1Approaches to establish Q-markers for the quality standards of traditional Chinese medicines显示文摘Traditional Chinese medicine(TCM) has played a pivotal role in maintaining the health of Chinese people and is now gaining increasing acceptance around the global scope. However, TCM is confronting more and more concerns with respect to its quality. The intrinsic 'multicomponent and multitarget' feature of TCM necessitates the establishment of a unique quality and bioactivity evaluation system, which is different from that of the Western medicine. However, TCM is investigated essentially as'herbal medicine' or 'natural product', and the pharmacopoeia quality monographs are actually chemicalmarkers-based, which can ensure the consistency only in the assigned chemical markers, but, to some extent, have deviated from the basic TCM theory. A concept of 'quality marker'(Q-marker), following the 'property-effect-component' theory, is proposed. The establishment of Q-marker integrates multidisciplinary technologies like natural products chemistry, analytical chemistry, bionics, chemometrics,pharmacology, systems biology, and pharmacodynamics, etc. Q-marker-based fingerprint and multicomponent determination conduce to the construction of more scientific quality control system of TCM.This review delineates the background, definition, and properties of Q-marker, and the associated technologies applied for its establishment. 4Novel C-17 spirost protostane-type triterpenoids from Alisma plantago-aquatica with antiinflammatory activity in Caco-2 cells显示文摘Twenty-one protostane-type triterpenoids with diverse structures, including nine new compounds(1 e9), were isolated from the of Alisma plantago-aquatica Linn. Structurally, alisolides A-F(1 e6), composed of an oxole group coupled to a five-membered ring, represent unusual C-17 spirost protostane-type triterpenoids. Alisolide H(8) is a novel triterpenoid with an unreported endoperoxide bridge. Alisolide I(9) represents the first example of 23,24-acetal triterpenoid. Their structures were elucidated based on spectroscopic analysis, wherein the absolute configurations of 4-6, 8 were further confirmed by the Mo2(OAc)4-induced ECD method. 5Polymer coated nanodiamonds as gemcitabine prodrug with enzymatic sensitivity for pancreatic cancer treatment显示文摘The fabrication of Gemcitabine(GEM) prodrug was reported to be an effective method to enhance its pancreatic cancer treatment efficiency. Here, a kind of nanocarbon-based materials, nanodiamond(ND), was selected as the nanocarrier of GEM, owing to its outstanding surface properties and non-cytotoxicity. The polyelectrolytes,polyethyleneimine and polyacrylic acid, were used to self-assemble outside ND surface through electrostatic forces, followed by attachment of polyethylene glycol to address better biocompatibility. GEM was conjugated with an enzyme-sensitive peptide gly-phe-leu-gly to build up the controlled release platform. From characterization results of dynamic laser scattering, zeta potential and transmission electron microscope, the significant improvement of ND stability in physiological condition was proved. Non-cytotoxicity of this functionalized ND carriers and cytotoxicity of the prodrug against Bx PC-3 pancreatic cancer cells were indicated by methylthiazolyl tetrazolium(MTT) assay. In vivo experiments also revealed its superior anticancer effect compared with free GEM treatment. 12GmGPDH12,a mitochondrial FAD-GPDH from soybean, increases salt and osmotic stress resistance by modulating redox state and respiration显示文摘In plants,glycerol-3-phosphate dehydrogenase(GPDH)catalyzes the interconversion of glycerol-3-phosphate(G3P)and dihydroxyacetone phosphate(DHAP)coupled to the reduction/oxidation of the nicotinamide adenine dinucleotide(NADH)pool,and plays a central role in glycerolipid metabolism and stress response.Previous studies have focused mainly on the NAD+-dependent GPDH isoforms,neglecting the role of flavin adenine dinucleotide(FAD)-dependent GPDHs.We isolated and characterized three mitochondrialtargeted FAD-GPDHs in soybean,of which one isoform(GmGPDH12)showed a significant transcriptional response to NaCl and mannitol treatments,suggesting the existence of a major FAD-GPDH isoform acting in soybean responses to salt and osmotic stress.An enzyme kinetic assay showed that the purified GmGPDH12 protein possessed the capacity to oxidize G3P to DHAP in the presence of FAD.Overexpression and RNA interference of GmGPDH12 in