| 2 | Overexpression of a dominant-negative mutant of SIRT1 in mouse heart causes cardiomyocyte apoptosis and early-onset heart failure显示文摘SIRT1,a mammalian ortholog of yeast silent information regulator 2(Sir2),is an NAD+-dependent protein deacetylase that plays a critical role in the regulation of vascular function.The current study aims to investigate the functional significance of deacetylase activity of SIRT1 in heart.Here we show that the early postnatal hearts expressed the highest level of SIRT1deacetylase activity compared to adult and aged hearts.We generated transgenic mice with cardiac-specific expression of a dominant-negative form of the human SIRT1(SIRT1H363Y),which represses endogenous SIRT1 activity.The transgenic mice displayed dilated atrial and ventricular chambers,and died early in the postnatal period.Pathological,echocardiographic and molecular phenotype confirmed the presence of dilated cardiomyopathy.Terminal deoxynucleotidyl transferase-mediated dUTP nick-end-labeling analysis revealed a greater abundance of apoptotic nuclei in the hearts of transgenic mice.Furthermore,we show that cardiomyocyte apoptosis caused by suppression of SIRT1 activity is,at least in part,due to increased p53acetylation and upregulated Bax expression.These results indicate that dominant negative form of SIRT1(SIRT1H363Y)overexpression in mouse hearts causes cardiomyocyte apoptosis and early-onset heart failure,suggesting a critical role of SIRT1 in preserving normal cardiac development during the early postnatal period. | MU WenLi ZHANG QingJun TANG XiaoQiang FU WenYan ZHENG Wei LU YunBiao LI HongLiang WEI YuSheng LI Li SHE ZhiGang CHEN HouZao LIU DePei | 2014 | Science China(Life Sciences)2014,57,9: | 12 |

| 4 | Correlation of serum vitamin D, adipose tissue vitamin D receptor, and peroxisome proliferator-activated receptor γ in women with gestational diabetes mellitus显示文摘Background:Gestational diabetes mellitus(GDM)is a common complication during pregnancy.Obesity and overweight are closely related to metabolic diseases and diabetes.However,the role of adipose tissue in the pathogenesis of GDM remains to be studied.The aim of this study was to investigate the correlation of vitamin D(VD)levels,VD receptor(VDR),and peroxisome proliferatoractivated receptor g(PPARg)expression with GDM in overweight or obese women.Methods:One hundred and forty pregnant women with full-term single-birth cesarean-section were selected as the study subjects and grouped(70 GDM women,including 35 non-overweight/non-obese women[group G1]and 35 women with overweight or obesity[group G2];70 pregnant women with normal glucose tolerance,including 35 non-overweight/non-obese women[group N1]and 35 overweight/obese women[group N2]).The levels of serum VD,blood biochemistry,and adiponectin were compared in these women.Subcutaneous adipose tissue was isolated from the abdominal wall incision.VDR and PPARg messenger RNA(mRNA)transcript levels in these adipose tissues were quantified by real-time polymerase chain reaction.The differences between the levels of PPARg protein and phosphorylated PPARg Ser273 were detected by Western blotting.Results:The serum VD level ofGDM women was lower in comparison to that of women with normal glucose tolerance(G1 vs.N1:20.62±7.87 ng/mL vs.25.85±7.29 ng/mL,G2 vs.N2:17.06±6.74 ng/mL vs.21.62±7.18 ng/mL,P<0.05),and the lowest in overweight/obese GDM women.VDR and PPARg mRNA expression was higher in the adipose tissues of GDM women in comparison to that of women with normal glucose tolerance(VDR mRNA:G1 vs.N1:210.00[90.58–311.46]vs.89.34[63.74–159.92],G2 vs.N2:298.67[170.84–451.25]vs.198.28[119.46–261.23],PPARg mRNA:G1 vs.N1:100.72[88.61–123.87]vs.87.52[66.37–100.04],G2 vs.N2:117.33[100.08–149.00]vs.89.90[76.95–109.09],P<0.05),and their expression was the highest in GDM+overweight/obese women.VDR mRNA levels positively correlated with the pre-pregnancy body mass index(BMI),pre-delivery BMI,fasting blood glucose(FBG),homeostasis model assessment of insulin resistance(HOMA-IR),and PPARg mRNA while it negatively correlated with the VD and the adiponectin levels(r=0.395,0.336,0.240,0.190,0.235,–0.350,–0.294,respectively,P<0.05).The degree of PPARg Ser273 phosphorylation increased in obese and GDM pregnant women.PPARg mRNA levels positively correlated with pre-pregnancy BMI,pre-delivery BMI,FBG,HOMA-IR,serum total cholesterol,triglyceride,free fatty acid,and VDR mRNA,while it negatively correlated with the VD and adiponectin levels(r=0.276,0.199,0.210,0.230,0.182,0.214,0.270,0.235,–0.232,–0.199,respectively,P<0.05).Conclusions:Both GDM and overweight/obese women had decreased serum VD levels and up-regulated VDR and PPARg mRNA expression in adipose tissue,which was further higher in the overweight or obese women with GDM.VDmay regulate the formation and differentiation of adipocytes through the VDR and PPARg pathways and participate in the occurrence of GDM. | Hui-Yan Wang Guang-Tong She Li-Zhou Sun Hao Lu Yin-Pin Wang Jun Miao Ke-Zhuo Liu Cai-Feng Sun Hui-Hui Ju | 2019 | Chinese Medical Journal2019,,21: | 9 |
