4China land soil moisture EnKF data assimilation based on satellite remote sensing data显示文摘Soil moisture plays an important role in land-atmosphere interactions. It is an important geophysical parameter in research on climate, hydrology, agriculture, and forestry. Soil moisture has important climatic effects by influencing ground evapotranspi ration, runoff, surface reflectivity, surface emissivity, surface sensible heat and latent heat flux. At the global scale, the extent of its influence on the atmosphere is second only to that of sea surface temperature. At the terrestrial scale, its influence is even greater than that of sea surface temperatures. This paper presents a China Land Soil Moisture Data Assimilation System (CLSMDAS) based on EnKF and land process models, and results of the application of this system in the China Land Soil Moisture Data Assimilation tests. CLSMDAS is comprised of the following components: 1) A land process mo del—Community Land Model Version 3.0 (CLM3.0)—developed by the US National Center for Atmospheric Research (NCAR); 2) Precipitation of atmospheric forcing data and surface-incident solar radiation data come from hourly outputs of the FY2 geostationary meteorological satellite; 3) EnKF (Ensemble Kalman Filter) land data assimilation method; and 4) Observa tion data including satellite-inverted soil moisture outputs of the AMSR-E satellite and soil moisture observation data. Results of soil moisture assimilation tests from June to September 2006 were analyzed with CLSMDAS. Both simulation and assimila tion results of the land model reflected reasonably the temporal-spatial distribution of soil moisture. The assimilated soil mois ture distribution matches very well with severe summer droughts in Chongqing and Sichuan Province in August 2006, the worst since the foundation of the People’s Republic of China in 1949. 62018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents显示文摘Syncope belongs to the transient loss of consciousness(TLOC), characterized by a rapid onset, short duration, and spontaneous complete recovery. It is common in children and adolescents, accounting for 1% to 2% of emergency department visits.Recurrent syncope can seriously affect children's physical and mental health, learning ability and quality of life and sometimes cardiac syncope even poses a risk of sudden death. The present guideline for the diagnosis and treatment of syncope in children and adolescents was developed for guiding a better clinical management of pediatric syncope. 8Molecular and Biochemical Evidence for Phenylpropanoid Synthesis and Presence of Wall-linked Phenolics in Cotton Fibers显示文摘The mature cotton (Gossypium hirsutum L.) fiber is a single cell with a typically thickened secondary cell wall. The aim of this research was to use molecular, spectroscopic and chemical techniques to investigate the possible occurrence of previously overlooked accumulation of phenolics during secondary cell wall formation in cotton fibers. Relative quantitative reverse transcription-polymerase chain reaction analysis showed that GhCAD6 and GhCAD1 were predominantly expressed among seven gene homologs, only GhCAD6 was up-regulated during secondary wall formation in cotton fibers. Phylogenic analysis revealed that GhCAD6 belonged to Class I and was proposed to have a major role in monolignol biosynthesis, and GhCAD1 belonged to Class Ⅲ and was proposed to have a compensatory mechanism for monolignol biosynthesis. Amino acid sequence comparison showed that the cofactor binding sites of GhCADs were highly conserved with high similarity and identity to bona fide cinnamyl alcohol dehydrogenases. The substrate binding site of GhCAD1 is different from GhCAD6. This difference was confirmed by the different catalytic activities observed with the enzymes. Cell wall auto-fluorescence, Fourier transform infrared spectroscopy (FTIR), high-performance liquid chromatography (HPLC) and chemical analyses confirmed that phenolic compounds were bound to the cell walls of mature cotton fibers. 