| 1 | Identification of a Stable Quantitative Trait Locus for Percentage Grains with White Chalkiness in Rice (Oryza sativa)显示文摘High chalkiness is a major problem in many rice-producing areas of the world, especially in hybrid rice (Oryza sativa L.) in China. We previously showed a major quantitative trait locus for the percentage of grains with white chalkiness (QTLqPGWC-8) in the interval G1149-R727 on chromosome 8 using a chromosome segment substitution line (CSSL). Here, we selected the line-CSSL50 harboring the QTLqPGWC-8 allele from the CSSLs derived from a cross between Asominori (as a recurrent parent) and IR24 (as a donor parent), which had higher percentage chalkiness, markedly different from that of Asominori. There were also significant differences in starch granules, appearance of amylose content (AAC) and milling qualities between Asominori and CSSL50, but not in grain size or thousand grain weight (TGW). The BC4F2 and BC4F3 populations from a cross between CSSL50 and Asominori were used for fine mapping of qPGWC-8. We narrowed down the location of this QTL to a 142 kb region between Indel markers 8G-7 and 8G-9. QTLqPGWC-8 accounted for 50.9% of the difference in PGWC between the parents. | 14 | A novel inhibitor of ARfl and ARv7 induces protein degradation to overcome enzalutamide resistance in advanced prostate cancer显示文摘Enzalutamide(ENZ) is a second-generation androgen receptor(AR) antagonist used for the treatment of castration-resistant prostate cancer(CRPC) and reportedly prolongs survival time within a year of starting therapy. However, CRPC patients can develop ENZ resistance(ENZR), mainly driven by abnormal reactivation of AR signaling, involving increased expression of the full-length AR(ARfl)or dominantly active androgen receptor splice variant 7(ARv7) and ARfl/ARv7 heterodimers. There is currently no efficient treatment for ENZR in CRPC. Herein, a small molecule LLU-206 was rationally designed based on the ENZ structure and exhibited potent inhibition of both ARfl and constitutively active ARv7 to inhibit PCa proliferation and suppress ENZR in CRPC. Mechanically, LLU-206 promoted ARfl/ARv7 protein degradation and decreased ARfl/ARv7 heterodimers through mouse double minute 2-mediated ubiquitination. Finally, LLU-206 exhibited favorable pharmacokinetic properties with poor permeability across the blood-brain barrier, leading to a lower prevalence of adverse effects, including seizure and neurotoxicity, than ENZ-based therapies. | 17 | Numerical studies on the radiation uniformity of Z-pinch dynamic hohlraum显示文摘Radiation uniformity is important for Z-pinch dynamic hohlraum driven fusion. In order to understand the radiation uniformity of Z-pinchdynamic hohlraum, the code MULTI-2D with a new developed magnetic field package is employed to investigate the related physical processeson Julong-I facility with drive current about 7e8 MA. Numerical simulations suggest that Z-pinch dynamic hohlraum with radiation temperaturemore than 100 eV can be created on Julong-I facility. Although some X-rays can escape out of the hohlraum from Z-pinch plasma and electrodes, the radiation field near the foam center is quite uniform after a transition time. For the load parameters used in this paper, the transitiontime for the thermal wave transports from r = 1 mm to r = 0 mm is about 2.0 ns. Implosion of a testing pellet driven by cylindrical dynamichohlraum shows that symmetrical implosion is hard to achieve due to the relatively slow propagation speed of thermal wave and the compressionof cylindrical shock in the foam. | 18 | Facile cyclic ammonium salt with the smallest size for high performance electric double layer capacitors显示文摘The smallest cyclic ammonium salt reported to date, N,N-dimethylpyrrolidinium tetrafluoroborate (P11-BF4), was successively synthesized using a synthesis route without metal ions and halogen ions, then investigated as the electrolyte with Propylene carbonate in EDLCs. The electrochemical characteristics of EDLCs assembled by 1 mol/L P11-BF4/PC paired with activated carbon electrodes were compared to traditional electrolytes. P11-BF4 has proven to have superior voltage resistance by using cyclic voltammetry and constant current charge-discharge testing. Moreover, P11-BF4 exhibits a more brilliant rate performance due to its high conductivity. | 19 | PA1426 regulates Pseudomonas aeruginosa quorum sensing and virulence:an in vitro study显示文摘Objective:Pseudomonas aeruginosa(P.aeruginosa)contains a hierarchy of quorum sensing(QS)network,consisting of the las,rhl and pqs systems,which play a key role in coordinating the expression of virulence factors.PA2146 was found to be associated with P.aeruginosa pathogenicity in macrophage and host Immune response.The aim of this study was to investigate the effects of PA2146 on the virulence of P.aeruginosa and explore its mechanism.Methods:PA2146 gene knockout strain and complement strain of P.aeruginosa PAO1 were constructed.The biomass of biofilm was detected by crystal violet staining;the virulence factors were measured,including pyocyanin,rhamnolipid,LasA elastase,LasB elastase and hemolytic activity;RNA-seq and label-free relative quantitative proteomics analyses were carried out to test the influence of PA2146 on transcriptomics and proteomics.This study was approved by the Institutional Review Board of the Third Xiangya Hospital,Central South University,China(approval No.2019-S021).Results:PA2146-deficient strains showed reduced biofilm formation and increased pyocyanin,rhamnolipid,LasA elastase,LasB elastase,and hemolytic activity,as well as increased motility,compared with the wild-type strain.RNA-seq and label-free relative quantitative proteomics analyses revealed that PA2146 repressed the transcription of several genes that are integral to the pqs system and to pyocyanin biosynthesis,and increased the expression of MexEF-OprN efflux pump components at the gene and protein level.Conclusion:PA2146 gene was found to inhibit the pqs system.PA2146 may affect quorum sensing by directly inhibiting the pqs system or by enhancing the expression of MexEF-OprN efflux pump components,thereby promoting efflux of 2-heptyl-4(1H)-quinolone,a Pseudomonas quinolone signal precursor,and thus affecting P.aeruginosa virulence. | Fang Tan Pengfei She Linying Zhou Shijia Li Xianghai Zeng Lanlan Xu Yaqian Liu Zubair Hussain Yong Wu | 2021 | Journal of Bio-X Research2021,4,1: | 0 |
