| 1 | NEAT1/miR-140-3p/MAPK1 mediates the viability and survival of coronary endothelial cells and affects coronary atherosclerotic heart disease显示文摘Studies have shown that long non-coding RNAs(lncRNA)play critical roles in coronary atherosclerotic heart disease(CAD).However,the function of lncRNA nuclear enriched abundant transcript 1(NEAT1)in CAD is unclear.In this study,we aimed to investigate the functions of lncRNA NEAT1 in CAD.RTPCR and western blot analysis were carried out to examine the expressions of related RNAs.Colony formation assay,cell proliferation assay,apoptosis assay,and dual-luc讦erase reporter assay were con ducted to investigate the abilities of colony migration,cell proliferatio n,apoptosis,and targeting.The results showed that NEAT1 was up-regulated in CAD blood samples and in human coronary endothelial cells(HCAECs).Transfection of pcNEATI sign讦icantly inhibited the survival rate of HCAECs and induced apoptosis of HCAECs.MiR-140-3p was down-regulated in HCAECs.NEAT1 directly targeted miR-140-3p,and the expression of miR-140-3p was inversely correlated with the expression of NEAT1 in CAD patients.In addition,co-transfection of NEAT1 with miR-140-3p mimic reversed the effect of pcNEATI on cell viability and apoptosis,mitogenactivated protein kinase 1(MAPK1)was proved to be a target gene of miR-140-3p,and the miR-140-3p mimic was shown to reduce the expression of MAPK1 in HCAECs.pcNEATI sigrdficantly increased the expression level of MAPK1,while shNEATI significantly reduced the expression level of MAPK1.Our results revealed that lncRNA NEAT1 increased cell viability and inhibited CAD cell apoptosis possibly by activating the miR-140-3p/MAPK1 pathway,and lncRNA NEAT1 might serve as a potential therapeutic target for CAD. | Hui Zhang Ningning Ji Xinyan Gong Shimao Ni Yu Wang | 2020 | Acta Biochimica et Biophysica Sinica2020,52,9: | 9 |
| 2 | Integration of A Deep Learning Classifier with A Random Forest Approach for Predicting Malonylation Sites显示文摘As a newly-identified protein post-translational modification, malonylation is involved in a variety of biological functions. Recognizing malonylation sites in substrates represents an initial but crucial step in elucidating the molecular mechanisms underlying protein malonylation. In this study, we constructed a deep learning(DL) network classifier based on long short-term memory(LSTM) with word embedding(LSTMWE) for the prediction of mammalian malonylation sites.LSTMWEperforms better than traditional classifiers developed with common pre-defined feature encodings or a DL classifier based on LSTM with a one-hot vector. The performance of LSTMWE is sensitive to the size of the training set, but this limitation can be overcome by integration with a traditional machine learning(ML) classifier. Accordingly, an integrated approach called LEMP was developed, which includes LSTMWEand the random forest classifier with a novel encoding of enhanced amino acid content. LEMP performs not only better than the individual classifiers but also superior to the currently-available malonylation predictors. Additionally, it demonstrates a promising performance with a low false positive rate, which is highly useful in the prediction application. | 3 | Interactions between bubble and particles of key minerals of diasporic bauxite through the extended DLVO theory显示文摘The flotation of diasporic bauxite is to separate diaspore(valuable mineral)from aluminosilicate minerals(gangue minerals,mainly including kaolinite,illite and pyrophyllite),and the microscopic interaction force between the two types of minerals and air bubbles determines the separation efficiency.In this paper,based on the extended Derjaguin-Landau-Verwey-Overbeek(DLVO)theory,the van der Waals,electrostatic and hydrophobic interaction between particles of the four minerals mentioned above and air bubbles in collectorless solution were calculated first,and then diaspore and kaolinite were taken as examples to analyze the influence of various factors such as electrolyte concentration,mineral particle size,air bubble size,collector type(dodecylamine hydrochloride(DAH)and sodium oleate(NaOL))and concentration,and pulp pH on the interactions between the particles of valuable mineral and gangue minerals and air bubbles.The results showed that the total extended DLVO interactions between the four minerals and air bubbles were repulsive in most cases in collectorless solution.The increase in electrolyte concentration reduced the interaction force or even changed the direction of the force under certain circumstances.The addition of DAH and NaOL can reduce the adhesion energy barrier of kaolinitebubble and diaspore-bubble respectively.Each type of minerals exhibited a specific interface interaction response with air bubbles in each collector with different pH values.The research results have theoretical guiding significance for the optimization and directional control of diasporic bauxite flotation conditions. | Ningning Zhang Tian Pang Rui Han Songjiang Chen Zhen Li Yuexian Yu Zhongyu Shi Lijun Liu Jinzhou Qu Anning Zhou | 2022 | International Journal of Mining Science and Technology2022,32,1: | 3 |
