2Recent advances in two-photon imaging: technology developments and biomedical applications显示文摘In the past two decades, two-photon microscopy (TPM) transforms biomedicalresearch, allowing nondestructive high-resolution fluorescent molecular imaging and label-free imaging in vivo and in real time. Here we review the recent advances of TPM technologyincluding novel laser sources, new image acquisition paradiams, and microendoscopicimaging systems. Then, we survey the capabilities of TPM imagingof biological relevant molecules such as nicotinamideadenine dinucleotide (NADH), flavin adenine dinucleotide (FAD),and reactive oxygen species (ROS). 4Dynamic degradation patterns of porous polycaprolactone/β-tricalcium phosphate composites orchestrate macrophage responses and immunoregulatory bone regeneration显示文摘Biodegradable polycaprolactone/β-tricalcium phosphate(PT)composites are desirable candidates for bone tissue engineering applications.A higherβ-tricalcium phosphate(TCP)ceramic content improves the mechanical,hydrophilic and osteogenic properties of PT scaffolds in vitro.Using a dynamic degradation reactor,we estab-lished a steady in vitro degradation model to investigate the changes in the physio-chemical and biological properties of PT scaffolds during degradation.PT46 and PT37 scaffolds underwent degradation more rapidly than PT scaffolds with lower TCP contents.In vivo studies revealed the rapid degradation of PT(PT46 and PT37)scaffolds disturbed macrophage responses and lead to bone healing failure.Macrophage co-culture assays and a subcutaneous implantation model indicated that the scaffold degradation process dynamically affected macro-phage responses,especially polarization.RNA-Seq analysis indicated phagocytosis of the degradation products of PT37 scaffolds induces oxidative stress and inflammatory M1 polarization in macrophages.Overall,this study reveals that the dynamic patterns of biodegradation of degradable bone scaffolds highly orchestrate immune responses and thus determine the success of bone regeneration.Therefore,through evaluation of the biological effects of biomaterials during the entire process of degradation on immune responses and bone regeneration are necessary in order to develop more promising biomaterials for bone regeneration.Hao Wu Xinghui Wei Yichao Liu Hui Dong Zhen Tang Ning Wang Shusen Bao Zhigang Wu Lei Shi Xiongfei Zheng Xiaokang Li Zheng Guo 2023Bioactive Materials2023,,3:1
5The coupling effect of slow-rate mechanical motion on the confined etching process in electrochemical mechanical micromachining显示文摘By introducing the mechanical motion into the confined etchant layer technique(CELT), we have developed a promising ultraprecision machining method, termed as electrochemical mechanical micromachining(ECMM), for producing both regular and irregular three dimensional(3 D) microstructures. It was found that there was a dramatic coupling effect between the confined etching process and the slow-rate mechanical motion because of the concentration distribution of electrogenerated etchant caused by the latter. In this article, the coupling effect was investigated systemically by comparing the etchant diffusion, etching depths and profiles in the non-confined and confined machining modes. A two-dimensional(2 D) numerical simulation model was proposed to analyze the diffusion variations during the ECMM process, which is well verified by the machining experiments. The results showed that, in the confined machining mode, both the machining resolution and the perpendicularity tolerance of side faces were improved effectively. 7Diagnosis of knee osteoarthritis by OCT and surface-enhanced Raman spectroscopy显示文摘In this paper,optical coherence tomography(OCT)and surface-enhanced Raman spectroscopy(SERS)were used to characterize normal knee joint(NKJ)tissue and knee osteoarthritis(KOA)tissue ex vivo.OCT images show that there is a clear hierarchical structure in NKJ tissue,including surface layer,transitional layer,radiation layer and cartilage matrix calcification layer tissue structure,while the hierarchical structure of KOA tissue is not clear and unevenly distributed,and the pathological tissues at different stages also show significant di®erences.SERS shows that NKJ tissue and mild osteoarthritic knee cartilage(MiKOA)tissue have strong characteristic Raman peaks at 964,1073(1086),1271,1305,1442,1660 and 1763 cm^(-1).Compared with the Raman spectrum of NKJ tissue,the Raman characteristic peaks of MiKOA tissue have some shifts,moving from 1073 cm^(-1)to 1086 cm^(-1)and from 1542 cm^(-1)to 1442 cm^(-1).There is a characteristic Raman peak of 1271 cm^(-1)in MiKOA tissue,but not in NKJ tissue.Compared with NKJ tissue,severely degenerated cartilage(SdKOA)tissues show some new SERS peaks at 1008,1245,1285,1311 and 1321 cm^(-1),which are not seen in SERS spectra of NKJ tissue.Principal component analysis(PCA)was used to analyze the Raman spectra of 1245–1345 cm^(-1)region.The results show that PCA can distinguish NKJ,MiKOA and SdKOA tissues and the accuracy is about 90%.These results indicate that OCT can clearly distinguish NKJ,MiKOA,moderate osteoarthritic knee cartilage(MoKOA)and SdKOA tissue,while SERS can provide further judgment basis.The results also prove that the contents of protein and polysaccharide in knee tissue are changed during the pathological process of knee tissue,which is the cause of pain caused by poor friction in knee joint during movement.Ran An Huaimin Gu Zhouyi Guo Huiqing Zhong Huajiang Wei Guoyong Wu Yonghong He Shusen Xie Hongqin Yang 2022Journal of Innovative Optical Health Sciences2022,15,5:0
