2CRISPR/Cas9-mediated PINK1 deletion leads to neurodegeneration in rhesus monkeys显示文摘Dear Editor,PINK1 mutations cause autosomal recessive and early-onset Parkinson’s disease(PD)with selective neurodegeneration.Unfortunately,current PINK1 knockout(KO)mouse1,2 and pig models3,4 are unable to recapitulate the selective and overt neurodegeneration seen in PD patients.Furthermore,endogenous Pink1 in the mouse brain is expressed at very low levels and can only be detected via immunoprecipitation,5 meaning that PINK1’s function in the mammalian brain needs to be assessed using larger animals that are closer to humans.We previously used CRISPR/Cas9 to target the monkey gene in one-cell stage embryos.6 Using the same approach,we designed two gRNAs to target exon 2(T1)and exon 4(T2),which encode a kinase domain in the PINK1 gene of rhesus monkeys(Fig.1a).CRISPR/Cas9 and PINK1 gRNAs were injected into one-cell stage rhesus monkey embryos.A T7E1 assay and sequencing of PCR products from the injected embryos showed high efficiency(61.5%)in targeting PINK1(Fig.1b and Supplementary information,Fig.S1).Transfer of 87 embryos to 28 surrogate rhesus monkeys resulted in 11 pregnancies(39.2%)(Fig.1b).Eleven fetuses developed to term and were born naturally.Of these live monkeys,eight carried PINK1 mutations(M),and three were wild type(WT).However,three mutant monkeys(M1,M3 and M4)were newborn triplets that struggled to survive and died 3-4 days after birth.One WT newborn monkey also died after a difficult labor.Another mutant monkey(M2)died 7 days after birth without noticeable warning signs or symptoms.The other three mutant monkeys(M6,M7 and M8)have lived for three years;M5,however,reduced its food intake and showed weakness at the age of 1.5 years,and died 30 days after anesthesia for MRI examination.Weili Yang Yunbo Liu Zhuchi Tu Chong Xiao Sen Yan Xishan Ma Xiangyu Guo Xiusheng Chen Peng Yin Zhengyi Yang Su Yang Tianzi Jiang Shihua Li Chuan Qin Xiao-Jiang Li 2019Cell Research2019,29,4:12
3Gut microbiome alterations and its link to corticosteroid resistance in immune thrombocytopenia显示文摘Quantitative metagenomic studies have linked the gut microbiota to autoimmune disorders.Here,we performed deep shotgun metagenomic sequencing of fecal samples from 99 immune thrombocytopenia(ITP)patients and 52 healthy controls.Dysbiosis in the gut microbiome of ITP was detected phylogenetically and functionally,and classifier based on species markers distinguished individuals with ITP from healthy controls.In particular,the abundance of Ruminococcus gnavus,Bifidobacterium longum and Akkermansia muciniphila was markedly increased in treatment-na?ve ITP patients,and the alterations of microbial species were correlated with clinical indices.Functionally,the secondary bile acid biosynthesis and flagellar assembly were depleted in the gut microbiota of ITP,which may contribute to the onset of ITP by affecting the immune system.Furthermore,we found that corticosteroid treatment affected the gut microbiome of ITP.Compared with corticosteroid-sensitive ITP patients,we identified that the corticosteroid-resistant ITP patients displayed a distinct gut microbiome,which was different from that of the treatment-na?ve ITP patients.Together,we provided support for the critical role of gut microbiota in the development of ITP and established a foundation for further research characterizing gut microbiota in relation to corticosteroid resistance of ITP.Yanan Wang Fengqi Liu Gaochao Zhang Yan Su Xueyan Sun Qi Chen Chencong Wang Haixia Fu Yun He Xiaolu Zhu Xiao Liu Meng Lv Xiangyu Zhao Xiaosu Zhao Yueying Li Qianfei Wang Xiaojun Huang Xiaohui Zhang 2021Science China(Life Sciences)2021,64,5:6