10Morphological study on colonic pathology in ulcerative colitis treated by moxibustion显示文摘AIM To observe the therapeutic effect ofmoxibustion on ulcerative colitis and itsinfluence on the colonic mucosal morphology.METHODS Forty-six patients with ulcerativecolitis were randomly divided into themoxibustion with herbal medicine underneathgroup and the western medicine group.Thirtypatients were treated with the abovemoxibustion and 16 patients with Salicylayefapyridine(SASP).The colonic mucosa of 13patients in the moxibustion group was observedby colonoscopy before and after the treatment.Mucin was also analyzed by H.E and AB-PASstaining.RESULTS Seventeen patients were clinicallycured,12 were improved and 1 unchanged in themoxibustion group.In the control group,5patients were clinically cured,7 improved and 4unchanged.Thirteen patients with active UCwere taken as the subjects for histopathologicanalysis in this study.The colonic mucosallesions were remarkably improved and thecharacteristic of the mucin also changed.Inmost sections,the chronic inflammation ofmucosa was geatly ameliorated(P<0.01).Theinflammatory cell infiltratation much decreased and neutrophils,disapeared in most sections(P<0.001).The goblet cells significantlyincreased(P<0.001);crypt paracrypt abscessor mucosal ulceration was seen(P<0.001).CONCLUSION The rate of cure of ulcerative colitis by moxibustion with herbal medicine beneath is superior to that by SASP. 11Oxymatrine liposome attenuates hepatic fibrosis via targeting hepatic stellate cells显示文摘AIM:To investigate the potential mechanism of ArgGly-Asp(RGD) peptide-labeled liposome loading oxymatrine(OM) therapy in CCl 4-induced hepatic fibrosis in rats.METHODS:We constructed a rat model of CCl 4 induced hepatic fibrosis and treated the rats with different formulations of OM.To evaluate the antifibrotic effect of OM,we detected levels of alkaline phosphatase,hepatic histopathology(hematoxylin and eosin stain and Masson staining) and fibrosis-related gene expression of matrix metallopeptidase(MMP)-2,tissue inhibitor of metalloproteinase(TIMP)-1 as well as type Ⅰ procollagen via quantitative real-time polymerase chain reaction.To detect cell viability and apoptosis of hepatic stellate cells(HSCs),we performed 3-(4,5)-dimethylthiahiazo(-z-y1)-3,5-diphenytetrazoliumromide assay and flow cytometry.To reinforce the combination of oxymatrine with HSCs,we constructed fluorescein-isothiocyanate-conjugated Arg-Gly-Asp peptide-labeled liposomes loading OM,and its targeting of HSCs was examined by fluorescent microscopy.RESULTS:OM attenuated CCl 4-induced hepatic fibrosis,as defined by reducing serum alkaline phosphatase(344.47 ± 27.52 U/L vs 550.69 ± 43.78 U/L,P < 0.05),attenuating liver injury and improving collagen deposits(2.36% ± 0.09% vs 7.70% ± 0.60%,P < 0.05) and downregulating fibrosis-related gene expression,that is,MMP-2,TIMP-1 and type Ⅰ procollagen(P < 0.05).OM inhibited cell viability and induced apoptosis of HSCs in vitro.RGD promoted OM targeting of HSCs and enhanced the therapeutic effect of OM in terms of serum alkaline phosphatase(272.51 ± 19.55 U/L vs 344.47 ± 27.52 U/L,P < 0.05),liver injury,collagen deposits(0.26% ± 0.09% vs 2.36% ± 0.09%,P < 0.05) and downregulating fibrosis-related gene expression,that is,MMP-2,TIMP-1 and type Ⅰ procollagen(P < 0.05).Moreover,in vitro assay demonstrated that RGD enhanced the effect of OM on HSC viability and apoptosis.CONCLUSION:OM attenuated hepatic fibrosis by inhibiting viability and inducing apoptosis of HSCs.The RGD-labeled formulation enhanced the targeting efficiency for HSCs and the therapeutic effect.Ning-Li Chai Qiang Fu Hui Shi Chang-Hao Cai Jun Wan Shi-Ping Xu Ben-Yan Wu 2012World Journal of Gastroenterology2012,18,31:27

13Mesenchymal stem cells alleviate TNBS-induced colitis by modulating inflammatory and autoimmune responses显示文摘AIM: To investigate the potential therapeutic effects of mesenchymal stem cells (MSCs) in inflammatory bowel disease (IBD), we transplanted MSCs into an experimental model of IBD. METHODS: A rectal enema of trinitrobenzene sulfonic acid (TNBS) (100 mg/kg body weight) was administered to female BALB/c mice. Bone marrow mesenchymal stem cells (BMSCs) were derived from male green fluorescent protein (GFP) transgenic mice and were transplanted intravenously into the experimental animals after disease onset. Clinical activity scores and histological changes were evaluated. GFP and Sex determining region Y gene (SRY ) expression were used for cell tracking. Ki67 positive cells and Lgr5-expressing cells were determined to measure proliferative