| 2 | Promotion of spinosad biosynthesis by chromosomal integration of the Vitreoscilla hemoglobin gene in Saccharopolyspora spinosa显示文摘To promote spinosad biosynthesis by improving the limited oxygen supply during high-density fermentation of Saccharopolyspora spinosa, the open reading frame of the Vitreoscilla hemoglobin gene was placed under the control of the promoter for the erythromycin resistance gene by splicing using overlapping extension PCR. This was cloned into the integrating vector pSET152, yielding the Vitreoscilla hemoglobin gene expression plasmid pSET152EVHB. This was then introduced into S. spinosa SP06081 by conjugal transfer, and integrated into the chromosome by site-specific recombination at the integration site ΦC31 on pSET152EVHB. The resultant conjugant, S. spinosa S078-1101, was genetically stable. The integration was further confirmed by PCR and Southern blotting analysis. A carbon monoxide differential spectrum assay showed that active Vitreoscilla hemoglobin was successfully expressed in S. spinosa S078-1101. Fermentation results revealed that expression of the Vitreoscilla hemoglobin gene significantly promoted spinosad biosynthesis under normal oxygen and moderately oxygen-limiting conditions (P<0.01). | 4 | Reproductive management through integration of PGD and MPS-based noninvasive prenatal screening/diagnosis for a family with GJB2-associated hearing impairment显示文摘A couple with a proband child of GJB2(encoding the gap junction protein connexin 26)-associated hearing impairment and a previous pregnancy miscarriage sought for a reproductive solution to bear a healthy child. Our study aimed to develop a customized preconception-to-neonate care trajectory to fulfill this clinical demand by integrating preimplantation genetic diagnosis(PGD), noninvasive prenatal testing(NIPT), and noninvasive prenatal diagnosis(NIPD) into the strategy. Auditory and genetic diagnosis of the proband child was carried out to identify the disease causative mutations. The couple then received in-vitro-fertilization treatment, and eight embryos were obtained for day 5 biopsy. PGD was performed by short-tandem-repeat linkage analysis and Sanger sequencing of GJB2 gene. Transfer of a GJB2 c.235del C heterozygous embryo resulted in a singleton pregnancy. At the 13 th week of gestation, genomic DNA(g DNA) from the trio family and cell-free DNA(cf DNA) from maternal plasma were obtained for assessment of fetal chromosomal aneuploidy and GJB2 mutations. NIPT and NIPD showed the absence of chromosomal aneuploidy and GJB2-associated disease in the fetus, which was later confirmed by invasive procedures and postnatal genetic/auditory diagnosis. | 7 | A Brief Introduction to BNU-HESM1.0 and Its Earth Surface Temperature Simulations显示文摘Integrated assessment models and coupled earth system models both have their limitations in understanding the interactions between human activity and the physical earth system. In this paper,a new human–earth system model,BNUHESM1.0,constructed by combining the economic and climate damage components of the Dynamic Integrated Model of Climate Change and Economy to the BNU-ESM model,is introduced. The ability of BNU-HESM1.0 in simulating the global CO2 concentration and surface temperature is also evaluated. We find that,compared to observation,BNU-HESM1.0underestimates the global CO2 concentration and its rising trend during 1965–2005,due to the uncertainty in the economic components. However,the surface temperature simulated by BNU-HESM1.0 is much closer to observation,resulting from the overestimates of surface temperature by the original BNU-ESM model. The uncertainty of BNU-ESM falls within the range of present earth system uncertainty,so it is the economic and climate damage component of BNU-HESM1.0 that needs to be improved through further study. However,the main purpose of this paper is to introduce a new approach to investigate the complex relationship between human activity and the earth system. | 8 | Aerodynamic performance enhancement forflapping airfoils by co-flow jet显示文摘Introducing active flow control into the design of flapping wing is an effective way to enhance its aerodynamic performance.In this paper,a novel active flow control technology called Co-Flow Jet(CFJ)is applied to flapping airfoils.The effect of CFJ on aerodynamic performance of flapping airfoils at low Reynolds number is numerically