| 2 | Exosomes derived from human amniotic epithelial cells accelerate diabetic wound healing via PI3K-AKT-mTOR-mediated promotion in angiogenesis and fibroblast function显示文摘Background:Diabetic wounds are one of the most common and serious complications of diabetes mellitus,characterized by the dysfunction of wound-healing-related cells in quantity and quality.Our previous studies revealed that human amniotic epithelial cells(hAECs)could promote diabetic wound healing by paracrine action.Interestingly,numerous studies demonstrated that exosomes derived from stem cells are the critical paracrine vehicles for stem cell therapy.However,whether exosomes derived from hAECs(hAECs-Exos)mediate the effects of hAECs on diabetic wound healing remains unclear.This study aimed to investigate the biological effects of hAECs-Exos on diabetic wound healing and preliminarily elucidate the underlying mechanism.Methods:hAECs-Exos were isolated by ultracentrifugation and identified by transmission electron microscopy,dynamic light scattering and flow cytometry.A series of in vitro functional analyses were performed to assess the regulatory effects of hAECs-Exos on human fibroblasts(HFBs)and human umbilical vein endothelial cells(HUVECs)in a high-glycemic microenvironment.Highthroughput sequencing and bioinformatics analyses were conducted to speculate the related mechanisms of actions of hAECs-Exos on HFBs and HUVECs.Subsequently,the role of the candidate signaling pathway of hAECs-Exos in regulating the function of HUVECs and HFBs,as well as in diabetic wound healing,was assessed.Results:hAECs-Exos presented a cup-or sphere-shaped morphology with a mean diameter of 105.89±10.36 nm,were positive for CD63 and TSG101 and could be internalized by HFBs and HUVECs.After that,hAECs-Exos not only significantly promoted the proliferation and migration of HFBs,but also facilitated the angiogenic activity of HUVECs in vitro.High-throughput sequencing revealed enriched miRNAs of hAECs-Exos involved in wound healing.Kyoto Encyclopedia of Genes and Genomes and Gene Ontology analyses have shown that the target genes of the top 15 miRNAs were highly enriched in the PI3K-AKT pathway.Further functional studies demonstrated that the PI3K-AKT-mTOR pathway was necessary for the induced biological effects of hAECs-Exos on HFBs and HUVECs,as well as on wound healing,in diabetic mice.Conclusions:Our findings demonstrated that hAECs-Exos represent a promising,novel strategy for diabetic wound healing by promoting angiogenesis and fibroblast function via activation of the PI3K-AKT-mTOR pathway. | Pei Wei Chenjian Zhong Xiaolan Yang Futing Shu Shichu Xiao Teng Gong Pengfei Luo Li Li Zhaohong Chen Yongjun Zheng Zhaofan Xia | 2020 | Burns & Trauma2020,8,1: | 12 |

| 6 | Ablative fractional CO_(2)laser surgery improving sleep quality,pain and pruritus in adult hypertrophic scar patients:a prospective cohort study显示文摘Background:Poor sleep quality is associated with a decrease in quality of life in patients with major burn scars,combined with pruritus and pain.Few interventions have been reported to improve the sleep quality of patients with scars.In the current prospective cohort study,we investigated the efficacy of CO_(2)-ablative fractional laser(AFL)surgery vs conventional surgery in post-burn patients with hypertrophic scars with sleep quality as the primary study outcome.Methods:In total 68 consecutive patients undergoing scar surgical treatment were recruited,including a CO_(2)-AFL surgery cohort(n=35)and a conventional surgery cohort(n=33).A subgroup from the AFL cohort was selected.Sleep quality,pain and pruritus were evaluated.Multiple linear regression analyses were performed to reveal the effect of CO_(2)-AFL surgery.Results:The CO_(2)-AFL surgery cohort had significantly lower Pittsburgh sleep quality index(PSQI)global scores than the conventional surgery cohort after the last surgical treatment.In the subgroup of patients receiving hardware sleep monitoring,CO_(2)-AFL markedly increased deep sleep