2N6-Methyladenine DNA Methylation in Japonica and Indica Rice Genomes and Its Association with Gene Expression,Plant Development, and Stress Responses显示文摘N6-Methyladenine (6mA)DNA methylation has recently been implicated as a potential new epigenetic marker in eukaryotes,including the dioot modelArabidopsis thaliana.However,the conservation and divergence of 6mA distribution patterns and functions in plants remain elusive.Here we report high-quality 6mA methylomes at single-nucleotide resolution in rice based on substantially improved genome sequences of two rice cultivars,Nipponbare (Nip;Japonica)and 93-11 (Indica).Analysis of 6mA genomic distribution and its association with transcription suggest that 6mA distribution and function is rather conserved between rice and Arabidopsis.We found that 6mA levels are positively correlated with the expression of key stressrelated genes,which may be responsible for the difference in stress tolerance between Nip and 93-11. Moreover,we showed that mutations in DDM1 cause defects in plant growth and decreased 6mA level. 3Diagnostic accuracy of transient elastography (Fibro Scan) in detection of esophageal varices in patients with cirrhosis: A meta-analysis显示文摘AIM To investigate the diagnostic accuracy of Fibro Scan(FS) in detecting esophageal varices(EV) in cirrhotic patients.METHODS Through a systemic literature search of multiple databases, we reviewed 15 studies using endoscopy as a reference standard, with the data necessary to calculate pooled sensitivity(SEN) and specificity(SPE), positive and negative LR, diagnostic odds ratio(DOR) and area under receiver operating characteristics(AUROC). The quality of the studies was rated by the Quality Assessment of Diagnostic Accuracy studies-2 tool. Clinical utility of FS for EV was evaluated by a Fagan plot. Heterogeneity was explored using meta-regression and subgroup analysis. All statistical analyses were conducted via Stata12.0, MetaD isc1.4 and RevM an5.RESULTS In 15 studies(n = 2697), FS detected the presence of EV with the summary sensitivities of 84%(95%CI: 81.0%-86.0%), specificities of 62%(95%CI: 58.0%-66.0%), a positive LR of 2.3(95%CI: 1.81-2.94), a negative LR of 0.26(95%CI: 0.19-0.35), a DOR of 9.33(95%CI: 5.84-14.92) and an AUROC of 0.8262. FS diagnosed the presence of large EV with the pooled SEN of 0.78(95%CI: 75.0%-81.0%), SPE of 0.76(95%CI: 73.0%-78.0%), a positive and negative LR of 3.03(95%CI: 2.38-3.86) and 0.30(95%CI: 0.23-0.39) respectively, a summary diagnostic OR of 10.69(95%CI: 6.81-16.78), and an AUROC of 0.8321. A meta-regression and subgroup analysis indicated different etiology could serve as a potential source of heterogeneity in the diagnosis of the presence of EV group. A Deek's funnel plot suggested a low probability for publication bias.CONCLUSION Using FS to measure liver stiffness cannot provide high accuracy for the size of EV due to the various cutoff and different etiologies. 4Sirt1 inhibits renal tubular cell epithelial–mesenchymal transition through YY1 deacetylation in diabetic nephropathy显示文摘Silent information regulator 1(Sirt1)is a deacetylase,which plays an important role in the occurrence and development of diabetic nephropathy(DN).Our previous study shows that Yin yang 1(YY1),a widely expressed zinc finger DNA/RNA-binding transcription factor,is a novel regulator of renal fibrosis in diabetic nephropathy.Since the activity of YY1 is regulated via acetylation and deacetylation modification,this study aimed to explore whether Sirt1-induced deacetylation of YY1 mediated high glucose(HG)-induced renal tubular epithelial–mesenchymal transition(EMT)and renal fibrosis in vivo and in vitro.We first confirmed that Sirt1 expression level was significantly decreased in the kidney of db/db mice and in HG-treated HK-2 cells.Diabetes-induced Sirt1 reduction enhanced the level of YY1 acetylation and renal tubular EMT.Then,we manipulated Sirt1 expression in vivo and in vitro by injecting resveratrol(50 mg·kg^(−1)·d^(−1).ip)to db/db mice for 2 weeks or application of SRT1720(2.5μM)in