| 1 | Network Pharmacology in Research of Chinese Medicine Formula: Methodology, Application and Prospective显示文摘Chinese medicine(CM) is usually prescribed as CM formula to treat disease. The lack of effective research approach makes it difficult to elucidate the molecular mechanisms of CM formula owing to its complicated chemical compounds. Network pharmacology is increasingly applied in CM formula research in recent years, which is identified suitable for the study of CM formula. In this review, we summarized the methodology of network pharmacology, including network construction, network analysis and network verification. The aim of constructing a network is to achieve the interaction between the bioactive compounds and targets and the interaction between various targets, and then find out and validate the key nodes via network analysis and network verification. Besides, we reviewed the application in CM formula research, mainly including targets discovery, bioactive compounds screening, toxicity evaluation, mechanism research and quality control research. | 2 | Resveratrol Induces Apoptosis and Autophagy in T-cell Acute Lymphoblastic Leukemia Cells by Inhibiting Akt/mTOR and Activating p38-MAPK显示文摘Objective To explore the effects of resveratrol-induced apoptosis and autophagy in T-cell acute lymphoblastic leukemia(T-ALL) cells and potential molecular mechanisms. Methods The anti-proliferation effect of resveratrol-induced, apoptosis and autophagy on T-ALL cells were detected by using MTT test, immunofluorescence, electronic microscope, and flow cytometry, respectively. Western blotting was performed for detecting changes of apoptosis-associated proteins, cell cycle regulatory proteins and state of activation of Akt, mTOR, p70S6K, 4E-BP1, and p38-MAPK. Results Resveratrol inhibited the proliferation and induced apoptosis and autophagy in T-ALL cells in a dose and time-dependent manner. It also induced cell cycle arrest at G0/G1 phase via up regulating cyclin-dependent kinase(CDK) inhibitors p21 and p27 and down regulating cyclin A and cyclin D1. Western blotting revealed that resveratrol significantly decreased the expression of antiapoptotic proteins(Mcl-1 and Bcl-2) and increased the expression of proapoptotic proteins(Bax, Bim, and Bad), and induced cleaved-caspase-3 in a time-dependent manner. Significant increase in ratio of LC3-II/LC3-I and Beclin 1 was also detected. Furthermore, resveratrol induced significant dephosphorylation of Akt, mTOR, p70S6K, and 4E-BP1, but enhanced specific phosphorylation of p38-MAPK which could be blocked by SB203580. When autophagy was suppressed by 3-MA, apoptosis in T-ALL cells induced by resveratrol was enhanced.Conclusion Our findings have suggested that resveratrol induces cell cycle arrest,apoptosis,and autophagy in T-ALL cells through inhibiting Akt/mTOR/p70S6K/4E-BP1 and activating p38-MAPK signaling pathways.Autophagy might play a role as a self-defense mechanism in T-ALL cells treated by resveratrol.Therefore,the reasonable inhibition of autophagy in T-ALL cells may serve as a promising strategy for resveratrol induced apoptosis and can be used as adjuvant chemotherapy for T-ALL. | GE Jiao LIU Yan LI Qiang GUO Xia GU Ling MA Zhi Gui ZHU Yi Ping | 2013 | Biomedical and Environmental Sciences2013,26,11: | 38 |

| 8 | Antibiotic Resistance of Probiotic Strains of Lactic Acid Bacteria Isolated from Marketed Foods and Drugs显示文摘Objective To identify the antimicrobial resistance of commercial lactic acid bacteria present in microbial foods and drug additives by analyzing their isolated strains used for fermentation and probiotics. Methods Antimicrobial susceptibility of 41 screened isolates was tested with disc diffusion and E-test methods after species-level identification. Resistant strains were selected and examined for the presence of resistance genes by PCR. Results Distribution of resistance was found in different species. All isolates were susceptible to chloramphenicol, tetracycline, ampicillin, amoxicillin/clavulanic acid, cephalothin, and imipenem. In addition, isolates resistant to vancomycin, rifampicin, streptomycin, bacitracin, and erythromycin were detected, although the incidence of resistance to these antibiotics was relatively low. In contrast, most strains were resistant to ciprofloxacin, amikacin, trimethoprim/sulphamethoxazole, and gentamycin. The genes msrC, vanX, and dfrA were detected in strains