time,deep sleep efficiency and reduced initial sleep latency.Compared to the conventional surgery cohort,the CO_(2)-AFL cohort presented significantly lower pain and pruritus scores.Correlation analysis showed pain and pruritus were significantly associated with PSQI scores,and there were also significant correlations between pain and pruritus scores.Multiple linear regression analysis showed that surgery method was negatively linearly correlated with visual analog scale(VAS)pain score,brief pain inventory(BPI)total,VAS pruritus score,5-D itch scale total,four-item itch questionnaire(FIIQ)total and PSQI total.Conclusions:CO_(2)-AFL surgery significantly improved sleep quality and reduced pain and pruritus of hypertrophic scar patients.The alleviation of sleep disorder was associated with improvement of deep sleep quality including deep sleep time and deep sleep deficiency.Trial registration:The Chinese Clinical Trial Registry(ChiCTR200035268)approved retrospectively registration on 5 Aug 2020. | Kaiyang Lv Huazhen Liu Haiting Xu Caixia Wang Shihui Zhu Xiaozhen Lou Pengfei Luo Shichu Xiao Zhaofan Xia | 2021 | Burns & Trauma2021,9,1: | 4 |
| 7 | A Virulent PEDV Strain FJzz1 with Genomic Mutations and Deletions at the High Passage Level Was Attenuated in Piglets via Serial Passage In Vitro显示文摘Highly virulent porcine epidemic diarrhea virus(PEDV)strains re-emerged and circulated in China at the end of 2010,causing significant economic losses in the pork industry worldwide.To understand the genetic dynamics of PEDV during its passage in vitro,the PEDV G2 strain FJzzl was serially propagated in Vero cells for up to 200 passages.The susceptibility and adaptability of the FJzzl strain increased gradually as it was serially passaged in vitro.Sequence analysis revealed that amino acid(aa)changes were mainly concentrated in the S glycoprotein,which accounted for 72.22%-85.71%of all aa changes.A continuous aa deletion(^(55)I^(56)G^(57)E→^(55)K^(56)Δ^(57)Δ)occurred in the N-terminal domain of S1(Sl-NTD).To examine how the aa changes affected its virulence,FJzzl-F20 and FJzzl-F200 were selected to simultaneously evaluate their pathogenicity in suckling piglets.All the piglets in the FJzzl-F20-infected group showed typical diarrhea at 24 h postinfection,and the piglets died successively by 48 h postinfection.However,the clinical signs of the piglets in the FJzzl-F200-infected group were significantly weaker,and no deaths occurred.The FJzzl-F200-infected group also showed a lower level of fecal viral shedding and lower viral loads in the intestinal tissues,and no obvious histopathological lesions.TypeⅠandⅢinterferon were induced in the FJzzl-F200 infection group,together with pro-inflammatory cytokines,such as TNF-α,IL-1βand IL-8.These results indicate that the identified genetic changes may contribute to the attenuation of FJzzl strain,and the attenuated FJzzl-F200 may have the potential for developing PEDV live-attenuated vaccines. | Pengfei Chen Xiongwei Zhao Shuting Zhou Tianxing Zhou Xiangmei Tan Xia Wu Wu Tong Fei Gao Lingxue Yu Yifeng Jiang Hai Yu Zhibiao Yang Guangzhi Tong Yanjun Zhou | 2021 | Virologica Sinica2021,36,5: | 4 |

| 10 | A new model for chromitite formation in ophiolites: Fluid immiscibility显示文摘Although the involvement of hydrous fluids has been widely invoked in formation of podiform chromitites in ophiolites, there is lack of natural evidence to signify the role and mechanism of fluids. In this study, a new model for the genesis of podiform chromitite is proposed on basis of revisits of comprehensive petrological, mineralogical and geochemical results of the well-preserved K?z?lda? ophiolite and the well-characterized Luobusa chromite deposit. In this model, ascending magmas intruding oceanic lithospheric mantle would presumably form a series of small magma chambers continuously connected by conduits. Tiny chromite nuclei would collect fluids dispersed in such magmas to form nascent droplets. They tend to float upward in the magma chamber and would be