1Ganoderic acid hinders renal fibrosis via suppressing the TGF- β/Smad and MAPK signaling pathways显示文摘Renal fibrosis is considered as the pathway of almost all kinds of chronic kidney diseases(CKD)to the end stage of renal diseases(ESRD).Ganoderic acid(GA)is a group of lanostane triterpenes isolated from Ganoderma lucidum,which has shown a variety of pharmacological activities.In this study we investigated whether GA exerted antirenal fibrosis effect in a unilateral ureteral obstruction(UUO)mouse model.After UUO surgery,the mice were treated with GA(3.125,12.5,and 50 mg·kg^−1·d^−1,ip)for 7 or 14 days.Then the mice were sacrificed for collecting blood and kidneys.We showed that GA treatment dose-dependently attenuated UUO-induced tubular injury and renal fibrosis;GA(50 mg·kg^−1·d^−1)significantly ameliorated renal disfunction during fibrosis progression.We further revealed that GA treatment inhibited the extracellular matrix(ECM)deposition in the kidney by suppressing the expression of fibronectin,mainly through hindering the over activation of TGF-β/Smad signaling.On the other hand,GA treatment significantly decreased the expression of mesenchymal cell markers alpha-smooth muscle actin(α-SMA)and vimentin,and upregulated E-cadherin expression in the kidney,suggesting the suppression of tubular epithelial-mesenchymal transition(EMT)partially via inhibiting both TGF-β/Smad and MAPK(ERK,JNK,p38)signaling pathways.The inhibitory effects of GA on TGF-β/Smad and MAPK signaling pathways were confirmed in TGF-β1-stimulated HK-2 cell model.GA-A,a GA monomer,was identified as a potent inhibitor on renal fibrosis in vitro.These data demonstrate that GA or GA-A might be developed as a potential therapeutic agent in the treatment of renal fibrosis.Xiao-qiang Geng Ang Ma Jin-zhao He Liang Wang Ying-li Jia Guang-ying Shao Min Li Hong Zhou Shu-qian Lin Jian-hua Ran Bao-xue Yang 2020Acta Pharmacologica Sinica2020,41,5:43
2Targeting ERK,an Achilles' Heel of the MAPK pathway,in cancer therapy显示文摘The mitogen-activated protein kinases(MAPK) pathway, often known as the RAS-RAFMEK-ERK signal cascade, functions to transmit upstream signals to its downstream effectors to regulate physiological process such as cell proliferation, differentiation, survival and death. As the most frequently mutated signaling pathway in human cancer, targeting the MAPK pathway has long been considered a promising strategy for cancer therapy. Substantial efforts in the past decades have led to the clinical success of BRAF and MEK inhibitors. However, the clinical benefits of these inhibitors are compromised by the frequently occurring acquired resistance due to cancer heterogeneity and genomic instability. This review briefly introduces the key protein kinases involved in this pathway as well as their activation mechanisms. We also generalize the correlations between mutations of MAPK members and human cancers, followed by a summarization of progress made on the development of small molecule MAPK kinases inhibitors. 3Atomically dispersed Au_1 catalyst towards efficient electrochemical synthesis of ammonia显示文摘Tremendous efforts have been devoted to explore energy-efficient strategies of ammonia synthesis to replace Haber–Bosch process which accounts for 1.4% of the annual energy consumption. In this study,atomically dispersed Au_1 catalyst is synthesized and applied in electrochemical synthesis of ammonia under ambient conditions. A high NH_4^+ Faradaic efficiency of 11.1% achieved by our Au_1 catalyst surpasses most of reported catalysts under comparable conditions. Benefiting from efficient atom utilization, an NH_4^+ yield rate of 1,305 lg h^(-1) mg_(Au)^(-1) has been reached, which is roughly 22.5 times as high as that by supported Au nanoparticles. We also demonstrate that by employing our Au_1 catalyst, NH4+can be electrochemically produced directly from N_2 and H_2 with an energy utilization rate of 4.02 mmol kJ^(-1). 