1First identification of kdr allele F1534S in VGSC gene and its association with resistance to pyrethroid insecticides in Aedes albopictus populations from Haikou City,Hainan Island,China显示文摘Background:Aedes albopictus is distributed widely in China,as a primary vector of Dengue fever and Chikungunya fever in south of China.Chemical insecticide control is one of the integrated programmes to prevent mosquito-borne diseases.Long-term applications of pyrethroids have resulted in the development of resistance in Ae.albopictus populations in China.However,the susceptibility of Ae.albopictus to pyrethroids in Hainan Island was unclear.Knockdown resistance(kdr),caused by point mutations in the VGSC gene,is one of the mechanisms that confer resistance to DDT and pyrethroids.This study was to investigate the resistance level of Ae.albopictus populations in Haikou City to three pyrethroid insecticides,and elucidate the relationship between the resistant phenotype and kdr mutations.Methods:The Aedes albopictus samples were collected in Xinbu Island(XI),Longtang Town(LT),Shishan Town(ST),Baishamen Park(BP),and Flower Market(FM)from Haikou City,Hainan Island,China.The larval susceptibility to deltamethrin,permethrin and beta-cypermethrin was tested by larval bioassays,and adult susceptibility to deltamethrin and DDT was determined by adult bioassays.The degree of resistance was determined by resistance ratio value(RR50>3)for larvae and by mortality for adult.The kdr alleles at codon 1534 of the VGSC gene were genotyped.The relationship between kdr genotypes and resistant phenotypes was analyzed by Chi-square test.Results:Out of five populations,assessed by larval bioassays,XI was susceptible to deltamethrin and permethrin;LT was susceptible to permethrin and beta-cypermethrin;and ST was susceptible to permithrin.FM and BP both were resistant to all of the three pyrethroids,and FM showed the highest degree of resistance,with RR50 values from 65.17 to 436.36.A total of 493 individuals from the larval bioassays were genotyped for kdr alleles.Five alleles were detected,including two wildtype alleles,TTC(F)(67.04%)and TTT(F)(0.41%),and three mutant alleles,TGC(C)(0.30%),TCC(S)(31.54%)and TTG(L)(0.71%).There was a clear correlation between mutant alleles(or F1534S)and resistant phenotypes(P<0.01).Conclusion:Two novel kdr mutant alleles F1534S and F1534L were detected in the pyrethroid resistant populations of Ae.albopictus in Haikou Hainan,China.For the first time,the mutant F1534S was associated with pyrethroid resistance in Ae.albopictus.Huiying Chen Kaili Li Xiaohua Wang Xinyan Yang Yi Lin Fang Cai Wenbin Zhong Chunyan Lin Zhongling Lin Yajun Ma 2016Infectious Diseases of Poverty2016,5,1:19
2Urban Expansion and Its Influencing Factors in Natural Wetland Distribution Area in Fuzhou City,China显示文摘This paper principally focuses on land use dynamics,urban expansion and underlying driving forces in the Natural Wetland Distribution Area(NWDA)of Fuzhou City in the southeastern China.Based on time series Landsat TM/ETM+imageries and historical data,relationships between urban land expansion and its influencing factors from 1989 to 2009 were analyzed by using an integrated approach of remote sensing(RS)and geographic information system(GIS)techniques.The results showed that built-up land increased from 151.16 km2in 1989 to 383.76 km 2in 2009. Approximately 64.25%of the newly emerging built-up land was converted from cropland(29.47%),forest and shrub (25.78%),water(3.73%),wetland(4.61%),and bare land(0.66%)during 1989 and 2009.With a remarkable decrease in cropland,the proportion of non-agricultural population increased by 23.6%.Moreover,rapid development of infrastructures,facilities,industrial parks,and urban and rural settlements along the Minjiang River resulted in the eastward and southward expansion of built-up land.Additionally,the growth pattern of built-up land in the NWDA is highly correlated with socio-economic factors,including the gross domestic product(GDP),GDP per capita,and structure of industry.As a result,the observed environmental degradation such as loss of cropland and wetland due to heavy pressure of rapid urbanization have greatly impaired the carrying capacity of city.Thus,in addition to scientific and rational policies towards minimizing the adverse effects of urbanization,coordination between the administrative agencies should be urgently strengthened to balance the conflicts between urban development and ecological conservation to make sure the sustainable land use.CAI Yuanbin ZHANG Hao PAN Wenbin CHEN Yanhong WANG Xiangrong 2012Chinese Geographical Science2012,22,5:8
