| 1 | Structural basis for bivalent binding and inhibition of SARS-CoV-2 infection by human potent neutralizing antibodies显示文摘Neutralizing monoclonal antibodies(nAbs)to severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)represent promising candidates for clinical intervention against coronavirus disease 2019(COVID-19).We isolated a large number of nAbs from SARS-CoV-2-infected individuals capable of disrupting proper interaction between the receptor binding domain(RBD)of the viral spike(S)protein and the receptor angiotensin converting enzyme 2(ACE2).However,the structural basis for their potent neutralizing activity remains unclear.Here,we report cryo-EM structures of the ten most potent nAbs in their native full-length IgG-form or in both IgG-form and Fab-form bound to the trimeric S protein of SARS-CoV-2.The bivalent binding of the full-length IgG is found to associate with more RBDs in the“up”conformation than the monovalent binding of Fab,perhaps contributing to the enhanced neutralizing activity of IgG and triggering more shedding of the S1 subunit from the S protein.Comparison of a large number of nAbs identified common and unique structural features associated with their potent neutralizing activities.This work provides a structural basis for further understanding the mechanism of nAbs,especially through revealing the bivalent binding and its correlation with more potent neutralization and the shedding of S1 subunit. | Renhong Yan Ruoke Wang Bin Ju Jinfang Yu Yuanyuan Zhang Nan Liu Jia Wang Qi Zhang Peng Chen Bing Zhou Yaning Li Yaping Shen Shuyuan Zhang Long Tian Yingying Guo Lu Xia Xinyue Zhong Lin Cheng Xiangyang Ge Juanjuan Zhao Hong-Wei Wang Xinquan Wang Zheng Zhang Linqi Zhang Qiang Zhou | 2021 | Cell Research2021,31,5: | 4 |

[Methods]The primers for heat resistant nuclease gene( nuc) of Staphylococcus aureus were designed for establishing the LAMP method for rapidly detecting Staphylococcus aureus. [Results] The results of LAMP detection on Staphylococcus aureus in various dairy products were completely identical with that by bacterial isolation test; meanwhile it has a high specificity and a 10-fold sensitivity over the hemi-nested PCR. Herein we found that a chemotherapeutic drug nanoformulation composed of irinotecan hydrochloride(CPT-11) and an amphiphilic molecule DSPE-mPEG_(2000) can interfere with the limulus amebocyte lysate assay(LAL). | 17 | Edible mushrooms as a potent therapeutics of subclinical thyroid dysfunction among adults,especially in obese individuals:a prospective cohort study显示文摘Background:Mushrooms are a good source of many nutrients which are potentially beneficial for chronic diseases.We speculated that due to its abundant nutrients edible mushrooms might have a beneficial effect on the prevention of subclinical thyroid dysfunction(SCTD).Therefore,we designed a large-scale cohort study to examine whether mushrooms consumption is a protective factor for SCTD in adults.Methods:This prospective cohort study investigated 6631 participants(mean age:(45.0±10.2)years;55.1%men).Edible mushrooms consumption was measured at baseline using a validated food frequency questionnaire.SCTD was defined as abnormal serum thyroid-stimulating hormone levels and normal free thyroxine.Cox proportional hazards regression models were used to examine the association of edible mushrooms consumption with incident SCTD.Results:During follow-up period,a total of 262 new cases of SCTD were identified,the incidence rate of subclinical hypothyroidism was 8.9/1000 person-years and subclinical hyperthyroidism was 7.2/1000 person-years.After adjusting potential confounding factors,the multivariable hazard ratios(95%confidence intervals)for subclinical hypothyroidism were 1.00(reference)for almost never,0.53(0.29,0.97)for 1-3 times/week and 0.30(0.10,0.87)for≥4 times/week(P for trend=0.02).It also showed edible mushrooms consumption was inversely associated with subclinical hypothyroidism in obese individuals but not non-obese individuals,the final hazard ratios(95%confidence intervals)were 0.14(0.03,0.73)(P for trend<0.01).Conclusions:This population-based prospective cohort study has firstly demonstrated that higher edible mushrooms consumption was significantly associated with lower incidence of subclinical hypothyroidism among general adult population,especially in obese individuals. | Juanjuan Zhang Sabina Rayamajhi Amrish Thapa Ge Meng Qing Zhang Li Liu Hongmei Wu Yeqing Gu Shunming Zhang Tingjing Zhang Xuena Wang Zhixia Cao Jun Dong Xiaoxi Zheng Xu Zhang Xinrong Dong Xing Wang Shaomei Sun Ming Zhou Qiyu Jia Kun Song Kaijun Niu | 2023 | Food Science and Human Wellness2023,12,1: | 0 |

