| 1 | Can Cadmium Uptake by Chinese Cabbage be Reduced After Growing Cd-Accumulating Rapeseed?显示文摘Rapeseed (Brassica campestris L.), a Cd-accumulating crop, is effective in reducing plant-available Cd from soil. A pot experiment was conducted to test the hypothesis that the Cd uptake by Chinese cabbage (Brassica pekinensis Rupr.) grown in rotation with rapeseed would be reduced due to the efficient removal of bioavailable Cd from soil solution by the rapeseed crops. The Cd removal by shoot for the rapeseed cultivar Zhucang Huazi ranged from 0.24% to 0.99% in natural Cd-contaminated soil (soil A) and from 0.63% to 1.23% in artificial Cd-contaminated soil (soil B) during the growth period of 4–7 weeks; whereas that for the rapeseed cultivar Chuanyou II-93 ranged from 0.25% to 0.66% and from 0.38% to 1.02%, respectively. Chinese cabbage grown in the pots with soil A for five weeks after harvesting rapeseed exhibited a significantly lower Cd concentration compared to that grown in the control pots (uncropped with rapeseed). However, the rotation of rapeseed did not lower the Cd concentration of Chinese cabbage on soil B. This study aimed to investigate the effects of TCDD on astrocytes proliferation and underlying molecular mechanism. Methods The cell proliferation was measured by EdU-based proliferation assay and PI staining by flow cytometry. Protein expression levels were detected by Western blotting. Immunofluorescence, cytoplasmic and nuclear fractions separation were used to assess the distribution of signal transducer and activator of transcription 3(STAT3). Results C6 cells treated with 10 and 50 nmol/L TCDD for 24 h showed significant promotion of the proliferation of. The exposure to TCDD resulted in the upregulation in the expression levels of phosphorylated protein kinase B(p-Akt), phosphorylated STAT3, and cyclin D1 in a dose-and time-dependent manner. The inhibition of Akt expression with LY294002 or STAT3 expression with AG490 abolished the TCDD-induced cyclin D1 upregulation and cell proliferation. Furthermore, LY294002 suppressed the activation of STAT3. Finally, TCDD promoted the translocation of STAT3 from the cytoplasm to the nucleus, and LY294002 treatment blocked this effect. | 6 | Could saline irrigation clear all residual common bile duct stones after lithotripsy?A self-controlled prospective cohort study显示文摘BACKGROUND A previous study showed that irrigation with 100 mL saline reduced residual common bile duct(CBD)stones,which potentially cause recurrent stones after endoscopic retrograde cholangiopancreatography.AIM To determine whether saline irrigation can improve CBD clearance after lithotripsy.METHODS This prospective self-controlled study enrolled patients receiving mechanical lithotripsy for large(>1.2 cm)CBD stones.After occlusion cholangiography confirmed CBD stone clearance,peroral cholangioscopy(POC)was performed to determine clearance scores based on the number of residual stones.The amounts of residual stones spotted via POC were graded on a 5-point scale(score 1,worst;score 5,best).Scores were documented after only stone removal(control)and after irrigation with 50 mL and 100 mL saline,respectively.The stone composition was analyzed using infrared spectroscopy.RESULTS Between October 2018 and January 2020,47 patients had CBD clearance scores of 2.4±1.1 without saline irrigation,3.5±0.7 with 50 mL irrigation,and 4.6±0.6 with 100 mL irrigation(P<0.001).Multivariate analysis showed that CBD diameter>15 mm[odds ratio(OR)=0.08,95%confidence interval(CI):0.01-0.49;P=0.007]and periampullary diverticula(PAD)(OR=6.51,95%CI:1.08-39.21;P=0.041)were independent risk factors for residual stones.Bilirubin pigment stones constituted the main residual stones found in patients with PAD(P=0.004).CONCLUSION Irrigation with 100 mL of saline may not clear all residual CBD stones after lithotripsy,especially in patients with PAD and/or a dilated(>15 mm)CBD.Pigment residual stones are soft and commonly found in patients with PAD.Additional saline irrigation may be required to remove retained stones. | Yan-Yan Lin Yu-Dong Wang Ping Yue Xian-Zhuo Zhang Joseph W Leung Pan-Pan Jiao Man Yang Hai-Ping Wang Bing Bai Ying Liu Jin-Duo Zhang Hong-Bo Chen Wen-Bo Meng Xun Li | 2021 | World Journal of Gastroenterology2021,27,4: | 4 |