HG-treated HK-2 cells,we found that activation of Sirt1 reversed the renal tubular EMT and YY1 acetylation induced by HG condition.On the contrary,Sirt1 was knocked down in db/m mice or EX527(1μM)was added in HK-2 cells,we found that inhibition of Sirt1 exacerbated renal fibrosis in diabetic mice and enhanced level of YY1 acetylation in HK-2 cells.Furthermore,knockdown of YY1 inhibited the ameliorating effect of resveratrol on renal tubular EMT and renal fibrosis in db/db mice.In conclusion,this study demonstrates that Sirt1 plays an important role in renal tubular EMT of DN through mediating deacetylation of YY1.Lei Du Xuan Qian Yuan Li Xi-zhi Li Lin-lin He Liu Xu Yi-qi Liu Cheng-cheng Li Pu Ma Fang-lin Shu Qian Lu Xiao-xing Yin 2021Acta Pharmacologica Sinica2021,42,2:8

6Integrating multisource RS data and GIS techniques to assist the evaluation of resource-environment carrying capacity in karst mountainous area显示文摘The karst mountainous area is an ecologically fragile region with prominent humanland contradictions.The resource-environment carrying capacity(RECC)of this region needs to be further clarified.The development of remote sensing(RS)and geographic information system(GIS)provides data sources and processing platform for RECC monitoring.This study analyzed and established the evaluation index system of RECC by considering particularity in the karst mountainous area of Southwest China;processed multisource RS data(Sentinel-2,Aster-DEM and Landsat-8)to extract the spatial distributions of nine key indexes by GIS techniques(information classification,overlay analysis and raster calculation);proposed the methods of index integration and fuzzy comprehensive evaluation of the RECC by GIS;and took a typical area,Guangnan County in Yunnan Province of China,as an experimental area to explore the effectiveness of the indexes and methods.The results showed that:(1)The important indexes affecting the RECC of karst mountainous area are water resources,tourism resources,position resources,geographical environment and soil erosion environment.(2)Data on cultivated land,construction land,minerals,transportation,water conservancy,ecosystem services,topography,soil erosion and rocky desertification can be obtained from RS data.GIS techniques integrate the information into the RECC results.The data extraction and processing methods are feasible on evaluating RECC.(3)The RECC of Guangnan County was in the mid-carrying level in 2018.The midcarrying and low-carrying levels were the main types,accounting for more than 80.00%of the total study area.The areas with high carrying capacity were mainly distributed in the northern regions of the northwest-southeast line of the county,and other areas have a low carrying capacity comparatively.The coordination between regional resource-environment status and socioeconomic development is the key to improve RECC.This study explores the evaluation index system of RECC in karst mountainous area and the application of multisource RS data and GIS techniques in the comprehensive evaluation.The methods can be applied in related fields to provide suggestions for data/information extraction and integration,and sustainable development.PU Jun-wei ZHAO Xiao-qing MIAO Pei-pei LI Si-nan TAN Kun WANG Qian TANG Wei 2020Journal of Mountain Science2020,17,10:6

8A flutter-effect-based triboelectric nanogenerator for breeze energy collection from arbitrary directions and self-powered wind speed sensor显示文摘Triboelectric nanogenerators(TENGs)have been developed rapidly into an efficient wind energy collection equipment.Reducing the frictionwear and energy loss in breeze energy collection is a research direction worthy of attention.Herein,a flutter-effect-based triboelectricnanogen erator(FE-TENG)is designed to collect the breeze energy at low wind speed from arbitrary directions.Distinguishing from previouswind-driven TENGs,the wind-driven part of this device is separated from the TENG units,which not only avoids the wear of friction layerscaused by direct wind contact but also reduces the energy