activity. Inflammatory response was determined by mea-suring the levels of different inflammatory mediators in the colon and serum. The inflammatory cytokines included tumor necrosis factor-α (TNF-α), interferon-γ (IFN-γ), interleukin-2 (IL-2), IL-6, IL-17, IL-4, IL-10, and transforming growth factor (TGF-β). Master regulators of Th1 cells (T-box expressed in T cells, T-bet), Th17 cells (retinoid related orphan receptor gamma(t), RORγt), Th2 cells (GATA family of transcription factors 3, GATA3) and regulatory T cells (forkhead box P3, Foxp3) were also determined. RESULTS: Systemic infusion of GFP-BMSCs ameliorated the clinical and histopathologic severity of colitis, including body weight loss, diarrhea and inflammation, and increased survival (P < 0.05). The cell tracking study showed that MSCs homed to the injured colon. MSCs promoted proliferation of intestinal epithelial cells and differentiation of intestinal stem cells (P < 0.01). This therapeutic effect was mainly mediated by downregulation of both Th1-Th17-driven autoimmune and inflammatory responses (IL-2, TNF-α, IFN-γ, T-bet; IL-6, IL-17, RORγt), and by up-regulation of Th2 activities (IL-4, IL-10, GATA-3) (P < 0.05). MSCs also induced activated CD4 + CD25 + Foxp3 + regulatory T cells (TGF-β, IL-10, Foxp3) with a suppressive capacity on Th1-Th17 effecter responses and promoted Th2 differentiation in vivo (P < 0.05). 14Effect of Lingguizhugan decoction on myocardial Nuclear factor kappa B protein expression in rats with chronic heart failure显示文摘OBJECTIVE: To investigate the effect of Lingguizhugan decoction (LGZGD) on changes of cardiac structure and function, and its putative mechanism of action, by investigating mRNA and protein expression of myocardial nuclear factor kappa B(NF-κB), and the plasma content of NF-κB in rats with chronic heart failure.METHODS: The chronic heart failure (CHF) model in rats was induced by coronary artery ligation.Sham operation was performed in control rats. Six weeks after the procedure, rats were randomly classified into the various treatment groups: model CHF, Captopril (4.4 mg/kg), low LGZGD dose (2.1 g/kg), medium LGZGD dose (4.2 g/kg), and high LG-ZGD dose (8.4 g/kg). Treatments continued for 4 consecutive weeks. Changes of hemodynamic indices were observed by the PowerLab data acquisition and analysis system. Morphological changes of myocardium were observed by hematoxylin and eosin staining, and Masson staining. The mRNA and protein expression of myocardial NF-κB were detected by reverse transcription-polymerase chain reaction and western blotting, respectively. The plasma content of NF-κB was detected by enzyme-linked immuno-sorbent assay.RESULTS: CHF rats showed significant dysfunction in hemodynamic indices and in cardiac structure.Compared with the sham operation group, mRNA expression of myocardial NF-κB and plasma content of NF-κB of the model group was significantly increased. All three doses of LGZGD, and Captopril,improved the hemodynamic dysfunction, and inhibited the change of cardiac structure while significantly improving the survival rate. 15Evaluation of the International Vehicle Emission (IVE) model with on-road remote sensing measurements显示文摘International Vehicle Emissions (IVE) model funded by U.S. Environmental Protection Agency (USEPA) is designed to estimate emissions from motor vehicles in developing countries. In this study, the IVE model was evaluated by utilizing a dataset available from the remote sensing measurements on a large number of vehicles at five different sites in Hangzhou, China, in 2004 and 2005. Average fuel-based emission factors derived from the remote sensing measurements were compared with corresponding emission factors derived from IVE calculations for urban, hot stabilized condition. The results show a good agreement between the two methods for gasoline passenger cars' HC emission for all IVE subsectors and technology classes. In the case of CO emissions, the modeled results were reasonably good, although systematically underestimate the emissions by almost 12%-50% for different technology classes. However, the model totally overestimated NOx emissions. The IVE NOx emission factors were 1.5-3.5 times of the remote sensing measured ones. The IVE model was also evaluated for light duty gasoline truck, heavy duty gasoline vehicles and motor cycles. A notable result was observed that the decrease in emissions from technology class State II to State I were overestimated by the IVE model compared to remote sensing measurements for all the three pollutants. 