2Single-cell transcriptome analysis reveals widespread monoallelic gene expression in individual rice mesophyll cells显示文摘Monoallelic gene expression refers to the phenomenon that all transcripts of a gene in a cell are expressed from only one of the two alleles in a diploid organism. Although monoallelic gene expression has been occasionally reported with bulk transcriptome analysis in plants, how prevalent it is in individual plant cells remains unknown. Here, we developed a single-cell RNA-seq protocol in rice and investigated allelic expression patterns in mesophyll cells of indica(93-11) and japonica(Nipponbare) inbred lines, as well as their F1 reciprocal hybrids. We observed pervasive monoallelic gene expression in individual mesophyll cells, which could be largely explained by stochastic and independent transcription of two alleles. By contrast, two mechanisms that were proposed previously based on bulk transcriptome analyses, parent-oforigin effects and allelic repression, were not well supported by our data. Furthermore, monoallelically expressed genes exhibited a number of characteristics, such as lower expression levels, narrower H3K4me3/H3K9ac/H3K27me3 peaks, and larger expression divergences between 93-11 and Nipponbare. 4Tuning charge density of chimeric antigen receptor optimizes tonic signaling and CAR-T cell fitness显示文摘Tonic signaling of chimeric antigen receptor(CAR),i.e.,the spontaneous CAR activation in the absence of tumor antigen stimulation,is considered to be a pivotal event controlling CAR-T efficacy.However,the molecular mechanism underlying the spontaneous CAR signals remains elusive.Here,we unveil that positively charged patches(PCPs)on the surface of the CAR antigenbinding domain mediate CAR clustering and result in CAR tonic signaling.For CARs with high tonic signaling(e.g.,GD2.CAR and CSPG4.CAR),reducing PCPs on CARs or boosting ionic strength in the culture medium during ex vivo CAR-T cell expansion minimizes spontaneous CAR activation and alleviates CAR-T cell exhaustion.In contrast,introducing PCPs into the CAR with weak tonic signaling,such as CD19.CAR,results in improved in vivo persistence and superior antitumor function.These results demonstrate that CAR tonic signaling is induced and maintained by PCP-mediated CAR clustering.Notably,the mutations we generated to alter the PCPs maintain the antigen-binding affinity and specificity of the CAR.Therefore,our findings suggest that the rational tuning of PCPs to optimize tonic signaling and in vivo fitness of CAR-T cells is a promising design strategy for the nextgeneration CAR.Jian Chen Shizhen Qiu Wentao Li Kun Wang Yu Zhang Han Yang Baichuan Liu Guangfei Li Li Li Min Chen Junjie Lan Jiahua Niu Peijie He Lei Cheng Gaofeng Fan Xin Liu Xianmin Song Chenqi Xu Haitao Wu Haopeng Wang 2023Cell Research2023,33,5:2
5RS and GIS-based Analysis of Landscape Pattern Changes in Urban-rural Ecotone:A Case Study of Daiyue District,Tai' an City,China显示文摘Based on RS data of Daiyue District,Tai'an City in 2000,2005 and 2010,changes of land use types in urban-rural ecotone of Tai'an City from 2000 to 2010 were analyzed.Ecological theories,ArcGIS techniques and landscape structure analysis software Fragstats were applied to select relevant landscape pattern indexes and analyze changes of landscape structure,and compare changes in two durations(2000-2005,2005-2010).The results showed that area of natural landscapes in the study area declined from 2000 to 2010,construction land expanded,land use types were mainly transferred from natural landscapes to man-made landscapes.In terms of landscape level,number of patches(NP),patch density(PD),patch shape index(SHAPE),Shannon's diversity index(SHDI) and Shannon's evenness index(SHEI) increased,the largest patch index(LPI) declined.In terms of type level,arable land were influenced by the most human interventions,large-scale patches turned fragmented,and landscape dominance degraded;woodland landscapes were concentrated in mountainous areas,waterscape indexes showed slight changes.Dominance of regional dominant landscape types degraded,landscape fragmentation and landscape heterogeneity increased,and landscape stability declined.SHI Haopeng YU Kaiqin FENG Yongjun DONG Xiaochang ZHAO Haiyan DENG Xiaomei 2012Journal of Landscape Research2012,4,9:2

7Object registration using an RGB-D camera for complex product augmented assembly guidance显示文摘Background Augmented assembly guidance aims to help users complete assembly operations more efficiently and quickly through augmented reality technology,breaking the limitations of traditional assembly guidance technology which is single in content and boring in way.Object registration is one of the key technologies in augmented assembly guidance process,which can affect the location and direction of virtual assembly guidance information in real assembly environment.Methods This paper presents an object registration method based on RGB-D camera,which combines Lucas-Kanade(LK)optical flow algorithm and Iterative Closet Point(ICP)algorithm.An augmented assembly guidance system for complex products through this method is built.Meanwhile,in order to compare the effectiveness of the proposed method,we also implemented object registration based on an open source augmented reality SDK Vuforia.Results An engine model and a complex weapon cabin equipment are taken as an case to verify this work.The result shows that the registration method proposed in this paper is more accurate and stable compared with that based on Vuforia and the augmented assembly guidance system through this method greatly improves the user's time compared with the traditional assembly.Conclusions Therefore,we can conclude that the object registration method mentioned in this paper can be well applied in the augmented assembly guidance system,which can do enhance the efficiency of assembly considerably.Kangkang YANG Yu GUO Pengzhou TANG Haopeng ZHANG Han LI 2020Virtual Reality & Intelligent Hardware2020,2,6:2

