3The combination of chidamide with the CHOEP regimen in previously untreated patients with peripheral T-cell lymphoma: a prospective, multicenter, single arm, phase 1b/2 study显示文摘Objective:To assess the efficacy and safety of the novel histone deacetylase inhibitor,chidamide,in combination with cyclophosphamide,doxorubicin,vincristine,etoposide,and prednisone(Chi-CHOEP)for untreated peripheral T-cell lymphoma(PTCL).Methods:A prospective,multicenter,single arm,phase 1 b/2 study was conducted.A total of 128 patients with untreated PTCL(18–70 years of age)were enrolled between March 2016 and November 2019,and treated with up to 6 cycles with the Chi-CHOEP regimen.In the phase 1 b study,3 dose levels of chidamide were evaluated and the primary endpoint was determination of the maximumtolerated dose and recommended phase 2 dose(RP2 D).The primary endpoint of the phase 2 study was 2-year progression-free survival(PFS).Results:Fifteen patients were enrolled in the phase 1 b study and the RP2 D for chidamide was determined to be 20 mg,twice a week.A total of 113 patients were treated at the RP2 D in the phase 2 study,and the overall response rate was 60.2%,with a complete response rate of 40.7%.At a median follow-up of 36 months,the median PFS was 10.7 months,with 1-,2-,and 3-year PFS rates of 49.9%,38.0%,and 32.8%,respectively.The Chi-CHOEP regimen was well-tolerated,with grade 3/4 neutropenia occurring in approximately two-thirds of the patients.No unexpected adverse events(AEs)were reported and the observed AEs were manageable.Conclusions:This large cohort phase 1 b/2 study showed that Chi-CHOEP was well-tolerated with modest efficacy in previously untreated PTCL patients.Wei Zhang Liping Su Lihong Liu Yuhuan Gao Quanshun Wang Hang Su Yuhuan Song Huilai Zhang Jing Shen Hongmei Jing Shuye Wang Xinan Cen Hui Liu Aichun Liu Zengjun Li Jianmin Luo Jianxia He Jingwen Wang O.A.O’Connor Daobin Zhou 2021Cancer Biology & Medicine2021,18,3:4

5Shale pore characteristics of Shahejie Formation: Implication for pore evolution of shale oil reservoirs in Dongying sag, north China显示文摘The pore characteristics of shale reservoirs in the lower submember of Member 3 to upper submember of Member 4 of Shahejie Formation in Dongying sag are analyzed,influences of mineral content and organic matter content on porosity and pore size are also investigated,and through the diagenetic thermal simulation experiment,the main pore evolution is further discussed.The results show that the pore structure of shale reservoirs is complex,the micron-nanometer pores can storage liquid hydrocarbons,and the free-phase crude oil is mainly distributed in intergranular dissolution pores of calcite,recrystallized intergranular pores,intergranular shrinkage fractures of clay mineral which have large pore size.Framework minerals and organic matter content directly influence porosity and pore size of shale reservoirs,relationship between porosity and content of felsic mineral as well as content of organic matter content is linear and positive,while relationship between content of carbonated mineral is negative.At the buried depth from 2500 to 3500 m,concentration of organic acid from hydrocarbon generation and expulsion of organic matter,increasing range of pressure coefficient,are well corresponding to highporosity intervals;pore formation in shale oil reservoirs are almost controlled by diagenetic evolution of clay minerals;framework storage spaces formed by carbonate grain crystals as well as intergranular and intergranular dissolution pores of carbonate increases porosity of shale oil reservoirs;local increase of porosity at the depth of 3500e3800 m is mainly caused by coupling of hydrocarbon-generating overpressure and dissolution,and size,distribution and connectivity of pores are enhanced obviously.Shun Zhang Huimin Liu Min Wang Xinjin Liu Huilai Liu Youshu Bao Weiqing Wang Runze Li Xin Luo Zhengwei Fang 2019Petroleum Research2019,4,2:2

