1Characterization of drug responses of mini patient-derived xenografts in mice for predicting cancer patient clinical therapeutic response显示文摘Background:Patient-derived organoids and xenografts(PDXs)have emerged as powerful models in functional diag-nostics with high predictive power for anticancer drug response.However,limitations such as engraftment failure and time-consuming for establishing and expanding PDX models followed by testing drug efficacy,and inability to subject to systemic drug administration for ex vivo organoid culture hinder realistic and fast decision-making in selecting the right therapeutics in the clinic.The present study aimed to develop an advanced PDX model,namely MiniPDX,for rapidly testing drug efficacy to strengthen its value in personalized cancer treatment.Methods:We developed a rapid in vivo drug sensitivity assay,OncoVee®MiniPDX,for screening clinically relevant regimens for cancer.In this model,patient-derived tumor cells were arrayed within hollow fiber capsules,implanted subcutaneously into mice and cultured for 7 days.The cellular activity morphology and pharmacokinetics were systematically evaluated.MiniPDX performance(sensitivity,specificity,positive and negative predictive values)was examined using PDX as the reference.Drug responses were examined by tumor cell growth inhibition rate and tumor growth inhibition rate in PDX models and MiniPDX assays respectively.The results from MiniPDX were also used to evaluate its predictive power for clinical outcomes.Results:Morphological and histopathological features of tumor cells within the MiniPDX capsules matched those both in PDX models and in original tumors.Drug responses in the PDX tumor graft assays correlated well with those in the corresponding MiniPDX assays using 26 PDX models generated from patients,including 14 gastric cancer,10 lung cancer and 2 pancreatic cancer.The positive predictive value of MiniPDX was 92%,and the negative predictive value was 81%with a sensitivity of 80%and a specificity of 93%.Through expanding to clinical tumor samples,Min-iPDX assay showed potential of wide clinical application.Conclusions:Fast in vivo MiniPDX assay based on capsule implantation was developed-to assess drug responses of both PDX tumor grafts and clinical cancer specimens.The high correlation between drug responses of paired MiniPDX and PDX tumor graft assay,as well as translational data suggest that MiniPDX assay is an advanced tool for personalized cancer treatment.Feifei Zhang Wenjie Wang Yuan Long Hui Liu Jijun Cheng Lin Guo Rongyu Li Chao Meng Shan Yu Qingchuan Zhao Shun Lu Lili Wang Haitao Wang Danyi Wen 2018Cancer Communications2018,38,1:20
2Model of nonlinear coupled thermo-hydro-elastodynamics response for a saturated poroelastic medium显示文摘Based on the Biot’s wave equation and theory of thermodynamic, Darcy law of fluid and the modified Fourier law of heat conduction, a nonlinear fully coupled thermo-hydro-elastodynamic response model (THMD) for saturated porous medium is derived. The compressibility of the medium, the influence of fluid flux on the heat flux, and the influence of change of temperature on the fluid flux are considered in this model. With some simplification, the coupled nonlinear thermo-hydro-elastodynamic response model can be reduced to the thermo-elastodynamic (TMD) model based on the traditional Fourier law and, further more, to the Biot’s wave equation without considering the heat phase. At last, the problem of one dimensional cylindrical cavity subjected to a time-dependent thermal/mechanical shock is analyzed by using the Laplace technique, the numerical results are used to discuss the influence of Biot’s modulus M and coefficient of thermoos-mosis on displacement and to compare with the results of thermo-elastodynamic response to ascertain the validity of this model.LIU GanBin XIE KangHe ZHENG RongYue 2009Science China(Technological Sciences)2009,52,8:9
