1Polycystic ovary syndrome patients with high BMI tend to have functional disorders of androgen excess:a prospective study显示文摘Biochemical or clinical changes of hyperandrogenism are important elements of polycystic ovary syndrome(PCOS).There is currently no consensus on the definition and diagnostic criteria of hyperandrogenism in PCOS.The aim of this study was to investigate the complex symptoms of hyperandrogenic disorders and the correlations between metabolism and hyperandrogenism in patients with PCOS from an outpatient reproductive medicine clinic in China.We conducted a case control study of 125 PCOS patients and 130 controls to evaluate differences in body mass index(BMI),total testosterone(TT),modified Ferriman-Gallwey hirsutism score,sex hormone binding globulin(SHBG),homeostasis model assessment-estimated insulin resistance(HOMA-IR) and free androgen index(FAI) between PCOS patients and controls and subgroups of PCOS.The prevalence of acne and hirsutism did not differ significantly between the hyperandrogenic and non-hyperandrogenic subgroup.Patients with signs of hyperandrogenism had significantly higher BMI(P<0.05),but differences in TT,SHBG,FAI and waist/hip ratio were insignificant.The odds ratio of overweight was calculated for all PCOS patients.Our results suggest that PCOS patients with high BMI tend to have functional disorders of androgen excess;therefore,BMI may be a strong predictor of hyperandrogenism in PCOS.Chun Yuan Xiaoqiang Liu Yundong Mao Feiyang Diao Yugui Cui Jiayin Liu 2016The Journal of Biomedical Research2016,30,3:8
2Single-cell Immune Landscape of Human Recurrent Miscarriage显示文摘t Successful pregnancy in placental mammals substantially depends on the establishment of maternal immune tolerance to the semi-allogenic fetus.Disorders in this process are tightly associated with adverse pregnancy outcomes including recurrent miscarriage(RM).However,an indepth understanding of the systematic and decidual immune environment in RM remains largely lacking.In this study,we utilized single-cell RNA-sequencing(scRNA-seq)to comparably analyze the cellular and molecular signatures of decidual and peripheral leukocytes in normal and unexplained RM pregnancies at the early stage of gestation.Integrative analysis identifies 22 distinct cell clusters in total,and a dramatic difference in leukocyte subsets and molecular properties in RM cases is revealed.Specifically,the cytotoxic properties of CD8^(+)effector T cells,nature killer(NK),and mucosal-associated invariant T(MAIT)cells in peripheral blood indicates apparently enhanced pro-inflammatory status,and the population proportions and ligand–receptor interactions of the decidual leukocyte subsets demonstrate preferential immune activation in RM patients.The molecular features,spatial distribution,and the developmental trajectories of five decidual NK(dNK)subsets have been elaborately illustrated.In RM patients,a dNK subset that supports embryonic growth is diminished in proportion,while the ratio of another dNK subset with cyto toxic and immune-active signature is significantly increased.Notably,a unique pro-inflammatory CD56^(+)CD16^(+)dNK subset substantially accumulates in RM decidua.These findings reveal a comprehensive cellular and molecular atlas of decidual and peripheral leukocytes in human early pregnancy and provide an in-depth insight into the immune pathogenesis for early pregnancy loss.Feiyang Wang Wentong Jia Mengjie Fan Xuan Shao Zhilang Li Yongjie Liu Yeling Ma Yu-Xia Li Rong Li Qiang Tu Yan-Ling Wang 2021Genomics, Proteomics & Bioinformatics2021,19,2:8

4First report on Guanshan Biota(Cambrian Stage 4) at the stratotype area of Wulongqing Formation in Malong County, Eastern Yunnan, China显示文摘Exceptionally preserved fossils,such as those from Cambrian Burgess Shale-type fossil-Lagerstatten are critical because of their unique contributions to knowledge of the phylogenetic radiation and palaeoecological expansion of metazoans during the Cambrian explosion.Critically,these deposits provide information that is usually unobtainable from shelly and skeletonized fossils alone.The Guanshan Biota(Cambrian Series 2,Stage 4)in the Yunnan Province of South China,has produced abundant and diverse,exquisitely preserved fossils that often retain soft tissues and organs.To date,most fossils from the Guanshan Biota have been collected from localities such as Gaoloufang and Gangtoucun,which have become inaccessible due to new urban expansions and constructions of residential buildings.Here we present the first report of soft bodied fossils from a new section at Kanfuqing,close to the Wulongqing village in Malong County,approximately 3 km east of the Wulongqing Formation stratotype section.Fossils