of Enterococcus faecium, Lactobacillus plantarum, Streptococcus thermophilus, and Lactococcus lactis. Conclusion Antibiotic resistance is present in different species of probiotic strains, which poses a threat to food safety. | 9 | Geothermal data analysis at the high-temperature hydrothermal area in Western Sichuan显示文摘The western Sichuan hydrothermal area is located at the northeastern margin of the eastern syntaxis of the Qinghai-Tibet Plateau, which is also the eastern end of the Mediterranean-Himalayan geothermal activity zone. There are 248 warm or hot springs in this area, and 11 have temperatures beyond the local boiling temperature. Most of these hot springs are distributed along the Jinshajiang, Dege-Xiangcheng, Ganzi-Litang, and Xianshuihe faults, forming a NW-SE hydrothermal belt. A geothermal analysis of this high-temperature hydrothermal area is an important basis for understanding the deep geodynamic process of the eastern syntaxis of the Qinghai-Tibet Plateau. In addition, this study offers an a priori view to utilize geothermal resources, which is important in both scientific research and application. We use gravity, magnetic, seismic, and helium isotope data to analyze the crust-mantle heat flow ratio and deep geothermal structure. The results show that the background terrestrial heat flow descends from southwest to northeast. The crustal heat ratio is not more than 60%. The high temperature hydrothermal active is related to crustal dynamics processes. Along the Batang-Litang-Kangding line, the Moho depth increases eastward, which is consistent with the changing Qc/Qm(crustal/mantle heat flow) ratio trend. The geoid in the hydrothermal zone is 4–6 km higher than the surroundings, forming a local 'platform'. The NW-SE striking local tensile stress zone and uplift structure in the upper and middle crust corresponds with the surface hydrothermal active zone. There is an average Curie Point Depth(CPD) of 19.5–22.5 km in Batang, Litang, and Kangding. The local shear-wave(S-wave) velocity is relatively low in the middle and lower crust. The S-wave shows a low velocity trap(Vs<3.2 km s.1) at 15–30 km, which is considered a high-temperature partial melting magma, the crustal source of the hydrothermal active zone. We conclude that the hydrothermal system in this area can be divided into Batang-type and Kangding-type, both of which rely on a crustal heating cycle of atmospheric precipitation and surface water along the fracture zone. | 14 | A minimally invasive alternative for the treatment of nutcracker syndrome using individualized three-dimensional printed extravascular titanium stents显示文摘Background: This study was designed to evaluate the clinical and radiographic outcomes of patients with nutcracker syndrome (NCS) who were treated with three-dimensional printing (3DP) extravascular titanium stents (EVTSs). The 3DP EVTS was expected to release the hypertension of the left renal vein (LRV) produced by its compression between the superior mesenteric artery (SMA) and the aorta without causing any complications. Method: The pre-operative kidney model of each patient was printed out to enable surgical planning. After that, the EVTS was designed based on the LRV’s primitive physiologic structure using computer-aided design software, and each stent was printed out with a precision setting of 20 μm. Seventeen patients who had been suffering from NCS underwent laparoscopic 3DP EVTS placement. The surgical procedure was designed for the placement of EVTS, taking great care in positioning and fixing the stent. Surgical data, which included patient demographic characteristics as well as pre- and post-operative test results, were collected and analyzed. Results: The mean duration of surgery was 75 ± 9 min, and the mean blood loss was 20 ± 5 mL. Computed tomography examinations revealed that the pre- and post-operative angle between the SMA and the aorta ranged from 18.7°± 4.3° to 48.0°± 8.8°(P < 0.05);in patients with left varicocele, the mean diameter of the left spermatic vein ranged from 3.7 ± 0.5 to 1.3 ± 0.2 mm (P < 0.05). Moreover, Doppler ultrasound examinations showed that the peak velocity of blood flow at the hilar area ranged from 12.4 ± 3.3 to 18.5 ± 3.4 cm/s (P < 0.05). No side effects were observed in the 24 to 42 months following surgery. | 17 | Data Logic Structure and Key Technologies on Intelligent High-precision Map显示文摘Taking autonomous driving