16Numerical simulation of GPS observed clockwise rotation around the eastern Himalayan syntax in the Tibetan Plateau显示文摘From Global Position System(GPS) measurements,there is a clockwise rotation around the eastern Himalayan syntax in the Tibetan Plateau.This phenomenon is difficult to be interpreted by simple two-dimensional modeling from a geodynamic point of view.Because of the extremely thick crust and the lower crust with relatively high temperature in the Tibetan Plateau,the lithospheric rheology in Tibet and surrounding areas present a complex structure.In general,the tectonic structure of the Tibetan Plateau consists of brittle upper crust,ductile lower crust,high viscosity lithospheric upper mantle,and low viscosity asthenosphere,the same as the case in many other continental regions.However,the lower crust in the Tibetan Plateau is much more ductile with a lower viscosity than those of its sur-roundings at the same depth,and the effective viscosity is low along the collision fault zone.In this study,we construct a three-dimensional Maxwell visco-elastic model in spherical coordinate system,and simulate the deformation process of the Tibetan Plateau driven by a continuous push from the Indian plate.The results show that the existence of the soft lower crust under the plateau makes the entire plateau uplift as a whole,and the Himalayas and the eastern Himalayan syntax uplift faster.Since the lower crust of surrounding blocks is harder except in the southeastern corner where the high-temperature material is much softer and forms an exit channel for material transfer,after the whole plateau reaches a certain height,the lower crustal and upper mantle material begins to move eastward or southeastward and drag the upper crust to behave same way.Thus,from the macroscopic point of view,a relative rigid motion of the plateau with a clockwise rotation around the eastern Himalayan syntax is developed.CAO JianLing SHI YaoLin ZHANG Huai WANG Hui 2009Chinese Science Bulletin2009,54,8:23

18Acacetin protects against cerebral ischemia-reperfusion injury via the NLRP3 signaling pathway显示文摘Acacetin(5,7-dihydroxy-4′-methoxyflavone), a potential neuroprotective agent, has an inhibitory effect on lipopolysaccharide-induced neuroinflammatory reactions. However, whether acacetin has an effect on inflammatory corpuscle 3(NLRP3) after cerebral ischemia-reperfusion injury has not been fully determined. This study used an improved suture method to establish a cerebral ischemia-reperfusion injury model in C57BL/6 mice. After ischemia with middle cerebral artery occlusion for 1 hour, reperfusion with intraperitoneal injection of 25 mg/kg of acacetin(acacetin group) or an equal volume of saline(0.1 mL/10 g, middle cerebral artery occlusion group) was used to investigate the effect of acacetin on cerebral ischemia-reperfusion injury. Infarct volume and neurological function scores were determined by 2,3,5-triphenyltetrazolium chloride staining and the Zea-Longa scoring method. Compared with the middle cerebral artery occlusion group, neurological function scores and cerebral infarction volumes were significantly reduced in the acacetin group. To understand the effect of acacetin on microglia-mediated inflammatory response after cerebral ischemia-reperfusion injury, immunohistochemistry for the microglia marker calcium adapter protein ionized calcium-binding adaptor molecule 1(Iba1) was examined in the hippocampus of ischemic brain tissue. In addition, tumor necrosis factor-α, interleukin-1β, and interleukin-6 expression in ischemic brain tissue of mice was quantified by enzyme-linked immunosorbent assay. Expression of Iba1, tumor necrosis factor-α, interleukin-1β and interleukin-6 was significantly lower in the acacetin group compared with the middle cerebral artery occlusion group. Western blot assay results showed that expression of Toll-like receptor 4, nuclear factor kappa B, NLRP3, procaspase-1, caspase-1, pro-interleukin-1β, and interleukin-1β were significantly lower in the acacetin group compared with the middle cerebral artery occlusion group.