3Research on Radiation Characteristics of Polymeric Plastic Fiber显示文摘The anti-radiation mechanism of polymeric plastic fiber is analyzed. The radiation characteristic of this type fiber is studied. Our experimental results indicate that under the low radiation dosage (below 1 kGy) the plastic optical fiber’s transmission rate rises instead of descends while this type of fiber is radiated by γ ray, electron beam and proton beam respectively. After a period of time, it gradually reaches a constant value. But under the high radiation dosage (above 1 kGy) the fiber’s transmission rate descends after radiation. 5Recent Advances and Applications Toward Emerging Lithium–Sulfur Batteries:Working Principles and Opportunities显示文摘Lithium–sulfur(Li-S)batteries have been considered as promising candidates for large-scale high energy density devices due to the potentially high energy density,low cost,and more pronounced ecological compatibility.However,the complex Li-S conversion reactions,unsatisfactory battery performance,and unsafe metallic Li anode restrict the development of Li-S batteries to achieve commercialization.This review mainly focuses on three aspects which are the remaining challenges,recent advances,and applications in Li-S batteries.Firstly,this review portrays Li-S conversion chemistry involving the multi-step and multi-electron reaction mechanism,as well as the remaining challenges.Then,the scientific strategies and very recent advances of the cathode,electrolyte,lithium anode,and other constituent parts of Li-S batteries are detailly summed up,as well as their advantages and limitations.For the sake of promoting the Li-S batteries practicalization,next section is primarily concerned with problems,the corresponding solutions,and application scenarios of practical pouch cells.Finally,the important findings as guidelines and some future directions as trends for developing emerging Li-S batteries are briefly summarized.Rongyu Deng Meng Wang Huanyu Yu Shunrui Luo Jinhui Li Fulu Chu Bin Liu Feixiang Wu 2022Energy & Environmental Materials2022,5,3:3
6FROST RESISTANCE OF POLYVINYL ALCOHOL FIBER REINFORCED CEMENTITIOUS COMPOSITES显示文摘This article studies the frost resistance of polyvinyl alcohol (PVA) fiber reinforced cementitious composites. The test samples contain different PVA fiber volume fractions such as 0, 0.5%, 1% and 2%. The freeze-thaw test is carried on in TDR type apparatus. 8Fabrication and characterization of implantable silicon neural probe with microfluidic channels显示文摘In this paper,a silicon-based neural probe with microfluidic channels was developed and evaluated.The probe can deliver chemicals or drugs to the target neurons while simultaneously recording the electrical action of these neurons extracellularly.The probe was fabricated by double-sided deep reactive ion etching(DRIE) from a silicon-on-insulator(SOI) wafer.The flu-idic channels were formed with V-shape groove etching on the silicon probe and sealed with silicon nitride and parylene-C.The shank of the probe is 4 mm long and 120 μm wide.The thickness of the probe is 100 μm.The probe has two fluidic chan-nels and two recording sites.The microfluidic channels can withstand a pressure drop as much as 30 kPa and the flow resistiv-ity of the microfluidic channel is 0.13 μL min-1 kPa-1.The typical impedance of the neural electrode is 32.3 kΩ at 1 kHz at room temperature.GUO Kai PEI WeiHua LI XiaoQian GUI Qiang TANG RongYu LIU Jian CHEN HongDa 2012Science China(Technological Sciences)2012,55,1:1

10Dynamic process of phagocytosis and forms of macrophage cell death induced by ingestion of apoptotic neutrophils显示文摘Clearance of apoptotic neutrophils by macrophages is important for both the successful resolution of acute inflammation and homeostasis.However,the dynamic process of phagocytosis of apoptotic neutrophils by macrophages and the fate of macrophages after the ingestion of apoptotic neutrophils has not been well documented.In the present study,we staged the recognition and tethering,internalization,digestion and exocytosis steps of phagocytosis of apoptotic neutrophils.Furthermore,we found that after the ingestion of apoptotic cells,a subset of macrophages underwent cell death by autophagy,apoptosis or oncosis as revealed by transmission electron microscopy and confocal microscopy combined with specific dyes.The percentage of autophagic,apoptotic and oncotic macrophages were 8.00%±2.00%,12.33%±2.08%,and 3.66%±1.50%,respectively.These results indicated that after ingestion of apoptotic neutrophils,a subset of macrophages undergoes autophagy and apoptosis.We propose that autophagy of macrophages after the ingestion of apoptotic cells may be a new mechanism present in the resolution of inflammation.WANG Jiong HUANG WeiLin WANG Cheng LIU RongYu 2014Science China(Life Sciences)2014,57,10:1
