2Lycorine inhibits breast cancer growth and metastasis via inducing apoptosis and blocking Src/FAK-involved pathway显示文摘Breast cancer is the most commonly diagnosed cancer type worldwide among women and more than 90% of patients die from tumor metastasis. Lycorine, a natural alkaloid, has been widely reported possessing potential efficacy against cancer proliferation and metastasis. In our study, the anti-tumor potency on breast cancer was evaluated in vitro and in vivo for the first time. Our results indicated that lycorine inhibited breast cancer cells growth, migration and invasion as well as induced their apoptosis.In in vivo study, lycorine not only suppressed breast tumor growth in xenograft models and inhibited breast tumor metastasis in MDA-MB-231 tail vein model. More importantly, we found lycorine had less toxicity than first-line chemotherapy drug paclitaxel at the same effective dose in vivo. Furthermore, on mechanism, lycorine inhibited tumor cell migration and invasion via blocking the Src/FAK(focal adhesion kinase)-involved pathway. 3Management of granulomatous lobular mastitis: an international multidisciplinary consensus(2021 edition)显示文摘Granulomatous lobular mastitis(GLM) is a rare and chronic benign inflammatory disease of the breast. Difficulties exist in the management of GLM for many front-line surgeons and medical specialists who care for patients with inflammatory disorders of the breast. This consensus is summarized to establish evidence-based recommendations for the management of GLM. Literature was reviewed using PubMed from January 1, 1971 to July 31, 2020. Sixty-six international experienced multidisciplinary experts from 11 countries or regions were invited to review the evidence.Levels of evidence were determined using the American College of Physicians grading system, and recommendations were discussed until consensus. Experts discussed and concluded 30 recommendations on historical definitions,etiology and predisposing factors, diagnosis criteria, treatment, clinical stages, relapse and recurrence of GLM. GLM was recommended as a widely accepted definition. In addition, this consensus introduced a new clinical stages and management algorithm for GLM to provide individual treatment strategies. In conclusion, diagnosis of GLM depends on a combination of history, clinical manifestations, imaging examinations, laboratory examinations and pathology.The approach to treatment of GLM should be applied according to the different clinical stage of GLM. 4Epidemiology,clinical and laboratory characteristics of currently alive HIV-1 infected former blood donors naive to antiretroviral therapy in Anhui Province,China显示文摘Background Unregulated commercial blood/plasma collection among farmers occurred between 1992 and 1995 in central China and caused the second major epidemic of human immunodeficiency virus type 1 (HIV-1) infection in China. It is important to characterize HIV-1-infected former blood donors and to study characteristics associated with disease progression for future clinical intervention and vaccine development. Methods A cross-sectional study was performed on HIV-1-infected former blood donors (FBDs) and age-matched HIV-seronegative local residents. Demographic, epidemiologic, clinical and key laboratory data were collected from all study participants. Both unadjusted and adjusted multivariate linear regressions were employed to analyze the association of the decrease of CD4+ T-cell counts with other characteristics. Results Two hundred and ninety-four HIV-1-infected FBDs and 59 age-matched HIV-seronegative local residents were enrolled in this study. The unregulated blood/plasma collection occurred more than a decade (10.8 -12.8 years) ago, which caused the rapid spread of HIV-1 infection and the high prevalence of co-infection with hepatitis C virus (HCV, 89.5%); hepatitis B virus (HBV) co-infection was observed in only 11 HIV+participants (3.7%). Deterioration in both clinical manifestation and laboratory parameters and increase of viral loads were observed in parallel with the decrease of CD4+ T-cell counts. The decrease of total lymphocyte counts (P<0.001) and hemoglobin levels (P<0.001) and the appearance of dermatosis (P=0.03) were observed in parallel with the decrease of CD4+ T-cell counts whereas viral loads (P<0.001) and CD8+ T-cell counts (P=0.01) were inverselyassociated with CD4+ T-cell counts. Conclusions Co-infection with HCV but not HBV is highly prevalent among HIV-1-infected FBDs. CD4+ T-cell counts is a reliable indicator for disease progression among FBDs. 5Comparison of the effects of aluminum and iron(Ⅲ)salts on ultrafiltration membrane biofouling in drinking water treatment显示文摘Coagulation plays an important role in alleviating membrane