1DEAP1 encodes a fasciclin-like arabinogalactan protein required for male fertility in rice显示文摘Arabinogalactan proteins(AGPs)are widely distributed in plant cells.Fasciclin-like AGPs(FLAs)belong to a subclass of AGPs that play important roles in plant growth and development.However,little is known about the biological functions of rice FLA.Herein,we report the identification of a male-sterile mutant of DEFECTIVE EXINE AND APERTURE PATTERNING1(DEAP1)in rice.The deap1 mutant anthers produced aberrant pollen grains with defective exine formation and a flattened aperture annulus and exhibited slightly delayed tapetum degradation.DEAP1 encodes a plasma membrane-associated member of groupⅢplant FLAs and is specifically and temporally expressed in reproductive cells and the tapetum layer during male development.Gene expression studies revealed reduced transcript accumulation of genes related to exine formation,aperture patterning,and tapetum development in deap1 mutants.Moreover,DEAP1 may interact with two rice D6 PROTEIN KINASE-LIKE3 s(OsD6PKL3s),homologs of a known Arabidopsis aperture protein,to affect rice pollen aperture development.Our findings suggested that DEAP1 is involved in male reproductive development and may affect exine formation and aperture patterning,thereby providing new insights into the molecular functions of plant FLAs in male fertility.Dan Zhou Ting Zou Kaixuan Zhang Pingping Xiong Fuxing Zhou Hao Chen Gongwen Li Kaiyou Zheng Yuhao Han Kun Peng Xu Zhang Shangyu Yang Qiming Deng Shiquan Wang Jun Zhu Yueyang Liang Changhui Sun Xiumei Yu Huainian Liu Lingxia Wang Ping Li Shuangcheng Li 2022Journal of Integrative Plant Biology2022,64,7:2

3Highly sensitive smartphone-based detection of Listeria monocytogenes using SYTO9显示文摘Listeriosis is caused by Listeria monocytogenes(LM) and is currently considered to be one of the leading food-borne diseases worldwide, with mortality rate of 20%~30%. Currently, detection methods for LM are time-consuming with low sensitivity, and delayed detection results. SYTO9 has a high affinity for DNA and exhibits enhanced fluorescence upon binding. Therefore, this study used SYTO9 staining and image processing to develop a rapid loop mediated isothermal amplification(LAMP) detection method for LM. Smartphone was successfully used for detecting the color change in different concentrations of LM. Besides, the optimized LAMP reaction temperature was 63 °C by color identification, and the limit of detection for LM was 6 copies/μL in the green channel. 4Salvinia-like slippery surface with stable and mobile water/air contact line显示文摘Superhydrophobic surfaces are widely used in many industrial settings,and mainly consist of rough solid protrusions that entrap air to minimize the liquid/solid area.The stability of the superhydrophobic state favors relatively small spacing between protrusions.However,this in turn increases the lateral adhesion force that retards the mobility of drops.Here we propose a novel approach that optimizes both properties simultaneously.Inspired by the hydrophobic leaves of Salvinia molesta and the slippery Nepenthes pitcher plants,we designed a Salvinia-like slippery surface(SSS)consisting of protrusions with slippery heads.We demonstrate that compared to a control surface,the SSS exhibits increased stability against pressure and impact,and enhanced lateral mobility of water drops as well as reduced hydrodynamic drag.We also systematically investigate the wetting dynamics on the SSS.With its easy fabrication and enhanced performance,we envision that SSS will be useful in a variety of fields in industry.Xiaomei Li Jinlong Yang Kaixuan Lv Periklis Papadopoulos Jing Sun Dehui Wang Yanhua Zhao Longquan Chen Dapeng Wang Zuankai Wang Xu Deng 2021National Science Review2021,8,5:1