5Emission factors of gaseous carbonaceous species from residential combustion of coal and crop residue briquettes显示文摘Experiments were performed to measure the emission factors (EFs) of gaseous carbonaceous species, such as CO2, CO, CH4, and non-methane volatile organic compounds (NMVOCs), from the combustion of five types of coal of varying organic maturity and two types of biomass briquettes under residential burning conditions. Samples were collected in stainless steel canisters and 2,4- dinitrophenylhydrazine (DNPH) cartridges and were analyzed by GC FID/MS and HPLC, respectively. The EFs from crop residue briquette burning were generally higher than those from coals, with the exception of CO2. The dominant NMVOC species identified in coal smoke were carbonyls (41.7%), followed by C2 unsaturated hydrocarbons (29.1%) and aromatics (12.1%), while C2 unsaturated hydrocarbons were the dominant species (68.9%) emitted from the combustion of crop residue briquettes, followed by aromatics (14.4%). A comparison of burning normal crop residues in stoves and the open field indicated that briquettes emitted a larger proportion of ethene and acetylene. Both combustion efficiency and coal organic maturity had a significant impact on NMVOC EFs from burning coal: NMVOC emissions increased with increasing coal organic maturity but decreased as the combustion efficiency improved. Emissions from the combustion of crop residue briquettes from stoves occurred mainly during the smoldering process, with low combustion efficiency. 6Involvement of aquaporins in a mouse model of rotavirus diarrhea显示文摘Rotavirus diarrhea is a major worldwide cause of infantile gastroenteritis; however, the mechanism responsible for intestinal fluid loss remains unclear. Water transfer across the intestinal epithelial membrane seems to occur because of aquaporins(AQPs). Accumulating evidence indicates that alterations in AQPs may play an important role in pathogenesis. Here, we focus on changes in AQPs in a mouse model of rotavirus diarrhea. In the present study, 32 of 35 mice developed diarrhea and mild dehydration within 24 hours after infection with rotavirus strain SA11. Intestinal epithelial cells demonstrated cytoplasmic vacuolation, malaligned villi, and atrophy. AQP1 expression was significantly attenuated in the ileum and colon in comparison with controls; likewise, AQP4 and-8 protein expression were significantly decreased in the colon of rotavirus diarrhea-infected mice. In contrast, AQP3 protein expression was significantly increased in the colon of rotavirus-infected mice in comparison with controls. These results indicate that rotavirus diarrhea is associated with the downregulation of AQP1,-4, and-8 expression. 9Zircon Hf Isotopes and Geochemistry of the Early Paleoproterozoic High-Sr Low-Y Quartz-Diorite in the Quanji Massif,NW China:Crustal Growth and Tectonic Implications显示文摘The metamorphosed Early Paleoproterozoic granitoids in the Quanji massif,Northwestern China provide constraints for relationship between the Tarim Craton and North China Craton.Among granitoids batholiths,rocks of the Mohe quartz-diorite show typically adakitic geochemical characteristics,with medium K2O/Na2O ratios(0.56–1.17)and high Sr(519–619 ppm)low Y(9.37–20.40 ppm)and Yb(0.97–1.77 ppm)concentrations.The rocks haveεNd(t)values between+2.4and+4.4 and depleted mantle Nd model ages of 2.43–2.59 Ga.The magmatic zircons have positiveεHf(t)values ranging from+0.40 to+7.60 and depleted mantle Hf model ages of 2.43–2.70 Ga,with major peaks at^2.54 and^2.65 Ga.The geochemical and Nd-Hf isotopic characteristics indicate that the Mohe quartz-dioritic rocks might be formed by partial melting of high-pressure metamorphosed juvenile crustal rocks in post-orogenic extensional regime in the Early Paleoproterozoic.It suggests that important crustal growth occurred in the Quanji massif and the Tarim Craton at^2.4 and 2.5–2.7 Ga.The Quanji massif and Tarim Craton might share a similar crustal evolution history with the North China Craton in the Neoarchean.Songlin Gong Nengsong Chen Hongyan Geng Min Sun Lu Zhang Qinyan Wang 2014Journal of Earth Science2014,25,1:8
