Special Section:SARS-CoV-2 Preliminary evidence from a multicenter prospective observational study of the safety and efficacy of chloroquine for the treatment of COVID-19显示文摘Effective therapies are urgently needed for the SARS-CoV-2 pandemic.Chloroquine has been proved to have antiviral effect against coronavirus in vitro. In this study,we aimed to assess the efficacy and safety of chloroquine with different doses in COVID-19.In this multicenter prospective observational study,we enrolled patients older than 18 years old with confirmed SARS-CoV-2 infection excluding critical cases from 12 hospitals in Guangdong and Hubei Provinces.Eligible patients received chloroquinephosphate s00 mg,orally, once(half dose)or twice(full dose)daily.Patients treated with non-chloroquine therapy were included as historical controls. The primary endpoint is the time to undetectable viral RNA.Secondary outcomes include the proportion of patients with undetectable viral RNA by day 10 and 14,hospitalization time, duration of fever, and adverse events.A total of 197 patients completed chloroquine treatment, and 176patients were included as historical controls. The median time to achieve an undetectable viral RNA was shorter in chloroquine than in non-chloroquine(absolute difference in medians-6.0 days;95%CI-6.0 to—4.0).The duration of fever is shorter in chloroquine(geometric mean ratio 0.6;95%CIO.5 to 0.8).No serious adverse events were observed in the chloroquine group.Patients treated with half dose experienced lower rate of adverse events than with full dose.Although randomized trials are needed for further evaluation, this study provides evidence for safety and efficacy of chloroquine in COVID-19 and suggests that chloroquine can be a cost-effective therapy for combating the COVID-19 pandemic. | Mingxing Huang Man Li Fei Xiao Pengfei Pang Jiabi Liang Tiantian Tang Shaoxuan Liu Binghui Chen Jingxian Shu Yingying You Yang Li Meiwen Tang Jianhui Zhou Guanmin Jiang Jingfen Xiang Wenxin Hong Songmei He Zhaoqin Wang Jianhua Feng Changqing Lin Yinong Ye Zhilong Wu Yaocai Li Bei Zhong Ruilin Sun Zhongsi Hong Jing Liu Huili Chen Xiaohua Wang Zhonghe Li Duanqing Pei Lin Tian Jinyu Xia Shanping Jiang Nanshan Zhong Hong Shan | 2020 | National Science Review2020,7,9: | 20
Underwater explosion in centrifuge partⅠ:Validation and calibration of scaling laws显示文摘Centrifuge is a promising tool for underwater explosion(UNDEX) research as both Mach and Froude similitudes could be satisfied with hyper-gravity in models, which would result in similarities in both shock wave and bubble oscillation. Scaling laws for UNDEX in centrifuge have been proposed based on dimensional analysis. Two dimensionless numbers, i.e.,π_3 and π_4, are used to characterize shock wave and bubble oscillation, respectively. To validate scaling laws, 17 UNDEX tests are designed by varying accelerations or explosive weights and positions in centrifuge. The tests are classified into different groups to validate scaling laws as well as calibrate coefficients in empirical formulae for both shock wave and bubble oscillation. The results show that changes of gravity acceleration or hydrodynamic pressure almost has no influence on shock wave peak pressure and time constant as long as π3 is constant. The dimensionless bubble period and maximum radius agreed with each other when π4 is constant. Based on the research, an example is exhibited to suggest method for the computation of initial loading conditions for a submerged obstacle subjected to UNDEX. | HU Jing CHEN ZuYu ZHANG XueDong WEI YingQi LIANG XiangQian LIANG JianHui MA GuoWei WANG QiuSheng LONG Yuan | 2017 | Science China(Technological Sciences)2017,60,11: | 14
Big Earth Data science:an information framework for a sustainable planet显示文摘The digital transformation of our society coupled with the increasing exploitation of natural resources makes sustainability challenges more complex and dynamic than ever before. These changes will unlikely stop or even decelerate in the near future. There is an urgent need for a new scientific approach and an advanced form of evidence-based decisionmaking towards the benefit of society, the economy, and the environment. To understand the impacts and interrelationships between humans as a society and natural Earth system processes,we propose a new engineering discipline,Big Earth Data science. This science is called to provide the methodologies and tools to generate knowledge from diverse, numerous, and