| 1 | Contrasting elevational diversity patterns for soil bacteria between two ecosystems divided by the treeline显示文摘Above- and below-ground organisms are closely linked, but how elevational distribution pattern of soil microbes shifting across the treeline still remains unknown. Sampling of 140 plots with transect, we herein investigated soil bacterial distribution pattern from a temperate forest up to a subalpine meadow along an elevational gradient using Illumina sequencing. Our results revealed distinct elevational patterns of bacterial diversity above and below the treeline in responding to changes in soil conditions:a hollow elevational pattern in the forest(correlated with soil temperature, pH, and C:N ratio) and a significantly decreasing pattern in the meadow(correlated with soil pH, and available phosphorus). The bacterial community structure was also distinct between the forest and meadow, relating to soil pH in the forest and soil temperature in the meadow. Soil bacteria did not follow the distribution pattern of herb diversity, but bacterial community structure could be predicted by herb community composition. These results suggest that plant communities have an important influence on soil characteristics, and thus change the elevational distribution of soil bacteria. | 7 | Identification of A-to-I RNA editing profiles and their clinical relevance in lung adenocarcinoma显示文摘Adenosine-to-inosine(A-to-I)RNA editing is a widespread posttranscriptional modification that has been shown to play an important role in tumorigenesis.Here,we evaluated a total of 19,316 RNA editing sites in the tissues of 80 lung adenocarcinoma(LUAD)patients from our Nanjing Lung Cancer Cohort(NJLCC)and 486 LUAD patients from the TCGA database.The global RNA editing level was significantly increased in tumor tissues and was highly heterogeneous across patients.The high RNA editing level in tumors was attributed to both RNA(ADAR1 expression)and DNA alterations(mutation load).Consensus clustering on RNA editing sites revealed a new molecular subtype(EC3)that was associated with the poorest prognosis of LUAD patients.Importantly,the new classification was independent of classic molecular subtypes based on gene expression or DNA methylation.We further proposed a simplified model including eight RNA editing sites to accurately distinguish the EC3 subtype in our patients.The model was further validated in the TCGA dataset and had an area under the curve(AUC)of the receiver operating characteristic curve of 0.93(95%CI:0.91-0.95).In addition,we found that LUAD cell lines with the EC3 subtype were sensitive to four chemotherapy drugs.These findings highlighted the importance of RNA editing events in the tumorigenesis of LUAD and provided insight into the application of RNA editing in the molecular subtyping and clinical treatment of cancer. | Cheng Wang Mingtao Huang Congcong Chen Yuancheng Li Na Qin Zijian Ma Jingyi Fan Linnan Gong Hui Zeng Liu Yang Xianfeng Xu Jun Zhou Juncheng Dai Guangfu Jin Zhibin Hu Hongxia Ma Fengwei Tan Hongbing Shen | 2022 | Science China(Life Sciences)2022,65,1: | 1 |

| 12 | Percolation-induced resistivity drop in lutetium dihydride with controllable electrical conductivity over six orders of magnitude显示文摘The recent report of near-ambient superconductivity in the nitrogen-doped lutetium hydride has attracted considerable attention.Subsequent follow-up studies confirmed the pressure-induced color changes in both N-free and N-doped LuH_(2) but failed to reproduce superconductivity. It remains a puzzle why the samples in the original report exhibited pronounced resistance anomaly reminiscent of the superconducting transition. Here, we show that percolation of metallic grains with high conductivity through the insulating surfaces in cold-pressed LuH_(2) samples can occasionally produce sharp resistance drops, which even display magnetic field and/or current dependences but stay far from zero resistance. The insulating surface of LuH2grain should be attributed to the modification of hydrogen stoichiometry or the contamination by oxygen/nitrogen, resulting in an increase of resistance by over six orders of magnitude. Such an effect is more significant than that discovered recently in LaH_(3±x), which may indicate that LuH_(2) can be a potential superionic conductor. | 17 | Monte Carlo simulation based thinning and calculating method for noninvasive blood glucose sensing by near-infrared spectroscopy显示文摘Previous results show that the floating reference theory(FRT)is an effective tool to reduce the infuence of interference factors on noninvasive blood glucose sensing by near infrared spectros-copy(NIRS).It is the key to measure the floating reference point(FRP)precisely for the application of FRT.Monte Carlo(MC)simulation has been introduced to quantitatively in-vestigate the effects of positioning errors and light source drifts on measuring FRP.In this article,thinning and calculating method(TCM)is proposed to quantify the positioning error.Mean-while,the normalization process(NP)is developed to significantly reduce the error induced by light source drift.The results according to TCM show that 7 purm deviations in positioning can generate about 10.63%relative error in FRP.It is more noticeable that 1%fluctuation in light source intensity may lead to 12.21%relative errors.Gratifyingly,the proposed NP model can effectively reduce the error caused by light source drift.Therefore,the measurement system for FRPs must meet that the positioning error is less than 7 purm,and the light source drift is kept within 1%.Furthermore,an improvement for measurement system is proposed in order to take advantage of the NP model. | Congcong Ma Jia Qin Qi Zhang Junsheng Lu Kexin Xu Jingying Jiang | 2017 | Journal of Innovative Optical Health Sciences2017,,2: | 0 |
| 18 | The impacts of ambiguity in preparation of 80% sulfuric acid solution and shaking time control of calibration solution on the determination of transparent exopolymer particles显示文摘The quantification of transparent exopolymer particles(TEP) by colorimetric method is of large error and low repeatability,one major reason of which is related to the absence of clear definition and evaluation for part steps of the original method.It is obscure that the 80% sulfuric acid solution,acted as the extraction solution in the determination of TEP,is prepared based on a volume ratio or mass ratio.Furthermore,the change of solubility of recently available Gum Xanthan(GX) from the market means that the original protocol is no longer applicable,and the grinding of GX stock solution with a tissue grinder is replaced by shaking with a rotating shaker in the study to prevent the excessive dissolution of GX.We found that different preparation techniques could result in the varied concentrations of 80% H_(2)SO_(4).The duration of shaking during the preparation of standard solution significantly affected the slope of the calibration curve,which caused different correction results of TEP.The impacts of different extraction solution concentrations and shaking time of GX solution on the quantification of TEP were investigated based on the field sampling and laboratory analysis.The extraction capacities of H_(2)SO_(4) with different concentrations for Alcian Blue were distinct,but had limited effect on the final measuring result of TEP.The change of the standard curve slope came along with the variation of shaking time,which markedly altered the detection limit and calibration result,and the extended shaking time was in favor of the determination of low-concentration TEP.It was suggested that the extraction solution concentration,shaking time and filtration volume of standard solution are required to be well controlled and selected to obtain more accurate results for TEP with different concentrations. | Congcong Guo Guicheng Zhang Shan Jian Wei Ma Jun Sun | 2023 | Acta Oceanologica Sinica2023,42,4: | 0 |
