Recent glacial retreat in High Asia in China and its impact on water resource in Northwest China显示文摘Under the impact of climatic warming, the glaciers in the High Asia in China have been retreating continuously with negative glacial mass balance in recent several decades. The retreat became more intensive in the past 10 years. The spatial pattern of the glacial retreat in the High Asia in China is that the smallest magnitude of retreat is in the inland of the Tibetan Plateau, the magnitude increases from the inland to the margin of the Tibetan Plateau, and the largest magnitude at the margin of the Tibetan Plateau. The glacial retreat in the High Asia in China has an important impact on the water resource of the arid regions in Northwest China. This study shows that the glacial retreat in the 1990s has caused an increase of 5.5% in river runoff in Northwest China. In the Tarim River basin, the increase of river runoff is higher than 5.5%.YAO Tandong 1,2 ,WANG Youqing 2 ,LIU Shiying 2,1 ,PU Jianchen 2,1 ,SHEN Yongping 2 & LU Anxin 2,1 1.Institute of Tibetan Plateau Research,Chinese Academy of Sciences,Beijing 100029,China 2.Cold and Arid Regions Environmental and Engineering Research Institute,Chinese Academy of Sciences,Lanzhou 730000,China 2004Science China Earth Sciences2004,47,12:117
Isolation and characterization of H7N9 viruses from live poultry markets--Implication of the source of current H7N9 infection in humans显示文摘On March 31, 2013, the National Health and Family Planning Commission announced that human infections with a previously undescribed influenza A (H7N9) virus had occurred in Shanghai and Anhui Province, China. To investigate the possible origins of the H7N9 viruses causing these human infections, we collected 970 samples, including drinking water, soil, and cloacal and tracheal swabs of poultry from live poultry markets and poultry farms in Shanghai and Anhui Province. Twenty samples were positive for the H7N9 influenza virus. Notably, all 20 viruses were isolated from samples collected from live poultry markets in Shanghai. Phylogenetic analyses showed that the six internal genes of these novel human H7N9 viruses were derived from avian H9N2 viruses, but the ancestor of their HA and NA genes is uncertain. When we examined the phylogenetic relationship between the H7N9 isolates from live poultry markets and the viruses that caused the human infections, we found that they shared high homology across all eight gene segments. We thus identified the direct avian origin of the H7N9 influenza viruses that caused the human infections. Importantly, we observed that the H7N9 viruses isolated from humans had acquired critical mutations that made them more 'human-like'. It is therefore imperative to take strong measures to control the spread of H7N9 viruses in birds and humans to prevent further threats to human health.SHI JianZhong DENG GuoHua LIU PeiHong ZHOU JinPing GUAN LiZheng LI WenHui LI XuYong GUO Jing WANG GuoJun FAN Jun WANG JinLiang LI YuanYuan JIANG YongPing LIU LiLing TIAN GuoBin LI ChengJun CHEN HuaLan 2013Chinese Science Bulletin2013,58,16:85

Whole-genome sequencing of 508 patients identifies key molecular features associated with poor prognosis in esophageal squamous cell carcinoma显示文摘Esophageal squamous cell carcinoma(ESCC)is a poor-prognosis cancer type with limited understanding of its molecular etiology.Using 508 ESCC genomes,we identified five novel significantly mutated genes and uncovered mutational signature clusters associated with metastasis and patients’outcomes.Several functional assays implicated that NFE2L2 may act as a tumor suppressor in ESCC and that mutations in NFE2L2 probably impaired its tumor-suppressive function,or even conferred oncogenic activities.Additionally,we found that the NFE2L2 mutations were significantly associated with worse prognosis of ESCC.We also identified potential noncoding driver mutations including hotspot mutations in the promoter region of SLC35E2 that were correlated with worse survival.Approximately 5.9%and 15.2%of patients had high tumor mutation burden