and driverless as the research object,we discuss and define intelligent high-precision map.Intelligent high-precision map is considered as a key link of future travel,a carrier of real-time perception of traffic resources in the entire space-time range,and the criterion for the operation and control of the whole process of the vehicle.As a new form of map,it has distinctive features in terms of cartography theory and application requirements compared with traditional navigation electronic maps.Thus,it is necessary to analyze and discuss its key features and problems to promote the development of research and application of intelligent high-precision map.Accordingly,we propose an information transmission model based on the cartography theory and combine the wheeled robot’s control flow in practical application.Next,we put forward the data logic structure of intelligent high-precision map,and analyze its application in autonomous driving.Then,we summarize the computing mode of“Crowdsourcing+Edge-Cloud Collaborative Computing”,and carry out key technical analysis on how to improve the quality of crowdsourced data.We also analyze the effective application scenarios of intelligent high-precision map in the future.Finally,we present some thoughts and suggestions for the future development of this field. | Jingnan LIU Jiao ZHAN Chi GUO Tingting LEI Ying LI | 2020 | Journal of Geodesy and Geoinformation Science2020,3,3: | 11 |
| 18 | Protective Effects of Chaihu Shugan San(柴胡疏肝散) on Nonalcoholic Fatty Liver Disease in Rats with Insulin Resistance显示文摘Objective: To investigate the protective effects of Chinese medicine formulation Chaihu Shugan San(柴胡疏肝散, CHSGS) on nonalcoholic fatty liver disease(NAFLD) in rats with insulin resistance(IR) and its molecular mechanisms. Methods: Male Sprague-Dawley rats were randomly divided into six groups: the control group, the model group, Dongbao Gantai group(东宝肝泰, DBGT, 0.09 g methionine/kg), CHSGS high-dose group(CHSG-H, 12.6 g crude drug/kg), CHSGS medium-dose group(CHSG-M, 6.3 g crude drug/kg), and CHSGS low-dose group(CHSG-L, 3.15 g crude drug/kg). After establishing the NAFLD rat model and treatment for 8 weeks, total cholesterol(TC), triglyceride(TG), high-density lipoprotein cholesterol(HDL-C), free fatty acid(FFA), fasting blood glucose(FBG), fasting insulin(FINS) contents in blood serum, and TC, TG contents in the hepatic homogenate were measured by an automatic biochemical analyzer, and a homeostasis model assessment was applied to assess the status of IR, insulin sensitivity index(ISI), and homeostasis model assessment for insulin secretion(HOMA-IS). The expression levels of adiponectin and leptin mRNA in liver tissue were analyzed by reverse transcription polymerase chain reaction. Pathological changes of livers were observed by hematoxylineosin staining of paraffin section. Results: Compared with the model group, the serum levels of TC, TG, FFA, FBG, FINS, IRI, ISI, and the liver levels of TC and TG in CHSG-H, CHSG-M, CHSG-L groups showed significant declines(P<0.01 or P<0.05); the serum levels of HDL-C, HOMA-IS were significantly increased(P<0.01 or P<0.05); the expression of leptin mRNA was dramatically decreased and the expression of adiponectin mRNA was increased in the hepatic tissue(P<0.01 or P<0.05). The fatty deposition of liver cells could also be alleviated. | 19 | ABNORMAL FLOWER AND GRAIN 1 encodes OsMADS6 and determines palea identity and affects rice grain yield and quality显示文摘The palea and lemma are floral organ structures unique to grasses;these structures form the hull and directly affect grain size and quality. However, the molecular mechanisms controlling the development of the hull are not well understood. In this study, we characterized the rice(Oryza sativa) abnormal flower and grain1(afg1) mutant, a new allele of OsMADS6. Similar to previously characterized osmads6 alleles, in the afg1 floret, the palea lost its marginal region and acquired the lemma identity. However, in contrast to other osmads6 alleles, the afg1 mutant showed altered grain size and grain quality, with decreased total starch and amylose contents, and increased protein and soluble sugar contents. The analysis of transcriptional activity suggested that AFG1 is a transcriptional activator and may affect grain size by regulating the expression levels of several genes related to cell expansion and proliferation in the afg1 mutant.
