ion-gel gated artificial synapse显示文摘We demonstrate a synaptic transistor that uses a thermally crosslinked three-dimensional network to accommodate ionic liquid to form an ion gel layer. The synaptic transistor successfully emulated important synaptic plasticity, such as paired-pulse facilitation, spike-number dependent plasticity, spike-voltage dependent plasticity, and spike-rate dependent plasticity;these responses imply successful use of the ion gel. Moreover, the device realized “OR” and “AND” logic operations, and high-pass filtering behavior. Energy consumption of the device can be reduced to sub-femtojoule level, which is below that of biological synapses. Compared with traditional physical cross-linking using block copolymers, this method provides a facile strategy to prepare ion gels with tunable properties by altering the polymers and crosslinkers,and to enormously reduce the price by replacing expensive block copolymers or eliminating additional synthesis processes. 18Cav3.2 channel regulates cerebral ischemia/reperfusion injury:a promising target for intervention显示文摘Calcium influx into neurons triggers neuronal death during cerebral ischemia/reperfusion injury.Various calcium channels are involved in cerebral ischemia/reperfusion injury.Cav3.2 channel is a main subtype of T-type calcium channels.T-type calcium channel blockers,such as pimozide and mibefradil,have been shown to prevent cerebral ischemia/reperfusion injury-induced brain injury.However,the role of Cav3.2 channels in cerebral ischemia/reperfusion injury remains unclear.Here,in vitro and in vivo models of cerebral ischemia/reperfusion injury were established using middle cerebral artery occlusion in mice and high glucose hypoxia/reoxygenation exposure in primary hippocampal neurons.The results showed that Cav3.2 expression was significantly upregulated in injured hippocampal tissue and primary hippocampal neurons.We further established a Cav3.2 gene-knockout mouse model of cerebral ischemia/reperfusion injury.Cav3.2 knockout markedly reduced infarct volume and brain water content,and alleviated neurological dysfunction after cerebral ischemia/reperfusion injury.Additionally,Cav3.2 knockout attenuated cerebral ischemia/reperfusion injury-induced oxidative stress,inflammatory response,and neuronal apoptosis.In the hippocampus of Cav3.2-knockout mice,calcineurin overexpression offset the beneficial effect of Cav3.2 knockout after cerebral ischemia/reperfusion injury.These findings suggest that the neuroprotective function of Cav3.2 knockout is mediated by calcineurin/nuclear factor of activated T cells 3 signaling.Findings from this study suggest that Cav3.2 could be a promising target for treatment of cerebral ischemia/reperfusion injury.Feibiao Dai Chengyun Hu Xue Li Zhetao Zhang Hongtao Wang Wanjun Zhou Jiawu Wang Qingtian Geng Yongfei Dong Chaoliang Tang 2024Neural Regeneration Research2024,19,11:0
19Ginkgo biloba extract alleviates fatty liver hemorrhagic syndrome in laying hens via reshaping gut microbiota显示文摘Background Ginkgo biloba extract(GBE)is evidenced to be effective in the prevention and alleviation of metabolic disorders,including obesity,diabetes and fatty liver disease.However,the role of GBE in alleviating fatty liver hemorrhagic syndrome(FLHS)in laying hens and the underlying mechanisms remain to be elucidated.Here,we investigated the effects of GBE on relieving FLHS with an emphasis on the modulatory role of GBE in chicken gut microbiota.Results The results showed that GBE treatment ameliorated biochemical blood indicators in high-fat diet(HFD)-induced FLHS laying hen model by decreasing the levels of TG,TC,ALT and ALP.The lipid accumulation and pathological score of liver were also relieved after GBE treatment.Moreover,GBE treatment enhanced the antioxidant activity of liver and serum by increasing GSH,SOD,T-AOC,GSH-PX and reducing MDA,and downregulated the expression of genes related to lipid synthesis(FAS,LXRα,GPAT1,PPARγand Ch REBP1)and inflammatory cytokines(TNF-α,IL-6,TLR4 and NF-κB)in the liver.Microbial profiling analysis revealed that GBE treatment reshaped the HFD-perturbed gut microbiota,particularly elevated the abundance of Megasphaera in the cecum.Meanwhile,targeted metabolomic analysis of SCFAs revealed that GBE treatment significantly promoted the production of total SCFAs,acetate and propionate,which were positively correlated with the GBE-enriched gut microbiota.Finally,we confirmed that the GBE-altered gut microbiota was sufficient to alleviate FLHS by fecal microbiota transplantation(FMT).Conclusions We provided evidence that GBE alleviated FLHS in HFD-induced laying hens through reshaping the composition of gut microbiota.Our findings shed light on mechanism underlying the anti-FLHS efficacy of GBE and lay foundations for future use of GBE as additive to prevent and control FLHS in laying hen industry.Xinyue Yang Depeng Li Meihong Zhang Yuqing Feng Xiaolu Jin Dan Liu Yuming Guo Yongfei Hu 2024Journal of Animal Science and Biotechnology2024,15,1:0
20Discovery of IHMT-337 as a potent irreversible EZH2 inhibitor targeting CDK4 transcription for malignancies显示文摘Enhancer of zeste homolog 2(EZH2),an enzymatic subunit of PRC2 complex,plays an important role in tumor development and progression through its catalytic and noncatalytic activities.Overexpression or gain-of-function mutations of EZH2 have been significantly associated with tumor cell proliferation of triple-negative breast cancer(TNBC)and diffuse large B-cell lymphoma(DLBCL).As a result,it has gained interest as a potential therapeutic target.The currently available EZH2 inhibitors,such as EPZ6438 and GSK126,are of benefit for clinical using or reached clinical trials.However,certain cancers are resistant to these enzymatic inhibitors due to its noncatalytic or transcriptional activity through modulating nonhistone proteins.Thus,it may be more effective to synergistically degrade EZH2 in addition to enzymatic inhibition.Here,through a rational design and chemical screening,we discovered a new irreversible EZH2 inhibitor,IHMT-337,which covalently bounds to and degrades EZH2 via the E3 ligase CHIP-mediated ubiquitination pathway.Moreover,we revealed that IHMT-337 affects cell cycle progression in TNBC cells through targeting transcriptional regulating of CDK4,a novel PRC2 complex-and enzymatic activity-independent function of EZH2.More significantly,our compound inhibits both DLBCL and TNBC cell proliferation in different preclinical models in vitro and in vivo.Taken together,our findings demonstrate that in addition to enzymatic inhibition,destroying of EZH2 by IHMT-337 could be a promising therapeutic strategy for TNBC and other malignancies that are independent of EZH2 enzymatic activity.Husheng Mei Hong Wu Jing Yang Bin Zhou Aoli Wang Chen Hu Shuang Qi Zongru Jiang Fengming Zou Beilei Wang Feiyang Liu Yongfei Chen Wenchao Wang Jing Liu Qingsong Liu 2023Signal Transduction and Targeted Therapy2023,8,2:0