fouling, and a noticeable problem is the development of microorganisms after long-time operation, which gradually secrete extracellular polymeric substances(EPS). To date, few studies have paid attention to the behavior of microorganisms in drinking water treatment with ultrafiltration(UF)membranes. Herein, the membrane biofouling was investigated with different aluminum and iron salts. We found that Al_2(SO_4)_3·18 H_2O performed better in reducing membrane fouling due to the slower growth rate of microorganisms. In comparison to Al_2(SO_4)_3·18H_2O,more EPS were induced with Fe_2(SO_4)_3·x18H_2O, both in the membrane tank and the sludge on the cake layer. We also found that bacteria were the major microorganisms, of which the concentration was much higher than those of fungi and archaea. Further analyses showed that Proteobacteria was dominant in bacterial communities, which caused severe membrane fouling by forming a biofilm, especially for Fe_2(SO_4)_3·x18H_2O. Additionally, the abundances of Bacteroidetes and Verrucomicrobia were relatively higher in the presence of Al_2(SO_4)_3·18 H_2O,resulting in less severe biofouling by effectively degrading the protein and polysaccharide in EPS. 6Efficacy and safety of Xinglouchengqi decoction for acute ischemic stroke with constipation:study protocol for a randomized con-trolled trial显示文摘OBJECTIVE: To investigate the efficacy and safety of Xinglouchengqi(XLCQ) decoction in treatment of acute ischemic stroke with constipation.METHODS: In this prospective, multicenter, assessor-blinded, randomized controlled trial, 360 eligible participants will be randomized to the XLCQ group or the control group. Participants in the XLCQ group will receive XLCQ decoction, while those in the control group will undergo clysis therapy using glycerin enemas or oral administration of lactulose solution. Both groups will undergo a treatment period of(5 ± 2) d and a 1-month follow-up.The primary outcome measure will be the Constipation Scale score. The secondary outcome measures will include scores on the National Institutes of Health Stroke Scale, the Traditional Chinese Medicine(TCM) Stroke Scale, the Diagnostic Scale for TCM Syndromes of Ischemic Stroke and TCM Scale for Syndrome of Phlegm-heat and Fu-organ Excess.Therapeutic mechanism outcomes and safety outcomes will also be assessed. Assessments will be conducted at baseline, at the end of the treatment period, and at the follow-up. In this review,we update the studies on the BiCuSeO thin films first.Then, we focus on the most recent progress of multiple approaches that enhance the thermoelectric performance including advanced synthesized technologies,notable mechanisms for higher power factor(optimizing carrier concentration, carrier mobility, Seebeck coefficient)and doping effects predicted by calculation. 8Medical expenditure for esophageal cancer in China:a 10-year multicenter retrospective survey(2002-2011)显示文摘Background: Esophageal cancer is associated with substantial disease burden in China, and data on the economic burden are fundamental for setting priorities in cancer interventions. The medical expenditure for the diagnosis and treatment of esophageal cancer in China has not been fully quantified. This study aimed to examine the medical expenditure of Chinese patients with esophageal cancer and the associated trends.Methods: From 2012 to 2014, a hospital-based multicenter retrospective survey was conducted in 37 hospitals in 13 provinces/municipalities across China as a part of the Cancer Screening Program of Urban China. For each esophageal cancer patient diagnosed between 2002 and 2011, clinical information and expense data were extracted by using structured questionnaires. All expense data were reported in Chinese Yuan(CNY; 1 CNY = 0.155 USD) based on the2011 value and inflated using the year-specific health care consumer price index for China.Results: A total of 14,967 esophageal cancer patients were included in the analysis. It was estimated that the overall average expenditure per patient was 38,666 CNY, and an average annual increase of 6.27% was observed from 2002(25,111 CNY) to 2011(46,124 CNY). The average expenditures were 34,460 CNY for stage Ⅰ,39,302 CNY for stage Ⅱ,40,353 CNY for stage Ⅲ, and 37,432 CNY for stage IV diseases(P < 0.01). 11The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. 