or actionable mutations, respectively, and may benefit from immunotherapy or targeted therapies. We found clinically relevant coding and noncoding genomic alterations and revealed three major subtypes that robustly predicted patients’outcomes.Collectively,we report the largest dataset of genomic profiling of ESCC useful for developing ESCC-specific biomarkers for diagnosis and treatment.Yongping Cui Hongyan Chen Ruibin Xi Heyang Cui Yahui Zhao Enwei Xu Ting Yan Xiaomei Lu Furong Huang Pengzhou Kong Yang Li Xiaolin Zhu Jiawei Wang Wenjie Zhu Jie Wang Yanchun Ma Yong Zhou Shiping Guo Ling Zhang Yiqian Liu Bin Wang Yanfeng Xi Ruifang Sun Xiao Yu Yuanfang Zhai Fang Wang Jian Yang Bin Yang Caixia Cheng Jing Liu Bin Song Hongyi Li Yi Wang Yingchun Zhang Xiaolong Cheng Qimin Zhan Yanhong Li Zhihua Liu-Show 2020Cell Research2020,30,10:19
Eco-environment range in the source regions of the Yangtze and Yellow rivers显示文摘Based on geographical and hydrological extents delimited, four principles are identified, as the bases for delineating the ranges of the source regions of the Yangtze and Yellow rivers in the paper. According to the comprehensive analysis of topographical characteristics, climate conditions, vegetation distribution and hydrological features, the source region ranges for eco-environmental study are defined. The eastern boundary point is Dari hydrological station in the upper reach of the Yellow River. The watershed above Dari hydrological station is the source region of the Yellow River which drains an area of 4.49×10 4 km 2 . Natural environment is characterized by the major topographical types of plateau lakes and marshland, gentle landforms, alpine cold semi-arid climate, and steppe and meadow vegetation in the source region of the Yellow River. The eastern boundary point is the convergent site of the Nieqiaqu and the Tongtian River in the upstream of the Yangtze River. The watershed above the convergent site is the source region of the Yangtze River, with a watershed area of 12.24×10 4 km 2 . Hills and alpine plain topography, gentle terrain, alpine cold arid and semi-arid climate, and alpine cold grassland and meadow are natural conditions in the source region of the Yangtze River.DING Yongjian,YANG Jianping,LIU Shiyin,CHEN Rensheng,WANG Genxu,SHEN Yongping,WANG Jian,XIE Changwei,ZHANG Shiqing(Cold and Arid Regions Environmental and Engineering Research Institute, CAS, Lanzhou 730000, China) 2003Journal of Geographical Sciences2003,13,2:18
MiR-181b suppresses proliferation of and reduces chemoresistance to temozolomide in U87 glioma stem cells显示文摘MicroRNAs regulate self renewal and differentiation of cancer stem cells. There,we sought to identify the expression of miR-181b in glioma stem cells and investigate the biological effect of miR-181b on glioma stem cells in this study.MiR-181b expression was measured by real-time PCR in glioma stem cells isolated from U87 cells by FACS sorting. After miR-181b was overexpressed in U87 glioma stem cells by miR-181b lentiviral expression vector and/or treatment of temozolomide, secondary neurosphere assay, soft agar colony assay and MTT assay were performed.Compared with U87 cells, the expression of miR-181b was significantly decreased in U87 glioma stem cells.Overexpression of miR-181b decreased neurosphere formation by U87 glioma stem cells in vitro and suppressed colony formation in soft agar, and the cell growth inhibition rates increased in a time-dependent manner in U87 glioma stem cells infected with miR-181b lentivirus.Furthermore,miR-181b had a synergistic effect on temozolomide-induced inhibition of secondary neurosphere and soft agar colony, and on cell growth inhibition rates.MiR-181b functions as a tumor suppressor that suppresses proliferation and reduces chemoresistance to temozolomide in glioma stem cells.Ping Li Xiaoming Lu Yingyi Wang Lihua Sun Chunfa Qian Wei Yan Ning Liu Yongping You Zhen Fu 2010The Journal of Biomedical Research2010,24,6:14
