2CMHX008,a PPAR γ partial agonist,enhances insulin sensitivity with minor influences on bone loss显示文摘Traditional thiazolidinediones(TZDs),such as rosiglitazone,are peroxisome proliferator-activated receptor g(PPARg)potent agonists that can be used to treat type 2 diabetes but carry unwanted effects,including increased risk for fracture.The present work aimed to compare the insulin-sensitizing efficacies and bone-loss side effects of CMHX008,a novel TZDs-like PPARg partial agonist,with those of rosiglitazone.A TR-FRET PPARg competitive binding assay was used to compare the binding affinity between CMHX008 and rosiglitazone.Mice were administered vehicle,CMHX008 or rosiglitazone for 16 weeks.Mesenchymal stem cells(MSCs)were used to examine differences in differentiation into osteoblasts after compounds treatment.TR-FRET showed lower affinity to PPARg by CMHX008 compared with rosiglitazone.Mice treated with CMHX008 showed insulin sensitization similar to that of mice treated with rosiglitazone,which was related to the significant inhibition of PPARg Ser273 phosphorylation and improved insulin sensitivity by facilitating the phosphorylation of insulin receptor and Akt in adipose tissues.Micro-CT and histomorphometric analyses demonstrated that the degree of trabecular bone loss after treatment with CMHX008 was weaker than that observed with rosiglitazone,as evidenced by consistent changes in BV/TV,Tb.N,Tb.Th,Tb.Sp,and the mineral apposition rate.MSCs treated with CMHX008 showed higher ALP activity and mRNA levels of bone formation markers than did cells treated with rosiglitazone in the osteoblast differentiation test.Thus,CMHX008 showed insulin-sensitizing effects similar to those of rosiglitazone with a lower risk of bone loss,suggesting that PPARg sparing eliminates the skeletal side effects of TZDs while maintaining their insulin-sensitizing properties.Yi Hou Xuemei Cao Xiangnan Hu Xinyu Li Xiaoqin Shi Hongying Wang Chuan Peng Jiayu Li Jibin Li Qifu Li Chaodong Wu Xiaoqiu Xiao 2018Genes & Diseases2018,5,3:6

4Sedimentary filling and sequence structure dominated by T-R cycles of the Nenjiang Formation in the Songliao Basin显示文摘The Nenjiang Formation in the Songliao Basin is a symmetrical sequence of progradation-retrogradation,and is formed in the transgression cycle and regression cycle(T-R cycles)of the base level of deposition.We analyzed the drilling,well logging,core data,and seismic profiles of basin level,and by using the sequence stratigraphy,identified one secondary sequence boundary,two third-order sequence boundaries,and eight fourth-order sequence boundaries in the Nenjiang Formation.These eleven sequence boundaries can be divided into structural unconformity,depositional unconformity,flooding surface,and forced regression surface.Therefore the Nenjiang Formation can be subdivided into one secondary sequence,three third-order sequences,and ten fourth-order sequences.We have restored the sedimentary filling evolution within the stratigraphic framework of fourth-order sequences in the Nenjiang Formation.The sedimentary period of the first member of the Nenjiang Formation was corresponded to the global transgression period,which is also the development period of transgression cycle(T cycle),when the lake basin had the largest scope and deepest sedimentary water,the SB07(the maximum flooding surface)was formed on the top of strata during this period;covering above the SB07,there developed a set of condensation layer-oil shale,which is distributed in the whole basin and is the important source bed and regional cover.Therefore,a retrogradation sequence was formed in the T cycle of the first member of the Nenjiang Formation,characterized by the retrograding delta at a low angle.The delta has a giant front,a small plain,and many underwater distributary channels.Meanwhile,large gravity flow channels and sublacustrine fans are developed in the front of the delta.During the depositional period of the second member of the Nenjiang Formation,the R cycle began to develop due to the compression of the pacific tectonic domain;the source direction rotated 90°along with the eastern uplifting of the basin,and formed a series of east-west prograded and forced retrogradations.The prograding delta at high angel was developed in the interior of the sequence;the delta had a small front,a giant plain,and fewer underwater distributary channels,with the collapse at the foreslope,forming a series of slump fans.The slump fans can be divided into three types:discrete type,superimposed type,and fluid type.We built a whole'triad model'of the slump fan.Pointed out that the sequence of forced retrogradation formed by R cycle is a good structural mark of basin optimization,and rejected the viewpoint of'transgression'in the Nenjiang Formation of the Songliao Basin.ZHANG ChenChen ZHANG Shun WEI Wei WU ChaoDong LIANG JiangPing NIU Wen DU JinXia FU XiuLi CUI KunNing WANG Chao WANG Hui 2014Science China Earth Sciences2014,57,2:5