12A highly potent and stable pan-coronavirus fusion inhibitor as a candidate prophylactic and therapeutic for COVID-19 and other coronavirus diseases显示文摘The development of broad-spectrum antivirals against human coronaviruses(HCoVs)is critical to combat the current coronavirus disease 2019(COVID-19)pandemic caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)and its variants,as well as future outbreaks of emerging CoVs.We have previously identified a polyethylene glycol-conjugated(PEGylated)lipopeptide,EK1C4,with potent pan-CoV fusion inhibitory activity.However,PEG linkers in peptide or protein drugs may reduce stability or induce anti-PEG antibodies in vivo.Therefore,we herein report the design and synthesis of a series of dePEGylated lipopeptide-based pan-CoV fusion inhibitors featuring the replacement of the PEG linker with amino acids in the heptad repeat 2 C-terminal fragment(HR2-CF)of HCoV-OC43.Among these lipopeptides,EKL1C showed the most potent inhibitory activity against infection by SARS-CoV-2 and its spike(S)mutants,as well as other HCoVs and some bat SARS-related coronaviruses(SARSr-CoVs)tested.The dePEGylated lipopeptide EKL1C exhibited significantly stronger resistance to proteolytic enzymes,better metabolic stability in mouse serum,higher thermostability than the PEGylated lipopeptide EK1C4,suggesting that EKL1C could be further developed as a candidate prophylactic and therapeutic for COVID-19 and other coronavirus diseases.Jie Zhou Wei Xu Zezhong Liu Chao Wang Shuai Xia Qiaoshuai Lan Yanxing Cai Shan Su Jing Pu Lixiao Xing Youhua Xie Lu Lu Shibo Jiang Qian Wang 2022Acta Pharmaceutica Sinica B2022,12,4:5
13Evolution of urban black and odorous water:The characteristics of microbial community and driving-factors显示文摘Urban black blooms that are primarily caused by organic carbon are deleterious environmental problems.However,detailed studies on the microbial characteristics that form urban black blooms are lacking.In this study,we observed the composition,diversity,and function of bacterial community in the overlying water and sediments during the occurrence and remediation of urban black blooms using high-throughput 16S rRNA gene amplicon sequencing analysis.First,we found that pivotal consortia in the overlying water increased significantly during the formation of black blooms,including the genera Acidovorax,Brevundimonas,Pusillimonas,and Burkholderiales involved in the degradation of refractory organics,as well as the genera Desulfovibrio,Dechloromonas,and Rhizobium related to the production of black and odorous substances.An RDA analysis revealed that chemical oxygen demand,dissolved oxygen,and oxidation reduction potential were related to the changes in microbial community composition.Furthermore,aeration was found to accelerate the removal of ammonia nitrogen and enhance the function of microbial community by stimulating the growth of order Planktomycetes during the remediation of black blooms,but aeration substantially damaged the microbial diversity and richness.Therefore,the health of the aquatic ecosystem should be comprehensively considered when aeration is applied to restore polluted waterbodies.Notably,we observed a large number of pathogenic bacteria in urban black blooms,which emphasizes the importance of treating domestic sewage so that it is harmless.Together,these findings provide new insights and a basis to prevent and manage urban black blooms.Dan Zhang Huilan Yang Shuhuan Lan Chen Wang Xudong Li Yunxiao Xing Hua Yue Qiulin Li Ling Wang Yifei Xie 2022Journal of Environmental Sciences2022,34,2:4
14Efficacy and safety of anti-PD-1/anti-PD-L1 antibody therapy in treatment of advanced gastric cancer or gastroesophageal junction cancer: A meta-analysis显示文摘BACKGROUND Faced with limited and inadequate treatment options for patients with advanced gastric cancer or gastroesophageal junction cancer(GC/GEJC), researchers have turned toward, with the support of promising clinical trials, anti-PD-1/anti-PD-L1 antibody therapy. But there are also different clinical trial results. To better assess its efficacy and safety, we integrated data from 13 eligible studies for a systematic review and meta-analysis.AIM To comprehensively evaluate the efficacy and safety of anti-PD-1/anti-PD-L1 antibody therapy in the treatment of advanced GC/GEJC patients.METHODS PubMed, Web of Science, Cochrane Library,and EMBASE databases were searched to identify eligible articles with outcomes including objective response rate(ORR), disease control rate(DCR), overall survival(OS), progression-free survival(PFS), and adverse events(AEs) of anti-PD-1/anti-PD-L1 antibody therapy.RESULTS Our study encompassed a total of 13 trials totaling 1618 patients. The outcomes showed a pooled ORR and DCR of 15%(95% confidence interval [CI]: 14%-18%) and 40%(95%CI: 33%-46%), respectively. The pooled 6-mo OS and PFS were 54%(95%CI: 45%-64%) and 26%(95%CI: 20%-32%), respectively, and the 12-mo OS and PFS were 42%(95%CI: 21%-62%) and 11%(95%CI: 8%-13%), respectively. In addition, the incidence of any-grade AEs and grade ≥ 3 AEs was 64%(95%CI: 54%-73%) and 18%(95%CI: 16%-20%), respectively. Most importantly, PD-L1 