| 1 | Genetic diversity analysis of spawner and recaptured populations of Chinese shrimp(Fenneropenaeus chinensis)during stock enhancement in the Bohai Bay based on an SSR marker显示文摘Eight microsatellite markers were used to analyze genetic diversity, level of inbreeding, and effective population size of spawner and recaptured populations of Chinese shrimp(Fenneropenaeus chinensis) during stock enhancement in the Bohai Bay in 2013. A total of 254 and 238 alleles were identified in the spawner and recaptured populations, respectively, and the numbers of alleles(N_a) were 8–63 and 6–60, respectively. The numbers of effective alleles(N_e) were 2.52–21.60 and 2.67–20.72, respectively. The polymorphism information content ranged from 0.529 to 0.952. The observed heterozygosity(H_o) values(0.638–0.910 and 0.712–0.927) were lower than the expected heterozygosity(H_e) values(0.603–0.954 and 0.625–0.952), which indicated that the two populations possessed a rich genetic diversity. In 16 tests(2 populations×8 loci), 13 tests deviated from the HardyWeinberg equilibrium. F_(is) values were positive at seven loci and the inbreeding coefficients(F) of the two populations estimated by trio ML were 13.234% and 11.603%, suggesting that there was a relatively high degree of inbreeding. A certain level of inbreeding depression had occurred in the Chinese shrimp population. F_(st) values ranged from 0 to 0.059, with a mean of 0.028, displaying a low level of genetic differentiation in the two populations. Effective population sizes(3 060.2 and 3 842.8) were higher than the minimum number suggested for retaining the evolutionary potential to adapt to new environmental conditions. For enhancement activity in 2014,the ideal number of captured shrimp spawners should have ranged from 7 686 to 19 214 to maintain genetic diversity and effective population size. | 2 | Efficacy of rigosertib, a small molecular RAS signaling disrupter for the treatment of KRAS-mutant colorectal cancer显示文摘Objective:Mutant KRAS,the principal isoform of RAS,plays a pivotal role in the oncogenesis of colorectal cancer by constitutively activating the RAF/MEK/ERK and PI3K/AKT pathways.Effective targeted therapies are urgently needed.We investigated whether rigosertib,a benzyl styryl sulfone RAS signaling disruptor,could selectively kill KRAS-mutant colorectal cancer cells.Methods:CCK-8 was used to determine the cell viability.Patient-derived tumor and cancer cell xenograft models were used to detect the inhibitory efficacy of rigosertib.Flow cytometry was used to evaluate the apoptosis and cell cycle progression.Apoptosis and cell cycle arrest markers were detected by Western blot.DCFH-DA was used to determine the reactive oxygen species.Immunohistochemistry staining and Western blot were performed to characterize RAS signaling markers in colorectal cancer tissues and cells.Results:Rigosertib(RGS)exhibited a cytotoxic effect against colorectal cancer cells,which was greater in KRAS-mutant cells.Furthermore,RGS induced mitotic arrest and oxidative stress-dependent apoptosis in KRAS-mutant DLD1 and HCT116 cells.Besides,RGS disrupted RAS signaling,and the inhibition of RAS/MEK/ERK was independent of cellular oxidative stress.Using patient-derived xenograft models,the response and tumor inhibition of RGS were significantly higher in the KRAS-mutant subgroup,while p-MEK,p-ERK,and p-AKT levels of RGS-treated tumors were significantly decreased.Finally,in a KRAS-mutant,chemotherapy-resistant patient-derived xenograft model,RGS showed a stronger therapeutic effect than the combination standard therapy involving fluoropyrimidine+oxaliplatin/irinotecan+bevacizumab.Conclusions:These data showed that targeting RAS signaling using RGS could be a therapeutic treatment for KRAS-mutant colorectal cancer patients. | Xinyi Zhou Qian Xiao Dongliang Fu Haochen Zhang Yang Tang Jinjie He Yeting Hu Xiangxing Kong Fei Teng Xiangrui Liu Ying Yuan Kefeng Ding | 2022 | Cancer Biology & Medicine2022,19,2: | 2 |
| 3 | Suggestions on the Effective Development of Shale Gas in China显示文摘Shale gas is a practical resource with great potential that could be economically explored and developed in the near future. It has been commercially developed in North America and other places. Featuring a vast distribution, a low abundance,unfavorable reservoir conditions, ultra-low permeability and a fast production decline, shale gas is more difficult to develop than conventional natural gas, as it needs advanced drilling and completion technology to develop. During the early stage of shale gas exploration and development, the United States offered enterprises necessary incentives such as tax reductions and exemptions and subsidies to achieve sustainable shale gas development and encourage technological progresses. | 4 | Impact of Initial Storm Intensity and Size on the Simulation of Tropical Cyclone Track and Western Pacific Subtropical High Extent显示文摘Typhoon Megi, the 13 th typhoon of the 