| 1 | Clinical Characteristics of Children with Coronavirus Disease 2019 in Hubei, China显示文摘Since December 2019,COVID-19 has occurred unexpectedly and emerged as a health problem worldw ide.Despite the rapidly increasing number of cases in subsequent weeks,the clinical characteristics of pediatric cases are rarely described.A cross-sectional multicenter study was carried out in 10 hospitals across Hubei province.A total of 25 confirmed pediatric cases of COVID-19 were collected.The demographic data,epidemiological history,underlying discascs,clinical mani festations,laboratory and radiological data,treatments,and outcomes were analyzcd.Of 25 hospitalized patients with COVID-19,the boy to girl ratio was 1.27:1.The median age was 3 years.COVID-19 cases in children aged<3 years,3-6 years,and≥6-years patients were 10(40%),6(24%),and 9(36%),respectively.The most common symptoms at onset of ilness were fever(13[52%]),and dry cough(11[44%]).Chest CT images showed essential normal in 8 cases(33.3%),unilateral involvement of lungs in 5 cases(20.8%),and bilateral involvement in 11 cases(45.8%).Clinical diagnoses included upper respiratory tract infection(n=8),mild pneumonia(n=15),and critical cases(n=2).Two critical cases(8%)were given invasive mechanical ventilation,corticosteroids,and immunoglobulin.The symptoms in 24(96%)of 25 patients were alleviated and one patient had been discharged.It was concluded that children were susceptible to COVID-19 like adults,while the clinical presentations and outcomes were more favorable in children.However,children less than 3 years old accounted for majority cases and critical cases lied in this age group,which demanded extra attentions during home caring and hospitalization treatment. | Fang ZHENG Chun LIAO Qi-hong FAN Hong-bo CHEN Xue-gong ZHAO Zhong-guo XIE Xi-lin LI Chun-xi CHEN Xiao-xia LU Zhi-sheng LIU Wei LU Chun-bao CHEN Rong JIAO Ai-ming ZHANG Jin-tang WANG Xi-wei DING Yao-guang ZENG Li-ping CHENG Qing-feng HUANG Jiang WU Xi-chang LUO Zhu-jun WANG Yan-yan ZHONG Yan BAI Xiao-yan WU Run-ming JIN | 2020 | Current Medical Science2020,40,2: | 10 |
| 2 | Generation of GGTA1 biallelic knockout pigs via zinc-finger nucleases and somatic cell nuclear transfer显示文摘Genetically modified pigs are valuable models of human disease and donors of xenotransplanted organs.Conventional gene targeting in pig somatic cells is extremely inefficient.Zinc-finger nuclease(ZFN)technology has been shown to be a powerful tool for efficiently inducing mutations in the genome.However,ZFN-mediated targeting in pigs has rarely been achieved.Here,we used ZFNs to knock out the porcineα-1,3-galactosyl-transferase(GGTA1)gene,which generates Gal epitopes that trigger hyperacute immune rejection in pig-to-human transplantation.Primary pig fibroblasts were transfected with ZFNs targeting the coding region of GGTA1.Eighteen mono-allelic and four biallelic knockout cell clones were obtained after drug selection with efficiencies of 23.4%and 5.2%,respectively.The biallelic cells were used to produce cloned pigs via somatic cell nuclear transfer(SCNT).Three GGTA1 null piglets were born,and one knockout primary fibroblast cell line was established from a cloned fetus.Gal epitopes on GGTA1 null pig cells were completely eliminated from the cell membrane.Functionally,GGTA1 knockout cells were protected from complement-mediated immune attacks when incubated with human serum.This study demonstrated that ZFN is an efficient tool in creating gene-modified pigs.GGTA1 null pigs and GGTA1 null fetal fibroblasts would benefit research and pig-to-human transplantation. | BAO Lei CHEN HaiDe JONG UiMyong RIM CholHo LI WenLing LIN XiJuan ZHANG Dan LUO Qiong CUI Chun HUANG HeFeng ZHANG Yan XIAO Lei FU ZhiXin | 2014 | Science China(Life Sciences)2014,57,2: | 9 |

| 4 | Validity of the Chinese Version of the Patient-Generated Subjective Global Assessment (PG-SGA) in Lung Cancer Patients显示文摘The prevalence of malnutrition is high among oncology patients in China. Although the Patient- Generated Subjective Global Assessment (PG-SGA) is widely recognized as an important nutritional assessment tool, it has not been validated for Chinese cancer patients. The purpose of this study was to conduct an analysis of nutrition screening and assessment data to validate the Chinese version of the PG-SGA in Chinese patients with lung cancer. This was an observational, cross-sectional study of 2,000 consecutive adult patients with lung cancer treated at several tertiary hospitals in China. Anthropometric and patient descriptive data were collected. The PG-SGA generated a score for the nutritional risk and a global rating for the nutritional status. The internal consistency, external consistency, and construct validity were evaluated, and a known-groups comparison and principal component analyses were conducted to evaluate the reliability and validity of the PG-SGA. The known-groups comparison demonstrated the ability of the PG-SGA to differentiate lung