| 1 | Melatonin Augments the Effects of Fluoxetine on Depression-Like Behavior and Hippocampal BDNF–TrkB Signaling显示文摘Depression is a debilitating psychiatric disorder with a huge socioeconomic burden, and its treatment relies on antidepressants including selective serotonin reuptake inhibitors(SSRIs). Recently, the melatonergic system that is closely associated with the serotonergic system has been implicated in the pathophysiology and treatment of depression. However, it remains unknown whether combined treatment with SSRI and melatonin has synergistic antidepressant effects. In this study, we applied a sub-chronic restraint stress paradigm, and evaluated the potential antidepressant effects of combined fluoxetine and melatonin in adult male mice. Sub-chronic restraint stress(6 h/day for 10 days) induced depression-like behavior as shown by deteriorated fur state, increased latency to groom in the splash test, and increased immobility time in the forced-swim test. Repeated administration of either fluoxetine or melatonin at 10 mg/kg during stress exposure failed to prevent depression-like phenotypes. However,combined treatment with fluoxetine and melatonin at theselected dose attenuated stress-induced behavioral abnormalities. Moreover, we found that the antidepressant effects of combined treatment were associated with the normalization of brain-derived neurotrophic factor(BDNF)–tropomyosin receptor kinase B(Trk B) signaling in the hippocampus, but not in the prefrontal cortex. | 4 | Expression of miRNA-140 in Chondrocytes and Synovial Fluid of Knee Joints in Patients with Osteoarthritis显示文摘Objective To investigate the expression of miRNA-140 in chondrocytes and synovial fluid of osteoarthritis(OA) patients, and explore the relationship between the miRNA-140 expression and OA severity. Methods This study enrolled 30 OA patients who underwent total knee arthroplasty for chondrocytes sampling and 30 OA patients who underwent intra-articular injection for synovial fluid sampling. All OA patients were grouped into mild [Kellgren and Lawrence(KL) grade 1-2], moderate(KL grade 3) and severe(KL grade 4), with 10 in each subgroups for each sampling purposes. 7 non-OA patients and 10 patients with knee injury were collected for cartilage and synovial fluid sampling respectively as control groups. Chondrocytes were isolated from the cartilage tissue and cultured in vitro. Quantitative real time PCR for miRNA-140 in chondrocytes and synovial fluid were performed, and the U6 sn RNA was used as internal control. The expression difference of miRNA-140 among groups and correlation between the expression and the KL grade of OA were analysed using one-way ANOVA and Spearman test respectively. Results The expression of miRNA-140 in chondrocytes of knees in OA patients was reduced than that in normal knees, and the between-group difference was statistically significant(F=305.464, P<0.001). miRNA-140 could be detected in synovial fluid of both normal knees and OA knees, its relative expression level was reduced in synovial fluid of OA group compared with normal group, and the between-group difference was statistically significant as well(F=314.245, P<0.001). The relative expression level of miRNA-140 in both chondrocytes and synovial fluid were negatively correlated with the KL grade of OA(r=-0.969, P<0.001; r=-0.970, P<0.001). | 9 | A common sense geographic knowledge base for GIR显示文摘As background knowledge of geographic information retrieval (GIR),the gazetteers have their limitations. In this paper we propose to develop and implement a com-mon sense geographic knowledge base (CSGKB) instead of the gazetteers. We define that CSGKB is concerned with the representation of geographic knowledge in human brain and the simulation of geographic reasoning in daily life. Traditional geographic information system (GIS) is based on the model of map with its data based on geographic coordinates and its computation based on geometry. However,CSGKB,which is made up of geographic features and relationships and is based on qualitative spatio-temporal reasoning,can be viewed as the direct model of geographic world. This paper also discusses the characters of CSGKB and pre-sents its structure which is composed of knowledge base,inference engine,geo-graphic ontology and learner. The applications using CSGKB include geographic information retrieval (GIR),natural language processing (NLP),named entity rec-ognition (NER),Semantic Web,etc. At present,our work focuses on the design of geographic ontology and the implementation of the CSGKB knowledge base. | 17 | Evaluation of Snow Depth and Snow Cover Fraction Simulated by Two Versions of the Flexible Global Ocean–Atmosphere–Land System Model显示文摘Based on historical runs, one of the core experiments of the fifth phase of the Coupled Model Intercomparison Project(CMIP5), the snow depth(SD) and snow cover fraction(SCF) simulated by two versions of the Flexible Global Ocean– Atmosphere–Land System(FGOALS) model, Grid-point Version 2(g2) and Spectral Version 2(s2), were validated against observational data. The results revealed that the spatial pattern of SD and SCF over the Northern Hemisphere(NH) are simulated well by both models, except over the Tibetan Plateau, with the average spatial correlation coefficient over all months being around 0.7 and 0.8 for SD and SCF, respectively. Although the onset of snow accumulation is captured well by the two models in terms of the annual cycle of SD and SCF, g2 overestimates SD/SCF over most mid- and high-latitude areas of the NH. Analysis showed that g2 produces lower temperatures than s2 because it considers the indirect effects of aerosols in its atmospheric component, which is the primary driver for the SD/SCF difference between the two models. In addition, both models simulate the significant decreasing trend of SCF well over(30°–70°N) in winter during the period 1971–94. | 18 | Factors associated with upstaging in patients preoperatively diagnosed with ductal carcinoma in situ by core needle biopsy显示文摘Objective: Patients preoperatively diagnosed with ductal carcinoma in situ(DCIS) by core needle biopsy(CNB) exhibit a significant risk for upstaging on final pathology, which leads to major concerns of whether axillary staging is required at the primary operation. The present study aimed to identify clinicopathological factors associated with upstaging in patients preoperatively diagnosed with DCIS by CNB.Methods: The present study enrolled 604 patients(cN0 M0) with a preoperative diagnosis of pure DCIS by CNB, who underwent axillary staging between August 2006 and December 2015, at Fudan University Shanghai Cancer Center(Shanghai, China).Predictive factors of upstaging were analyzed retrospectively.Results: Of the 604 patients, 20.03%(n = 121) and 31.95%(n = 193) were upstaged to DCIS with microinvasion(DCISM) and invasive breast cancer(IBC) on final pathology, respectively. Larger tumor size on ultrasonography(> 2 cm) was independently associated with upstaging [odds ratio(OR) 1.558, P = 0.014]. Additionally, patients in lower breast imaging reporting and data system(BI-RADS) categories were less likely to be upstaged(4 B vs. 5: OR 0.435, P = 0.002;4 C vs. 5: OR 0.502, P = 0.001). Overall,axillary metastasis occurred in 6.79%(n = 41) of patients. Among patients with axillary metastasis, 1.38%(4/290), 3.31%(4/121)and 17.10%(33/193) were in the DCIS, DCISM, and IBC groups, respectively.Conclusions: For patients initially diagnosed with DCIS by CNB, larger tumor size on ultrasonography(> 2 cm) and higher BIRADS category were independent predictive factors of upstaging on final pathology. Previously, it was demonstrated that various microRNAs (miRNAs) play important roles in osteogenesis and regulate the complex process of osteogenic differentiation. However, it remains unclear how miRNA expression changes during Foxci-promoted osteogenic differentiation. In this study, we successfully overexpressed the Foxci gene in MC3T3-E1 cells and investigated the alterations in the miRNA expression profile on day 3 after osteogenic induction by using a miRNA microarray. Nine downregulated miRNAs and eight upregulated miRNAs were found to be differentially expressed. Among these miRNAs, miR-103-3p was consistently downregulated in the Foxc7-overexpressing MC3T3-E1 cells and was identified as a negative regulator of osteogenic differentiation by using a gain-and Iose-of-funotion assay. The special AT-rich sequence-binding protein 2 (Satb2), a pivotal osteogenic transcription factor, was identified as the miR-103-3p targeting gene and was verified by real-time polymerase chain reaction, western blot analysis, and luciferase assay. Overexpression of miR-103-3p markedly inhibited the expression of Satb2 and attenuated Foxci-promoted osteogenic differentiation.