2010 typhoon season, was selected for case study by utilizing the Weather Research and Forecasting(WRF) model. Twelve sensitivity experiments with various initial tropical cyclone(TC) intensities and sizes were conducted to investigate their impacts on the simulation of typhoon track. Interaction between TC and the western Pacific subtropical high(WPSH) was also analyzed to explore the mechanism for the impact on TC track of the initial TC intensity and size. Numerical results indicate that the simulated TC size and TC track are sensitive to initial TC intensity and size. Stronger initial TC intensity and larger initial TC size often lead to larger simulated TC size and make TC turn northward earlier. Further analysis suggests that, with the increase of initial TC intensity and size, more air mass enters into the TC region, which subsequently reduces the extent of WPSH.As a result, the steering flow changes significantly and eventually causes the TC to turn northward earlier. | 5 | Measurement and influencing factors of urban traffic ecological resilience in developing countries: A case study of 31 Chinese cities显示文摘An urban traffic ecosystem is a spatial structure composed of air,population,vehicles,roads,green spaces,and regions.Traffic ecological resilience is a critical issue in high-quality urban development.From the perspective of system optimization,it is important to study the level of urban traffic ecological resilience and analyze its influencing factors.In this study,we evaluated traffic ecological resilience,characterized its spatio-temporal differentiation,and explored its influencing factors by constructing a system of urban traffic ecological resilience and by analyzing the environmental protection and urban construction data in 31 Chinese cities during 2011-2018.By conducting Kernel density analysis,standard deviation ellipse,comprehensive weight determination,panel data regression analysis,andχ2test,we found that traffic ecological resilience was low on the whole and exhibited the temporal trend of“decreasing first and then increasing”and the spatial characteristic of“high in the east,second in the middle,and low in the west”.The cities with high traffic ecological resilience density values were located in Southeast China and tended to move from northwest to southeast.Governance capability,market activity,technological innovation capability,opening degree,and financial resources had significant effects on urban traffic ecological resilience.Finally,we gave some suggestions for improving the urban traffic ecological resilience in Chinese cities as well as other developing countries in the world. | HU Xiwu SU Yunqing REN Kefeng SONG Fang XUE Ruixiang | 2021 | Regional Sustainability2021,2,3: | 1 |

| 8 | Junction plakoglobin regulates and destabilizes HIF2α to inhibit tumorigenesis of renal cell carcinoma显示文摘Background:Increased hypoxia-inducible factor 2α(HIF2α)activation is a common event in clear cell renal cell carcinoma(ccRCC)progression.However,the function and underlying mechanism of HIF2α in ccRCC remains uninvestigated.We conducted this study to access the potential link between junction plakoglobin(JUP)and HIF2αin ccRCC.Methods:Affinity purification and mass spectrometry(AP-MS)screening,glutathione-s-transferase(GST)pull-down and co-immunoprecipitation(Co-IP)assays were performed to detect the interacting proteins of HIF2α.Quantitative PCR(qPCR)and Western blotting were used to detect the expression of JUP in human ccRCC samples.Luciferase reporter assays,chromatin immunoprecipitation(ChIP),cycloheximide chase assays,and ubiquitination assays were conducted to explore the regulation of JUP on the activity of HIF2α.Cell Counting Kit-8(CCK-8)assays,colony formation assays,transwell assays,and xenograft tumor assays were performed to investigate the effect of JUP knockdown or overexpression on the tumorigenicity of renal cancer cells.Results:We identified JUP as a novel HIF2α-binding partner and revealed an important role of JUP in recruiting von Hippel-Lindau(VHL)and histone deacetylases 1/2(HDAC1/2)to HIF2α to regulate its stability and transactivation.JUP knockdown promoted and overexpression suppressed the tumorigenicity of renal cell carcinoma in vitro and in vivo.Importantly,the low expression of JUP was found in clinical ccRCC samples and correlated with enhanced hypoxia scores and poor treatment outcomes.Conclusion:Taken together,these data support a role of JUP in modulating HIF2α signaling during ccRCC progression and identify JUP as a potential therapeutic target. | Ke Chen Jin Zeng Yi Sun Wei Ouyang Gan Yu Hui Zhou Yangjun Zhang Weimin Yao Wei Xiao Junhui Hu Jinchun Xing Kefeng Xiao Lily Wu Zhiqiang Chen Zhangqun Ye Hua Xu | 2021 | Cancer Communications2021,41,4: | 1 |