| 4 | Significance of combined detection of LunX mRNA and tumor markers in diagnosis of lung carcinoma显示文摘Objective:To evaluate the significance of combined detection of LunX mRNA,carcinoembryonic antigen(CEA),neuron-specific enolase(NSE),and cytokeratin 21-1 fragment(CYFRA21-1) in clinical diagnosis of lung carcinoma.Methods:Based on the quantitative RT-PCR and chemiluminescence immunoassay,the expression levels of LunX mRNA,CEA,NSE,and CYFRA21-1 in 113 patients with lung carcinoma(case group) and 30 healthy participants(control group) were detected.Meantime,the sensitivity,specificity,and accuracy of the combination detection were also explored.Results:The positive rates of LunX mRNA in peripheral blood and CEA,NSE,and CYFRA21-1 in serum were significantly higher in case group than those in control group(χ2=17.295,16.825,19.148,and 17.450;P<0.05).There was no statistical significance when positive rate of LunX mRNA was evaluated among different pathological types(χ2=0.047,P>0.05).The positive rate of LunX mRNA in stage Ⅰ+Ⅱ,Ⅲ,and Ⅳ had a significantly increasing tendency(χ2=10.565,32.462,P<0.05).The positive rate of CYFRA21-1 was highest in squamous carcinoma(78.5%),the positive rate of NSE was highest in small cell carcinoma(86.7%),and the positive rate of CEA wag highest in lung adenocarcinoma(80.4%).The sensitivity and accuracy of the combination detection were 91.1% and 88.1%,respectively.Conclusions:The combined detection of LunX mRNA and tumor markers(TMs) including CEA,NSE,and CYFRA21-1 in peripheral blood is helpful to increase the diagnostic accuracy of lung cancer.Also,it can inform the pathological typing of lung carcinoma. | Hui Yu Xiuying Huang Zhihua Zhu Yi Hu Wei Ou Lanjun Zhang Ningning Zhou | 2014 | Chinese Journal of Cancer Research2014,26,1: | 3 |
| 5 | Regulation of Leaf Longevity by DML3-Mediated DNA Demethylation显示文摘Leaf senescence is driven by the expression of senescence-associated genes(SAGs).Development-specific genes often undergo DNA demethylation in their promoter and other regions,which regulates gene expression.Whether and how DNA demethylation regulates the expression of SAGs and thus leaf senescence remain elusive.Whole-genome bisulfite sequencing(WGBS)analyses of wild-type(WT)and demeter-like 3(dml3)Arabidopsis leaves at three developmental stages revealed hypermethylation during leaf senescence in dml3 compared with WT,and 20556 differentially methylated regions(DMRs)were identified by comparing the methylomes of dml3 and WT in the CG,CHG,and CHH contexts.Furthermore,we identified that 335 DMR-associated genes(DMGs),such as NAC016 and SEN1,are upregulated during leaf senescence,and found an inverse correlation between the DNA methylation levels(especially in the promoter regions)and the transcript abundances of the related SAGs in WT.In contrast,in dml3 the promoters of SAGs were hypermethylated and their transcript levels were remarkably reduced,and leaf senescence was significantly delayed.Collectively,our study unraveled a novel epigenetic regulatory mechanism underlying leaf senescence in which DML3 is expressed at the onset of and during senescence to demethylate promoter,gene body or 3'UTR regions to activate a set of SAGs. | Lu Yuan Dan Wang Liwen Cao Ningning Yu Ke Liu Yongfeng Guo Susheng Gan Liping Chen | 2020 | Molecular Plant2020,13,8: | 3 |
| 6 | DEM simulation of particle flow and separation in a vibrating flip-flow screen显示文摘Vibrating flip-flow screens(VFFS)with stretchable polyurethane sieve mats have been widely used in screening fine-grained materials in recent years.In this work,the discrete element method(DEM)is used to study the screening process in VFFS to explain particle flow and separation behavior at the particle scale.Unlike traditional vibrating screens,for VFFS,the amplitude response of each point on the elastic sieve mat is different everywhere.This study measures the kinematics of the elastic sieve mat under different conditions such as different stretched lengths and material loads.To establish the elastic sieve mat model in a DEM simulation,the continuous elastic sieve mat is discretized into multiple units,and the displacement signal of each unit tested is analyzed by Fourier series.The Fourier series analysis results of each unit are used as the setting parameters for motion.In this way,the movement of the elastic sieve mat is approximately simulated,and a DEM model of VFFS is produced.Through the simulation,the flow and separation of different-sized particles in VFFS are studied,and the reasonability of the simulation is verified by a pilot-scale screening experiment.The present study demonstrates the potential of the DEM method for the analysis of screening processes in VFFS. | Chi Yu Dongdong Lin Ningning Xu Xinwen Wang Kunwei Pu Zhenghao Wang Guofeng Zhao Runhui Geng Sanpeng Gong | 2023 | Particuology2023,,2: | 2 |