6Association between gene polymorphisms of vitamin D receptor and pulmonary tuberculosis susceptibility:a meta-analysis显示文摘Background:The vitamin D receptor(VDR) gene is a primary candidate gene for tuberculosis susceptibility,but results of previous studies are somewhat contradictory and underpowered.Thus,it is essential to further explore the association between VDR gene polymorphisms and risk of pulmonary tuberculosis(PTB).Methods:A systematic review and meta-analysis about the association between FokI,TaqI,ApaI and BsmI polymorphisms and PTB susceptibility was conducted.Statistical Package for Social Science(Version 13.0) and Review Manager(Version 4.2,The Cochrane Collaboration) were used to analyze the data reported in studies.Results:A total of 13 studies with 2 262 cases and 2 833 controls were involved in the FokI polymorphism,and the results showed FokI polymorphism was associated with PTB susceptibility(allele f vs F:OR=1.12,95% CI=[1.02,1.23];the additive effect model ff vs FF:OR=1.40,95%CI=[1.10,1.77];the recessive genetic model ff vs Ff+FF:OR=1.39,95%CI=[1.12,1.71]).No significant associations were observed between TaqI(15 studies with 3 031 cases and 3 132 controls),ApaI(7 studies with 1 495 cases and 1 922 controls),BsmI(6 studies with 919 cases and 1 250 controls) variants and PTB susceptibility.Conclusion:We found variant FokI polymorphism of VDR gene may play a risky role in PTB development,and the genetic model was presumed to be recessive.Su Qian Ma Xiangyu Lin Hui Li Ying Hu Daiyu Xiong Hongyan Xu Rufu Li Yafei 2011Journal of Medical Colleges of PLA(China)2011,26,2:2
7Bioinformatic analysis of chromatin organization and biased expression of duplicated genes between two poplars with a common whole-genome duplication显示文摘The nonrandom three-dimensional organization of chromatin plays an important role in the regulation of gene expression.However,it remains unclear whether this organization is conserved and whether it is involved in regulating gene expression during speciation after whole-genome duplication(WGD)in plants.In this study,high-resolution interaction maps were generated using high-throughput chromatin conformation capture(Hi-C)techniques for two poplar species,Populus euphratica and Populus alba var.pyramidalis,which diverged~14 Mya after a common WGD.We examined the similarities and differences in the hierarchical chromatin organization between the two species,including A/B compartment regions and topologically associating domains(TADs),as well as in their DNA methylation and gene expression patterns.We found that chromatin status was strongly associated with epigenetic modifications and gene transcriptional activity,yet the conservation of hierarchical chromatin organization across the two species was low.The divergence of gene expression between WGD-derived paralogs was associated with the strength of chromatin interactions,and colocalized paralogs exhibited strong similarities in epigenetic modifications and expression levels.Thus,the spatial localization of duplicated genes is highly correlated with biased expression during the diploidization process.This study provides new insights into the evolution of chromatin organization and transcriptional regulation during the speciation process of poplars after WGD.Le Zhang Jingtian Zhao Hao Bi Xiangyu Yang Zhiyang Zhang Yutao Su Zhenghao Li Lei Zhang Brian J.Sanderson Jianquan Liu Tao Ma 2021Horticulture Research2021,8,1:2

11Enhancing the adsorption function of biochar by mechanochemical graphitization for organic pollutant removal显示文摘Biochar(BC)has been extensively studied as adsorbent for the treatment of water pollution.Despite the distinct advantages,the high calcination temperature and low adsorption capacity of pristine BC limit its practical applications.Most of the former studies focused on the structure and/or surface modification to improve the adsorption capacity of BC.However,the harsh experiment conditions involved in the biochar modification limited the application in industrial level.Herein,we introduced mechanical treatment into BC preparation to reduce the calcination temperature and improve the adsorption capacity simultaneously.The results indicated that the calcination temperature was reduced and the adsorption capacity of the