complex data sources necessary to ensure a sustainable human society essential for the preservation of planet Earth.Big Earth Data science aims at utilizing data from Earth observation and social sensing and develop theories for understanding the mechanisms of how such a social-physical system operates and evolves. The manuscript introduces the universe of discourse characterizing this new science, its foundational paradigms and methodologies, and a possible technological framework to be implemented by applying an ecosystem approach.CASEarth and GEOSS are presented as examples of international implementation attempts. Conclusions discuss important challenges and collaboration opportunities. | Huadong Guo Stefano Nativi Dong Liang Max Craglia Lizhe Wang Sven Schade Christina Corban Guojin He Martino Pesaresi Jianhui Li Zeeshan Shirazi Jie Liu Alessandro Annoni | 2020 | International Journal of Digital Earth2020,13,7: | 12
Underwater explosion in centrifuge Part Ⅱ:Dynamic responses of defensive steel plate显示文摘Part II of this paper is focused on the dynamic response of a steel plate of the centrifuge model which is fixed on the container. This plate is 5 cm thick, allowing for non-destructive elastic response to all the 19 blasts. Three accelerators and 10 sets of strain gauges are installed on the plate to monitor the acceleration and deformation histories induced by shock waves and bubble oscillations. This article presents the characteristic values measured for shock waves and bubble oscillations response. The detailed information can be found in a database available on the web. In order to obtain the correct acceleration time history, the cut-off frequency of low-pass filtering by Fourier transformation was applied. The results indicated that the acceleration and strain induced by shock wave was nearly unaffected by the increasing gravity, while the effect of gravity has remarkable influence on those induced by bubble oscillation. A preliminary study of comparing the measured dynamic response with numerical simulations has been carried out using ANSYS LS-DYNA. The results captured the dynamic response in terms of characteristics and trends, but deviated in magnitude due the theoretical and technological limitations involved in both the physical and numerical models. It certainly deserves further studies. | LONG Yuan ZHOU HongYuan LIANG XiangQian SONG Ge CHEN ZuYu HU Jing WANG QiuSheng ZHANG XueDong LIANG JianHui HUANG ZhiJie | 2017 | Science China(Technological Sciences)2017,60,12: | 8

Numerical study on aero-optical imaging deviations of vehicles at different altitudes显示文摘When the vehicle is flying in the atmosphere at high speed, the optical head and the atmosphere will have severe friction, thus forming a complex flow field, which makes the target image shift in the optical imaging system. The influence of altitude on aero-optical imaging deviation is studied in this paper. The geometric modeling and mesh generation of a typical blunt nosed high-speed vehicle were carried out, and the three-dimensional(3 D) flow field density was obtained by a large amount of computational fluid dynamic calculation. In order to complete the optical calculation, the backward ray tracing method and the backward ray tracing stop criterion were used. The results show that as the height increases, the imaging deviation decreases gradually, and the imaging deviation slope increases and tends to be flat and close to zero. | XU Liang ZHANG Ziye WANG Tao WU Jianhui CHEN Xi | 2022 | Optoelectronics Letters2022,18,2: | 4
Rheb promotes brown fat thermogenesis by Notch-dependent activation of the PKA signaling pathway显示文摘Increasing brown and beige fat thermogenesis have an anti-obesity effect and thus great metabolic benefits. However, the molecular mechanisms regulating brown and beige fat thermogenesis remain to be further elucidated. We recently found that fat-specific knockout of Rheb promoted beige fat thermogenesis. In the current study,we show that Rheb has distinct effects on thermogenic gene expression in brown and beige fat.Fat-specific knockout of Rheb decreased protein kinase A(PKA)activity and thermogenic gene expression in brown adipose tissue of high-fat diet-fed mice.On the other hand, overexpression of Rheb activated PKA and increased uncoupling protein 1 expression in brown adipocytes.Mechanistically,Rheb overexpression in brown adipocytes increased Notch expression, leading to disassociation of the regulatory subunit from the catalytic subunit of PKA and subsequent PKA activation. Our study demonstrates that Rheb,by selectively modulating thermogenic gene expression in brown and beige adipose tissues, plays an important role in regulating energy homeostasis. | Wen Meng Xiuci Liang Ting Xiao Jing Wang Jie Wen Hairong Luo Jianhui Teng Yanquan Fei Qinghai Zhang Bilian Liu Fang Hu juli Bai Meilian Liu Zhiguang Zhou Feng Liu | 2019 | Journal of Molecular Cell Biology2019,11,9: | 2