cancer patients of different nutritional and body mass index (BMI) groups. A confirmatory factor analysis supported the original four-factor structure of the PG-SGA. In terms of the internal and external consistency, the PG-SGA demonstrated intra-class correlation coefficients (ICC) up to 0.987. Alternative form validity was also demonstrated between Box 4 of the PG-SGA and the Karnofsky Performance Status score, with excellent external consistency (r = 0.972, P < 0.001). The construct validity was supported, and selected questionnaire dimensions were evident in the principal component analysis. Significant Spearman correlations (P < 0.001) were demonstrated. The Chinese version of the PG-SGA demonstrated significant reliability and sufficient exploratory properties to support its validity. It seems to be a valid tool that can be used to access the nutritional status of Chinese lung cancer patients. | 5 | Validity of the Chinese Version of the Patient-Generated Subjective Global Assessment (PG-SGA) in Gastric Cancer Patients显示文摘Purpose Malnutrition is severe among gastric cancer patients in China. While the Patient-Generated Subjective Global Assessment (PG-SGA) is a widely used nutritional assessment tool, the validation for Chinese patients with gastric cancer has not been performed yet. The aim of this research is to validate the Chinese version of the PGSGA in Chinese gastric cancer patients by conducting analyses of nutrition screening and assessment data. Methods Two thousand consecutive adult patients with gastric cancer were included in this observational, cross-sectional study in several tertiary hospitals in China. A survey was made among those patients, which included several questionnaires and some anthropometric parameters. We calculated the parameters to validate the use of PG-SGA categorizing the nutritional status of gastric cancer patients. Then we performed statistical analysis to evaluate the internal consistency, external consistency, and construct validity. Results This study verified the ability of the PG-SGA to differentiate gastric cancer patients into different nutritional groups. The confirmatory factor analysis verified the original four-factor structure of the PG-SGA. The internal and external consistency was measured by a concordance analysis which showed an intraclass correlation coefficients (ICC) of 0.987. The correlations of the Box 4 score with the Karnofsky performance status score demonstrated alternative form validity with good external consistency (Pearson, r = 0.643, P < 0.001). All three anthropometric dimensions of the PG-SGA were supported by principal component analysis. Spearman correlations (P < 0.001) were significant in the majority of tests. | 15 | Environmental microcystin exposure triggers the poor prognosis of prostate cancer: Evidence from case-control, animal, and in vitro studies显示文摘Microcystin-leucine-arginine(MC-LR)is positively linked with multiple cancers in humans.However,the association between MC-LR and the risk and prognosis of prostate cancer has not been conducted in epidemiological studies.No reported studies have linked MC-LR exposure to the poor prognosis of prostate cancer by conducting experimental studies.The content of MC-LR was detected in most of the aquatic food in wet markets and supermarkets in Nanjing and posed a health risk for consumers.MC-LR levels in both prostate cancer tissues and serum were significantly higher than controls.The adjusted odds ratio(OR)for prostate cancer risk by serum MC-LR was 1.75(95%CI:1.21-2.52)in the whole subjects,and a positive correlation between MC-LR and advanced tumor stage was observed.Survival curve analysis indicated patients with higher MC-LR levels in tissues exhibited poorer overall survival.Human,animal,and cell studies confirmed that MC-LR exposure increases the expression of estrogen receptor-α(ERα)and promotes epithelial-mesenchymal transition(EMT)in prostate cancer.Moreover,MC-LR-induced decreased E-cadherin levels,increased vimentin levels,and increased migratory and invasive capacities of prostate cancer cells were markedly suppressed upon ERαknockdown.MC-LR-induced xenograft tumor growth and lungmetastasis in BALB/c nude mice can be effectively alleviated with ERαknockdown.Our data demonstrated that MC-LR upregulated vimentin and downregulated E-cadherin through activating ERα,promoting migration and invasion of prostate cancer cells.Our findings highlight the role of MC-LR in prostate cancer,providing new perspectives to understand MC-LR-induced prostatic toxicity. | Chun Pan Haixiang Qin Minghao Yan Xuefeng Qiu Wenyue Gong Wenxin Luo Hongqian Guo Xiaodong Han | 2023 | Journal of Environmental Sciences2023,,5: | 1 |