6Telomere-to-telomere genome of the allotetraploid legume Sesbania cannabina reveals transposon-driven subgenome divergence and mechanisms of alkaline stress tolerance显示文摘Alkaline soils pose an increasing problem for agriculture worldwide,but using stress-tolerant plants as green manure can improve marginal land.Here,we show that the legume Sesbania cannabina is very tolerant to alkaline conditions and,when used as a green manure,substantially improves alkaline soil.To understand genome evolution and the mechanisms of stress tolerance in this allotetraploid legume,we generated the first telomere-to-telomere genome assembly of S.cannabina spanning~2,087 Mb.The assembly included all centromeric regions,which contain centromeric satellite repeats,and complete chromosome ends with telomeric characteristics.Further genome analysis distinguished A and B subgenomes,which diverged approximately 7.9 million years ago.Comparative genomic analysis revealed that the chromosome homoeologs underwent large-scale inversion events(>10 Mb)and a significant,transposon-driven size expansion of the chromosome 5A homoeolog.We further identified four specific alkali-induced phosphate transporter genes in S.cannabina;these may function in alkali tolerance by relieving the deficiency in available phosphorus in alkaline soil.Our work highlights the significance of S.cannabina as a green tool to improve marginal lands and sheds light on subgenome evolution and adaptation to alkaline soils.Haofei Luo Xiaofei Wang Changqing You Xuedan Wu Duofeng Pan Zhiyao Lv Tong Li Dongmei Zhang Zhongbao Shen Xiaodong Zhang Guodao Liu Kaixuan He Qingtong Ye Yajun Jia Qinghua Zhao Xian Deng Xiaofeng Cao Xianwei Song Gai Huang 2024Science China(Life Sciences)2024,67,1:0
7Development and evaluation of a thermostatic nucleic acid testing device based on magnesium pyrophosphate precipitation for detecting Enterocytozoon hepatopenaei显示文摘Enterocytozoon hepatopenaei(EHP)infection has seriously affected prawn culture globally.The symptoms of the infection are not apparent,and traditional detection methods are time consuming and low in accuracy.We developed a new onsite rapid testing device(size 18.8×16.7×6.6 cm^(3))for EHP based on magnesium pyrophosphate precipitation and facilitated by loop mediated isothermal amplification(LAMP).The design and fabrication of the device enables efficient light absorbance.The device has a highly sensitive detector,high-precision thermal controller,and humanized touch screen.The temperature control precision of the device is 0.2-0.3℃ at 60℃,63℃,and 65℃.The coefficients of variation values(CVV)of the luminous power in one channel at light on and off were found to be 0.0097 and 0.0014,respectively,within 1 h.The CVV of the background,luminous power,and values of eight PCR tubes filled with pure water were all less than 5%.In the EHP experiment,eight samples(including seven positive and one negative)confirmed the effectiveness of the device,and four positive and four negative samples verified whether cross-contamination exists.Among them,the rise time of the curve was about 15 min.These results assert that the developed device exhibits enhanced stability and uniformity and has excellent performance with high sensitivity,good specificity,and low testing time.Moreover,the optimal and minimum absorbance range was 555-655 nm for monitoring the production of LAMP.Zhu Chen Kaixuan Zhao Ziyu He Xiaofang Luo Zuodong Qin Yimin Tan Xiangming Zheng Zuozhong Wu Yan Deng Hui Chen Yuan Guo Song Li 2022Chinese Chemical Letters2022,33,8:0
8Enzootic epidemiology of Brucella in livestock in central Gansu Province after the National Brucellosis Prevention and Control Plan显示文摘Brucellosis remains one of the most common zoonoses spread worldwide,inducing enormous economic losses to the livestock industry and posing serious health threats to humans.Brucellosis re-emerged in China in the mid-1990s and reached a historically high level in 2015.The National Brucellosis Prevention and Control Plan(NBPCP)was initiated from 2016 to 2020.However,the present epidemiological status in livestock has not been elucidated,and whether Brucella variation occurred remains unclear.This study performed an extensive serological investigation in ruminant livestock from 2019 to 2021 in central Gansu Province,China.In total,11,296 samples from 337 farms were collected to detect the specific antibodies of Brucella.The yearly average serological prevalence of Brucella at the flock level and individual level declined from 11.32%to 8.26%and 1.17%to 0.57%,respectively.The apparent individuallevel seroprevalence of small and large ruminants was 0.89%and 0.52%,respectively.The brucellosis distribution has shifted from pastoral areas to agro-pastoral areas.Flock size and gender may be major risks of Brucella infection.Then,the