treated BC was improved after mechanochemical treatment.Characterization of the samples disclosed that BCs were graphitized with the particle size reduced to nanoscale after treatment.Adsorption tests indicated that the mechanochemically treated BCs showed much better removal performance of organic contaminants than that of pristine BCs.For instance,among four pristine BCs(BC600,BC700,BC800,and BC900),only BC900 has strong adsorption capacity for MB,while BC600 has low adsorption capacity(1.2 mg/g).By comparison,the adsorption capacity of MB increased greatly to 173.96 mg/g by BC600-500/1(treated at 500 r/min for 1 hour).To optimize the mechanochemical treatment,the effects of rotation speed and agitation duration were also investigated.Peidong Su Xiangyu Gao Junke Zhang Ridha Djellabi Bo Yang Qi Wu Zhen Wen 2021Frontiers of Environmental Science & Engineering2021,15,6:1

18Smart palm-size optofluidic hematology analyzer for automated imaging-based leukocyte concentration detection显示文摘A critical function of flow cytometry is to count the concentration of blood cells,which helps in the diagnosis of certain diseases.However,the bulky nature of commercial flow cytometers makes such tests only available in hospitals or laboratories,hindering the spread of point-of-care testing(POCT),especially in underdeveloped areas.Here,we propose a smart Palm-size Optofluidic Hematology Analyzer based on a miniature fluorescence microscope and a microfluidic platform to lighten the device to improve its portability.This gadget has a dimension of 35×30×80 mm and a mass of 39 g,less than 5%of the weight of commercially available flow cytometers.Additionally,automatic leukocyte concentration detection has been realized through the integration of image processing and leukocyte counting algorithms.We compared the leukocyte concentration measurement between our approach and a hemocytometer using the Passing-Bablok analysis and achieved a correlation coefficient of 0.979.Through Bland-Altman analysis,we obtained the relationship between their differences and mean measurement values and established 95%limits of agreement,ranging from−0.93×10^(3)to 0.94×10^(3)cells/μL.We anticipate that this device can be used widely for monitoring and treating diseases such as HIV and tumors beyond hospitals.Deer Su Xiangyu Li Weida Gao Qiuhua Wei Haoyu Li Changliang Guo Weisong Zhao 2023Opto-Electronic Science2023,2,12:1

20Alkylation of benzene with carbon dioxide to low-carbon aromatic hydrocarbons over bifunctional Zn-Ti/HZSM-5 catalyst显示文摘Alkylation of benzene to value-added,high octane number and low toxic toluene and xylenes provides a way to lower benzene content in gasoline pool,and is hence a method to promote fuel quality.On the other hand,CO_(2) accumulation in the atmosphere causes global warming and requires effective route for its valorization.Utilization of CO_(2) as a carbon source for benzene alkylation could achieve both goals.Herein,alkylation of benzene with CO_(2) and H2 was realized by a series of low-cost bifunctional catalysts containing zinc/titanium oxides(Zn/Ti oxides)and HZSM-5 molecular sieves in a fixed-bed reactor.By regulating and controlling oxygen vacancies of Zn/Ti oxides and the acidities of HZSM-5,benzene conversion and CO_(2) conversion reached 28.7%and 29.9%respectively,along with a total selectivity of toluene and xylene higher than 90%.In this process,more than 25%CO_(2) was effectively utilized and incorporated into the target products.Moreover,the mechanism of the reaction was analyzed and the course was simultaneously traced.CO_(2) was transformed into methanol firstly,and then methanol reacted with benzene generating toluene and xylene.The innovation provides a new method for upgrading of fuels and upcycling the emissions of CO_(2),which is of great environmental and economic benefits.Xiangyu Liu Yanling Pan Peng Zhang Yilin Wang Guohao Xu Zhaojie Su Xuedong Zhu Fan Yang 2022Frontiers of Chemical Science and Engineering2022,16,3:0