An improved CFD model of gas flow and particle interception in a fiber material显示文摘An improved CFD model of gas flow and particle interception in a fiber material which fiber size is Y-shape was developed in this work. The porous medium model was used to build the model of the whole size of fiber filter medium. Mixture model was adopted. The algorithm of particle interception in the whole size of fiber filter medium was derived and UDF(User Defined Function) that described kinds of particle filtering mechanisms in filter fibrous media was added to the Fluent default conservation equation as source term for simulation. The inertial resistance of the filter was taken into consideration, which provided a more precise measurement of the smoke flow and the particle interception in the filter under higher smoke speed conditions. The commercial software, Fluent 6.3, was used to simulate the smoke flow and particle interception in the filter in a single suction. The velocity and pressure profiles of smoke or nicotine particle in the filter, as well as nicotine particle volume fraction profile were well simulated. Finally, the comparisons of nicotine particle filtration efficiency between Fluent simulation results in this work and experimental results, as well as the model prediction in the literature were made to validate the simulation model. The comparisons showed that the particle entrapment model from simulation results was in good agreement with that from the experimental results. In addition, the Fluent simulation results are closer to reality both at the beginning and the end of the smoke process comparing with the model predicted results in the literature. | Zhiwei Sun Jianhui Wen Xiao Luo Wen Du Zhiwu Liang Kaiyun Fu | 2017 | Chinese Journal of Chemical Engineering2017,25,3: | 2

The antigenicity of SARS-CoV-2 Delta variants aggregated 10 high-frequency mutations in RBD has not changed sufficiently to replace the current vaccine strain显示文摘Emerging SARS-CoV-2 variants are the most serious problem for COVID-19 prophylaxis and treatment. To determine whether the SARS-CoV-2 vaccine strain should be updated following variant emergence like seasonal flu vaccine, the changed degree on antigenicity of SARS-CoV-2 variants and H3N2 flu vaccine strains was compared. The neutralization activities of Alpha,Beta and Gamma variants’spike protein-immunized sera were analysed against the eight current epidemic variants and 20 possible variants combining the top 10 prevalent RBD mutations based on the Delta variant, which were constructed using pseudotyped viruses.Meanwhile, the neutralization activities of convalescent sera and current inactivated and recombinant protein vaccine-elicited sera were also examined against all possible Delta variants.Eight HA protein-expressing DNAs elicited-animal sera were also tested against eight pseudotyped viruses of H3N2 flu vaccine strains from 2011–2019.Our results indicate that the antigenicity changes of possible Delta variants were mostly within four folds, whereas the antigenicity changes among different H3N2 vaccine strains were approximately 10–100-fold.Structural analysis of the antigenic characterization of the SARS-CoV-2 and H3N2 mutations supports the neutralization results. This study indicates that the antigenicity changes of the current SARS-CoV-2 may not be sufficient to require replacement of the current vaccine strain. | Jiajing Wu Jianhui Nie Li Zhang Hao Song Yimeng An Ziteng Liang Jing Yang Ruxia Ding Shuo Liu Qianqian Li Tao Li Zhimin Cui Mengyi Zhang Peng He Youchun Wang Xiaowang Qu Zhongyu Hu Qihui Wang Weijin Huang | 2022 | Signal Transduction and Targeted Therapy2022,7,2: | 1