investigated using Unsteady Reynolds Averaged Navier-Stokes(URANS)simulation with Spalart-Allmaras(SA)turbulence model.Numerical methods are validated by a NACA6415-based CFJ airfoil case and a S809 pitching airfoil case.Then NACA6415 baseline airfoil and NACA6415-based CFJ airfoil with jet-off and jet-on are simulated in flapping motion,with Reynolds number 70,000 and reduced frequency 0.2.As a result,CFJ airfoils with jet-on generally have better lift and thrust characteristics than baseline airfoils and jet-off airfoil when Cμgreater than 0.04,which results from the CFJ effect of reducing flow separation by injecting high-energy fluid into boundary layer.Besides,typical kinematic and geometric parameters,including the reduced frequency and the positions of the suction and injection slot,are systematically studied to figure out their influence on aerodynamic performance of the CFJ airfoil.And a variable Cμjet control strategy is proposed to further improve effective propulsive efficiency.Compared with using constant Cμ,an increase of effective propulsive efficiency by22.6%has been achieved by using prescribed variable CμNACA6415-based CFJ airfoil at frequency 0.2.This study may provide some guidance to performance enhancement for Flapping wing Micro Air Vehicles(FMAV). | Tao WU Bifeng SONG Wenping SONG Wenqing YANG Zhonghua HAN | 2020 | Chinese Journal of Aeronautics2020,33,10: | 7 |

| 10 | Short-term effects of biochar and gypsum on soil hydraulic properties and sodicity in a saline-alkali soil显示文摘Salt and sodicity of saline-alkali soil adversely affect the construction of ecological landscapes and negatively impact crop production.The reclamation potential of biochar(BC,wheat straw biochar applied at\%by weight),gypsum(G,0.4%by weight),and gypsum coupled with biochar(GBC)was examined in this laboratory-based study by evaluating their effects on a saline-alkali soil(silt loam)with no amendment as a control(CK).Saline ice and fresh water(simulated rainfall)were leached through soil columns to investigate changes in salt content,sodium adsorption ratio(SAR),alkalinity,and pH of the leachate and the soil.Results showed that saturated water content and field water capacity(FWC)significantly increased by 4.4%and 5.6%,respectively,in the BC treatment after a short incubation time.Co-application of biochar and gypsum(GBC)increased soil saturated hydraulic conductivity(Ks)by 58.4%,which was also significantly higher than the sole addition.Electrical conductivity(EC)of the leachate decreased sharply after saline ice leaching;subsequent freshwater leaching accelerated the removal of the rest of the salts,irrespective of the amendment application.However,the application of gypsum(G and GB)significantly enhanced the removal of exchangeable Na^+and reduced leachate SAR.After leaching,the soil salt content decreased significantly for all treatments.The application of gypsum resulted in a significantly lower soil pH,exchangeable sodium percentage(ESP),SAR,and alkalinity values than those recorded for the CK and BC treatments.These results demonstrated that the co-application of gypsum and biochar could improve saline-alkali soil hydraulic conductivity and decrease leaching-induced sodicity over a short period. | Yue ZHANG Jingsong YANG Rongjiang YAO Xiangping WANG Wenping XIE | 2020 | Pedosphere2020,30,5: | 6 |

| 13 | Multi-infill strategy for kriging models used in variable fidelity optimization显示文摘In this paper, a computationally efficient optimization method for aerodynamic design has been developed. The low-fidelity model and the multi-infill strategy are utilized in this approach.Low-fidelity data is employed to provide a good global trend for model prediction, and multiple sample points chosen by different infill criteria in each updating cycle are used to enhance the exploitation and exploration ability of the optimization approach. Take the advantages of lowfidelity model and the multi-infill strategy, and no initial sample for the high-fidelity model is needed. This approach is applied to an airfoil design case and a high-dimensional wing design case.It saves a large number of high-fidelity function evaluations for initial model construction. What's more, faster reduction of an aerodynamic function is achieved, when compared to ordinary kriging using the multi-infill strategy and variable-fidelity model using single infill criterion. | 15 | Interaction