| 7 | 定量分析乳腺癌术后化疗前外周血多药耐药基因的检测及其临床意义的研究(英文)显示文摘Objective: To explore expressions of multidrug resistance gene (MDR) product P170 and peripheral blood MDR1 mRNA in breast cancer tissue and their clinical significances. Methods: Fluorescent quantitative RT-PCR and immunohis-tochemistry were used to detect peripheral blood MDR1 mRNA before chemotherapy in breast cancer tissue samples from 32 cases with P170 positive (trial group) and 11 cases with P170 negative (control group). Results: There were 14 cases (43.75%) peripheral blood MDR1 mRNA positive and 18 cases (56.25%) negative in 32 cases P170 positive breast cancer pa-tients, while there were 6 cases (54.55%) peripheral blood MDR1 mRNA positive and 5 cases (45.45%) negative in 11 cases P170 negative breast cancer patients; there wasn't a significance difference with peripheral blood MDR1 mRNA express rate in P170 positive (43.75%) group and P170 negative (54.55%) group (χ2 = 0.383, P > 0.05). There were 12 cases (37.50%) Her-2 positive and 20 cases (62.50%) negative in P170 positive group; there were 2 cases (18.18%) Her-2 positive and 9 cases (81.82%) negative in P170 negative group; there wasn't a significance difference with Her-2 positive rate in P170 posi-tive group (37.50%) and P170 negative (18.18%) group (χ2 = 1.391, P > 0.05). There were 13 cases (40.63%) ER positive and 19 cases (59.37%) negative in P170 positive group; there were 7 cases (63.64%) ER positive and 4 cases (36.36%) negative in P170 negative group; there wasn't a significance difference with ER positive rate in P170 positive group (40.63%) and P170 negative (63.64) group (χ2 = 1.742, P > 0.05). | 13 | Down-staging depth score could be a survival predictor for locally advanced gastric cancer patients after preoperative chemoradiotherapy显示文摘Objective:The predictive effect of preoperative chemoradiotherapy(CRT)is low and difficult in guiding individualized treatment.We examined a surrogate endpoint for long-term outcomes in locally advanced gastric cancer patients after preoperative CRT.Methods:From April 2012 to April 2019,95 patients with locally advanced gastric cancer who received preoperative concurrent CRT and who were enrolled in three prospective studies were included.All patients were stage T_(3/4) N_(+).Local control,distant metastasis-free survival(DMFS),disease-free survival(DFS)and overall survival(OS)were evaluated.Clinicopathological factors related to long-term prognosis were analyzed using univariate and multivariate analyses.The down-staging depth score(DDS),which is a novel method of evaluating CRT response,was used to predict long-term outcomes.Results:The median follow-up period for survivors was 30 months.The area under the curve(AUC)of the receiver operating characteristic(ROC)curve predicted by the DDS was 0.728,which was better than the pathological complete response(pCR),histological response and ypN0.Decision curve analysis further affirmed that DDS had the largest net benefit.The DDS cut-off value was 4.pCR and ypN0 were associated with OS(P=0.026 and 0.049).Surgery and DDS are correlated with DMFS,DFS and OS(surgery:P=0.001,<0.001 and<0.001,respectively;and DDS:P=0.009,0.013 and 0.032,respectively).Multivariate analysis showed that DDS was an independent prognostic factor of DFS(P=0.021).Conclusions:DDS is a simple,short-term indicator that was a better surrogate endpoint than pCR,histological response and ypN0 for DFS. | Ning Li Xin Wang Yuan Tang Dongbin Zhao Yihebali Chi Lin Yang Liming Jiang Jun Jiang Jinming Shi Wenyang Liu Hua Ren Hui Fang Yu Tang Bo Chen Ningning Lu Hao Jing Shunan Qi Shulian Wang Yueping Liu Yongwen Song Yexiong Li Jing Jin | 2021 | Chinese Journal of Cancer Research2021,33,4: | 1 |