Comparative proteomics analysis of Pichia pastoris cultivating in glucose and methanol显示文摘The methylotrophic yeast Pichia pastoris(syn.Komagataella phaffii)has been extensively engineered for protein production, and is attracting attention as a chassis cell for methanol biotransformation toward production of small molecules. However, the relatively unclear methanol metabolism hampers the metabolic rewiring to improve the biosynthetic efficiency. We here performed a label-free quantitative proteomic analysis of Pichia pastoris when cultivated in minimal media containing methanol and glucose, respectively. There were 243,158 up-regulated proteins and 244,304 down-regulated proteins in log and stationary phase, respectively, when cultivated in methanol medium compared with that of glucose medium.Peroxisome enrichment further improved the characterization of more differentially expressed proteins(481 proteins in log phase and 524 proteins in stationary phase). We demonstrated the transaldolase isoenzyme(Tal2,Protein ID:C4R244)was highly up-regulated in methanol medium cultivation, which plays an important role in methanol utilization. Our work provides important information for understanding methanol metabolism in methyltrophic yeast and will help to engineer methanol biotransformation in P.pastoris. | Rui Hou Linhui Gao Jianhui Liu Zhen Liang Yongjin J.Zhou Lihua Zhang Yukui Zhang | 2022 | Synthetic and Systems Biotechnology2022,7,3: | 1

UAV-based spatial pattern of three-dimensional green volume and its influencing factors in Lingang New City in Shanghai,China显示文摘Three-dimensional green volume(TDGV)reflects the quality and quantity of urban green space and its provision of ecosystem services;therefore, its spatial pattern and the underlying influential factors play important roles in urban planning and management. However, little is known about the factors contributing to the spatial pattern of TDGV.In this paper,TDGV and land use intensity(LUI)extracted from high spatial resolution(0.05 m)remotely sensed data acquired by an unmanned aerial vehicle(UAV),anthropogenic factors^(1))and natural factors^(2))were utilized to identify the spatial pattern of TDGV and the potential influencing factors in Lingang New City,a rapidly developed coastal town in Shanghai. The results showed that most of the TDGV was distributed in the western part of this new city and that its spatial variations were significantly axial.TDGV corresponded well with the chronologies of land formation, urban planning, and construction in the city.Generalized least squares(GLS)analysis of TDGV(grid cell size:100×100 m)and its influencing factors showed that the TDGV in this new city was significantly negatively correlated with both LUI and distance from roads and significantly positively correlated with land formation time and distance from water.Distance from buildings did not affect TDGV.Additionally, the degree of influence decreased in the following order:distance from water>land formation time>distance from roads>LUI.These results indicate that the spatial pattern of TDGV in this new town was mainly affected by natural factors(i.e.,the distance from water and land formation time)and that the artificial disturbances caused by rapid urbanization did not decrease the regional TDGV.The main factors shaping the spatial distribution of TDGV in this city were local natural factors. Our findings suggest that the improvement in local soil and water conditions should be emphasized in the construction of new cities in coastal areas to ensure the efficient provision of ecological services by urban green spaces. | Sijun ZHENG Chen MENG Jianhui XUE Yongbo WU Jing LIANG Liang XIN Lang ZHANG | 2021 | Frontiers of Earth Science2021,15,3: | 1

Electron-ion collider in China显示文摘Lepton scattering is an established ideal tool for studying inner structure of small particles such as nucleons as well as nuclei. As a future high energy nuclear physics project,an Electron-ion collider in China(EicC)has been proposed. It will be constructed based on an upgraded heavy-ion accelerator,High Intensity heavy-ion Accelerator Facility(HIAF)which is currently under construction, together with a new electron ring. The proposed collider will provide highly polarized electrons(with a po-larization of 80%)and protons(with a polarization of 70%)with variable center of mass energies from 15 to 20 GeV and the luminosity of(2–3)×1033 cm^(−2)·s^(−1).Polarized deuterons and Helium-3,as well as unpolarized ion beams from Carbon to Uranium, will be also available at the EicC.The main foci of the EicC will be precision measurements of the structure of the nucleon in the sea quark region, including 3D tomography of nucleon;the partonic structure of nuclei and the parton interaction with the nuclear environment;the