mechanism of the interface between a deep buried sand and a paleo-weathered rock mass using a high normal stress direct shear apparatus显示文摘In order to evaluate the feasibility of safe mining close to the contact zone under reduced security coal pillar conditions at a coal mine in eastern China, the interaction mechanism of the interface between deep buried sand and a paleo-weathered rock mass was investigated in the laboratory by direct shear testing. A DRS-1 high pressure soil shear testing machine and orthogonal design method were used in the direct shear tests. Variance and range methods were applied to analyze the sensitivity of each factor that has an influence on the mechanical characters of the interface. | 19 | Aerodynamic performance of owl-like airfoil undergoing bio-inspired flapping kinematics显示文摘Natural flyers have extraordinary flight skills and their prominent aerodynamic performance has attracted a lot of attention.However,the aerodynamic mechanism of birds’flapping wing kinematics still lacks in-depth understanding.In this paper,the aerodynamic performance of owl-like airfoil undergoing bio-inspired flapping kinematics extracted from a free-flying owl wing has been numerically investigated.The overset mesh technique is used to deal with the large range movements of flapping airfoils.The bio-inspired kinematics consist of plunging and pitching movement.A pure sinusoidal motion and a defined motion composed of plunging of sinusoidal motion and pitching of the bio-inspired kinematics are selected for comparison.The other two NACA airfoils are also selected to figure out the advantages of the owl-like airfoil.It is found that the cambered owl-like airfoil can enhance lift during the downstroke.The bio-inspired kinematics have an obvious advantage in lift generation with a presence of higher peak lift and positive lift over a wider proportion of the flapping cycle.Meanwhile,the bio-inspired motion is more economical for a lower power consumption compared with the sinusoidal motion.The sinusoidal flapping motion is better for thrust generation for a higher peak thrust value in both upstroke and downstroke,while the bio-inspired kinematics mainly generate thrust during the downstroke but produce more drag during the upstroke.The defined motion has similar lift performance with the bio-inspired kinematics,while it consumes more energy and generates less thrust.The unsteady flow field around airfoils is also analyzed to explain the corresponding phenomenon.The research in this paper is helpful to understand the flight mechanism of birds and to design a micro air vehicle with higher performance. | Xinyu LANG Bifeng SONG Wenqing YANG Wenping SONG | 2021 | Chinese Journal of Aeronautics2021,34,5: | 2 |
| 20 | Local delivery of arsenic trioxide nanoparticles for hepatocellular carcinoma treatment显示文摘Hepatocellular carcinoma(HCC)is a malignancy with a poor prognosis.Surgery combined with chemotherapy has been recommended as a curative regimen for HCC.Nevertheless,the anticancer mechanisms of chemicals in hepatocellular carcinoma remain unclear.Pyroptosis is a type of programmed necrosis,and its mechanism in hepatocellular carcinoma is poorly understood.The efficacy and mechanism of arsenic trioxide nanoparticles in the treatment of HCC were explored in this research.Arsenic trioxide alone and arsenic trioxide nanoparticles were conveniently administered to mice intratumorally using a needle.Compared with As_(2)O_(3),As_(2)O_(3)nanoparticles(As_(2)O_(3)-NPs)showed better inhibition,promoted greater LDH release,and induced cell morphology indicative of pyroptosis in vitro.Compared with the free drug,As_(2)O_(3)-NPs increased GSDME-N expression and decreased Dnmt3a,Dnmt3b,and Dnmt1 expression in Huh7 cells.In vivo,As_(2)O_(3)-NPs induced a significant decrease in the expression of Dnmt3a,Dnmt3b and Dnmt1,but significantly upregulated the expression of GSDME-N(gasdermin E(GSDME)was originally found to be related to deafness;recently,it has been defined as a gasdermin family member associated with pyroptosis).As_(2)O_(3)-NPs inhibited tumor growth more strongly than As_(2)O_(3)or control,a finding likely attributed to the downregulation of PCNA and DNMT-related proteins and the upregulation of GSDME-N. | Jian Hu Yi Dong Li Ding Yang Dong Zhihua Wu Wenping Wang Ming Shen Yourong Duan | 2019 | Signal Transduction and Targeted Therapy2019,4,1: | 2 |