loss,therefore,relatively stable electric outputs are obtained with Voc-281 V,Isc-13.4μA,Qsc-143 nC,and output power-4 mW at the wind speed of 4.5 m/s,respectively.In addition,a real-time wind speed monitoringsystem based on LabVIEW software with high sen sitivity and fast response to wind is achieved relying on the excellent linear relation shipbetween wind speed and electrical output signal.Furthermore,it has been successfully applied as power sources for portable electronics,about 170 commercial light-emitting devices(LEDs)are lighted and a digital watch is successfully driven at the wind speed of 2.9 m/s.This worknot only provides a new structure and idea for the future collection of clean and sustainable breeze energy from arbitrary directions but also hasgreat potential in the field of self-powered systems.Jie Hu Xianjie Pu Hongmei Yang Qixuan Zeng Qian Tang Dazhi Zhang Chenguo Hu Yi Xi 2019Nano Research2019,12,12:6

11New insights on the enhanced non-hydroxyl radical contribution under copper promoted TiO_(2)/GO for the photodegradation of tetracycline hydrochloride显示文摘TiO_(2)/graphene oxide(GO)as photocatalyst in the photo-degradation of multitudinous pollutants has been extensively studied.But its low photocatalytic efficiency is attributed to the high band gap energy which lead to low light utilization.Cu-TiO_(2)/GO was synthesized via the impregnation methods to enhance the catalytic performance.The Cu-TiO_(2)/GO reaction rate constant for photo-degradation of pollutants(tetracycline hydrochloride,TC)was about 1.4 times that of TiO_(2)/GO.In 90 min,the removal ratio of Cu-TiO_(2)/GO for TC was 98%,and the maximum degradation ratio occurred at p H 5.After five cycles,the removal ratio of Cu-Ti O_(2)/GO still exceeded 98%.UV-visible adsorption spectra of Cu-Ti O_(2)/GO showed that its band gap was narrower than TiO_(2)/GO.Electron paramagnetic resonance(EPR)spectra test illustrated the generation rate of·O_(2)^(-)and·OH was higher in Cu-TiO_(2)/GO system than TiO_(2)/GO and TiO_(2) system.The contribution sequence of oxidative species was·O_(2)^(-)>holes(h+)>·OH in both TiO_(2)/GO and Cu-Ti O_(2)/GO system.Interestingly,the contribution of·OH in Cu-TiO_(2)/GO was less than that in TiO_(2)/GO during the photo-degradation process.This phenomenon was attributed to the better adsorption performance of Cu-Ti O_(2)/GO which could reduce the accessibility of TC to·OH in liquid.The enhanced non-hydroxyl radical contribution could be attributed to that the more other active species or sites on(nearby)the surface of Cu-TiO_(2)/GO generated after doping Cu.These results provide a new perspective for the tradition metal-doped conventional catalysts to enhance the removal of organic pollutants in the environment.Sijia Qian Shengyan Pu Ying Zhang Peng Wang Yingchen Bai Bo Lai 2021Journal of Environmental Sciences2021,33,2:4
12Prevalence and clinical significance of pathogenic germline BRCA1/2 mutations in Chinese non-small cell lung cancer patients显示文摘Objective: Germline alterations in the breast cancer susceptibility genes type 1 and 2, BRCA1 and BRCA2, predispose individuals to hereditary cancers, including breast, ovarian, prostate, pancreatic, and stomach cancers.Accumulating evidence suggests inherited genetic susceptibility to lung cancer.The present study aimed to survey the prevalence of pathogenic germline BRCA mutations(gBRCAm) and explore the potential association between gBRCAm and disease onset in Chinese advanced non-small cell lung cancer(NSCLC) patients.Methods: A total of 6,220 NSCLC patients were screened using capture-based ultra-deep targeted sequencing to identify patients harboring germline BRCA1/2 mutations.Results: Out of the 6,220 patients screened, 1.03%(64/6,220) of the patients harbored the pathogenic gB RCAm, with BRCA2 mutations being the most predominant mutations(49/64, 76.5%).Patients who developed NSCLC before 50 years of age were more likely to carry gBRCAm(P = 0.036).Among the patients harboring classic lung cancer driver mutations, those with concurrent gBRCAm were