5Hippocampal overexpression of TREM2 ameliorates high fat diet induced cognitive impairment and modulates phenotypic polarization of the microglia显示文摘Type 2 diabetes mellitus(T2DM)and Alzheimer's disease(AD)share several common pathophysiological features.Rare variants of triggering receptor expressed on myeloid cells 2(TREM2)increase the risk of developing AD,suggesting the involvement of TREM2 and innate immunity in AD development.It is still unknown whether TREM2 is related to cognitive impairment in T2DM.Here,we investigated the effects of the hippocampal overexpression of TREM2 on cognitive in long-term high-fat diet(HFD)-fed mice.Male C57BL/6J mice were maintained on HFD for 50 weeks.TREM2 was overexpressed in the hippocampus 36 weeks after HFD feeding using adeno-associated virus vector(AAV)-mediated gene delivery.The results showed that the HFD feeding induced rapid and persistent weight gain,glucose intolerance and significant impairments in learning and memory.Compared with AAV-con,AAV-TREM2 significantly ameliorated cognitive impairment without altering body weight and glucose homeostasis in HFD mice.The overexpression of TREM2 upregulated the synaptic proteins spinophilin,PSD95 and synaptophysin,suggesting the improvement in synaptic transmission.Dendritic complexity and spine density in the CA1 region were rescued after TREM2 overexpression.Furthermore,TREM2 markedly increased the number of iba-1/Arg-1-positive microglia in the hippocampus,suppressed neuroinflammation and microglial activation.In sum,hippocampal TREM2 plays an important role in improving HFD-induced cognitive dysfunction and promoting microglial polarization towards the M2 anti-inflammatory phenotype.Our study also suggests that TREM2 might be a novel target for the intervention of obesity/diabetes-associated cognitive decline.Min Wu Maolin Liao Rongfeng Huang Chunxiu Chen Tian Tian Hongying Wang Jiayu Li Jibin Li Yuxiang Sun Chaodong Wu Qifu Li Xiaoqiu Xiao 2022Genes & Diseases2022,9,2:4
6Glycolysis in the control of blood glucose homeostasis显示文摘Glycolysis,a simple pathway of glucose metabolism,critically regulates insulin secretion and metabolic functions of various cells.Depending on cell types,rates of glycolysis are determined at various steps of glycolysis that are subjected to the control of key metabolic and regulatory enzyme(S),which include glucokinase,6-phosphofructo-I-kinase,and 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase.These enzymes are regulated by both nutritional and hormonal signals at the levels of transcription,translation,and post-translational modifications.In hepatocytes,glycolysis is involved in the control of hepatic glucose production.The latter,when excessive,contributes to hyperglycemia in diabetes.In pancreatic β cells,glycolysis couples glucose-stimulated insulin secretion.Absolute or relatively low levels of circulating insulin causes hyperglycemia.In adipocytes,glycolysis generates metabolites for lipogenesis and channels fatty acids from excessive oxidation to triglyceride synthesis,thereby reducing oxidative stress.With increased proinflammatory status,adipocytes produce pro-hyperglycemic factors and bring about hyperglycemia and insulin resistance.In hypotha lamic neurons,glycolysis conveys nutrient sensing that is related to feeding control.Dysregulation of glycolysis occurs in conditions of insulin deficiency or resistance,and is attributable to inappropriate amount and/or activities of metabolic and regulatory enzymes of glycolysis.Targeting key metabolic and regulatory enzymes to enhance glycolysis may offer viable approaches for treatment of diabetes.Xin Guo Honggui Li Hang Xu Shihlung Woo Hui Dong Fuer Lu Alex J.Lange Chaodong Wu 2012Acta Pharmaceutica Sinica B2012,2,4:3
7Pyrite morphology and episodic euxinia of the Ediacaran Doushantuo Formation in South China显示文摘The Ediacaran Doushantuo Formation with well-preserved fossil record in South China provides a rare window for our understanding of biological evolution,global carbon cycle,and oceanic redox states.Prominent negativeδ^(13)C anomalies(i.e.,the Shuram excursion)in Ediacaran successions worldwide fundamentally challenge the traditional models of isotopic mass balance.Additionally,conflicting opinions of both oxic and anoxic conditions have been proposed for the deep waters during this period.Here,we present a detailed study of pyrite morphology and carbonate carbon isotope data documented from drill core samples at Songtao