| 5 | Characterization of Aroma-Active Components and Antioxidant Activity Analysis of E-jiao(Colla CoriiAsini)from Different Geographical Origins显示文摘E-jiao(Colla Corii Asini,CCA)has been widely used as a healthy food and Chinese medicine.Although authentic CCA is characterized by its typical sweet and neutral fragrance,its aroma components have been rarely investigated.This work investigated the aroma-active components and antioxidant activity of 19 CCAs from different geographical origins.CCA extracts obtained by simultaneous distillation and extraction were analyzed by gas chromatography-mass spectrometry(GC-MS),gas chromatography-olfactometry(GC-O)and sensory analysis.The antioxidant activity of CCAs was determined by ABTS and DPPH assays.A total of 65 volatile compounds were identified and quantified by GC-MS and 23 aroma-active compounds were identified by GC-O and aroma extract dilution analysis.The most powerful aroma-active compounds were identified based on the flavor dilution factor and their contents were compared among the 19 CCAs.Principal component analysis of the 23 aroma-active components showed 3 significant clusters.Canonical correlation analysis between antioxidant assays and the 23 aroma-active compounds indicates strong correlation(r=0.9776,p=0.0281).Analysis of aroma-active components shows potential for quality evaluation and discrimination of CCAs from different geographical origins. | Shan Zhang Lu Xu Yang-Xi Liu Hai-Yan Fu Zuo-Bing Xiao Yuan-Bin She | 2018 | Natural Products and Bioprospecting2018,8,2: | 9 |

| 8 | The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. | 9 | Expression of reconstructive hF Ⅷ in the hrDNA by using hrDNA targeting vector显示文摘Our lab has constructed a new nonviral vec-tor—hrDNA targeting vector(pHrneo). pHrneo is a human derived vector that can target gene into human ribosomal DNA(hrDNA) locus. In this study, we inserted expression cassette of reconstructive hF Ⅷ (hFVIII-BDDAK39) to pHrneo to construct targeting vector: pHrneo-BDDAK39. Through electroporation of pHrneo-BDDAK39 into HT1080 cells, we identified the homologous recombinants by PCR and Southen blotting, and tested the expression of hFVIII- BDDAK39 in the hrDNA locus. The hFⅧ-BDDAK39 was successfully targeted into hrDNA locus of HT-1080 by pHrneo-BDDAK39, and the efficiency of site-specific inte-gration was 2.0×10?5. hFⅧ-BDDAK39 in hrDNA locus of HT-1080 is found to be able to express efficiently (32±5 ng·106 cells?1·24 h?1). | 12 | Phaeocystis globosa Bloom Monitoring: Based on P. globosa Induced Seawater Viscosity Modification Adjacent to a Nuclear Power Plant in Qinzhou Bay, China显示文摘The mucus produced by the outbreak of Phaeocystis globosa in the adjacent waters of the Fangchenggang Nuclear Power Plant(FCGNPP) in China has blocked the entrance of the cooling water filtration system of the FCGNPP, and posed a threat to the safe operation of the FCGNPP. At present, there is no related research on whether the changes in seawater viscosity could be used as a new method for monitoring and providing early warning of P. globosa tide. During a complete red tide cycle, the temporal and spatial changes in the hydrological conditions(temperature, salinity, dissolved oxygen), chlorophyll-a(Chl-a), composition and abundance of phytoplankton, number and size of P. globose colonies, concentration of transparent exopolymer particles(TEP) and the seawater viscosity were measured in this study. The results indicate that there was an extremely significant negative correlation between the physical seawater viscosity η_(T,S) and temperature, and the correlation coefficient reached-0.998. The biological seawater viscosity η_(Bio) was positively correlated with the number of P. globosa colonies, and the correlation coefficients is 0.747. Because the increase in phytoplankton abundance, especially during the outbreak of P. globosa and a large amount of mucus produced by the colonies could significantly increase seawater viscosity, we suggest that biological factors were the main reasons for the increase in seawater viscosity. | 15 | Photocrosslinking silver nanoparticles–aloe vera–silk fibroin composite hydrogel for treatment of full-thickness cutaneous wounds显示文摘Damage to the skin causes physiological and functional issues.The most effective treatment approach is the use of wound dressings.Silk fibroin(SF)is a promising candidate biomaterial for regulating wound healing;however,its antibacterial properties and biological activity must be further improved.In this study,a photocrosslinking hydrogel was developed to treat full-thickness cutaneous wounds.The composite hydrogel(Ag-AV-SF hydrogel)was prepared by introducing the silver nanoparticles(AgNPs)and aloe vera(AV)as the modifiers.In vitro study exhibited great antibacterial ability,biocompatibility and cell-proliferation and-migration-promoting capacities.It also showed the pH-response releasing properties which release more AgNPs in a simulated chronic infection environment.The healing effect evaluation in vivo showed the healing-promoting ability of the Ag-AV-SF hydrogel was stronger than the