soybean hairy roots resulted in elevated tolerance and sensitivity to salt and osmotic stress,respectively.G3P contents were significantly lower in GmGPDH12-overexpressing hair roots and higher in knockdown hair roots,indicating that GmGPDH12 was essential for G3P catabolism.A significant perturbation in redox status of NADH,ascorbic acid(ASA)and glutathione(GSH)pools was observed in GmGPDH12-knockdown plants under stress conditions.The impaired redox balance was manifested by higher reactive oxygen species generation and consequent cell damage or death;however,overexpressing plants showed the opposite results for these traits.GmGPDH12 overexpression contributed to maintaining constant respiration rates under salt or osmotic stress by regulating mRNA levels of key mitochondrial respiratory enzymes.This study provides new evidence for the roles of mitochondria-localized GmGPDH12 in conferring resistance to salt or osmotic stress by maintaining cellular redox homeostasis,protecting cells and respiration from oxidative injury.Ying Zhao Xin Li Zexin Zhang Wenjing Pan Sinan Li Yun Xing Wanying Xin Zhanguo Zhang Zhenbang Hu Chunyan Liu Xiaoxia Wu Zhaoming Qi Dawei Xin Qingshan Chen 2021The Crop Journal2021,9,1:1
13Generating superposed terahertz perfect vortices via a spin-multiplexed all-dielectric metasurface显示文摘Perfect optical vortices(POVs),characterized as a ring radius independent of topological charge(TC),possess extensive application in particle manipulation and optical communication.At present,the complex and bulky optical device for generating POVs has been miniaturized by leveraging the metasurface,and either spindependent or spin-independent POV conversions have been further accomplished.Nevertheless,it is still challenging to generate superposed POVs for incidences with orthogonal circular polarization.Here,a spinmultiplexed all-dielectric metasurface method for generating superposed POVs in the terahertz frequency range is proposed and demonstrated.By using the multiple meta-atom comprised structure as the basic unit,the complex amplitude of two superposed POVs is modulated,decoupled,and subsequently encoded to left-and righthanded circular polarization incidences.Furthermore,two kinds of metasurfaces are fabricated and characterized to validate this controlling method.It is demonstrated that the measured intensity and phase distributions match well with the calculation of the Rayleigh–Sommerfeld diffraction integral,and the radius of superposed POVs is independent of TCs.This work provides promising opportunities for developing ultracompact terahertz functional devices applied to complex structured light generation and terahertz communication,and exploring sophisticated spin angular momentum and orbital angular momentum interactions like the photonic spin-Hall effect.FAN HUANG QUAN XU WANYING LIU TONG WU JIANQIANG GU JIAGUANG HAN WEILI ZHANG 2023Photonics Research2023,11,3:1

15Coat protein of Chinese wheat mosaic virus upregulates and interacts with cytosolic glyceraldehyde-3-phosphate dehydrogenase,a negative regulator of plant autophagy,to promote virus infection显示文摘Autophagy is an intracellular degradation mechanism involved in antiviral defense,but the strategies employed by plant viruses to counteract autophagy-related defense remain unknown for the majority of the viruses.Herein,we describe how the Chinese wheat mosaic virus(CWMV,genus Furovirus)interferes with autophagy and enhances its infection in Nicotiana benthamiana.Yeast two-hybrid screening and in vivo/in vitro assays revealed that the 19 k Da coat protein(CP19 K)of CWMV interacts with cytosolic glyceraldehyde-3-phosphate dehydrogenases(GAPCs),negative regulators of autophagy,which bind autophagy-related protein 3(ATG3),a key factor in autophagy.CP19 K also directly interacts with ATG3,possibly leading to the formation of a CP19 K–GAPC–ATG3 complex.CP19 K–GAPC interaction appeared to intensify CP19 K–ATG3 binding.Moreover,CP19 K expression upregulated GAPC gene transcripts and reduced autophagic activities.Accordingly,the silencing of GAPC genes in transgenic N.benthamiana reduced CWMV accumulation,whereas CP19 K overexpression enhanced it.Overall,our results suggest that CWMV CP19 K interferes with autophagy through the promotion and utilization of the GAPC role as a negative regulator of autophagy.Erbo Niu Chaozheng Ye Wanying Zhao Hideki Kondo Yunfeng Wu Jianping Chen Ida Bagus Andika Liying Sun 2022Journal of Integrative Plant Biology2022,64,8:0

