| 2 | A draft sequence of the rice (Oryza sativa ssp. indica) genome显示文摘The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a 'whole genome shotgun' approach, we have pro-duced a draft rice genome sequence of Oryza sativa ssp. in-dica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from an indica variant cultivar 93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. | 3 | Mechanism and control technology of strong ground pressure behaviour induced by high-position hard roofs in extra-thick coal seam mining显示文摘This work aimed at revealing the mechanism of strong ground pressure behaviour(SGPB)induced by high-position hard roof(HHR).Based on the supporting structures model of HHR,a modified voussoir beam mechanical model for HHR was established by considering the gangue support coefficient,through which the modified expressions of limit breaking span and breaking energy of HHR were deduced.Combined with the relationship between the dynamic-static loading stress of supporting body(hydraulic support and coal wall)and its comprehensive supporting strength,the criteria of ground pressure behaviour(GPB)induced by HHR were discussed.The types of Ⅰ_(1),Ⅰ_(2),Ⅱ_(1),andⅡ_(2) of GPB were interpreted.Results showed that types Ⅰ_(1) and Ⅰ_(2) were the main forms of SGPB in extra-thick coal seam mining.The main manifestation of SGPB was static stress,which was mainly derived from the instability of HHR rather than fracture.Accordingly,an innovative control technology was proposed,which can weaken static load by vertical-well separated fracturing HHR.The research results have been successfully applied to the 8101 working face in Tashan coal mine,Shanxi Province,China.The results of a digital borehole camera observation and stress monitoring proved the rationality of the GPB criteria.The control technology was successful,paving the way for new possibilities to HHR control for safety mining. | Chao Pan Binwei Xia Yujun Zuo Bin Yu Changnan Ou | 2022 | International Journal of Mining Science and Technology2022,32,3: | 6 |
| 4 | PUM1 represses CDKN1B translation and contributes to prostate cancer progression显示文摘Posttranscriptional regulation of cancer gene expression programs plays a vital role in carcinogenesis;identifying the critical regulators of tumorigenesis and their molecular targets may provide novel strategies for cancer diagnosis and therapeutics.Highly conserved RNA-binding protein Pumilio-1(PUM1)regulates mouse growth and cell proliferation,propelling us to examine its role in cancer.We found human PUM1 is highly expressed in a diverse group of cancer,including prostate cancer;enhanced PUM1 expression is also correlated with reduced survival among prostate cancer patients.Detailed expression analysis in twenty prostate cancer tissues showed enhanced expression of PUM1 at mRNA and protein levels.Knockdown of PUM1 reduced prostate cancer cell proliferation and colony formation,and subcutaneous injection of PUM1 knockdown cells led to reduced tumor size.Downregulation of PUM1 in prostate cancer cells consistently elevated cyclin-dependent kinase inhibitor 1B(CDKN1B)protein expression through increased translation but did not impact its mRNA level,while overexpression of PUM1 reduced CDKN1B protein level.Our finding established a critical role of PUM1 mediated translational control,particularly the PUM1-CDKN1B axis,in prostate cancer cell growth and tumorigenesis.We proposed that PUM1-CDKN1B regulatory axis may represent a novel mechanism for the loss of CDKN1B protein expression in diverse cancers and potential targets for therapeutics development. | Xin Li Jian Yang Xia Chen Dandan Cao Eugene Yujun Xu | 2021 | The Journal of Biomedical Research2021,35,5: | 3 |

| 20 | Tanshinone IIA protects intestinal epithelial cells from ferroptosis through the upregulation of GPX4 and SLC7A11显示文摘Background:Inflammatory bowel disease(IBD)is a chronic inflammatory disease of the gastrointestinal tract.The destruction of the intestinal epithelial barrier is one of the major pathological processes in IBD pathology.Growing evidence indicated that epithelial cell ferroptosis is linked to IBD and is considered a target process.Methods:RAS-selective lethal 3(RSL3)was used to induce ferroptosis in intestinal epithelial cell line No.6(IEC-6)cells,and cell ferroptosis and the effects of tanshinone IIA(Tan IIA)were determined by cell counting kit-8(CCK-8),reactive oxygen species(ROS)staining,Giemsa staining and transmission electron microscope(TEM).The cell viability of natural product library compounds was determined by CCK-8.The expression of ferroptosis-related genes were detected by real-time quantitative polymerase chain reaction(RT-qPCR)and western blot.Results:Treatment of IEC-6 cells results in the accumulation of ROS and typical morphological characteristics of ferroptosis.RSL3 treatment caused rapid cellular cytotoxicity which could be reversed by ferrostatin-1(Fer-1)in IEC-6 cells.Natural product library screening revealed that Tan IIA is a potent inhibitor of IEC-6 cell ferroptosis.Tan IIA could significantly protect the RSL3-induced ferroptosis of IEC-6 cells.Furthermore,the ferroptosis suppressors,glutathione peroxidase 4(GPX4),solute carrier family 7 member 11(SLC7A11),and miR-17-92 were found to be early response genes in RSL3-treated cells.Treatment of IEC-6 cells with Tan IIA resulted in upregulation of GPX4,SLC7A11,and miR-17-92.Conclusion:Our study demonstrated that Tan IIA protects IEC-6 cells from ferroptosis through the upregulation of GPX4,SLC7A11,and miR-17-92.The findings might provide a theoretical grounding for the future application of Tan IIA to treat or prevent IBD. | HAN WANG YANG SUN XIAOXU ZHANG XIAOYING WANG YUJUN XIA LISHENG WANG | 2023 | BIOCELL2023,47,5: | 0 |