retain soft morphology,and include brachiopods with delicate marginal setae,priapulids with well-preserved sclerites and vetulicolians with entire sections of body.In addition,this fauna includes rare occurrences of trilobites preserved with soft tissues replicated as pyrite pseudomorphs after weathering.This discovery represents an important palaeogeographical extension of soft-bodied fossils of the Guanshan fauna to the east of the Xiaojiang Fault(related to Tsinning tectonic movements ca.700 Ma).The fauna from the new Kanfuqing section is similar to that reported from the Wulongqing Formation west of the Xiaojiang Fault,and thus has significant implications for early Cambrian palaeogeography,faunal successions and palaeoenvironments of eastern Yunnan.Feiyang Chen Zhifei Zhang Marissa J. 5Genome-wide identification of agronomically important genes in outcrossing crops using OutcrossSeq显示文摘Many important crops(e.g.,tuber,root,and tree crops)are cross-pollinating.For these crops,no inbred lines are available for genetic study and breeding because they are self-incompatible,clonally propagated,or have a long generation time,making the identification of agronomically important genes difficult,particularly in crops with a complex autopolyploid genome.In this study,we developed a method,OutcrossSeq,for mapping agronomically important loci in outcrossing crops based on whole-genome low-coverage resequencing of a large genetic population,and designed three computation algorithms in OutcrossSeq for different types of outcrossing populations.We applied OutcrossSeq to a tuberous root crop(sweet potato,autopolyploid),a tree crop(walnut tree,highly heterozygous diploid),and hybrid crops(double-cross populations)to generate high-density genotype maps for the outcrossing populations,which enable precise identification of genomic loci underlying important agronomic traits.Candidate causative genes at these loci were detected based on functional clues.Taken together,our results indicate that OutcrossSeq is a robust and powerful method for identifying agronomically important genes in heterozygous species,including polyploids,in a cost-efficient way.The OutcrossSeq software and its instruction manual are available for downloading at www.xhhuanglab.cn/tool/OutcrossSeq.html.Mengjiao Chen Weijuan Fan Feiyang Ji Hua Hua Jie Liu Mengxiao Yan Qingguo Ma Jiongjiong Fan Qin Wang Shufeng Zhang Guiling Liu Zhe Sun Changgeng Tian Fengling Zhao Jianli Zheng Qi Zhang Jiaxin Chen Jie Qiu Xin Wei Ziru Chen Peng Zhang Dong Pei Jun Yang Xuehui Huang 2021Molecular Plant2021,14,4:4
6Association of assisted reproductive technology, germline de novo mutations and congenital heart defects in a prospective birth cohort study显示文摘Emerging evidence suggests that children conceived through assisted reproductive technology(ART)have a higher risk of congenital heart defects(CHDs)even when there is no family history.De novo mutation(DNM)is a well-known cause of sporadic congenital diseases;however,whether ART procedures increase the number of germline DNM(gDNM)has not yet been well studied.Here,we performed whole-genome sequencing of 1137 individuals from 160 families conceived through ART and 205 families conceived spontaneously.Children conceived via ART carried 4.59 more gDNMs than children conceived spontaneously,including 332 paternal and 1.26 maternal DNMs,after correcting for parental age at conception,cigarette smoking,alcohol drinking,and exercise behaviors.Paternal DNMs in offspring conceived via ART are characterized by C>T substitutions at CpG sites,which potentially affect protein-coding genes and are significantly associated with the increased risk of CHD.In addition,the accumulation of non-coding functional mutations was independently associated with CHD and 87.9% of the mutations were originated from the father.Among ART offspring,infertility of the father was associated with elevated paternal DNMs;usage of both recombinant and urinary follicle-stimulating hormone and high-dosage human chorionic gonadotropin trigger was associated with an increase of maternal DNMs.In sum,the increased gDNMs in offspring conceived by ART were primarily originated from fathers,indicating that ART itself may not be a major reason for the accumulation of gDNMs.Our findings emphasize the importance of evaluating the germline status of the fathers in families with the use of ART.Cheng Wang Hong Lv Xiufeng Ling Hong Li Feiyang Diao Juncheng Dai Jiangbo Du Ting Chen Qi Xi Yang Zhao Kun Zhou Bo Xu Xiumei Han Xiaoyu Liu Meijuan Peng Congcong Chen Shiyao Tao Lei Huang Cong Liu Mingyang Wen Yangqian Jiang Tao Jiang Chuncheng Lu Wei Wu Di Wu Minjian Chen Yuan Lin Xuejiang Guo Ran Huo Jiayin Liu Hongxia Ma Guangfu Jin Yankai Xia Jiahao Sha Hongbing Shen Zhibin Hu 2021Cell Research2021,31,8:2