17Effect of magnetic field on double layer in argon helicon plasma显示文摘The electric double layer in argon helicon plasma by using floating electrostatic probe and local optical emission spectroscopy under different external magnetic field was investigated.Results show that electrons generated in the plasma source can be magnetized by the magnetic field and transported by the gradient of diverging magnetic field in the diffusion chamber,giving rise to the hollowed distribution of electron temperature.The electron temperature on axis has a sudden decrease,leading to the sudden decrease of plasma density as well as the plasma potential,forming a double layer structure.The position of double layer moves along with the position of diverging magnetic field,and the potential drop increases along with the strength of magnetic field.The structure of the external magnetic field plays the decisive factor for the double layer in helicon plasma.Kaiyi Yang Ruilin Cui Wanying Zhu Zhiwen Wu Jiting Ouyang 2021High Voltage2021,6,2:0
18Enzymatic pretreatment mitigates the dissemination of antibiotic resistance genes via regulating microbial populations and gene expressions during food waste fermentation显示文摘Food waste(FW)has been recognized as essential reservoir for resource recovery via anaerobic fermentation,which could also bring the potential risk of antibiotic resistance genes(ARGs)dissemination.Although the structural deficiency of FW could be stimulated by enzymatic pretreatment to enhance fermentation efficiency,the influences of enzymatic pretreatment on ARGs fate and microbial metabolic pathways involved in ARGs dissemination have rarely been reported.This work proved that enzymatic pretreatment could effectively decrease the total abundance of ARGs(reduced by 13.8%-24.5%)during long-term FW fermentation.It was found that enzymatic pretreatment significantly reduced the ARGs belonging to the efflux pump,which might be ascribed to its ability to increase membrane permeability.Furthermore,enzymatic pretreatment was in favor of reducing microbial diversity and various potential ARGs host(e.g.,Methanosarcina,Clostridium,Prevotella,Parabacteroides).Also,this pretreatment remarkably up-regulated the genetic expressions involved in ABC transporter(e.g.,eryF and mntA)and down-regulated the genetic expressions that participated in DNA replication,two-component systems(e.g.,uphA and cckA),and quorum sensing(e.g.,rpfF and lsrG),thereby decreasing ARGs transmission.This study would expand the insight of the influences of pretreatment method on ARGs fate during FW fermentation,and offer practical guidance on the sustainable management of FW.Yang Wu Wanying Hu Haining Huang Xiong Zheng Lei Dong Yinguang Chen 2023Chinese Chemical Letters2023,34,8:0

20Comprehensive evaluation and selection of the potential complex medium for industrial glutamate decarboxylase (GAD) production by Escherichia coli显示文摘To address the need for producing large quantities of an enzyme,glutamate decarboxylase(GAD),this study evaluated the performance of the currently used complex and defined culture media in terms of biomass production,GAD yield,and byproducts accumulation,and selected the candidate(s)that could maximize the production of GAD by Escherichia coli(AS 1.505,the China General Microbiological Culture Collection Center,Beijing,China).The highest yield of the enzyme per biomass in the final broth(4431 U/g)corresponded to the Luria Bertani medium(LBM),which was 1.16 times the second highest from the Terrific Broth medium(TBM),while the highest biomass production(4.5 g/L)was achieved in the cultivation with TBM.For the byproducts,the profiles of all the media except the TK medium displayed a distinct exponential phase after six hour cultivation with a rapid byproducts generation,which could be attributed to the mixed acid fermentation caused by oxygen limitation.In addition,the subsequent re-assimilation of acetate could reduce the loss of GAD accumulated by stabilizing the media pH.The comprehensive analysis showed that although the highest biomass production was achieved by TBM,the fastest intracellular accumulation of GAD occurred in the cultivation with LBM(184.64 U/(g·h)),which also was the cheapest medium(0.56$/103 U).Therefore,LBM was considered the potentially competitive candidate medium for large-scale production of GAD by E.coli.Wanying Yao Xiao Wu Jun Zhu Bo Sun 2011International Journal of Agricultural and Biological Engineering2011,4,2:0