significantly younger than those harboring the wild-type gBRCA(P = 0.029).By contrast, the age of patients with or without concurrent gBRCAm was comparable to those of patients without the driver mutations(P = 0.972).In addition, we identified EGFR-mutant patients with concurrent gBRCAm who showed comparable progression-free survival but significantly longer overall survival(P = 0.002) compared to EGFR-mutant patients with wild-type germline BRCA.Conclusions: Overall, our study is the largest survey of the prevalence of pathogenic gBRCAm in advanced Chinese NSCLC patients.Results suggested a lack of association between germline BRCA status and treatment outcome of EGFR-TKI.In addition,results showed a positive correlation between pathogenic gB RCAm and an early onset of NSCLC.Xingsheng Hu Dongyong Yang Yalun Li Li Li Yan Wang Peng Chen Song Xu Xingxiang Pu Wei Zhu Pengbo Deng Junyi Ye Hanhan Zhang Analyn Lizaso Hao Liu Xinru Mao Hai Huang Qian Chu Chengping Hu 2019Cancer Biology & Medicine2019,16,3:4
13Growth Conditions and 3-D Seismic Delineation of Carboniferous Barrier Reefs in the Southwestern Tarim Basin显示文摘The Tarim Basin was a craton block during the Carboniferous period. Except for its open southwestern(SWW) deepening basin margin, the central part of the basin was surrounded by northern, eastern, and southern uplifts. In most parts of the basin, there are three layers of carbonate rocks called Biocalcarenite member, standard limestone member and Xiaohaizi Formation. The present-day Maigeti slope, located in the southwestern area of the basin, was ever a paleogeographic carbonate platform margin to clinoform. Boreholes in the area reveal over ten meters of high-energy biospararenite of beach deposition in Xiaohaizi Formation. This feature is quite different from the low-energy micritic limestone of equivalent horizon in the middle and northern basins. Such typical carbonate beach and platform margin backgrounds indicate an environment conducive for reef growth. According to an interpretation of 3-D seismic data on a 600 km2 Xinbaza survey, a 25 km-long barrier reef orientated in an east-westward direction is delineated with a hummocky geometry, a 1–2 km wide south-dip reef front, and a 3–6 km wide north-dip backreef. The reef buttress and outlines are also shown on an isopach map of the Xiaohaizi Formation as well as horizon slices of coherency, amplitude, and frequency attributes. The reef extends beyond the range of the rectangular 3-D seismic survey. The thickness of the Xiaohaizi Formation is over 60 m at the central reef, and 12 m larger than the outside of the reef. The barrier reef consists of segments separated by narrow tidal inlets. The neighborhood of the reef is mainly beach deposition of bioclasticgrainstones. Due to Late Permier and Late Tertial tectonic movements, the barrier reef was tilted to dip in a SWW direction. Its updip part was broken and misplaced by a set of reverse faults. 15Genome-wide analysis indicates diverse physiological roles of the turnip (Brassica rapa var. rapa) oligopeptide transporters gene family显示文摘Oligopeptide transporters(OPTs) encode integral membrane-localized proteins and have a broad range of substrate transport capabilities. Here, 28 BrrOPT genes were identified in the turnip. Phylogenetic analyses revealed two well-supported clades in the OPT family, containing 15 BrrOPTs and 13 BrrYSLs.The exon/intron structure of OPT clade are conserved but the yellow stripe-like(YSL) clade was different.The exon/intron of the YSL clade possesses structural differences, whereas the YSL class motifs structure are conserved. The OPT genes are distributed unevenly among the chromosomes of the turnip genome.Phylogenetic and chromosomal distribution analyses revealed that the expansion of the OPT gene family is mainly attributable to segmental duplication. For the expression profiles at different developmental stages, a comprehensive analysis provided insights into the possible functional divergence among members of the paralog OPT gene family. Different expression levels under a variety of ion deficiencies also indicated that the OPT family underwent functional divergence during long-term evolution.Furthermore. BrrOPT8.1, BrrYSL1.2, BrrYSL1.3, BrrYSL6 and BrrYSL9 responded to Fe(Ⅱ) treatments and BrrYSL7 responded to calcium treatments, BrrYSL6 responded to multiple treatments in root, suggesting that turnip OPTs may be involved in mediating cross-talk among different ion deficiencies. 