7Follicular hyperandrogenism and insulin resistance in polycystic ovary syndrome patients with normal circulating testosterone levels显示文摘Polycystic ovary syndrome(PCOS) is a common reproductive disease with high heterogeneity. The role of excess androgen in PCOS etiology remains disputed, since around 20%-50% of PCOS women do not display hyperandrogenemia. The microenvironment of the ovary critically influences follicular development. In the present study, we assessed the role of androgen in PCOS by investigating whether excessive follicular fluid androgen was present in PCOS patients with normal serum androgen levels and influenced by follicular fluid insulin resistance(IR).Follicular fluid samples of 105 women with PCOS and 105 controls were collected. Levels of steroid hormones,glucose and insulin in the follicular fluid were examined and compared with data from serum biochemistry tests. We found that 64.9%(63/97) of PCOS patients with normal serum androgen levels displayed abnormally high follicular fluid androgen level. The follicular fluid androgen level was positively correlated with follicular fluid IR within a certain range and follicular fluid estrogen-to-testosterone(E2/T) ratio was significantly reduced in these patients.These results indicated that there existed a subgroup of PCOS patients who displayed excessive follicular fluid androgen and IR despite their normal circulating testosterone(T) levels. 8The appliances and prospects of aurum nanomaterials in biodiagnostics, imaging, drug delivery and combination therapy显示文摘Aurum nanomaterials(ANM), combining the features of nanotechnology and metal elements, have demonstrated enormous potential and aroused great attention on biomedical applications over the past few decades. Particularly, their advantages, such as controllable particle size, flexible surface modification, higher drug loading, good stability and biocompatibility, especially unique optical properties, promote the development of ANM in biomedical field. In this review, we will discuss the advanced preparation process of ANM and summarize their recent applications as well as their prospects in diagnosis and therapy. 10Effects of hydrological connection and human disturbance on genetic variation of submerged Vallisneria natans populations in four lakes in China显示文摘With the increase in the need for flood prevention and lake resource used by humans,the construction of floodgates and sluices has changed the hydrological connection between rivers and lakes,and between adjacent lakes.In river-disconnected lakes,exploitation and use of lake resources have resulted in water quality decline and mechanical disturbance intensification to a different degree.Of the large number of river-disconnected lakes in the middle-lower reaches of the Changjiang(Yangtze)River,the Futou Lake,and the Xiliang Lake lie close together and are,historically,directly connected,and so do Liangzi Lake and Baoan Lake.The extent of human disturbance is severe in the Futou Lake and the Baoan Lake,but relatively mild in the Xiliang Lake and Liangzi Lake.The freshwater rosette-forming submerged plant Vallisneria natans is one of the dominant species in the four lakes.Using microsatellite markers,we studied the genetic variation of V.natans subpopulations in lakes with different intensities of human disturbance and historical direct hydrological connections.Our results showed that human disturbance decreased plant density and clonal growth in V.natans,but might increase genetic and clonal diversity at a subpopulation level and enhance gene flow among subpopulations by sexual propagule movement.Under similar climatic conditions,different intensities of disturbance seem to have such a high selective potential to differentiate genetically adjacent lake populations that they outperform the forces of gene flow through historical direct hydrological interconnection,which tends to produce genetic homogeneity.Our findings imply that human disturbance has a profound eff ect on the evolutionary process of natural populations of submerged plants.Moreover,increased subpopulation genetic diversity can enhance resistance and resilience to environmental disturbances.To a certain degree,we could expect that disturbed populations have the possibility of restoring spontaneously if humans cease to perturb natural ecosystems in the future.Qianjin CAO Beibei LIU Feiyang HU 2021Journal of Oceanology and Limnology2021,39,4:1