3Folate receptor-mediated boron-10 containing carbon nanoparticles as potential delivery vehicles for boron neutron capture therapy of nonfunctional pituitary adenomas显示文摘Invasive nonfunctional pituitary adenomas(NFPAs) are difficult to completely resect and often develop tumor recurrence after initial surgery.Currently,no medications are clinically effective in the control of NFPA.Although radiation therapy and radiosurgery are useful to prevent tumor regrowth,they are frequently withheld because of severe complications.Boron neutron capture therapy(BNCT) is a binary radiotherapy that selectively and maximally damages tumor cells without harming the surrounding normal tissue.Folate receptor(FR)-targeted boron-10 containing carbon nanoparticles is a novel boron delivery agent that can be selectively taken up by FR-expressing cells via FR-mediated endocytosis.In this study,FR-targeted boron-10 containing carbon nanoparticles were selectively taken up by NFPAs cells expressing FR but not other types of non-FR expressing pituitary adenomas.After incubation with boron-10 containing carbon nanoparticles and following irradiation with thermal neutrons,the cell viability of NFPAs was significantly decreased,while apoptotic cells were simultaneously increased.However,cells administered the same dose of FR-targeted boron-10 containing carbon nanoparticles without neutron irradiation or received the same neutron irradiation alone did not show significant decrease in cell viability or increase in apoptotic cells.The expression of Bcl-2 was down-regulated and the expression of Bax was up-regulated in NFPAs after treatment with FR-mediated BNCT.In conclusion,FR-targeted boron-10 containing carbon nanoparticles may be an ideal delivery system of boron to NFPAs cells for BNCT.Furthermore,our study also provides a novel insight into therapeutic strategies for invasive NFPA refractory to conventional therapy,while exploring these new applications of BNCT for tumors,especially benign tumors.DAI CongXin CAI Feng HWANG Kuo Chu ZHOU YongMao ZHANG ZiZhu LIU XiaoHai MA SiHai YANG YaKun YAO Yong FENG Ming BAO XinJie LI GuiLin WEI JunJi JIAO YongHui WEI ZhenQing MA WenBin WANG RenZhi 2013Science China(Life Sciences)2013,56,2:8
4SERK Receptor-like Kinases Control Division Patterns of Vascular Precursors and Ground Tissue Stem Cells during Embryo Development in Arabidopsis显示文摘During embryo development, the vascular precursors and ground tissue stem cells divide to renew them-selves and produce the vascular tissue, endodermal cells, and cortical cells. However, the molecular mech-anisms regulating division of these stem cells have remained largely elusive. In this study, we show that loss of function of SOMATIC EMBRYOGENESIS RECEPTOR-UKE KINASE (SERK) genes results in aberrant em-bryo development. Fewer cortical, endodermal, and vascular cells are generated in the embryos of serk1 serk2bak1 triple mutants. WUSCHEL-RELATED HOMBOBOX5 (WOXS) is ectopically expressed in vascular cells of serkl serk2 bak1 embryos. The first transverse division of vascular precursors in mid-globular em-bryos and second asymmetric division of ground tissue stem cells in early-heart embryos are abnormally altered to a longitudinal division. The embryo defects can be partially rescued by constitutively activated mitogen-activated protein kinase (MAPK) kinase kinase YODA (YDA) and MAPK kinase MKK5. 6The Influence of Key Environmental Variables on Phytoplankton Community Structure in the Estuary of Tidal Rivers Around Luoyuan Bay,China显示文摘A total of 348 species belonging to 8 phyla and 125 genera were observed in seasonally sampled phytoplankton of tidal rivers from 13 sampling sites around Luoyuan Bay, and all field samplings were carried out in productive period(March/June/August/December) at ebb tide. Bacillariophyta species were the most abundant species, followed by Chlorophyta, Cyanophytes, Euglenophyta, Cryptophyta, Dinophyta, Xanthophyta and Chrysophytas. Seasonal distribution index(SDI) value ranged from 0.63 to 0.86, which meant that species found at those sites in 4 seasons tended to be largely different. Phytoplankton individuals ranged from 5.939×10~4 ind L^(-1) in winter to 75.31×10~4 ind L^(-1) in autumn. Phytoplankton biomass ranged from 0.620 mg L^(-1) in summer to 2.373 mg L^(-1) in autumn. The grey correlation analysis(GCA) showed that the nutrient variables played an important role in the influence on phytoplankton community in every season. The canonical correspondence analysis(CCA) revealed impact of environmental variables on the different species, most of Bacillariophyta species were negative correlation with nutrients(TP and NH_3-N) in the four seasons, Chlorophyta species and Cyanophyta species did not show obvious correlation with environment variables in every season. 