County,northeastern Guizhou.Framboid aggregates are the dominant pyrite form in black shale and they can transfer to euhedral crystals through continuous growth of the constituent microcrystals.A positive correlation between microcrystal sizes(d)and framboid diameters(D)is observed,while the different D/d ratios of framboids in argillaceous dolostone and black shale reflect different substrate availability.Electron microprobe analyses reveal no consistent compositional patterns between framboidal and euhedral pyrites.Framboid size distributions of the investigated drill core,in combination with previously published redox data from the intra-shelf Jiulongwan section,shelf margin Zhongling section,and lower slope Wuhe section,suggest that three episodes of marine euxinia have been established throughout the deposition of the Doushantuo Formation.The time lag between the uppermost euxinic interval and the Shuram excursion may arise from the depression of sulfate reduction maintained by other oxidants.YE YunTao WU ChaoDong ZHAI LiNa AN ZhengZe 2017Science China Earth Sciences2017,60,1:2

12A joint call for actions to advance taxonomy in China显示文摘Taxonomy plays an important role in understanding the origin, evolution, and ecological functionality of biodiversity. There are large number of unknown species yet to be described by taxonomists, which together with their ecosystem services cannot be effectively protected prior to description. Despite this, taxonomy has been increasingly underrated insufficient funds and permanent positions to retain young talents. Further, the impact factordriven evaluation systems in China exacerbate this downward trend, so alternative evaluation metrics are urgently necessary. When the current generation of outstanding taxonomists retires,there will be too few remaining taxonomists left to train the next generation. 13Adoptive transfer of Pfkfb3-disrupted hematopoietic cells to wild-type mice exacerbates diet-induced hepatic steatosis and inflammation显示文摘Background and objectives:Hepatic steatosis and inflammation are key characteristics of non-alcoholic fatty liver disease(NAFLD).However,whether and how hepatic steatosis and liver inflammation are differentially regulated remains to be elucidated.Considering that disruption of 6-phosphofructo-2-kinase/fructose-2,6-biphosphatase 3(Pfkfb3/iPfk2)dissociates fat deposition and inflammation,the present study examined a role for Pfkfb3/iPfk2 in hematopoietic cells in regulating hepatic steatosis and inflammation in mice.Methods:Pfkfb3-disrupted(Pfkfb3^(+/-))mice and wild-type(WT)littermates were fed a high-fat diet(HFD)and examined for NAFLD phenotype.Also,bone marrow cells isolated from Pfkfb3^(+/-)mice andWT mice were differentiated into macrophages for analysis of macrophage activation status and for bone marrow transplantation(BMT)to generate chimeric(WT/BMT-Pfkfb3^(+/-))mice in which Pfkfb3 was disrupted only in hematopoietic cells and control chimeric(WT/BMT-WT)mice.The latter were also fed an HFD and examined for NAFLD phenotype.In vitro,hepatocytes were co-cultured with bone marrowderived macrophages and examined for hepatocyte fat deposition and proinflammatory responses.Results:After the feeding period,HFD-fed Pfkfb3^(+/-)mice displayed increased severity of liver inflammation in the absence of hepatic steatosis compared with HFD-fed WT mice.When inflammatory activation was analyzed,Pfkfb3^(+/-)macrophages revealed increased proinflammatory activation and decreased anti-proinflammatory activation.When NAFLD phenotype was analyzed in the chimeric mice,WT/BMT-Pfkfb3^(+/-) mice displayed increases in the severity of HFD-induced hepatic steatosis and inflammation compared with WT/BMT-WT mice.At the cellular level,hepatocytes co-cultured with Pfkfb3^(+/-) macrophages revealed increased fat deposition and proinflammatory responses compared with hepatocytes co-cultured with WT macrophages.Conclusions:Pfkfb3 disruption only in hematopoietic cells exacerbates HFD-induced hepatic steatosis and inflammation whereas the Pfkfb3/iPfk2 in nonhematopoietic cells appeared to be needed for HFD feeding to induce hepatic steatosis.As such,the Pfkfb3/iPfk2 plays a unique role in regulating NAFLD pathophysiology.Xin Guo Bilian Zhu Hang Xu Honggui Li Boxiong Jiang Yina Wang Benrong Zheng Shannon Glaser Gianfranco Alpini Chaodong Wu 2020Liver Research2020,4,3:1