exotic states, especially those with heavy flavor quark contents. In addition, issues fundamental to understanding the origin of mass could be addressed by measurements of heavy quarkonia near-threshold production at the EicC.In order to achieve the above-mentioned physics goals,a hermetical detector system will be constructed with cutting-edge technologies. This document is the result of collective contributions and valuable inputs from experts across the globe. The EicC physics program complements the ongoing scientific programs at the Jefferson Laboratory and the future EIC project in the United States. The success of this project will also advance both nuclear and particle physics as well as accelerator and detector technology in China. | Daniele PAnderle Valerio Bertone Xu Cao Lei Chang Ningbo Chang Gu Chen Xurong Chen Zhuojun Chen Zhufang Cui Lingyun Dai Weitian Deng Minghui Ding Xu Feng Chang Gong Longcheng Gui Feng-Kun Guo Chengdong Han Jun He Tie-Jiun Hou Hongxia Huang Yin Huang KrešImir KumeričKi LPKaptari Demin Li Hengne Li Minxiang Li Xueqian Li Yutie Liang Zuotang Liang Chen Liu Chuan Liu Guoming Liu Jie Liu Liuming Liu Xiang Liu Tianbo Liu Xiaofeng Luo Zhun Lyu Boqiang Ma Fu Ma Jianping Ma Yugang Ma Lijun Mao Cédric Mezrag HervéMoutarde Jialun Ping Sixue Qin Hang Ren Craig DRoberts Juan Rojo Guodong Shen Chao Shi Qintao Song Hao Sun PawełSznajder Enke Wang Fan Wang Qian Wang Rong Wang Ruiru Wang Taofeng Wang Wei Wang Xiaoyu Wang Xiaoyun Wang Jiajun Wu Xinggang Wu Lei Xia Bowen Xiao Guoqing Xiao Ju-Jun Xie Yaping Xie Hongxi Xing Hushan Xu Nu Xu Shusheng Xu Mengshi Yan Wenbiao Yan Wencheng Yan Xinhu Yan Jiancheng Yang Yi-Bo Yang Zhi Yang Deliang Yao Zhihong Ye Peilin Yin C-PYuan Wenlong Zhan Jianhui Zhang Jinlong Zhang Pengming Zhang Yifei Zhang Chao-Hsi Chang Zhenyu Zhang Hongwei Zhao Kuang-Ta Chao Qiang Zhao Yuxiang Zhao Zhengguo Zhao Liang Zheng Jian Zhou Xiang Zhou Xiaorong Zhou Bingsong Zou Liping Zou | 2021 | Frontiers of physics2021,16,6: | 1
Structural Characteristics and its Significances on Hydrocarbon Accumulation in the Yunkai Low Uplift,Pearl River Mouth Basin显示文摘The Yunkai low uplift with low exploration degree is close to the Baiyun sag,and has hydrocarbon exploration potential in the deepwater area of the Pearl River Mouth Basin.Based on seismic and drilling data, balanced profiles and growth strata, this paper mainly discusses geological structures and formation processes of the Yunkai low uplift, and also analyzes the characteristics of fault system and their influence on hydrocarbon migration and accumulation. The EWtrending basement faults divide the Yunkai low uplift into two parts,i.e.the southern sector and the northern sector. The northern sector is a relatively wide and gentle uplift, while the southern sector is composed of two secondary half-grabens with faulting in the south and overlapping in the north. The Yunkai low uplift experienced three major formation stages, including the rapid uplifting stage during the deposition period of the Eocene Wenchang Formation, the slow uplifting stage during the deposition period of the Late Eocene-Middle Miocene Enping-Hanjiang formations, and the whole burial stage from the Middle Miocene to present. The extensional faults in the Yunkai low uplift and its adjacent areas strike mainly along the NW,NWW and near-EW directions. Also, the strikes of faults present a clockwise rotation from the deep to the shallow strata.According to effects of faults on hydrocarbon accumulation the faults in the Yunkai low uplift and its adjacent areas can be divided into trap-controlled faults and source-controlled faults. The trap-controlled faults control trap development and can effectively seal oil and gas.The source-controlled faults connect directly source rocks and reservoirs, which are highly active during the rifting stage and weakly active since the Miocene. This activity features of the source-controlled faults is beneficial to migration of the early crude oil from the Baiyun sag to the high part of the Yunkai low uplift, but is not good for migration of the late natural gas.In the Yunkai low uplift and its adjacent areas, the traps in the deep Zhuhai and Enping formations that are close to source rocks in the Baiyun sag should be the favorable exploration objectives. | YU Yixin ZHANG Tianlong ZHANG Zhongtao ZHANG Gongcheng ZENG Jianhui YANG Haizhang ZHAO Zhao LIANG Weiwei | 2021 | Acta Geologica Sinica(English Edition)2021,95,1: | 1