8Source identification, contamination status and health risk assessment of heavy metals from road dusts in Dhaka, Bangladesh显示文摘In this study, concentrations of Cr, Mn, Ni, Cu, Zn, Cd and Pb were determined in road dusts collected from different locations in Dhaka to assess source, contamination status and health risk. Energy-dispersive X-ray fluorescence spectroscopy and energy-dispersive X-ray spectroscopy were used to determine Cr, Mn, Ni, Cu, Zn, Cd and Pb and their mean concentrations were 162.27 ± 29.46, 721.18 ± 180.14, 35.65 ± 12.55, 104.56 ± 128.33, 515.32 ± 321.90,BDL, and 342.82 ± 591.20 mg/kg, respectively. Among the heavy metals, highest concentrations of Cu, Zn and Pb were found at urban sites-7(municipal waste dumping) and 8(medical waste incineration). Highest concentration of Cr followed by Cu and Zn was found at site-5(Tejgaon, urban). Principal component analysis revealed that anthropogenic activities are the potential sources for Cr, Ni, Cu, Zn and Pb while earth crust for Mn. Pollution index and pollution load index results suggested that all the sites were contaminated and/or degraded by Cr, Cu, Zn and Pb except sites-9(urban), 10(sub-urban), 11(rural) while sites-7 and 8(urban) were extremely degraded. For noncarcinogenic health risk, hazard quotient values for dermal were higher compared to that of inhalation/ingestion. Though hazard index values were less than 1 at all the sites, these were at least one order of magnitude higher for children group than that of adult group, thus the children group may face more noncarcinogenic health risk at sites -7 and 8. 9Overexpression of miR-101 suppresses collagen synthesis by targeting EZH2 in hypertrophic scar fibroblasts显示文摘Background:MicroRNA-101(miR-101)is a tumor suppressor microRNA(miRNA)and its loss is associated with the occurrence and progression of various diseases.However,the biological function and target of miR-101 in the pathogenesis of hypertrophic scars(HS)remains unknown.Methods:We harvested HS and paired normal skin(NS)tissue samples from patients and cultured their fibroblasts(HSF and NSF,respectively).We used quantitative reverse transcriptase polymerase chain reaction(qRT-PCR),fluorescence in situ hybridization(FISH),enzyme-linked immunosorbent assays(ELISA)and Western blot analyses to measure mRNA levels and protein expression of miR-101,enhancer of zeste homolog 2(EZH2),collagen 1 and 3(Col1 and Col3)andα-smooth muscle actin(α-SMA)in different in vitro conditions.We also used RNA sequencing to evaluate the relevant signaling pathways and bioinformatics analysis and dual-luciferase reporter assays to predict miR-101 targets.We utilized a bleomycin-induced fibrosis mouse model in which we injected miR-101 mimics to evaluate collagen deposition in vivo.Results:We found low expression of miR-101 in HS and HSF compared to NS and NSF.Overexpressing miR-101 decreased Col1,Col3 andα-SMA expression in HSF.We detected high expression of EZH2 in HS and HSF.Knockdown of EZH2 decreased Col1,Col3 andα-SMA in HSF.Mechanistically,miR-101 targeted the 3-untranslated region(3UTR)of EZH2,as indicated by the decreased expression of EZH2.Overexpressing EZH2 rescued miR-101-induced collagen repression.MiR-101 mimics effectively suppressed collagen deposition in the bleomycin-induced fibrosis mouse model.Conclusions:Our data reveal that miR-101 targets EZH2 in HS collagen production,providing new insight into the pathological mechanisms underlying HS formation.Jie Li Yan Li Yunchuan Wang Xiang He Jing Wang Weixia Cai Yanhui Jia Dan Xiao Jian Zhang Ming Zhao Kuo Shen Zichao Li Wenbin Jia Kejia Wang Yue Zhang Linlin Su Huayu Zhu Dahai Hu 2021Burns & Trauma2021,9,1:3