However, the small size of the microLED chip (1–100 µm) makes it extremely challenging for high efficiency and low cost to accurately, selectively, integrate millions of microLED chips. Recent impressive technological advances have overcome the drawbacks of traditional pick-and-place techniques when they were utilized in the assembly of microLED display, including the most broadly recognized laser lift-off technique, contact micro-transfer printing (µTP) technique, laser non-contact µTP technique, and self-assembly technique. Herein, we firstly review the key developments in mass transfer technique and highlight their potential value, covering both the state-of-the-art devices and requirements for mass transfer in the assembly of the ultra-large-area display and virtual reality glasses. We begin with the significant challenges and the brief history of mass transfer technique, and expand that mass transfer technique is composed of two major techniques, namely, the epitaxial Lift-off technique and the pick-and-place technique. The basic concept and transfer effects for each representative epitaxial Lift-off and pick-and-place technique in mass transfer are then overviewed separately. | 18 | Influence of traditional Chinese medicines on the in vivo metabolism of lopinavir/ritonavir based on UHPLC-MS/MS analysis显示文摘A fast,reliable,and cost-effective liquid chromatography-tandem mass spectrometry method was established to determine the effects of the traditional Chinese medicine employed to treat coronavirus disease 2019,namely,Lianhua Qingwen granules,Huoxiang Zhengqi capsules,Jinhua Qinggan granules,Shufeng Jiedu capsules,and Angong Niuhuang pills,on the pharmacokinetics of lopinavir/ritonavir in rats.Blood samples were prepared using the protein precipitation method and atazanavir was selected as the internal standard(IS).Separation was performed on an Agilent ZORBAX eclipse plus C18(2.1 mm×50 mm,1.8 μm)column using acetonitrile and water containing 0.1%formic acid as the mobile phase for gradient elution.The flow rate was 0.4 mL/min and the injection volume was 2 μL.Agilent Jet Stream electrospray ionization was used for mass spectrometry detection under positive ion multiple reaction monitoring mode at a transition of m/z 629.3→447.3 for lopinavir,m/z 721.3→296.1 for ritonavir,and m/z 705.4→168.1 for the IS.The method showed good linearity in the concentration range of 25→2500 ng/mL(r=0.9981)for lopinavir and 5e500 ng/mL(r=0.9984)for ritonavir.The intra-day and inter-day precision and accuracy were both within±15%.Items,such as dilution reliability and residual effect,were also within the acceptable limits.The method was used to determine the effects of five types of traditional Chinese medicines on the pharmacokinetics of lopinavir/ritonavir in rats.The pharmacokinetic results showed that the half-life of ritonavir in the groups administered Lianhua Qingwen granules and Huoxiang Zhengqi capsules combined with lopinavir/ritonavir was prolonged by approximately 1.5-to 2-fold relative to that in the control group.Similarly,the pharmacokinetic parameters of lopinavir were altered.Overall,the results of this study offer important theoretical parameters for the effective clinical use of five types of traditional Chinese medicines combined with lopinavir/ritonavir to reduce the occurrence of clinical adverse reactions. | Linlin Li Xinxiang Yu Dongmin Xie Ningning Peng Weilin Wang Decai Wang Binglong Li | 2022 | Journal of Pharmaceutical Analysis2022,12,2: | 1 |
| 19 | The heterogeneous immune landscape between lung adenocarcinoma and squamous carcinoma revealed by single-cell RNA sequencing显示文摘A thorough interrogation of the immune landscape is crucial for immunotherapy strategy selection and prediction of clinical responses in non-small-cell lung cancer(NSCLC)patients.Single-cell RNA sequencing(scRNA-seq)techniques have prompted the opportunity to dissect the distinct immune signatures between lung adenocarcinoma(LUAD)and lung squamous cell carcinoma(LUSC),the two major subtypes of NSCLC.Here,we performed scRNA-seq on 72,475 immune cells from 40 samples of tumor and matched adjacent normal tissues spanning 19 NSCLC patients,and drew a systematic immune cell transcriptome atlas.Joint analyses of the distinct cellular compositions,differentilly expressed genes(DEGs),cell-cell interactions,pseudotime trajectory,transcriptomic factors and prognostic factors based on The Cancer Genome Atlas(TCGA),revealed the central roles of cytotoxic and effector T and NK cells and the distinct functional macrophages(Mo)subtypes in the immune microenvironment heterogeneity between LUAD and LUSC.The dominant subtype of Mo was FABP4-Mp in LUAD and SPP1-Mo in LUSC.Importantly,we identifieda novel lymphocyte-related Mo cluster,which we named SELENOP-Mo,and further established its antitumor role in both types,especially in LUAD.Our comprehensive depiction of the immune heterogeneity and definition of Mp clusters could help design personalized treatment for lung cancer patients in clinical practice. | Chengdi Wang Qiuxiao Yu Tingting Song Zhoufeng Wang Lujia Song Ying Yang Jun Shao Jingwei Li Yinyun Ni Ningning Chao Li Zhang Weimin Li | 2022 | Signal Transduction and Targeted Therapy2022,7,9: | 1 |