B.melitensis TZ strain was isolated from female Tibetan sheep blood cell lysates.Phonotypical characterization demonstrated that it belongs to B.melitensis.biovar 3,and multilocus sequencing typing results indicated that it belongs to ST8.The whole genome and subsequent phylogenetic analysis demonstrated that the B.melitensis TZ strain is genetically more closely related to the B.melitensis QH61 strain.The B.melitensis TZ strain has similar growth characteristics to the B.melitensis 16 M strain.Overall,our study suggests that after strengthening control and prevention measures based on the NBPCP,there is a very low prevalence or absence of B.melitensis in the central Gansu Province of China,and the genotype of an epidemic strain of Brucella in Northwest China is relatively stable.Yupeng Fang Jianjun Wang Guanyin Zhang Fengdong Zhu Chaoyue Guo Jiandong Zhang Kaixuan Guo Yun Deng Jinxue Zhang Huanchun Chen Zhengfei Liu 2023Animal Diseases2023,3,2:0
9The histone H3K27 demethylase REF6/JMJ12 promotes thermomorphogenesis in Arabidopsis显示文摘Dynamic trimethylation of histone H3 at Lys27(H3 K27 me3)affects gene expression and controls plant development and environmental responses.In Arabidopsis thaliana,RELATIVE OF EARLY FLOWERING 6(REF6)/JUMONJI DOMAIN-CONTAINING PROTEIN 12 demethylates H3 K27 me3 by recognizing a specific DNA motif.However,little is known about how REF6 activates target gene expression after recognition,especially in environmental responses.In response to warm ambient temperature,plants undergo thermomorphogenesis,which involves accelerated growth,early flowering and changes in morphology.Here we show that REF6 regulates thermomorphogenesis and cooperates with the transcription factor PHYTOCHROME INTERACTING FACTOR 4 to synergistically activate thermoresponsive genes under warm ambient temperature.The ref6 loss-of-function mutants exhibited attenuated hypocotyl elongation at warm temperature,partially due to downregulation of GIBBERELLIN20-OXIDASE 2 and BASIC HELIX-LOOP-HELIX 87.REF6 enzymatic activity is necessary for warm ambient temperature responses.Together,our results provide direct evidence of an epigenetic modifier and a transcription factor working together to respond to the environment.Kaixuan He Hailiang Mei Jiaping Zhu Qi Qiu Xiaofeng Cao Xian Deng 2022National Science Review2022,9,5:0

11Pepper(Capsicum annuum)xylogen-like arabinogalactan protein(XYLP)1 and XYLP2 promote synthesis of lignin during stem development to cope with stresses显示文摘Pepper stems exhibit a high level of strength and lignin deposition to support plant growth,which direct the cultivation style without binding sticks or scaffolding.However,regulation of lignin synthesis and accumulation in pepper stem has not been extensively studied.Herein,we first investigated the pepper stem developmental process and confirmed that increasing lignin accumulation occurs during stem growth.We then performed genome-wide identification and characterization of xylogen-like arabinogalactan protein(XYLP)family members and obtained 10,22,and 19 XYLPs in pepper,tomato,and potato respectively.Evaluation of the phylogenetic relationship among the identified XYLPs suggested that these proteins are conserved in Solanaceae.Thereafter,we analyzed the 10 CanXYLP genes and observed that these genes exhibit differential expressing patterns at different stages of pepper stem development.Among these genes,two XYLPs,namely CanXYLP1 and CanXYLP2,exhibited an increased expression pattern and a strong correlation with lignin accumulation in pepper stem.We further found that CanXYLP1 and CanXYLP2 play a role in pepper stem lignification by positively regulating the lignin synthesis pathway genes in pepper,and the CanXYLP1/2-silenced plants displayed a blocked lignification phenotype.Finally,we confirmed that CanXYLP1/2 expression is upregulated in response to some abiotic and biotic signals,suggesting that these two genes enhance the tolerance of pepper stem to unfavorable conditions.These results contribute to our understanding of the molecular mechanism controlling pepper stem lignification,and the relationship between the lignin content of pepper stem and XYLPs.Min Zhang Qianwen Zhang Long Cheng Qianying Li Xinyue He Kaixuan Wang Jianan Liu Feng Li Yingtian Deng 2022Vegetable Research2022,2,1:0