15p75 neurotrophin receptor signal pathway influence on apoptosis in anterior horn neurons of the spinal cord in a rat model of cauda equina compression injury显示文摘BACKGROUND: Studies have demonstrated that cauda equina compression results in apoptosis ofmotor neurons in the spinal cord. The combination of p75 neurotrophin receptor (p75NTR) andprecursor of nerve growth factor (pro-NGF) expression initiates the apoptotic pathway and inducesneuronal apoptosis. However, few reports have focused on the p75-mediated mechanism ofneuronal apoptosis following cauda equine compression injuryOBJECTIVE: To determine apoptosis of spinal cord neurons and activation of thepro-NGF-p75NTR-JNK(c-Jun N-terminal kinase) signal pathway in rats following cauda equinacompression, and to verify experimental outcomes.DESIGN, TIME AND SETTING: A randomized, controlled, in vivo experiment was performed at theMedical Experimental Center of Xi'an Jiaotong University between April and November in 2008.MATERIALS: Streptavidin-perosidase kit was purchased from Wuhan Boster, China; in situ endlabeling detection kit was provided by Promega, USA; type AEG-220G electron microscope waspurchased from Hitachi, Japan.METHODS: A total of 48 healthy, adult, female, Sprague Dawley rats were randomly assigned tothree groups: normal (n = 6), sham-surgery (n = 6), and compression (n = 36). The compressiongroup was randomly assigned to six subsets at 1,3, 5, 7, 14, and 28 days, respectively, with 6 rats ineach subset. A cylindrical silica gel stick was implanted into the rats to compress 75% of thevertebral canal in the compression group; in the sham-surgery group, only vertebral resection wasperformed; and no procedures were performed in the normal group.MAIN OUTCOME MEASURES: At 1,3, 5, 7, 14, and 28 days following compression, L_(2-3) spinalcord segments were processed for immunohistochemistry, in situ cell apoptosis detection, andtransmission electron microscopy observation. Nissl staining was used to observe neuronal survivalin the L_2 spinal cord segment. Immunohistochemistry was applied to detect expressions of pro-NGF,p75NTR, and JNK in the L_2 segment. TUNEL fluorometric method was used to observe apoptosis ofneurons in the L_2 segment.RESULTS: In the normal and sham-surgery groups, little neuronal apoptosis was observed in theL_(2-3) spinal cord segment. At 3 days after compression injury, pro-NGF, p75NTR and JNK expressionwas observed in the spinal cord. Expression levels reached a peak at 7 days, and then graduallydecreased. In the compression and sham-surgery groups, neurons primarily expressed pro-NGFand p75NTR. The number of JNK-positive neurons in the compression group was dramaticallyincreased compared with the sham-surgery group (P < 0.05). A few neurons were apoptotic in thespinal cord 1 day after compression injury. The number of apoptotic neurons gradually increasedand reached a peak at 7 days, and subsequently decreased. Apoptosis was still detectable at 28days. There was a positive correlation between p75NTR expression and neuronal apoptosis(r= 0.75, P< 0.05).CONCLUSION: Following cauda equina compression injury, apoptosis of spinal cord neurons wasobserved. 18Adenovirus-mediated short hairpin RNA interference against p75 neurotrophin receptor in pheochromocytoma cells显示文摘Previous studies have confirmed that motor neuron apoptosis in the anterior horn of the lumbosacral spinal cord is positively correlated with p75 neurotrophin receptor (p75NTR) expression in rat models of cauda equina syndrome. This study used adenovirus to carry a short hairpin RNA (shRNA) for p75NTR gene silencing, to reduce p75NTR expression in the damaged phase and to decrease motor neuron apoptosis. Three p75 siRNA template oligonucleotide segments (shRNA) were designed, and cloned into the 1.0 CMV shuttle vector. HEK293 cells were cotransfected with shuttle vector (carrying shRNA) and an adenovirus vector framework expressing enhanced green fluorescent protein. Thus, this study successfully obtained adenovirus carrying p75shRNA. The obtained viruses were named Ad.shRNA1, Ad.shRNA2, and Ad.shRNA3. The recombinant adenoviruses were separately used to infect cultured pheochromocytoma cells (PC12). Forty-eight hours later, p75NTR mRNA and total protein were analyzed from the PC12 cells. Compared with the negative controls, RNA interference rates were separately 98.49 ± 0.68%, 95.08 ± 1.79% and 96.60 ± 1.14% at the mRNA level, and 72.89 ± 2.17%, 58.83 ± 1.15% and 59.88 ± 0.44% at the protein level in the Ad.shRNA1, Ad.shRNA2, and Ad.shRNA3 groups, respectively.