10Surface plasmons interference nanogratings:wafer-scale laser direct structuring in seconds显示文摘It is always a great challenge to bridge the nano-and macro-worlds in nanoscience,for instance,manufacturing uniform nanogratings on a whole wafer in seconds instead of hours even days.Here,we demonstrate a single-step while extremely high-throughput femtosecond laser scanning technique to obtain wafer-scale,highly regular nanogratings on semiconductor-on-metal thin films.Our technique takes advantage of long-range surface plasmons-laser interference,which is regulated by a self-initiated seed.By controlling the scanning speed,two types of nanogratings are readily manufactured,which are produced by either oxidation or ablation.We achieve a record manufacturing speed(>1 cm^(2)s^(-1)),with tunable periodicity of∧<1μm.The fractional variation of their periodicity is evaluated to be as low as△∧/∧≈0.5%.Furthermore,by utilizing the semiconductor-on-metal film-endowed interference effects,an extremely high energy efficiency is achieved via suppressing light reflection during femtosecond laser nano-processing.As the fabricated nanogratings exhibit multi-functionality,we exemplify their practical applications in highly sensitive refractive index sensing,vivid structural colors,and durable superhydrophilicity.liao Geng Wei Yan Liping Shi Min Qiu 2022Light(Science & Applications)2022,11,7:8
11Prediction of 10-year Atherosclerotic Cardiovascular Disease Risk among Adults Aged 40-79 Years in China:a Nationally Representative Survey显示文摘Objective To establish the distribution of 10-year atherosclerotic cardiovascular disease(ASCVD) risk among Chinese adults.Methods We estimated the 10-year ASCVD risk by applying the 2013 American College of Cardiology/American Heart Association pooled cohort equations(PCEs) to the data obtained from the 2010 China Chronic Disease and Risk Factor Surveillance that involved 61,541 participants(representing 520,158,652 Chinese adults) aged 40-79 years.We also compared the ASCVD risk with the 10-year ischemic cardiovascular disease(ICVD) risk,which was calculated using the simplified scoring tables recommended by the Chinese Guidelines for Prevention of Cardiovascular Diseases(Chinese model).Results Based on the PCEs,the average 10-year ASCVD risk among adults without self-reported stroke or myocardial infraction was 12.5%.Approximately 247 million(47.4%) and 107 million(20.6%) adults had ≥ 7.5% and > 20% 10-year ASCVD risks,respectively.The 10-year ASCVD risk > 20% was higher among men,less educated individuals,smokers,drinkers,and physically inactive individuals than among their counterparts.Overall,29.0% of adults categorized using the Chinese model were overclassified with the PCEs.Conclusion Our results define the distribution of 10-year ASCVD risk among Chinese adults.The 10-year ASCVD risk predicted by the PCEs was higher than the ICVD risk predicted by the Chinese model.ZHANG Mei JIANG Yong WANG Li Min LI Yi Chong HUANG Zheng Jing LI Jian Hong ZHOU Mai Geng ZHAO Wen Hua 2017Biomedical and Environmental Sciences2017,30,4:7
12Rescue of premature aging defects in Cockayne syndrome stem cells by CRISPR/Cas9-mediated gene correction显示文摘Cockayne syndrome(CS)is a rare autosomal recessive inherited disorder characterized by a variety of clinical features,including increased sensitivity to sunlight,progressive neurological abnormalities,and the appearance of premature aging.However,the pathogenesis of CS remains unclear due to the limitations of current disease models.Here,we generate integration-free induced pluripotent stem cells(iPSCs)from fibroblasts from a CS patient bearing mutations in CSB/ERCC6 gene and further derive isogenic genecorrected CS-iPSCs(GC-iPSCs)using