easily transported upward by flowing magmas. Chromite-rich droplets would be enlarged via coalescence of dispersed droplets during mingling and circulation in the magma chamber and/or transport in magma conduits. Crystallization of the chromite-rich liquid droplets would proceed from the margin of the droplet inward, leaving liquid entrapped within grains as precursor of mineral inclusions. With preferential upward transportation, immiscible chromite-rich liquids would coalesce to a large pool in a magma chamber. Large volumes of chromite would crystallize in situ, forming podiform chromitite and resulting in fluid enrichment in the chamber. The fluids would penetrate and compositionally modify ambient dunite and harzburgite, leading to significant fractionations of elemental and isotopic compositions between melts and fluids from which dunite and chromitite respectively formed. | 14 | Experimental and density functional theory computational evaluation of poly(N-vinyl caprolactam-co-butyl methacrylate) kinetic hydrate inhibitors显示文摘Natural gas hydrate inhibitor has been serving the oil and gas industry for many years. The development and search for new inhibitors remain the focus of research. In this study, the solution polymerization method was employed to prepare poly(N-vinyl caprolactam-co-butyl methacrylate)(P(VCap-BMA)), as a new kinetic hydrate inhibitor(KHI). The inhibition properties of P(VCap-BMA) were investigated by tetrahydrofuran(THF) hydrate testing and natural gas hydrate forming and compared with the commercial KHIs. The experiment showed that PVCap performed better than copolymer P(VCap-BMA). However,low doses of methanol or ethylene glycol are compounded with KHIs. The compounding inhibitors show a synergistic inhibitory effect. More interesting is the P(VCap-BMA)-methanol system has a better inhibitory effect than the PVCap-methanol system. 1% P(VCap-BMA) + 5% methanol presented the best inhibiting performance at subcooling 10.3 °C, the induction time of natural gas hydrate was 445 min.Finally, the interaction between water and several dimeric inhibitors compared by natural bond orbital(NBO) analyses and density functional theory(DFT) indicated that inhibitor molecules were able to form the hydrogen bond with the water molecules, which result in gas hydrate inhibition. | 18 | Probing the active sites of 2D nanosheets with Fe-N-C carbon shell encapsulated Fe_(x)C/Fe species for boosting sodium-ion storage performances显示文摘Developing stable but high active metal-nitrogen-carbon(M-N-C)-based hard carbon anode is a promising way to be the alternatives to graphene and blank hard carbon for sodium-ion batteries(SIBs),requiring the precise tailoring of the electronic structure for optimizing the Na+intercalation behavior,yet is greatly challenging.Herein,Fe-N-C graphitic layer-encapsulating Fe3C species within hard carbon nanosheets(Fe-N-C/Fe3C@HCNs)are rationally engineered by pyrolysis of self-assembled polymer.Impressively,the Fe-N-C/Fe3C@HCNs exhibit outstanding rate capacity(242 mAh·g^(−1)at 2,000 mA·g^(−1)),which is 2.1 and 4.2 times higher than that of Fe-N-C and N-doped carbon(N-C),respectively,and prolonged cycling stability(176 mAh·g^(−1)at 2,000 mA·g^(−1)after 2,000 cycles).Theoretical calculations unveil that the Fe3C species enhance the electronic transfer from Na to Fe-N-C,resulting in the charge redistribution between the interfaces of Fe3C and Fe-N-C.Thus,the optimized adsorption behavior towards Na+reduces the thermodynamic energy barriers.The synergistic effect of Fe3C and Fe-N-C species maintains the structural integrity of electrode materials during the sodiation/desodiation process.The in-depth insight into the advanced Na+storage mechanisms of Fe3C@Fe-N-C offers precise guidance for the rational establishment of confinement heterostructures in SIBs. | Huicong Xia Pengfei Yuan Lingxing Zan Gan Qu Yunchuan Tu Kaixin Zhu Yifan Wei Zeyu WeiFangying Zheng Mo Zhang Yongfeng Hu Dehui Deng Jianan Zhang | 2022 | Nano Research2022,15,8: | 1 |