| 12 | A 2.09 Mb fragment translocation on chromosome 6 causes abnormalities during meiosis and leads to less seed watermelon显示文摘Seedlessness is a valuable agronomic trait in watermelon(Citrullus lanatus)breeding.Conventional less seed watermelons are mainly triploid,which has many disadvantages due to unbalanced genome content.Less seed watermelon can be achieved at the diploid level when certain reproductive genes are mutated or by chromosome translocation,which leads to defects during meiosis.However,the formation mechanism of diploid less seed watermelons remains largely unknown.Here,we identified a spontaneous mutant line,watermelon line“148”,which can set seeds normally when self-pollinated.A total of 148×JM F_(1) hybrid plants exhibited seed number reductions to 50.3%and 47.3%of those of the two parental lines,respectively,which are considered to be less seed.Examination of pollen viability and hybridization experiments revealed that F 1 hybrids produce semisterile pollen and ovules.Further cytological observations indicated that semisterility was a result of a reciprocal translocation of chromosomes,which exhibited one quadrivalent ring of four chromosomes at prometaphase I during meiosis.RT-qPCR analysis indirectly confirmed that the semisterile phenotype is caused by chromosome translocation rather than disruption of specific meiotic gene expression.F 2 population genetic analysis indicated that the“148”watermelon line is a homozygous translocation and that the less seed phenotype of the F_(1) hybrid is prompted by one chromosome fragment translocation.The translocated fragment was further fine mapped to a 2.09 Mb region on chromosome 6 by whole-genome resequencing and genetic map cloning procedures.Our work revealed that a 2.09Mb chromosome fragment translocation on chromosome 6,causing meiotic defects at metaphase I during meiosis,leads to diploid less seed watermelon.Our findings provide a new promising method for less seed watermelon breeding at the diploid level,as well as a fragment size reference for breeding less seed watermelon through artificially induced chromosome translocation. | Shujuan Tian Jie Ge Gongli Ai Jiao Jiang Qiyan Liu Xiner Chen Man Liu Jianqiang Yang Xian Zhang Li Yuan | 2021 | Horticulture Research2021,8,1: | 1 |

| 19 | RBM4 dictates ESCC cell fate switch from cellular senescence to glutamine-addiction survival through inhibiting LKB1-AMPK-axis显示文摘Cellular senescence provides a protective barrier against tumorigenesis in precancerous or normal tissues upon distinct stressors.However,the detailed mechanisms by which tumor cells evade premature senescence to malignant progression remain largely elusive.Here we reported that RBM4 adversely impacted cellular senescence to favor glutamine-dependent survival of esophageal squamous cell carcinoma(ESCC)cells by dictating the activity of LKB1,a critical governor of cancer metabolism.The level of RBM4 was specifically elevated in ESCC compared to normal tissues,and RBM4 overexpression promoted the malignant phenotype.RBM4 contributed to overcome H-RAS-or doxorubicin-induced senescence,while its depletion caused P27-dependent senescence and proliferation arrest by activating LKB1-AMPK-mTOR cascade.Mechanistically,RBM4 competitively bound LKB1 to disrupt the LKB1/STRAD/MO25 heterotrimeric complex,subsequently recruiting the E3 ligase TRIM26 to LKB1,promoting LKB1 ubiquitination and degradation in nucleus.Therefore,such molecular process leads to bypassing senescence and sustaining cell proliferation through the activation of glutamine metabolism.Clinically,the ESCC patients with high RBM4 and low LKB1 have significantly worse overall survival than those with low RBM4 and high LKB1.The RBM4 high/LKB1 low expression confers increased sensitivity of ESCC cells to glutaminase inhibitor CB-839,providing a novel insight into mechanisms underlying the glutamine-dependency to improve the efficacy of glutamine inhibitors in ESCC therapeutics. | Lei Chen Wenjing Zhang Dan Chen Quan Yang Siwen Sun Zhenwei Dai Zhengzheng Li Xuemei Liang Chaoqun Chen Yuexia Jiao Lili Zhi Lianmei Zhao Jinrui Zhang Xuefeng Liu Jinyao Zhao Man Li Yang Wang Yangfan Qi | 2023 | Signal Transduction and Targeted Therapy2023,8,5: | 0 |