the CRISPR/Cas9 system.CS-associated phenotypic defects are recapitulated in CS-iPSC-derived mesenchymal stem cells(MSCs)and neural stem cells(NSCs),both of which display increased susceptibility to DNA damage stress.Premature aging defects in CS-MSCs are rescued by the targeted correction of mutant ERCC6.We next map the transcriptomic landscapes in CS-iPSCs and GC-iPSCs and their somatic stem cell derivatives(MSCs and NSCs)in the absence or presence of ultraviolet(UV)and replicative stresses,revealing that defects in DNA repair account for CS pathologies.Moreover,we generate autologous GC-MSCs free of pathogenic mutation under a cGMP(Current Good Manufacturing Practice)-compliant condition,which hold potential for use as improved biomaterials for future stem cell replacement therapy for CS.Collectively,our models demonstrate novel disease features and molecular mechanisms and lay a foundation for the development of novel therapeutic strategies to treat CS.Si Wang Zheying Min Qianzhao Ji Lingling Geng Yao Su Zunpeng Liu Huifang Hu Lixia Wang Weiqi Zhang Keiichiro Suzuiki Yu Huang Puyao Zhang Tie-Shan Tang Jing Qu Yang Yu Guang-Hui Liu Jie Qiao 2020Protein & Cell2020,11,1:7
13Altered Neuronal Activity in the Central Nucleus of the Amygdala Induced by Restraint Water-Immersion Stress in Rats显示文摘Restraint water-immersion stress(RWIS), a compound stress model, has been widely used to induce acute gastric ulceration in rats. A wealth of evidence suggests that the central nucleus of the amygdala(CEA) is a focal region for mediating the biological response to stress. Different stressors induce distinct alterations of neuronal activity in the CEA; however, few studies have reported the characteristics of CEA neuronal activity induced by RWIS. Therefore, we explored this issue using immunohistochemistry and in vivo extracellular single-unit recording. Our results showed that RWIS and restraint stress(RS) differentially changed the c-Fos expression and firing properties of neurons in the medial CEA. In addition,RWIS, but not RS, induced the activation of corticotropinreleasing hormone neurons in the CEA. 15Initial detection of the quorum sensing autoinducer activity in the rumen of goats in vivo and in vitro显示文摘Quorum sensing(QS) is a type of microbe-microbe communication system that is widespread among the microbial world, particularly among microorganisms that are symbiotic with plants and animals. Thereby, the cell-cell signalling is likely to occur in an anaerobic rumen environment, which is a complex microbial ecosystem. In this study, using six ruminally fistulated Liuyang black goats as experimental animals, we aimed to detect the activity of quorum sensing autoinducers(AI) both in vivo and in vitro and to clone the lux S gene that encoded autoinducer-2(AI-2) synthase of microbial samples that were collected from the rumen of goats. Neutral detergent fiber(NDF) and soluble starch were the two types of substrates that were used for in vitro fermentation. The fermented fluid samples were collected at 0, 2, 4, 6, 8, 12, 24, 36, and 48 h of incubation. The acyl-homoserine lactones(AHLs) activity was determined using gas chromatography-mass spectrometer(GC-MS) analysis. However, none of the rumen fluid extracts that were collected from the goat rumen showed the same or similar fragmentation pattern to AHLs standards. Meanwhile, the AI-2 activity, assayed using a Vibrio harveyi BB170 bioassay, was negative in all samples that were collected from the goat rumen and from in vitro fermentation fluids. Our results indicated that the activities of AHLs and AI-2 were not detected in the ruminal contents from six goats and in ruminal fluids obtained from in vitro fermentation at different sampling time-points. However, the homologues of lux S in Prevotella ruminicola were cloned from in vivo and in vitro ruminal fluids. We concluded that AHLs and AI-2 could not be detected in in vivo and in vitro ruminal fluids of goats using the current detection techniques under current dietary conditions. 