8Stratified Analysis of Survival Benefit for ABO-incompatible Deceased-donor Liver Transplantation:Multicenter Propensity Score-matched Study显示文摘Background and Aims:Liver transplantation(LT)using ABO-incompatible(ABOi)grafts can extend the donor pool to a certain extent and hence reduce the waiting time for transplantation.However,concerns of the impending prognosis associated with this option,especially for patients with liver failure and higher model for end-stage liver disease(MELD)scores,who tend to be more fragile during the waiting period before LT.Methods:Recipients undergoing LT for acute-onchronic liver failure or acute liver failure were retrospectively enrolled at four institutions.Overall survival was compared and a Cox regression analysis was performed.Propensity score matching was performed for further comparison.Patients were stratified by MELD score and cold ischemia time(CIT)to determine the subgroups with survival benefits.Results:Two hundred ten recipients who underwent ABOi LT and 1,829 who underwent ABO compatible(ABOc)LT were enrolled.The 5-year overall survival rate was significantly inferior in the ABOi group compared with the ABOc group after matching(50.6%vs.75.7%,p<0.05).For patients with MELD scores≤30,using ABOi grafts achieved a comparable overall survival rate as using ABOc grafts(p>0.05).Comparison of the survival rates revealed no statistically significant difference for patients with MELD scores≥40(p>0.05).For patients with MELD scores of 31-39,the overall survival rate was significantly inferior in the ABOi group compared with the ABOc group(p<0.001);however,the rate was increased when the liver graft CIT was<8 h.Conclusions:For recipients with MELD scores≤30,ABOi LT had a prognosis comparable to that of ABOc LT and can be regarded as a feasible option.For recipients with MELD scores≥40,ABOi should be adopted with caution in emergency cases.For recipients with MELD scores of 31-39,the ABOi LT prognosis was worse.However,those patients benefited from receiving ABOi grafts with a CIT of<8 h.Mengfan Yang Xuyong Wei Abdul Rehman Khan Qiang Wei Rui Wang Binhua Pan Kun Wang Zhisheng Zhou Di Lu Beini Cen Shuijun Zhang Wenzhi Guo Shusen Zheng Yang Yang Xiao Xu 2023Journal of Clinical and Translational Hepatology2023,11,4:0
9Elimination of Voids at Interface of β-SiC Films and Si Substrate by Laser CVD显示文摘Void-free β-SiC films were deposited on Si(001) substrates by laser chemical vapor deposition using hexamethyldisilane(HMDS) as the precursor. The effect of the time of introducing HMDS, i e, the substrate temperature when HMDS introduced(T_(in)), on the preferred orientation, surface microstructure and void was investigated. The orientation of the deposited SiC films changed from <001> to random to <111> with increasing T_(in). The surface showed a layer-by-layer microstructure with voids above T_(in) ≥ 773 K, and then transformed into mosaic structure without voids at T_(in)= 298 K. The mechanism of the elimination of voids was discussed. 10Magnesium surface-activated 3D printed porous PEEK scaffolds for in vivo osseointegration by promoting angiogenesis and osteogenesis显示文摘Polyetheretherketone(PEEK)has been an alternative material for titanium in bone defect repair,but its clinical application is limited by its poor osseointegration.In this study,a porous structural design and activated surface modification were used to enhance the osseointegration capacity of PEEK materials.Porous PEEK scaffolds were manufactured via fused deposition modeling and a polydopamine(PDA)coating chelated with magnesium ions(Mg^(2+))was utilized on the surface.After surface modification,the hydrophilicity of PEEK scaffolds was significantly enhanced,and bioactive Mg^(2+)could be released.In vitro results showed that the activated surface could promote cell proliferation and adhesion and contribute to osteoblast differentiation and mineralization;the released Mg^(2+)promoted angiogenesis and might contribute to the formation of osteogenic H-type vessels.Furthermore,porous PEEK scaffolds were implanted in rabbit femoral condyles for in vivo evaluation of osseointegration.The results showed that the customized three-dimensional porous structure facilitated vascular ingrowth and bone ingrowth within the PEEK scaffolds.The PDA coating enhanced the interfacial osseointegration of porous PEEK scaffolds and the released Mg^(2+)accelerated early bone ingrowth by promoting early angiogenesis during the coating degradation process.This study provides an efficient solution for enhancing the osseointegration of PEEK materials,which has high potential for translational clinical applications.Xinghui Wei Wenhao Zhou Zhen Tang Hao Wu Yichao Liu Hui Dong Ning Wang Hai Huang Shusen Bao Lei Shi Xiaokang Li Yufeng Zheng Zheng Guo 2023Bioactive Materials2023,,2:0
11Research and application of uniform material counting method based on machine vision显示文摘Manufacturing and agricultural industries use manual methods to count materials. This leads to low accuracy and inefficiency. This paper proposes a secondary counting method that combines main and differential counting. The area-fill identification algorithm is applied to mark the counted materials. To verify the effectiveness of the proposed counting algorithm, numbers of countings are conducted for different materials, such as the screws, hole gaskets, beans, jujube, etc. The results show that the counting accuracy reaches 98% for materials with size of 2—20 mm.