11A nomogram predicting clinical pregnancy in the first fresh embryo transfer for women undergoing in vitro fertilization and intracytoplasmic sperm injection(IVF/ICSI) treatments显示文摘The extent to which factors affect the probability of clinical pregnancy in the first fresh embryo transfer after assisted conception is unknown.In order to examine the predictors of clinical pregnancy,a retrospective cohort study was launched between January 1,2013 and December 31,2016 in four infertility clinics including 19837 in vitro fertilization and intracytoplasmic sperm injection(IVF/ICSI)fresh cycles with known outcomes and relevant records.A multivariable logistic regression was used to select the most significant predictors in the final nomogram for predicting clinical pregnancy.Furthermore,the model was validated by an independent validation set and the performance of the model was evaluated by the receiver operating characteristic(ROC)curves along with the area under the ROC curve(AUC)and calibration plots.In a training set including 17854 participants,we identified that female age,tubal factor,number of embryos transferred,endometrial thickness and number of good-quality embryos were independent predictors for clinical pregnancy.We developed a nomogram using these five factors and the predictive ability was 0.66 for AUC(95%CI=0.64−0.68),which was independently validated in the validation set(AUC=0.66,95%CI=0.65−0.68).Our results show that some specific factors can be used to provide infertile couples with an accurate assessment of clinical pregnancy following assisted conception and facilitate to guide couples and clinicians.Fang Wu Feng Liu Yichun Guan Jiangbo Du Jichun Tan Hong Lv Qun Lu Shiyao Tao Lei Huang Kun Zhou Yankai Xia Xinru Wang Hongbing Shen Xiufeng Ling Feiyang Diao Zhibin Hu Guangfu Jin 2019The Journal of Biomedical Research2019,33,6:1

133个小型高原湖泊水陆交错带中植物叶片pH值变异及其潜在影响因素显示文摘陆生植物叶片pH值是与植物生理功能和养分利用状况息息相关的重要植物属性,并随植物功能型和环境的变化而变化。然而,水生植物叶片pH值的变异特征及其与陆生植物间的差异性仍不清楚。在3个自然形成的小型高原湖泊的湖滨植物群落中,我们采集植物、土壤和湖水样品,检测了植物叶片pH值与碳、氮含量以及环境条件(水或土壤pH值、土壤水位状况)。从植物功能型和群落两个水平分析了湖滨植物叶pH值沿水分梯度的变化及其潜在影响因素。研究结果表明,不同类型的水生植物,以及水生、沼生和陆生环境中生长的植物的叶pH存在较大差异;浮叶植物叶片pH值(4.21±0.05)显著低于挺水植物(5.71±0.07)和沉水植物(5.82±0.06);水生草本植物叶片pH值(5.43±0.10)显著低于沼生草本植物(6.12±0.07)和陆生草本植物(5.74±0.05);陆生草本植物叶片pH值显著高于木本植物。植物功能群间叶片pH值的变异可能与叶片的结构、光利用特性和养分特性有关。群落水平的结果与植物功能型水平相一致:水生群落叶片pH值显著低于陆生群落。群落水平叶片pH值可能主要受物种组成、水分条件和环境pH值的影响。本研究首次探索了水生植物的pH值,并且在景观尺度上对比了不同功能型植物的叶片pH值,为深入探索植物pH值变异机制以及其在水生和湿地生态系统中的生态意义提供参考。Sining Liu Shenqun An Zhengbing Yan Junpeng Ren Xiaoqian Lu Feiyang Ge Wenxuan Han 2021Journal of Plant Ecology2021,14,3:1

15Discovery of IHMT-337 as a potent irreversible EZH2 inhibitor targeting CDK4 transcription for malignancies显示文摘Enhancer of zeste homolog 2(EZH2),an enzymatic subunit of PRC2 complex,plays an important role in tumor development and progression through its catalytic and noncatalytic activities.Overexpression or gain-of-function mutations of EZH2 have been significantly associated with tumor cell proliferation of triple-negative breast cancer(TNBC)and diffuse large B-cell lymphoma(DLBCL).As a result,it has gained interest as a potential therapeutic target.The currently available EZH2 inhibitors,such as EPZ6438 and GSK126,are of benefit for clinical using or reached clinical trials.However,certain cancers are resistant to these enzymatic inhibitors due to its noncatalytic or transcriptional activity through modulating nonhistone proteins.Thus,it may be more effective to synergistically degrade EZH2 in addition to enzymatic inhibition.Here,through a rational design and chemical screening,we discovered a new irreversible EZH2 inhibitor,IHMT-337,which covalently bounds to and degrades EZH2 via the E3 ligase CHIP-mediated ubiquitination pathway.Moreover,we revealed that IHMT-337 affects cell cycle progression in TNBC cells through targeting transcriptional regulating of CDK4,a novel PRC2 complex-and enzymatic activity-independent function of EZH2.More significantly,our compound inhibits both DLBCL and TNBC cell proliferation in different preclinical models in vitro and in vivo.Taken together,our findings demonstrate that in addition to enzymatic inhibition,destroying of EZH2 by IHMT-337 could be a promising therapeutic strategy for TNBC and other malignancies that are independent of EZH2 enzymatic activity.Husheng Mei Hong Wu Jing Yang Bin Zhou Aoli Wang Chen Hu Shuang Qi Zongru Jiang Fengming Zou Beilei Wang Feiyang Liu Yongfei Chen Wenchao Wang Jing Liu Qingsong Liu 2023Signal Transduction and Targeted Therapy2023,8,2:0