10Osteoporosis-decreased extracellular matrix stiffness impairs connexin 43-mediated gap junction intercellular communication in osteocytes显示文摘Osteocytes are the main sensitive and responsive cells for mechanical stimuli in bone.The connexin family enables them to communicate with each other via forming functional gap junctions.However,how osteoporosis-impaired extracellular mechanical property modulates gap junction intercellular communication in osteocytes remains elusive.In this study,we established an ovariectomy(OVX)-induced osteoporosis mouse model in vivo and a polydimethylsiloxane(PDMS)-based cell culture substrate model in vitro to explore the influence of extracellular matrix(ECM)stiffness on cell-to-cell communication in osteocytes.Firstly,we established an OVX-induced osteoporosis mouse model by characterizing the changes in radiography,morphology and histochemistry of femurs.Our results showed that osteoporosis decreased the bone matrix stiffness together with the changes including the loss of osteocytes and the decrease of protein markers.Meanwhile,the dendritic process interconnection and channel-forming protein,Cx43,were reduced in osteoporosis mice.Next we mimicked ECM stiffness changes in vitro by using PDMS substrates at ratios 1:5 for normal stiffness and 1:45 for osteoporosis stiffness.Our results showed that the decreased ECM stiffness reduced the number of dendritic processes in a single cell and gap junctions between adjacent osteocytes.We further detected the decreased expression of Cx43,in the substrate with decreased stiffness.Finally,we found that gap junction-based intercellular communication was reduced in living osteocytes in the substrate with decreased stiffness.This study demonstrates the correlation between ECM mechanical property and cell-to-cell communication in osteocytes and might pave the way for further exploration of osteoporosis in terms of biomechanics.Demao Zhang Xin Li Caixia Pi Linyi Cai Yang Liu Wei Du Wenbin Yang Jing Xie 2020Acta Biochimica et Biophysica Sinica2020,52,5:3
11Optical visualization and polarized light absorption of the single-wall carbon nanotube to verify intrinsic thermal applications显示文摘The predicted extraordinary properties of carbon nanotubes(CNTs)from theoretical calculations have great potential for many applications.However,reliable experimental determination of intrinsic properties at the single-tube level is currently a matter of concern,and many challenges remain because of the unhandled and nanoscale size of individual nanotubes.Here,we demonstrated a prototype to detect the intrinsic thermal conductivity of the single-wall carbon nanotube(SWCNT)and verify the significant non-resonant optical absorption behavior on tiny nanotubes by integrating the nanotube and ice into a new core-shell design.In particular,a reversible optical visualization method based on the individual suspended ultra-long SWCNT was first developed by wrapping a nanotube with ice in the cryogenic air environment.The light-induced thermal effect on the hybrid core-shell structure was used tomelt the ice shell,which subsequently acted as a temperature sensor to verify the intrinsic thermal conductivity of the core-like nanotube.More interestingly,we successfully determined for the first time the thermal response phenomenon of the tiny absorption cross section in SWCNT in the vertical-polarization configuration and the significant non-resonant absorption behavior in the parallel-polarization configuration.These investigations will provide a better understanding for the unique optical behaviors of CNT and enable the detection of intrinsic properties of various one-dimensional nanostructures such as nanotubes,nanowires,and nanoribbons.Xiao Zhang Li Song Le Cai Xuezeng Tian Qiang Zhang Xiaoying Qi Wenbin Zhou Nan Zhang Feng Yang Qingxia Fan Yanchun Wang Huaping Liu Xuedong Bai Weiya Zhou Sishen Xie 2015Light(Science & Applications)2015,4,1:2

13Clinical manifestations and pathogen characteristics in children admitted for suspected COVID-19显示文摘Coronavirus disease 2019(COVID-19),which is caused by severe acute respiratory syndrome coronavirus-2(SARS-CoV-2),has spread around the world.However,approaches to distinguish COVID-19 from pneumonia caused by other pathogens have not yet been reported.We retrospectively analyzed the clinical data of 97 children with probable COVID-19.A total of 13(13.4%)patients were confirmed positive for SARS-CoV-2 infection by nucleic acid RT-PCR testing,and 41(42.3%)patients were found to be infected with other pathogens.Notably,no pathogen was detected in 43(44.3%)patients.Among all patients,25(25.8%)had familial cluster exposure history,and 52(53.6%)had one or more coexisting conditions.Fifteen(15.5%)patients were admitted or transferred to the PICU.In the 11 