16A supplemental description of the genus Lista Walker from China(Lepidoptera: Pyralidae), with two new and a newly record species显示文摘Three species of the genus Lista Walker are recognized from China, as a supplement of the former work. Among them, two species are described as new to science, namely L. furcillatusa Wang, Chen & Wu sp. nov. and L. menglaensis Wang, Chen & Wu sp. nov. One species, L. monticola Yamanaka, 2000, is reported from China for the first time. Adults and genitalia of the three species are illustrated. 17Rock-magnetic properties of Neogene sediments, northern flank of Tianshan Mountains显示文摘Analyses of rock-magnetic properties of Neogene sediments of the Taxihe section, northern Tianshan Mountains, show that the section can be classified into three categories including lacustrine facies, fluvial facies and alluvial facies, which correspond to the lower, middle and upper of the Taxihe section respectively. The magnetic minerals of the lacustrine facies may be affected by the process of weath- ering, lithogenesis and biolithogenesis besides the source of the sediments. The natural remanence intensities are between 10-3 A/m and 10-2 A/m. The minerals are dominated by magnetite and the high coercive magnetic mineral may be goethite. The magnetic grains are the mixture of PSD+SD or SD+SP. The natural remanence intensities of the strata of fluvial facies are between 10-2 A/m and 10-1 A/m, about ten times that of the lacustrine facies. The magnetic minerals are mainly magnetite and hematite, and the magnetic grains are mainly PSD. The characteristic remanence (ChRM) carriers are magnetites. In the alluvial facies, the natural remanence intensities are mostly less than 1×10-2 A/m. The magnetic minerals of the series are dominated by magnetite and hematite, almost the same as the fluvial facies. But the difference is that most of the stepwise demagnetization can reveal two components and the ChRM carriers are hematites. 19New insights into multiple provenances evolution of the Jurassic from heavy minerals characteristics in southern Junggar Basin, NW China显示文摘The method of random forest was used to classify the heavy mineral assemblages of 2 418 Jurassic samples in the southern Junggar Basin, and determine the distribution of the heavy mineral assemblages from the same provenance systems. Based on the analysis of heavy minerals assemblages, ZTR index, sedimentary characteristics, U-Pb zircon ages, whole-rock geochemical and paleocurrent direction analysis, the study reveals that five important provenances were providing sediments to the southern Junggar Basin in the Jurassic period: The North Tianshan(NTS), Central Tianshan(CTS), Bogda Mountains, Zhayier Mountains and Kalamaili Mountains. During the Early Jurassic, NTS-CTS, Kalamaili Mountains and Zhayier Mountains are primary provenances, Bogda Mountains started to uplift and supply clastic materials in the Middle Jurassic. There are three sedimentary area in the Jurassic of southern Junggar Basin: the western part, the central part and the eastern part. In the western part, the clastic materials of the Early Jurassic was mainly from NTS blocks and Zhayier Mountains, and the sediments were dominantly derived from the Zhayier Mountains during the Middle–Late Jurassic. In the central part, the main provenance of the Early Jurassic switched from NTS to CTS. In the Xishanyao Formation, the main source went back to NTS again. The NTS was the primary provenance during the sedimentary periods of Toutunhe Formation and Qigu Formation. In the eastern part, the contribution of CTS and Kalamaili Mountains were considered as major provenances in the Early Jurassic-Xishanyao Formation, small proportion of sediments were from NTS. The Bogda mountains uplifted and started to provide sediments to the Junggar Basin in the sedimentary period of Xishanyao Formation, and became the major source during the Toutunhe Formation period, with small amount of sediments from CTS. 20Electroaeroelastic Modeling and Analysis for Flow Energy Piezoelectric Harvester显示文摘An electroaeroelastic model for wind energy harvesting using piezoelectric generators is presented.The flow field is mapped in detail.The force which the fluid flow exerts on the generator is formulated.The output voltage levels generated from the mechanical strain within the piezoelectric elements are determined.An analytical model is developed with consideration of the interactions between the fluid,solid and electric.Various analytical results are obtained,such as flow velocity contour and pressure contour for the flow,moving trajectories,stress contour and output voltage of the harvester.A prototype is fabricated and tested.The simulation result is close to the experimental result.The model developed in this paper can predict the performance and behavior of different energy harvesters.And it also can be used as a design tool for optimizing the performance of the harvester.Zhang Jiantao Wu Song Shu Chang Li Chaodong 2017Transactions of Nanjing University of Aeronautics and Astronautics2017,34,1:0