single-modifiers groups,and the healing rate of it reached 97.02%on Day 21,higher than the commercial wound dressing,silver sulfadiazine(SS)cream on sale.Additionally,the histological and protein expression results showed that the Ag-AV-SF hydrogel has a greater effect on the pro-healing regenerative phenotype with M2 macrophages at the early stage,reconstructing the blood vessels networks and inhibiting the formation of scars.In summary,the Ag-AV-SF hydrogel developed in this study had good physical properties,overwhelming antibacterial properties,satisfactory biocompatibility and significantly promoting effect on cell proliferation,migration and wound healing.Overall,our results suggest that the Ag-AV-SF hydrogel we developed has great potential for improving the wound healing in clinical treatment. | Yangkun Liu JinChuan Fan MingQi Lv Kepeng She Jiale Sun Qingqing Lu Changhao Han SongTao Ding Shuang Zhao GuiXue Wang YuChan Zhang GuangChao Zang | 2021 | Regenerative Biomaterials2021,8,6: | 2 |
| 16 | Sensitivity of P300 auditory event-related potentials for assessing cognitive impairment in elderly type 2 diabetic patients显示文摘BACKGROUND:In previous studies, cognitive function in elderly type 2 diabetic patients was evaluated by psychometric tests. These studies have confirmed that P300 event-related potential is an objective way of assessing cognitive function. OBJECTIVE: To analyze the objectivity of P300 for assessment of cognitive function in elderly type 2 diabetic patients. DESIGN, TIME AND SETTING: This case-control experiment was performed at the Department of Endocrinology of the Fourth Affiliated Hospital, Guangxi Medical University from January 2004 to December 2006. PARTICIPANTS: Seventy-two patients (38 males and 34 females) with type 2 diabetes mellitus were enrolled in this study. The patients were divided according to those with diabetes alone (diabetes alone group) (n=38) and those with diabetes and cerebral ischemia (diabetes and cerebral ischemia group) (n=34). A further 31 healthy individuals (16 males and 15 females), who received health examinations over the same period, were included as normal controls (normal control group). METHODS: All subjects were assessed by Mini-Mental State Examination (MMSE). Abnormalities in cognitive functions were identified by analyzing the auditory P300 event-related potentials. MAIN OUTCOME MEASURES: Auditory event-related potentials and MMSE scores. Multiple linear regression analysis was conducted using the 'enter method' with the 72 elderly patients with type 2 diabetes mellitus. P3 latency, P3 amplitude and N2 latency served as dependent variables. Age, sex, education, course of the disease, glycosylated hemoglobin, and ischemic brain damage were used as independent variables. RESULTS: No significant difference in scores of MMSE was detected between the diabetes alone and normal control groups (P > 0.05). MMSE score was significantly lower in the diabetes and cerebral ischemia group (P < 0.01) than in the normal control group. N2 and P3 latencies of auditory event-related potential were significantly longer, and P3 amplitude was significantly lower in the diabetes alone and diabetes and cerebral ischemia groups (P < 0.01) than in the normal control group. N2 and P3 latencies were significantly longer in the diabetes and cerebral ischemia group than in the diabetes alone group (P < 0.01), but amplitude was not significantly different. N2 and P3 latencies were negatively correlated with MMSE score in elderly type 2 diabetic patients (r=–0.421, –0.604; both P < 0.01). P3 amplitude was positively related to the score of MMSE (r =0.517; P < 0.01). P3 latency was positively associated with age, course of disease, glycosylated hemoglobin and ischemic brain damage in elderly type 2 diabetic patients (t=2.186 to 3.490; all P < 0.05). P3 amplitude was negatively correlated with age, course of disease and glycosylated hemoglobin (t=–2.220, –2.491, and –2.024, respectively; all P < 0.05). N2 latency was positively correlated with age, course of disease and ischemic brain damage (t=2.946, 2.511, and 2.331, respectively; P < 0.05). CONCLUSION: The course of disease, glycosylated hemoglobin and ischemic brain damage are key influential factors for cognitive impairment in elderly type 2 diabetic patients. | 17 | Interference of Hepatitis B Virus with Cellular Signaling显示文摘The presence of hepatitis B virus (HBV) proteins leads to changes in the cellular gene expression. As a consequence, the cellular signaling processes are influenced by the actions of HBV proteins. It has been shown that HBV nucleocapsid protein and the amino-terminal part of polymerase termed as terminal protein (TP) could inhibit interferon signaling. Further, the global gene expression profiles differ in hepatoma cells with and without HBV gene expression and replication. The expression of interferon (IFN) stimulated genes (ISGs) was differently regulated in cells with HBV replication and could be modulated by antiviral treatments. The HBV TP has been found to modulate the ISG expression and enhance the HBV replication.