| 13 | Hierarchical Capillary Coating to Biofunctionlize Drug-Eluting Stent for Improving Endothelium Regeneration显示文摘The drug-eluting stent(DES)has become one of the most successful and important medical devices for coronary heart disease,but yet suffers from insufficient endothelial cell(EC)growth and intima repair,eventually leading to treatment failure.Although biomacromolecules such as vascular endothelial growth factor(VEGF)would be promising to promote the intima regeneration,combining hydrophilic and vulnerable biomacromolecules with hydrophobic drugs as well as preserving the bioactivity after harsh treatments pose a huge challenge.Here,we report on a design of hierarchical capillary coating,which composes a base solid region and a top microporous region for incorporating rapamycin and VEGF,respectively.The top spongy region can guarantee the efficient,safe,and controllable loading of VEGF up to 1μg/cm^(2) in 1 minute,providing a distinctive real-time loading capacity for saving the bioactivity.Based on this,we demonstrate that our rapamycin-VEGF hierarchical coating impressively promoted the competitive growth of endothelial cells over smooth muscle cells(ratio of EC/SMC~25)while relieving the adverse impact of rapamycin to ECs.We further conducted the real-time loading of VEGF on stents and demonstrate that the hierarchical combination of rapamycin and VEGF showed remarkable endothelium regeneration while maintaining a very low level of in-stent restenosis.This work paves an avenue for the combination of both hydrophobic and hydrophilic functional molecules,which should benefit the next generation of DES and may extend applications to diversified combination medical devices. | Jing Wang Yunfan Xue Jun Liu Mi Hu He Zhang Kefeng Ren Yunbing Wang Jian Ji | 2020 | Research2020,,1: | 1 |
| 14 | The Impact of Satellite Radiance Data Assimilation within a Frequently Updated Regional Forecast System Using a GSI-based Ensemble Kalman Filter显示文摘A regional ensemble Kalman filter (EnKF) data assimilation (DA) and forecast system was recently established based on the Gridpoint Statistical Interpolation (GSI) analysis system. The EnKF DA system was tested with continuous threehourly updated cycles followed by 18-h deterministic forecasts from every three-hourly ensemble mean analysis. Initial tests showed negative to neutral impacts of assimilating satellite radiance data due to the improper bias correction procedure. In this study, two bias correction schemes within the established EnKF DA system are investigated and the impact of assimilating additional polar-orbiting satellite radiance is also investigated. Two group experiments are conducted. The purpose of the first group is to evaluate the bias correction procedure. Two online bias correction methods based on GSI 3DVar and EnKF algorithms are used to assimilate AMSU-A radiance data. Results show that both variational and EnKF-based bias correction procedures effectively reduce the observation and background radiance differences, achieving positive impacts on forecasts. With proper bias correction, we assimilate full radiance observations including AMSU-A, AMSU-B, AIRS, HIRS3/4, and MHS in the second group. The relative percentage improvements (RPIs) for all forecast variables compared to those without radiance data assimilation are mostly positive, with the RPI of upper-air relative humidity being the largest. | 16 | Glypican-3 versus alpha-fetoprotein as a biomarker for hepatocellular carcinoma: a diagnostic meta-analysis显示文摘Objective:To assess the diagnostic accuracy of serum GPC3 versus alpha-fetoprotein(AFP)for HCC by using the method of system review.Methods:PubMed and EMBASE were searched from its inception to 20,April 2014 for studies that compared diagnostic accuracy of serum GPC3 with AFP for HCC.Sensitivity,specificity and other measures were pooled using random-effects models.Summary receiver operating characteristic curve analysis was used to summarize the overall test performance.Results:Fourteen studies were included in this meta-analysis.Summary estimates for serum GPC3 and AFP in diagnosing HCC were as follows:sensitivity,69%(95%confidence interval(CI),56-80%)vs.60%(95%CI,50-69%);specificity,91%(95%CI,76-97%)vs.92%(95%CI,84-98%);diagnostic odds ratio(DOR),22(95%CI,6-83)vs.18(95%CI,8-41);and area under sROC,0.85(95%CI,0.81-0.88)vs.0.80(95%CI,0.76-0.83).The pooled sensitivity and specificity for(GPC3+AFP)and AFP were:sensitivity 80%(95%CI,75-85%)vs.64%(95%CI,53-73%)and specificity 86%(95%CI,74-93%)vs.96%(95%CI,86-99%).A significant heterogeneity was found among the ten studies,and meta-regression and subgroup meta-analysis suggested that race and assay type were probably responsible for the heterogeneity.Conclusions:Serum GPC3 may be a promising diagnostic marker of HCC and it was helpful for early detection of primary hepatocellular carcinoma when combined with AFP.More studies for specific race of patients,and using certain methods for detecting GPC3 are required to further confirm the diagnostic value of GPC3 for HCC. | Weiyu FU Hongyu LU Li LI Kefeng WU Yanping LI Yi LIU Jianli HU | 2015 | BIOCELL2015,39,2: | 0 |