Monitoring of frozen soil hydrology in macro-scale in the Qinghai-Xizang Plateau显示文摘Monitoring of frozen soil hydrology in macro-scale was performed by Chinese and Japanese scientists from 1997 to 1998. Quality measured data were obtained. Measured data on soil moisture and temperature are preliminarily analyzed. Based on profiles of soil temperature and moisture in individual measured sites, intra-annual freezing and melting process of soil is discussed. Maximum frozen and thawed depths and frozen days in various depths are estimated. The work emphasized the spatial distribution on soil temperature and moisture in macro-scale and the effect of topography on conditions of soil water and heat.Yongjian Ding Baisheng Ye Shiying Liu Yongping Shen Shaoling Wang Meixie Yang 2000Chinese Science Bulletin2000,45,12:13

FHY3 and FAR1 Integrate Light Signals with the miR156-SPL Module-Mediated Aging Pathway to Regulate Arabidopsis Flowering显示文摘In response to competition for light from their neighbors,shade-intolerant plants flower precociously to ensure reproductive success and survival. However, the molecular mechanisms underlying this key developmental switch are not well understood. Here,we show that a pair of Arabidopsis transcription factors essential for phytochrome A signaling,FAR-RED ELONGATED HYPOCOTYL3(FHY3)and FAR-RED IMPAIRED RESPONSE1(FAR1),regulate flowering time by integrating environmental light signals with the miR156-SPL module-mediated aging pathway. We found that FHY3 and FAR1 directly interact with three flowering-promoting SQUAMOSA-PROMOTER BINDING PROTEIN-LIKE(SPL)transcription factors,SPL3,SPL4,and SPL5,and inhibit their binding to the promoters of several key flowering regulatory genes, including FRUITFUL(FUL),LEAFY(LFY),APETALA1(AP1),and MIR172C,thus downregulating their transcript levels and delaying flowering.Under simulated shade conditions, levels of SPL3/4/5 proteins increase, whereas levels of FHY3 and FAR1 proteins decline, thus releasing SPL3/4/5 from FHY3/FAR1 inhibition to allow activation of FUL,LFY,AP1,and MIR172C and, consequently, early flowering.Taken together, these results unravel a novel mechanism whereby plants regulate flowering time by integrating environmental cues(such as light conditions)and an internal developmental program(the miR156-SPL module-mediated aging pathway).Yurong Xie Qin Zhou Yongping Zhao Quanquan Li Yang Liu Mengdi Ma Baobao Wang Rongxin Shen Zhigang Zheng Haiyang Wang 2020Molecular Plant2020,13,3:8

Effect of vernalization on tuberization and flowering in the Tibetan turnip is associated with changes in the expression of FLC homologues显示文摘The turnip(Brassica rapa var. rapa) is a biennial crop that is planted in late summer/early fall and forms fleshy tubers for food in temperate regions. The harvested tubers then overwinter and are planted again the next spring for flowering and seeds. FLOWERING LOCUS C(FLC) is a MADS-box transcription factor that acts as a major repressor of floral transition by suppressing the flowering promoters FT and SOC1. Here we show that vernalization effectively represses tuber formation and promotes flowering in Tibetan turnip. We functionally characterized four FLC homologues(BrrFLC1,FLC2, FLC3, and FLC5), and found that BrrFLC2 and BrrFLC1 play a major role in repressing flowering in turnip and in transgenic Arabidopsis. In contrast, tuber formation was correlated with BrrFLC1 expression in the hypocotyl and was repressed under cold treatment following the quantitative downregulation of BrrFLC1. Grafting experiments of non-vernalized and vernalized turnips revealed that vernalization independently suppressed tuberization in the tuber or hypocotyl of the rootstock or scion, which occurred in parallel with the reduction in BrrFLC1 activity. Together, our results demonstrate that the Tibetan turnip is highly responsive to cold exposure, which is associated with the expression levels of BrrFLC genes.Yan Zheng Landi Luo Yuanyuan Liu Yunqiang Yang Chuntao Wang Xiangxiang Kong Yongping Yang 2018Plant Diversity2018,40,2:7