16Sustainable Investment Forecasting of Power Grids Based on theDeep Restricted Boltzmann Machine Optimized by the Lion Algorithm显示文摘This paper proposes a new power grid investment prediction model based on the deep restricted Boltzmann machine(DRBM)optimized by the Lion algorithm(LA).Firstly,two factors including transmission and distribution price reform(TDPR)and 5G station construction were comprehensively incorporated into the consideration of influencing factors,and the fuzzy threshold method was used to screen out critical influencing factors.Then,the LA was used to optimize the parameters of the DRBM model to improve the model’s prediction accuracy,and the model was trained with the selected influencing factors and investment.Finally,the LA-DRBM model was used to predict the investment of a power grid enterprise,and the final prediction result was obtained by modifying the initial result with the modifying factors.The LA-DRBMmodel compensates for the deficiency of the singlemodel,and greatly improves the investment prediction accuracy of the power grid.In this study,a power grid enterprise was taken as an example to carry out an empirical analysis to prove the validity of the model,and a comparison with the RBM,support vector machine(SVM),back propagation neural network(BPNN),and regression model was conducted to verify the superiority of the model.The conclusion indicates that the proposed model has a strong generalization ability and good robustness,is able to abstract the combination of low-level features into high-level features,and can improve the efficiency of the model’s calculations for investment prediction of power grid enterprises.Qian Wang Xiaolong Yang Di Pu Yingying Fan 2022Computer Modeling in Engineering & Sciences2022,,1:2
17On-demand assembly of polymeric nanoparticles for longer-blood-circulation and disassembly in tumor for boosting sonodynamic therapy显示文摘Sonodynamic therapy (SDT) is one of the promising strategies for tumor therapy, but its application is usually hindered by fast clearance in blood-circulation, abnormal tumor microenvironment, and inefficient generation of reactive oxygen species. To solve these problems, we proposed an on-demand assembly-disassembly strategy, where the assembly is favorable for longer-blood-circulation and then the disassembly in tumor is favorable for boosting SDT. Hematoporphyrin monomethyl ether (HMME) as the model of organic sonosensitizers were conjugated with hyaluronic acid (HA). Then HA-HMME was mixed with catalase (CAT) and assembled into polymeric nanoparticles (CAT@HA-HMME NPs) with size of ~80 nm. CAT@HA-HMME NPs exhibit good biocompatibility and a longer blood half-time (t1/2 = 4.17 h) which is obviously longer than that (~0.82 h) of HMME molecules. After HA receptor-mediated endocytosis of cancer cells, CAT@HA-HMME NPs can be cleaved by endogenous hyaluronidase, resulting in the on-demand disassembly in tumor to release HA-HMME molecules and CAT. The CAT catalyzes the endogenous H_(2)O_(2) into O_(2) to relieve the hypoxic microenvironment, and the released HA-HMME exhibits a higher ROS generation ability, greatly boosting SDT for the inhibition of tumor growth. 