16Genetic characteristics and pathogenicity of human hepatitis E virus in Nanjing,China显示文摘AIM:To investigate the genetic characteristics and pathogenicity of hepatitis E virus (HEV) and assess the potential risk factors for sporadic hepatitis E.METHODS:Sixty-two serum samples from the patients with acute hepatitis E were collected,including 23 cases coinfected with hepatitis B virus.Anti-HEV detection and partial HEV RNA amplification were performed by enzyme immunoassays and reverse transcriptionnested polymerase chain reaction (RT-nPCR) method,respectively,and PCR products were sequenced.The isolated human HEV sequences were analyzed phylogenetically.RESULTS:The positive rate of serum HEV RNA were 21.0% (13/62),including 5 cases of liver failure.All the 13 isolates shared a 82.1%-98.0% nucleotide homology with each other and had identities of 74.7%-81.0%,75.3%-78.6%,75.3%-80.0% and 82.1%-96.1% with the corresponding regions of HEV genotypes 1-4,respectively.The human HEV strain GS-NJ-12 shared a 100% nucleotide identity with the swine HEV strain swIM6-43 isolated from Inner Mongolia,China.CONCLUSION:Swine may be a principal risk factor for occurrence of sporadic hepatitis E in eastern China,and genotype 4 HEV can induce acute liver failure.lia-Bao Geng Mao-Rong Wang Jie Wang Zhi-Guo Yang Yan Cheng Fei Qiao Min Wang 2012World Journal of Gastroenterology2012,18,9:5
17Genetic Diversity, Antimicrobial Resistance, and Virulence Genes of Aeromonas Isolates from Clinical Patients, Tap Water Systems, and Food显示文摘Objective This study aimed to evaluate the genetic diversity,virulence,and antimicrobial resistance of Aeromonas isolates from clinical patients,tap water systems,and food.Methods Ninety Aeromonas isolates were obtained from Ma’anshan,Anhui province,China,and subjected to multi-locus sequence typing(MLST)with six housekeeping genes.Their taxonomy was investigated using concatenated gyr B-cpn60 sequences,while their resistance to 12 antibiotics was evaluated.Ten putative virulence factors and several resistance genes were identified by PCR and sequencing.Results The 90 Aeromonas isolates were divided into 84 sequence types,80 of which were novel,indicating high genetic diversity.The Aeromonas isolates were classified into eight different species.PCR assays identified virulence genes in the isolates,with the enterotoxin and hemolysin genes act,aer A,alt,and ast found in 47(52.2%),13(14.4%),22(24.4%),and 12(13.3%)of the isolates,respectively.The majority of the isolates(≥90%)were susceptible to aztreonam,imipenem,cefepime,chloramphenicol,gentamicin,tetracycline,and ciprofloxacin.However,several resistance genes were detected in the isolates,as well as a new mcr-3 variant.Conclusions Sequence type,virulence properties,and antibiotic resistance vary in Aeromonas isolates from clinical patients,tap water systems,and food.MENG Shuang WANG Yong Lu LIU Chen Geng YANG Jing YUAN Min BAI Xiang Ning JIN Dong LIANG Jun Rong CUI Zhi Gang LI Juan 2020Biomedical and Environmental Sciences2020,33,6:5
18Refinement of primary Si in Cu-50Si alloys with novel Al-Zr-P master alloy显示文摘In this article, a novel Al-6Zr-2P master alloy with ZrP particles was successfully synthesized, and the refining performance of this novel master alloy for the primary Si in Cu-50Si alloys was also investigated. By means of the fracture plane observation, it is found that the ZrP phase would precipitate first in the solidification process, and then, the ZrAl 3 phase grows around them. Furthermore, it is observed that the refining effect can be remarkably improved by changing the addition sequence of the raw materials. After the melting of commercial Cu, the 2.0 wt% Al-6Zr-2P master alloy and crystalline Si were added in sequence, and the mean size of the primary Si in Cu-50Si alloy can be significantly refined from 255.7 to 75.3 lm.