16Development of a new cerebral ischemia reperfusion model of Mongolian gerbils and standardized evaluation system显示文摘Background:The Mongolian gerbil is an excellent laboratory animal for preparing the cerebral ischemia model due to its inherent deficiency in the circle of Willis.However,the low incidence and unpredictability of symptoms are caused by numerous complex variant types of the circle.Additionally,the lack of an evaluation system for the cer-ebral ischemia/reperfusion(I/R)model of gerbils has shackled the application of this model.Methods:We created a symptom-oriented principle and detailed neurobehavioral scoring criteria.At different time points of reperfusion,we analyzed the alteration in locomotion by rotarod test and grip force score,infarct volume by triphenyltetrazo-lium chloride(TTC)staining,neuron loss using Nissl staining,and histological charac-teristics using hematoxylin-eosin(H&E)straining.Results:With a successful model rate of 56%,32 of the 57 gerbils operated by our method harbored typical features of cerebral I/R injury,and the mortality rate in the male gerbils was significantly higher than that in the female gerbils.The suc-cessfully prepared I/R gerbils demonstrated a significant reduction in motility and grip strength at 1 day after reperfusion;formed obvious infarction;exhibited typi-cal pathological features,such as tissue edema,neuronal atrophy and death,and vacuolated structures;and were partially recovered with the extension of reperfu-sion time.Conclusion:This study developed a new method for the unilateral common carotid artery ligation I/R model of gerbil and established a standardized evaluation system for this model,which could provide a new cerebral I/R model of gerbils with more practical applications.Ying Wu Caijiao Hu Zhihui Li Feiyang Li Jianyi Lv Meng Guo Xin Liu Changlong Li Xueyun Huo Zhenwen Chen Lifeng Yang Xiaoyan Du 2024Animal Models and Experimental Medicine2024,7,1:0

18Local-scale patterns of genetic variation in coexisting floating-leaved Nymphoides peltata and submerged Myriophyllum spicatum in Donghu Lake显示文摘Coexisting floating-leaved and submerged plants experience similar environmental changes but may evolve different patterns of genetic variation.To compare local-scale genetic variation,we collected samples of floating-leaved Nymphoides peltata and submerged Myriophyllum spicatum coexisting in a disturbed urban lake in China.At the subpopulation level,using microsatellites,M.spicatum had higher clonal diversity than N.peltata.M.spicatum had 28.4%multilocus genotypes(MLGs)shared between subpopulations,but N.peltata had only one MLG shared between two adjacent subpopulations.N.peltata displayed more genetic variation between subpopulations than within subpopulations,but the reverse was true for M.spicatum.Principal components and Bayesian cluster analyses showed that individuals from each subpopulation of N.peltata tended to have relatively close genetic relationships.For M.spicatum,individuals from each subpopulation were genetically scattered with those from other subpopulations.Our results imply that in unpredictable adverse environments M.spicatum may be less subjected to local-deme extinction than N.peltata because of genetically diverse clones at the subpopulation level.This characteristic means that following adverse events,M.spicatum may rapidly restore subpopulation distributions via recolonization and intense gene flow among subpopulations.CAO Qianjin HU Feiyang LIU Na 2020Journal of Oceanology and Limnology2020,38,6:0
19Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND 20Soluble triarylamine functionalized symmetric viologen for all-solid-state electrochromic supercapacitors显示文摘Electrochromic supercapacitors have drawn enormous attention due to their ability to monitor the charge and discharge processes through color changes of electroactive materials.However,there are few work on small organic molecules as active materials for all-solid-state electrochromic supercapacitors.Herein,we reported two novel multifunctional symmetric viologens(TPA-bpy and CZ-bpy),which showed different solvatochromic,electrochromic,electroluminochromic and energy storage behaviors despite their similar chemical structures.The different performances between these two viologens were attributed to the difference in the intramolecular charge transfer capability and the solubility in organic solvents.Devices containing TPA-bpy displayed faster response time and higher coloration efficiency due to the introduction of packing-disruptive and three-dimensional triarylamine groups.Moreover,devices containing TPA-bpy also showed energy storage characteristics with an obvious color change from purple to yellow.It showed a wide voltage window(2.0 V),long discharge time(230.3 s at 0.01 mA cm−2),and excellent cycling stability with 90%capacitance retention after 6,000 cycles.The work provides a new and convenient strategy towards the development of novel electrochromic capacitive materials.Yanling Zhuang Weiwei Zhao Longlu Wang Feiyang Li Weikang Wang Shujuan Liu Wei Huang Qiang Zhao 2020Science China Chemistry2020,63,11:0