18Fourier-domain optical coherence tomography-guided phototherapeutic keratectomy for the treatment of anterior corneal scarring显示文摘AIM:To evaluate the safety,visual and anatomic outcomes of fourier-domain optical coherence tomography(FD-OCT)-guided excimer laser phototherapeutic keratectomy(PTK)combined with photorefractive keratectomy(PRK)surgery in treating anterior corneal scarring.METHODS:Clinical data of 23 eyes of 21 patients with anterior corneal scarring underwent FD-OCT-guided PTK and PRK from Dec.2014 to Jul.2016 were reviewed.Patients were assessed for preoperative and postoperative uncorrected visual acuity(UCVA),best spectacle-corrected visual acuity(BSCVA),contrast sensitivity(CS),FD-OCT,corneal topography and colour figures of anterior segments.RESULTS:The preoperative corneal pathologic conditions included viral keratitis(7 patients,7 eyes),band keratopathy(2 patients,4 eyes),corneal dystrophy(4 patients,4 eyes),traumatic corneal disease(2 patients,2 eyes)and corneal chemical injury(6 patients,6 eyes).Mean follow-up time was 10.65(range,3-19)mo.UCVA(in IogMAR)improved from a mean of 0.79(95%Cl,0.28­1.29)preoperatively to a mean of 0.45(95%Cl,0.29-0.62)postoperatively(P=0.021).BSCVA(in IogMAR)improved from 0.57(95%Cl,0.27-0.88)preoperatively to a mean of 0.28(95%Cl,0.15-0.41)postoperatively(P=0.001).Corneal topographic indices postoperatively showed significant improvement in corneal cylinder(P=0.009),the surface regularity index(P=0.007)and surface asymmetry index(P=0.00).Postoperative spherical equivalent averaged-0.53 diopters(-1.49 to 0.42).No complications were associated with the treatment.CONCLUSION:FD-OCT-guided PTK combined with PRK is safe and effective for the treatment of anterior corneal scarring by eliminating or reducing corneal opacities.Yu-Li Yang Qian Jian Bo Liu Ke Wang Yu-Juan Chen Lian Tan Mei-Jun Pu Yong Liu 2020International Journal of Ophthalmology(English edition)2020,13,11:2

20Exosomes secreted from cardiomyocytes suppress the sensitivity of tumor ferroptosis in ischemic heart failure显示文摘Heart failure(HF)patients in general have a higher risk of developing cancer.Several animal studies have indicated that cardiac remodeling and HF remarkably accelerate tumor progression,highlighting a cause-and-effect relationship between these two disease entities.Targeting ferroptosis,a prevailing form of non-apoptotic cell death,has been considered a promising therapeutic strategy for human cancers.Exosomes critically contribute to proximal and distant organ-organ communications and play crucial roles in regulating diseases in a paracrine manner.However,whether exosomes control the sensitivity of cancer to ferroptosis via regulating the cardiomyocyte-tumor cell crosstalk in ischemic HF has not yet been explored.Here,we demonstrate that myocardial infarction(MI)decreased the sensitivity of cancer cells to the canonical ferroptosis activator erastin or imidazole ketone erastin in a mouse model of xenograft tumor.Post-MI plasma exosomes potently blunted the sensitivity of tumor cells to ferroptosis inducers both in vitro in mouse Lewis lung carcinoma cell line LLC and osteosarcoma cell line K7M2 and in vivo with xenograft tumorigenesis model.The expression of miR-22-3p in cardiomyocytes and plasma-exosomes was significantly upregulated in the failing hearts of mice with chronic MI and of HF patients as well.Incubation of tumor cells with the exosomes isolated from post-MI mouse plasma or overexpression of miR-22-3p alone abrogated erastin-induced ferroptotic cell death in vitro.Cardiomyocyte-enriched miR-22-3p was packaged in exosomes and transferred into tumor cells.Inhibition of cardiomyocyte-specific miR-22-3p by AAV9 sponge increased the sensitivity of cancer cells to ferroptosis.ACSL4,a pro-ferroptotic gene,was experimentally established as a target of miR-22-3p in tumor cells.Taken together,our findings uncovered for the first time that MI suppresses erastin-induced ferroptosis through releasing miR-22-3p-enriched exosomes derived from cardiomyocytes.Therefore,targeting exosome-mediated cardiomyocyte/tumor pathological communication may offer a novel approach for the ferroptosis-based antitumor therapy.Ye Yuan Zhongting Mei Zhezhe Qu Guanghui Li Shuting Yu Yingqi Liu Kuiwu Liu Zhihua Shen Jiaying Pu Yanquan Wang Changhao Wang Zhiyong Sun Qian Liu Xiaochen Pang Ao Wang Zijing Ren Tong Wang Ying Liu Jinhuan Hong Jiajie Xie Xin Li Zhonghua Wang Weijie Du Baofeng Yang 2023Signal Transduction and Targeted Therapy2023,8,4:2
