| 1 | Humoral immune response to circulating SARS-CoV-2 variants elicited by inactivated and RBD-subunit vaccines显示文摘SARS-CoV-2 variants could induce immune escape by mutations on the receptor-binding domain(RBD)and N-terminal domain(NTD).Here we report the humoral immune response to circulating SARS-CoV-2 variants,such as 501Y.V2(B.1.351),of the plasma and neutralizing antibodies(NAbs)elicited by CoronaVac(inactivated vaccine),ZF2001(RBD-subunit vaccine)and natural infection.Among 86 potent NAbs identified by high-throughput single-cell VDJ sequencing of peripheral blood mononuclear cells from vaccinees and convalescents,near half anti-RBD NAbs showed major neutralization reductions against the K417N/E484K/N501Y mutation combination,with E484K being the dominant cause.VH3-53/VH3-66 recurrent antibodies respond differently to RBD variants,and K417N compromises the majority of neutralizing activity through reduced polar contacts with complementarity determining regions.In contrast,the 242–244 deletion(242–244Δ)would abolish most neutralization activity of anti-NTD NAbs by interrupting the conformation of NTD antigenic supersite,indicating a much less diversity of anti-NTD NAbs than anti-RBD NAbs.Plasma of convalescents and CoronaVac vaccinees displayed comparable neutralization reductions against pseudo-and authentic 501Y.V2 variants,mainly caused by E484K/N501Y and 242–244Δ,with the effects being additive.Importantly,RBD-subunit vaccinees exhibit markedly higher tolerance to 501Y.V2 than convalescents,since the elicited anti-RBD NAbs display a high diversity and are unaffected by NTD mutations.Moreover,an extended gap between the third and second doses of ZF2001 leads to better neutralizing activity and tolerance to 501Y.V2 than the standard three-dose administration.Together,these results suggest that the deployment of RBD-vaccines,through a third-dose boost,may be ideal for combating SARS-CoV-2 variants when necessary,especially for those carrying mutations that disrupt the NTD supersite. | Yunlong Cao Ayijiang Yisimayi Yali Bai Weijin Huang Xiaofeng Li Zhiying Zhang Tianjiao Yuan Ran An Jing Wang Tianhe Xiao Shuo Du Wenping Ma Liyang Song Yongzheng Li Xiang Li Weiliang Song Jiajing Wu Shuo Liu Xuemei Li Yonghong Zhang Bin Su Xianghua Guo Yangyang Wei Chuanping Gao Nana Zhang Yifei Zhang Yang Dou Xiaoyu Xu Rui Shi Bai Lu Ronghua Jin Yingmin Ma Chengfeng Qin Youchun Wang Yingmei Feng Junyu Xiao Xiaoliang Sunney Xie | 2021 | Cell Research2021,31,7: | 12 |
| 2 | TPX2 knockdown suppressed hepatocellular carcinoma cell invasion via inactivating AKT signaling and inhibiting MMP2 and MMP9 expression显示文摘Objective: Targeting protein for Xenopus kinesin-like protein 2(TPX2) is a nuclear proliferation-related protein that plays a critical role in the formation of mitotic spindle. High expression of TPX2 has been observed in several types of tumors. However, the role of TPX2 in hepatocellular carcinoma(HCC) remains unclear. Our study aimed to investigate the effect of TPX2 on HCC cell invasion.Methods: The immortalized normal human liver cell line L02 and six HCC cell lines including SMMC-7721, BEL-7402, Huh-7, HepG2, Hep3 B and SKHep1 were subjected to qRT-PCR and western blot for TPX2 mRNA and protein, respectively. Furthermore, TPX2 small interfering RNA(siRNA) was used to knock down TPX2 expression in SMMC-7721 and HepG2 cells. Cell proliferation and invasion were determined by MTT and transwell assays. Otherwise, expression of p-AKT, MMP2 and MMP9 were evaluated by western blot in SMMC-7721 cells.Results: The expression of TPX2 in HCC cell lines was markedly higher than that in normal human liver cell line. TPX2 knockdown using a specific TPX2-siRNA reduced the number of invaded cells and inhibited cell proliferation in SMMC-7721 and HepG2 cells. | 3 | Optimization of a new animal glue binder system cured by CO_2 for use in foundry显示文摘A new sand binder system cured by CO2 was prepared based on the animal bone glue.To overcome the disadvantages of animal glue such as agglomeration at room temperature,high energy consumption and low efficiency,an alkaline decomposition process was selected,and certain modifier was used to modify the performance of the animal glue binder.For the alkaline decomposition,NaOH was used as the catalyst with an addition of 4wt.% to the animal glue.A modifier was determined through the orthogonal experiment with a weight ratio of glycerin:glycol:dextrin:animal glue = 9:16:15:100,and the optimal modification reaction should be performed at 75 ℃ with a reaction time of 90 min.Ca(OH)2 was used as a promoter;the optimal CO2 gas flux blowing into the sand was 0.7 m3.h-1 for a duration of 60s under the experimental conditions.Results show that an original strength above 0.7 MPa and a final strength about 4.2 MPa can be achieved,which could meet the requirement of rapid moulding and core-making for foundry.The new binder was characterized and analyzed by means of IR,and the modification and CO2-cured mechanisms of this animal glue binder were also discussed. | Liu Weihua Zhang Yilin Li Yingmin Liu Xiuling | 2012 | China Foundry2012,9,4: | 5 |

Firstly, the data association of measurement-to-target is formulated as an integer programming problem. Through using the linear programming (LP) based branchand-bound method and adjusting the constraint conditions, an optimal set integer programming (OSIP) algorithm is then proposed for tracking multiple non-maneuvering targets in clutter. For the case of maneuvering targets, this paper introduces the OSIP algorithm into the filtering step of the interacting multiple model (IMM) algorithm resulting in the IMM based on OSIP algorithm. | 19 | Annotation and Joint Extraction of Scientific Entities and Relationships in NSFC Project Texts显示文摘Aiming at the lack of classification and good standard corpus in the task of joint entity and relationship extraction in the current Chinese academic field, this paper builds a dataset in management science that can be used for joint entity and relationship extraction, and establishes a deep learning model to extract entity and relationship information from scientific texts. With the definition of entity and relation classification, we build a Chinese scientific text corpus dataset based on the abstract texts of projects funded by the National Natural Science Foundation of China(NSFC) in 2018–2019. By combining the word2vec features with the clue word feature which is a kind of special style in scientific documents, we establish a joint entity relationship extraction model based on the Bi LSTM-CNN-CRF model for scientific information extraction. The dataset we constructed contains 13060 entities(not duplicated) and 9728 entity relation labels. In terms of entity prediction effect, the accuracy rate of the constructed model reaches 69.15%, the recall rate reaches 61.03%, and the F1 value reaches 64.83%.