Reduning injection for fever, rash, and ulcers in children with mild hand, foot, and mouth disease: a randomized controlled clinical study显示文摘OBJECTIVE: To assess the efficacy and safety of Reduning injection for fever, rash, and ulcers in children with mild hand, foot, and mouth disease(HFMD). METHODS: A stratified-block randomized, double-blind, parallel-controlled, and multicenter clinical trial was conducted with 360 patients in five hospitals across China: Quanzhou Children's Hospital, Shijiazhuang No. 5 Hospital, Shanghai Public Health Centre, Hunan Provincial Children's Hospital, and Kaifeng Children's Hospital. Patients were randomized into three groups with 120 in each. Group A was treated with Western Medicine, group B with Reduning injection, a Chinese herbal medicine, and group C with both Reduning injection and Western Medicine. Results were compared for treatment efficacy and safety on HFMD. The clinical outcomes were observed as follows: fever and onset time of antifebrile effect(time to bring the body temperature down ≥0.5℃ after medication); cumulative time for fever recovery(body temperature recovering to normal and lasting more than 24 h without medication); cumulative time for rash disappearance(without new rashes or ulcers appearing and the original ones fading away); and cumulative time for mouth ulcer disappearance. RESULTS: For the onset time of the antifebrile effect, there was no statistical difference between groups A and B(P>0.05) and groups B and C(P> 0.05). However, there was a statistical difference between groups A and C(P<0.05), and the effect in group C was the best. For the cumulative time for rash disappearance, there was no statistical difference between groups A and B(P>0.05). There were statistical differences between groups A and C, and groups B and C(P<0.05), and the effect in group C was the best. For the cumulative time for mouth ulcers disappearance, there were no statistical differences among the three groups(P>0.05). CONCLUSION: Reduning injection with Western Medicine for symptomatic treatment is most effective for mild HFMD. No adverse reactions were observed.Guoliang Zhang Jie Zhao Liyun He Shiyan Yan Ziqiang Zhuo Haojie Zheng Yongping Mu Shuangjie Li Xi Zhang Jihan Huang Xiuhui Li Jianping Liu Hehe Wan Chaoyu Wei Wei Xiao 2013Journal of Traditional Chinese Medicine2013,33,6:7
Effect of quercetin on the expression of Bcl-2/Bax apoptotic proteins in endometrial cells of lipopolysaccharide-induced-abortion mice显示文摘OBJECTIVE: To explore the effect of quercetin on the expressions of Bcl-2/Bax apoptotic proteins in endometrial cells in mice with abortion induced by lipopolysaccharide.METHODS: For in vivo experiment, twenty five Kunming mice were randomly divided into five groups at day 4 of pregnancy, with 5 mice per group. The mice were treated with lipopolysaccharide(LPS)through tail vein intravenous injection at day 4 of pregnancy, followed by different concentrations of quercetin by oral gavage consecutively at days 5 to6 of pregnancy. On day 7 of gestation, the mice were sacrificed and the histopathological changes of the uterus tissues were observed. Immunohistochemical staining was applied to the detection of Bcl-2/Bax apoptotic proteins in the endometrial cells. For in vitro experiment, the primary endometrial cells werecultured using a uterus tissue mass culturing method sampled at day 4.5 of pregnancy. The cells were treated with LPS with or without different dosages of quercetin, respectively, for 12 h after 80% confluence. The expression of Bcl-2/Bax apoptotic proteins were detected by western blotting.RESULTS: Both the in vivo and in vitro experiments showed decreased expression of Bcl-2 and enhanced expression of Bax after LPS treatment, leading to a decreased Bcl-2/Bax ratio. The expression of Bcl-2 significantly increased while the expression of Bax was significantly elevated, in the LPS plus quercetin group compared to the LPS only group.CONCLUSION: These results suggest that quercetin has protective effect by partially regulating the expression of Bcl-2/Bax proteins, which in turn inhibits endometrial cell apoptosis and benefits the embryo implantation.Wang Xiaodan Yan Yongping Yang Liu Li Mu Zhong Xiuhui 2016Journal of Traditional Chinese Medicine2016,36,6:7