| 16 | Haemorrhage risk of brain arteriovenous malformation during pregnancy and puerperium显示文摘Background This study aimed to assess whether pregnancy and puerperium were associated with the risk of brain arteriovenous malformation(bAVM)haemorrhage.Methods A retrospective review was conducted in Xiangya Hospital,Central South University from January 2012 to December 2021.A case-crossover design was adopted to calculate the incidence density of bAVM-related haemorrhage among female patients in risk(pregnancy and puerperium)and control(non-pregnancy and non-puerperium)periods,according to four scenarios observed in different populations scenario I:patients with haemorrhagic bAVM of all ages;scenario II:patients with haemorrhagic bAVM of all ages,with at least one previous pregnancy;scenario III:patients with haemorrhagic bAVM who are of reproductive age(15-45 years);scenario IV:patients with haemorrhagic bAVM of reproductive age(15-45 years),with at least one previous pregnancy.Next,a comprehensive literature aggregation(up to April 2022)was performed for evidence synthesis.Results Among the 311 female patients with haemorrhagic bAVM,a significant haemorrhage risk during pregnancy and puerperium was found in Scenarios I(relative risk[RR],2.08;95%CI,1.28 to 3.39),II(RR,3.21;95%CI,1.95 to 5.31)and IV(RR,2.92;95%CI,1.73 to 4.93);however,a suggestive risk was found in scenario III(RR,1.62;95%CI,0.99 to 2.67).Evidence synthesis revealed a consistent haemorrhage risk among patients of all ages(RR,3.15;95%CI,1.93 to 5.15)and those of reproductive age(RR,1.29;95%CI,0.89 to 1.86).Conclusion Compared with most previous studies,a higher but relatively moderate risk for bAVM-related haemorrhage was identified during pregnancy and puerperium.Individualised prevention and treatment strategies should be preferred when neurosurgeons make clinical decisions. | Junyu Liu Honghao Zhang Chun Luo Yuxin Guo Yifeng Li Dun Yuan Weixi Jiang Junxia Yan | 2023 | Stroke & Vascular Neurology2023,8,4: | 1 |
| 17 | Differential diagnosis of high-grade squamous intraepithelial lesions and benign atrophy in older women using p16 immunocytochemistry显示文摘Background For cervical cancer screening,routine cytology has a high specificity but a lower sensitivity.In older women,atrophy,which may mimic HSIL,presents a diagnostic challenge.p16 is a widely used biomarker for histological diagnosis of HSIL.Our objective was to evaluate PathCIN®p16INK4a immunocytology in identification of high grade dysplasia vs.benign atrophy.Methods As part of a multi-center screening program,3351 women were co-tested by p16 immunocytology.Among women referred for colposcopy on basis of cytology and high-risk HPV status,those with atrophy were older than the population screened(52 vs.43 years).Cases from older women with atrophy(n=116)and controls without atrophy(n=47)were identified by re-examination of Pap smears.The detection of CIN2+was compared for p16,cytology and HR-HPV results.Results The sensitivity of routine cytology(≥LSIL)was much lower for cases with atrophy(17%)than non-atrophic cases(75%).The sensitivity of p16 immunocytology and of HR-HPV testing was high(88%–100%)both with and without atrophy.The specificity of routine cytology(≥LSIL)was higher for cases with atrophy(79%)than non-atrophic cases(38%).The specificity of p16 immunocytology was high(88–95%)and the specificity of HR-HPV testing was low(31%–33%)both with and without atrophy.Combining p16 with HPV testing and/or routine cytology had no benefit,as compared to p16 staining alone.Conclusions p16 immunocytology compares favorably with routine cytology and HPV testing in the differential diagnosis of HSIL and benign atrophy.It is more sensitive than cytology for atrophic specimens,and is more specific than HPV testing.p16 immunocytology may decrease the need for colposcopy referrals and could be a useful tool for early detection of cervical cancer in peri-and post-menopausal women,who are more likely to have HSIL coexisting with atrophy. | Peisha Yan Hui Du Chun Wang Fangbin Song Xia Huang Yingjie Luo Ruifang Wu | 2021 | Gynecology and Obstetrics Clinical Medicine2021,1,1: | 0 |
| 18 | Lentiviral-mediated siRNA against osteopotin in U251 glioma cell line显示文摘Background:It has been reported that the expression of SPP1, the mRNA for osteopontin (OPN), is increased in a number of transformed cell lines and some tumors. Increased serum and plasma OPN levels were associated with advanced-stage carcinomas of the lung, breast, colon and prostate. The extent of OPN expression may be correlated with the malignancy degree of gliomas, but the specific role of OPN in the malignancy of gliomas remains to be clarified. Methods:In this study, we used a lentiviral system, which could induce a long-lasting down-regulation of gene expression, to knock down the OPN gene in human glioma cell line U251. And then we observed the proliferation and apoptosis of glioma cells. Results:OPN mRNA and protein were successfully knocked down in U251 cells, and the expression of caspase-3 and caspase-9 were increased about by 2 folds in OPN down-regulated U251 cells. 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium (MTT) assay showed that OPN down-regulation did not affect the proliferation of U251 cells.