7Targeting EZH1/2 induces cell cycle arrest and inhibits cell proliferation through reactivation of p57CDKN1C and TP53INP1 in mantle cell lymphoma显示文摘Objective: To explore the effect of dysregulation of epigenetic regulator EZH1 and EZH2 on the proliferation in MCL and the underlying mechanisms.Methods: In this study, we elucidated the role of EZH1 and EZH2 overexpression by immunohistochemistry and correlated them to clinical outcome in 41 MCL patients.Quantitative real-time PCR and Western blot were applied to confirm the level of EZH1 and EZH2 in well-characterized MCL cell lines which were compared to those of na?ve B cells.Then we manipulated the expression of EZH1 and EZH2 in MCL cells using CRISPR/Cas9 system to directly investigate their functional roles in MCL.We also evaluated the effect of two small molecule selective inhibitors, EPZ005687 and UNC1999, on MCL cell proliferation, cell cycle distribution and apoptosis in vitro.Finally, we performed RNA-sequencing(RNA-Seq) and Chromatin immunoprecipitation(ChIP) assay to further gain insight into the underlying molecular mechanisms.Results: We found that EZH2 protein is overexpressed in approximately half of this cohort of MCL cases.More importantly, the overexpression of EZH2 is associated with poor OS in the patients.Nevertheless, simple EZH2 depletion in vitro has little impact on the viability of MCL cells, predominantly because of the consequent up-regulation of EZH1.Consistently, UNC1999, a dual EZH1/2 inhibitor, unlike the EZH2 selective inhibitor EPZ005687, exerts a potent inhibitory effect on MCL cells.Furthermore, we discover CDKN1C and TP53 INP1 as the two important cell cycle regulators, the expression of which are repressed by EZH1/2 mediated epigenetic regulation and are restored by EZH1/2 dual inhibition.Conclusions: Our study suggests that EZH2 participates in the pathogenesis of MCL which may serve as a potential biomarker for prognosis prediction.The dual inhibition of EZH1/2 is a promising therapeutic strategy for MCL.Wei Li Chengfeng Bi Yating Han Tian Tian Xianhuo Wang Huijing Bao Xiaoying Xu Xuhan Zhang Lu Liu Weiwei Zhang Hai Gao Huaqing Wang Huilai Zhang Bin Meng Xi Wang Kai Fu 2019Cancer Biology & Medicine2019,16,3:2

11Global stability of a SEIR epidemic model with infectious force in latent period and infected period under discontinuous treatment strategy显示文摘We consider a SEIR epidemic model with infectious force in latent period and infected period under discontinuous treatment.The treatment rate has at most a finite number of jump discontinuities in every compact interval.By using Lyapunov theory for discontinuous differential equations and other techniques on non-smooth analysis,the basic reproductive number Ro is proved to be a sharp threshold value which completely determines the dynamics of the model.If Ro<1,then there exists a disease-free equilibrium which is globally stable.If Ro>1,the disease-free equilibrium becomes unstable and there exists an endemic equilibrium which is globally stable.We discuss that the disease will die out in a finite time which is impossible for the corresponding SEIR model with continuous treatment.Furthermore,the numerical simulations indicate that strengthening treatment measure after infective individuals reach some level is beneficial to disease control.Yanjun Zhao Huilai Li Wenxuan Li Yang Wang 2021International Journal of Biomathematics2021,14,5:0
12Immunosuppressive tumor microenvironment contributes to tumor progression in diffuse large B-cell lymphoma upon anti-CD19 chimeric antigen receptor T therapy显示文摘Anti-CD19 chimeric antigen receptor(CAR)-T cell therapy has achieved 40%–50%long-term complete response in relapsed or refractory diffuse large B-cell lymphoma(DLBCL)patients.However,the underlying mechanism of alterations in the tumor microenvironments resulting in CAR-T cell therapy failure needs further investigation.A multi-center phase I/II trial of anti-CD19 CD28z CAR-T(FKC876,ChiCTR1800019661)was conducted.Among 22 evaluable DLBCL patients,seven achieved complete remission,10 experienced partial remissions,while four had stable disease by day 29.Single-cell RNA sequencing results were obtained from core needle biopsy tumor samples collected from long-term complete