Regional difference of annual precipitation and discharge variation over west China during the last 50 years显示文摘Using annual precipitation and discharge data measured in the past five decades, this paper analyzed the regional differences over west China in terms of climate and discharge variations, and investigated the relationship between the regional characteristics and the activities of South and East Asian sum-mer monsoon. Results revealed that the precipitation and discharge in the upper reaches of the Yellow River (Central West China) have a negative correlation with those in Xinjiang (northwest China) and the Yarlung Zangbo River (the upper reaches of the Brahmaputra Rive, southwest China) regions. The geographical patterns of precipitation and discharge variations are different over west China,i.e. the regional climate displays the alteration of dry-wet-dry or wet-dry-wet from north to south in west China. The negative correlation of annual discharges between Xinjiang and the upper reaches of the Yellow River is found statistically significant in the decadal scale, and that between the Yarlung Zangbo River and the upper reaches of the Yellow River is found active in the interannual scale. The regional char-acteristics indicate that the discharge/precipitation variations in the upper reaches of the Yellow River are dominated by the East Asian summer monsoon while their variations in Xinjiang are affected by both the west wind and East Asian summer monsoon.DING YongJian YE BaiSheng HAN TianDing SHEN YongPing LIU ShiYin 2007Science China Earth Sciences2007,50,6:7
FAT1, a direct transcriptional target of E2F1, suppresses cell proliferation, migration and invasion in esophageal squamous cell carcinoma显示文摘Objective: Growing evidence indicates that FAT atypical cadherin 1(FAT1) has aberrant genetic alterations and exhibits potential tumor suppressive function in esophageal squamous cell carcinoma(ESCC). However, the role of FAT1 in ESCC tumorigenesis remains not well elucidated. The aim of this study was to further investigate genetic alterations and biological functions of FAT1, as well as to explore its transcriptional regulation and downstream targets in ESCC.Methods: The mutations of FAT1 in ESCC were achieved by analyzing a combined study from seven published genomic data, while the copy number variants of FAT1 were obtained from an analysis of our previous data as well as of The Cancer Genome Atlas(TCGA) and Cancer Cell Line Encyclopedia(CCLE) databases using the cBioPortal. The transcriptional regulation of FAT1 expression was investigated by chromatin immunoprecipitation(ChIP) and the luciferase reporter assays. In-cell western, Western blot and reverse transcription-quantitative polymerase chain reaction(RT-qPCR) were used to assess the indicated gene expression. In addition, colony formation and Transwell migration/invasion assays were employed to test cell proliferation, migration and invasion. Finally, RNA sequencing was used to study the transcriptomes. Results: FAT1 was frequently mutated in ESCC and was deleted in multiple cancers. Furthermore, the transcription factor E2 F1 occupied the promoter region of FAT1, and depletion of E2 F1 led to a decrease in transcription activity and mRNA levels of FAT1. Moreover, we found that knockdown of FAT1 promoted KYSE30 and KYSE150 cell proliferation, migration and invasion;while overexpression of FAT1 inhibited KYSE30 and KYSE410 cell proliferation, migration and invasion. In addition, knockdown of FAT1 led to enrichment of the mitogen-activated protein kinase(MAPK) signaling pathway and cell adhesion process. Conclusions: Our data provided evidence for the tumor suppressive function of FAT1 in ESCC cells and elucidated the transcriptional regulation of FAT1 by E2 F1, which may facilitate the understanding of molecular mechanisms of the progression of ESCC.Yu Wang Guangchao Wang Yunping Ma Jinglei Teng Yan Wang Yongping Cui Yan Dong Shujuan Shao Qimin Zhan Xuefeng Liu 2019Chinese Journal of Cancer Research2019,31,4:6