| 19 | An Edge-assisted, Object-oriented Random Forest Approach for Refined Extraction of Tea Plantations Using Multi-temporal Sentinel-2 and High-resolution Gaofen-2 Imagery显示文摘As a consumed and influential natural plant beverage,tea is widely planted in subtropical and tropical areas all over the world.Affected by(sub)tropical climate characteristics,the underlying surface of the tea distribution area is extremely complex,with a variety of vegetation types.In addition,tea distribution is scattered and fragmentized in most of China.Therefore,it is difficult to obtain accurate tea information based on coarse resolution remote sensing data and existing feature extraction methods.This study proposed a boundary-enhanced,object-oriented random forest method on the basis of high-resolution GF-2 and multi-temporal Sentinel-2 data.This method uses multispectral indexes,textures,vegetable indices,and variation characteristics of time-series NDVI from the multi-temporal Sentinel-2 imageries to obtain abundant features related to the growth of tea plantations.To reduce feature redundancy and computation time,the feature elimination algorithm based on Mean Decrease Accuracy(MDA)was used to generate the optimal feature set.Considering the serious boundary inconsistency problem caused by the complex and fragmented land cover types,high resolution GF-2 image was segmented based on the MultiResolution Segmentation(MRS)algorithm to assist the segmentation of Sentinel-2,which contributes to delineating meaningful objects and enhancing the reliability of the boundary for tea plantations.Finally,the object-oriented random forest method was utilized to extract the tea information based on the optimal feature combination in the Jingmai Mountain,Yunnan Province.The resulting tea plantation map had high accuracy,with a 95.38%overall accuracy and 0.91 kappa coefficient.We conclude that the proposed method is effective for mapping tea plantations in high heterogeneity mountainous areas and has the potential for mapping tea plantations in large areas. | Juanjuan YU Xiufeng HE Jia XU Zhuang GAO Peng YANG Yuanyuan CHEN Jiacheng XIONG | 2023 | Journal of Geodesy and Geoinformation Science2023,6,1: | 0 |
| 20 | Modified intratympanic steroid therapy for sudden sensorineural hearing loss via tympanic tube and gelfoam as a salvage treatment显示文摘Background: Sudden sensorineural hearing loss(SSNHL) is a prevalent emergency in ear, nose, and throat practice. Previous studies have demonstrated that intratympanic steroid therapy(IST) can serve as a salvage treatment for SSNHL after the failure of systemic steroid therapy(SST).Objective: This study aimed to analyze the efficacy of modified IST involving the insertion of a tympanic tube and gelfoam as a salvage treatment for patients with SSNHL, and to explore its associated factors.Methods: Totally, 74 patients who were aged 22–81 years with SSNHL were enrolled and allocated to either the control group(n = 25) or the treatment group(n = 49) based on their treatment modalities. All patients received SST lasting for at least 7 days. Subsequently, patients in the treatment group, after SST failure, underwent IST twice a week for 2–6 weeks, while the control group did not. Efficacy was assessed by the improvement in pure tone average at the affected frequency at the beginning and end of IST.Results: Hearing improvement in all patients after IST in the treatment group was 9.71 ± 14.84 dB, with significant improvement at affected frequencies(250-8000 Hz) compared with the control group(P < 0.05). The findings indicated the duration from the onset of SSNHL to the beginning of IST as an independent factor for pure tone average improvement after treatment(P = 0.002), whereas age, duration of SST, and time of IST were not(P > 0.05).Conclusion: The modified IST was demonstrated to be a safe and effective method as a salvage treatment for SSNHL. This study explored the efficacy of a modified IST approach, incorporating the utilization of tympanic tubes and gelfoam as key components. The findings underscore the advantages of gelfoam as a strategic drug carrier placed in the round window niche.