remission and early-progressed patients,and compared at different stages of treatment.M2-subtype macrophages were significantly involved in both in vivo and in vitro anti-tumor functions of CAR-T cells,leading to CAR-T cell therapy failure and disease progression in DLBCL.Immunosuppressive tumor microenvironments persisted before CAR-T cell therapy,during both cell expansion and disease progression,which could not be altered by infiltrating CAR-T cells.Aberrant metabolism profile of M2-subtype macrophages and those of dysfunctional T cells also contributed to the immunosuppressive tumor microenvironments.Thus,our findings provided a clinical rationale for targeting tumor microenvironments and reprogramming immune cell metabolism as effective therapeutic strategies to prevent lymphoma relapse in future designs of CAR-T cell therapy.Zixun Yan Li Li Di Fu Wen Wu Niu Qiao Yaohui Huang Lu Jiang Depei Wu Yu Hu Huilai Zhang Pengpeng Xu Shu Cheng Li Wang Sahin Lacin Muharrem Muftuoglu Weili Zhao 2023Frontiers of Medicine2023,17,4:0
13Multidisciplinary team for the diagnosis and treatment of 2cases of primary intestinal yolk sac tumor显示文摘Extragonadal primary yolk sac tumor of the intestinal tract origin is exceedingly rare. Through a multiple disciplinary team, the diagnosis and treatment of primary intestinal yolk sac tumor were further defined. We report 2 such cases with detailed histologic and immunohistochemical analysis. The two patients were a 7-year-old girl and a 29-year-old woman. Both of them preoperatively had an elevated serum alpha fetoprotein(AFP) level(≥ 1,210 ng/mL). The tumors are located in the intestine and imaging examination indicated the rectum as the primary site. Grossly the mass was grey-white and crisp texture. Microscopic examination featured reticular, microcystic, macrocystic, papillary, solid, and some glandular patterns. Immunohistochemically,tumor cells of both cases were positive for SALL4, AFP, pan-cytokeratin(AE1/AE3), and glypican-3. Simultaneously, a stain for EMA, OCT4, CD30, HCG, vimentin and CK20 were negative in all 2 neoplasms. 14Optimal Location and Sizing ofMulti-Resource Distributed Generator Based onMulti-Objective Artificial Bee Colony Algorithm显示文摘Distribution generation(DG)technology based on a variety of renewable energy technologies has developed rapidly.A large number of multi-type DG are connected to the distribution network(DN),resulting in a decline in the stability of DN operation.It is urgent to find a method that can effectively connect multi-energy DG to DN.photovoltaic(PV),wind power generation(WPG),fuel cell(FC),and micro gas turbine(MGT)are considered in this paper.A multi-objective optimization model was established based on the life cycle cost(LCC)of DG,voltage quality,voltage fluctuation,system network loss,power deviation of the tie-line,DG pollution emission index,and meteorological index weight of DN.Multi-objective artificial bee colony algorithm(MOABC)was used to determine the optimal location and capacity of the four kinds of DG access DN,and compared with the other three heuristic algorithms.Simulation tests based on IEEE 33 test node and IEEE 69 test node show that in IEEE 33 test node,the total voltage deviation,voltage fluctuation,and system network loss of DN decreased by 49.67%,7.47%and 48.12%,respectively,compared with that without DG configuration.In the IEEE 69 test node,the total voltage deviation,voltage fluctuation and system network loss of DN in the MOABC configuration scheme decreased by 54.98%,35.93%and 75.17%,respectively,compared with that without DG configuration,indicating that MOABC can reasonably plan the capacity and location of DG.Achieve the maximum trade-off between DG economy and DN operation stability.Qiangfei Cao Huilai Wang Zijia Hui Lingyun Chen 2024Energy Engineering2024,121,2:0