confirmed COVID-19 cases,5(45.5%)and 7(63.6%)were positive for IgM and IgG against SARS-CoV-2,respectively.In 22 patients with suspected COVID-19,1(4.5%)was positive for IgG but negative for IgM.The most frequently detected pathogen was Mycoplasma pneumonia(29,29.9%).One patient with confirmed COVID-19 died.Our results strongly indicated that the detection of asymptomatic COVID-19 or coexisting conditions must be strengthened in pediatric patients.These cases may be difficult to diagnose as COVID-19 unless etiologic analysis is conducted.A serologic test can be a useful adjunctive diagnostic tool in cases where SARS-CoV-2 infection is highly suspected but the nucleic acid test is negative.Xiaofang Cai Hanlan Jiang Simin Zhang Shengying Xia Wenhui Du Yaoling Ma Tao Yu Wenbin Li 2020Frontiers of Medicine2020,14,6:2
14The role of the MAP kinase−kinase protein StMKK1 in potato immunity to different pathogens显示文摘Mitogen-activated protein kinase(MAPK)cascades play important roles in plant immunity.Previously,we reported that the potato StMKK1 protein negatively regulates Nicotiana benthamiana resistance to Phytophthora infestans.However,the functions of StMKK1 in potato immunity are unknown.To investigate the roles of StMKK1 in potato resistance to different pathogens,such as the potato late-blight pathogen P.infestans,the bacterial wilt pathogen Ralstonia solanacearum,and the gray-mold fungal pathogen Botrytis cinerea,we generated StMKK1 transgenic lines and investigated the response of potato transformants to destructive oomycete,bacterial,and fungal pathogens.The results showed that overexpression and silencing of StMKK1 do not alter plant growth and development.Interestingly,we found that StMKK1 negatively regulated potato resistance to the hemibiotrophic/biotrophic pathogens P.infestans and R.solanacearum,while it positively regulated potato resistance to the necrotrophic pathogen B.cinerea.Further investigation showed that overexpression of StMKK1 suppressed potato pathogen-associated molecular pattern(PAMP)-triggered immunity(PTI)and salicylic acid(SA)-related responses,while silencing of StMKK1 enhanced PTI and SA-related immune responses.Taken together,our results showed that StMKK1 plays dual roles in potato defense against different plant pathogens via negative regulation of PTI and SA-related signaling pathways.Xiaokang Chen Wenbin Wang Pingping Cai Ziwei Wang Tingting Li Yu Du 2021Horticulture Research2021,8,1:2
15Association of human leukocyte antigen-DR-DQ-DP haplotypes with the risk of hepatitis B virus-related hepatocellular carcinoma显示文摘Aim:Genetic polymorphisms of human leukocyte antigen(HLA)class II molecules are associated with chronic hepatitis B virus(HBV)infection.We aimed to investigate the impacts of HLA-II haplotypes on viral evolution and the risks of HBV-caused liver diseases.Methods:HLA-DR-DQ-DP haplotypes were estimated in 1210 healthy controls,296 HBV clearance subjects,301 asymptomatic hepatitis B surface antigen carriers,770 chronic hepatitis B patients,443 HBV-related liver cirrhosis(LC)patients,and 1037 HBV-related hepatocellular carcinoma(HCC)patients.HBV mutations were determined by sequencing.The associations of HLA-DR-DQ-DP haplotypes with viral mutations and the risks of liver diseases were assessed by multivariate logistic regression.Results:Compared to HBV-free subjects,the haplotypes CCAACG,CCGACG,TCAATA,and TCGATA were associated with decreased HCC risk,with an odds ratio(OR)[95%confidence interval(CI)]of 0.62(0.40-0.95),0.60(0.39-0.92),0.73(0.54-0.98),and 0.58(0.42-0.78),respectively.CCAACG,CCGACG,and TCAATA were significantly associated with decreased frequencies of the HCC-risk HBV mutations:preS1 deletion,APOBECsignature HBV mutations in the core promoter and preS regions,A51C/T,G104C/T,and G146C/T.TCGATA and TTAACG were associated with increased LC risk,with an OR(95%CI)of 1.54(1.03-2.30)and 2.23(1.50-3.33),respectively.However,TCGATA and TTAACG were not consistently associated with the cirrhosis-risk HBV mutations.Conclusion:CCAACG,CCGACG,and TCAATA are inversely associated with HCC risk,possibly because they are involved in creating an immune microenvironment attenuating the generation of HCC-risk HBV mutations.TCGATA and TTAACG might predispose the polarity of immunity towards Th17 isotype related to LC.Yifan Chen Jiansheng Lin Yang Deng Wenbin Liu Zishuai Li Xinyu Zhou Shiliang Cai Rui Pu Jianhua Yin Xiaojie Tan Jun Zhao Xue Han Guangwen Cao 2022Hepatoma Research2022,8,1:2