15Expression of Preprotachykinin-I (PPT-I), Neurokinin-1 (NK-1) and Neurokinin-2 (NK-2) in Breast Cancer Cells Improves Tumor Cell Survival in Bone Marrow in the Early Stage of Metastasis显示文摘OBJECTIVE To study the potential relationship between theexpression of PPT-I, NK-1, NK2 and the development of breastcancer cells in bone marrow stroma and to provide evidence ofpotential molecular mechanisms of bone metastasis in early stageof breast cancer patients.METHODS The cocultures of breast cancer cell line T-47D andmarrow-derived mesenchymal stem cells (MSC) were establishedwith equal numbers. T-47D cells were separated from the coculturesystem at 48 h and 96 h after coculture by MACS magnetic cellsorting (MicroBeads). The expression of PPT-I, NK-1, NK-2 inT-47D was then examined before and after coculture by real-timePCR and by Western blot. Alterations in cellular ultrastructureof T-47D cells were detected before and after coculture underelectron microscope. Finally, changes in cell cycle distributionwere examined by flow cytometry, and growth curves from beforeand after coculture were drawn and analyzed.RESULTS Following coculture of T-47D and MSC, the expressionof PPT-I mRNA and protein was significantly upregulated, whilethe expression of NK-1 and NK-2 mRNA and protein was greatlydownregulated. The analysis of cell cycle distribution by flowcytometry showed that the proportion of T-47D during S phasewas increased, and the duration of the G_2/M phase was sharplydecreased. Under electron microscope, we observed that thesynthesis of hereditary material was increased, but the hepatingranules were shown prominent stacking in T-47D cells. Theseresults suggest that although the synthesis of DNA was increased,the proliferation of cells was inhibited after coculture. The cellgrowth curve confirmed the findings from the observation underthe electron microscope and flow cytometry.CONCLUSION Tumor cells could survive through theupregulation in expression of preprotachykinin-I gene duringearly bone metastasis in breast cancer. 16Systematic analysis of DNA polymerases as therapeutic targets in pan-cancers显示文摘Introduction:DNA polymerases are crucial for maintaining genome stability and influencing tumorigenesis.However,the clinical implications of DNA polymerases in tumorigenesis and their potential as anti-cancer therapy targets are not well understood.Methods:We conducted a systematic analysis using TCGA Pan-Cancer Atlas data and Gene Set Cancer Analysis results to examine the expression profiles of 15 DNA polymerases(POLYs)and their clinical correlations.We also evaluated the prognostic value of POLYs by analyzing their expression levels in relation to overall survival time(OS)using Kaplan-Meier survival curves.Additionally,we investigated the correlations between POLY expression and immune cells,DNA damage repair(DDR)pathways,and ubiquitination.Drug sensitivity analysis was performed to assess the relationship between POLY expression and drug response.Results:Our analysis revealed that 14 out of 15 POLYs exhibited significantly distinct expression patterns between tumor and normal samples across most cancer types,except for DNA nucleotidylexotransferase(DNTT).Specifically,POLD1 and POLE showed elevated expression in almost all cancers,while POLQ exhibited high expression levels in all cancer types.Some POLYs showed heightened expression in specific cancer subtypes,while others exhibited low expression.Kaplan-Meier survival curves demonstrated significant prognostic value of POLYs in multiple cancers,including PAAD,KIRC,and ACC.Cox analysis further validated these findings.Alteration patterns of POLYs varied significantly among different cancer types and were associated with poorer survival outcomes.Significant correlations were observed between the expression of POLY members and immune cells,DDR pathways,and ubiquitination.Drug sensitivity analysis indicated an inverse relationship between POLY expression and drug response.Conclusion:Our comprehensive study highlights the significant role of POLYs in cancer development and identifies them as promising prognostic and immunological biomarkers for various cancer types.Additionally,targeting POLYs therapeutically holds promise for tumor immunotherapy.ZHENHUA LI HUILAI LV FAN ZHANG ZIMING ZHU QIANG GUO MINGBO WANG CHAO HUANG LIJUAN CHEN WENPAN ZHANG YUN LI ZIQIANG TIAN 2024BIOCELL2024,48,1:0