positive patients exhibited a higher ORR rate than PD-L1 negative patients(odds ratio = 2.54, 95%CI: 1.56-4.15).CONCLUSION Anti-PD-1/anti-PD-L1 antibody therapy has shown promising anti-tumor efficacy with manageable AEs in advanced GC/GEJC patients, with PD-L1 overexpressing patients exhibiting a higher ORR. 15Effect of Shenqi Yangxin decoction on high mobility group box 1and inflammatory signal pathway in a rat model of dilated cardiomyopathy显示文摘OBJECTIVE: To investigate the effects of Shenqi Yangxin decoction(SQYXD) on heart function in a rat model of dilated cardiomyopathy(DCM) and its potential mechanisms.METHODS: Sprague-Dawley rats were randomly divided into normal(10 rats) and DCM(150 rats)groups. DCM was induced by an intraperitoneal injection of adriamycin. Then, DCM baseline group was randomly selected sixteen DCM rats. The remaining DCM rats were randomly divided into DCM control, perindopril, metoprolol, and SQYXD groups. Cardiac function and histological analysis plus biochemical measurement of serum levels of brain natriuretic peptide(BNP), and inflammatory factors were measured. The mRNA and protein expression levels of high-mobility group box 1(HMGB1), Toll-like receptor 4(TLR-4), receptor for advanced glycation end products(RAGE), and nuclear factor-κB(NF-κB) were determined. Myocardial metabolism imaging was performed on the normal, SQYXD and DCM control groups to evaluate the effectiveness of treatments.RESULTS: Rats in the DCM control group exhibited dilated left ventricular diameter, impaired cardiac function, disorganized sarcomere, impaired glucose metabolism, increased heart weight index,and increased levels of BNP, which were improved by treatment with SQYXD. In addition, hearts from rats in the DCM baseline group exhibited significantly higher levels of HMGB1, TLR-4, RAGE, NF-κB,tumor necrosis factor-α, interleukin-1, interleukin-6, interleukin-10, compared with the normal group. Interestingly, the mRNA level of HMGB1 in the DCM baseline group was positively correlated with that of TLR-4, RAGE, NF-κB, BNP, and LVEDD,but negatively correlated with LVEF. SQYXD inhibited the upregulation of HMGB1 expression and its downstream inflammatory factors.CONCLUSION: Shenqi Yangxin decoction effectively reduced the dilated left ventricular diameter and improved heart function in dilated cardiomyopathy. 16The effects of hydrogen peroxide pre-oxidation on ultrafiltration membrane biofouling alleviation in drinking water treatment显示文摘Pre-oxidation is widely used to reduce ultrafiltration membrane fouling.However,the variation in the composition of microbial communities and extracellular polymeric substances(EPSs) accompanying pre-oxidation in drinking water treatment has received little attention.In this study,hydrogen peroxide(H_2O_2) was used in a coagulationultrafiltration process with Al_2(SO_4)_3·18H_2O.A long-term reactor experiment(60 d) showed that pre-oxidation alleviated membrane fouling,mainly due to its inhibition of microbial growth,as observed by flow cytometry measurements of the membrane tank water.Further analysis of the formed cake layer demonstrated that the corresponding levels of EPS released from the microbes were lower with than without H_2O_2 treatment.In comparison to polysaccharides,proteins dominated the EPS.2 D-electrophoresis showed little difference(p > 0.05,Student's t-test) in the composition of proteins in the cake layer between the treatments with and without H_2O_2.The molecular weights of proteins ranged from approximately 30–50 kDa and the majority of isoelectric points ranged from 6 to 8.Highthroughput sequencing showed that the predominant bacteria were Proteobacteria,Bacteroidetes,and Verrucomicrobia in both cake layers.However,the relative abundance of Planctomycetes was higher in the cake layer with H_2O_2 pre-oxidation,which was likely probably due to the strong oxidative resistance of its cell wall.Overall,our findings clarify the fundamental molecular mechanism in H_2O_2 pre-oxidation for ultrafiltration membrane bio-fouling alleviation in drinking water treatment.Xing Wang Baiwen Ma Yaohui Bai Huachun Lan Huijuan Liu Jiuhui Qu 2018Journal of Environmental Sciences2018,30,11:4
17Multidecadal Trends in Large-Scale Annual Mean SATa Based on CMIP5 Historical Simulations and Future Projections显示文摘基于观测和第五次耦合模式比较计划(CMIP5)模式的模拟结果,本文对全球、半球、半球陆地及海洋尺度的年平均地面气温异常在过去一百多年及两个代表性浓度路径(RCPs)情景下的多年代际变化及趋势进行了评估分析。根据模式对全球平均地面气温异常的时间变率、长期趋势、多年代际变化及趋势的模拟能力,筛选出15个模式进行分析。观测结果表明,北半球陆地、北半球海洋和南半球海洋平均地面气温异常与全球平均地面气温异常具有相似的多年代际变化特征:在1900—1944年及1971—2000年呈现增暖趋势,并在1945—1970年和2001—2013年呈现增暖停滞甚至变冷趋势。模式能够基本再现以上观测特征。然而,与以上变化不同的是,南半球陆地的平均地面气温在1945—1970年呈现增暖趋势,并且模式不能很好模拟该特征。对于近期的增暖停滞阶段(2001—2013年),BCC-CSM1-1-m模式、CMCC-CM模式、GFDL-ESM2M模式及NorESM1-ME模式在RCP4.5和RCP8.5情景下预估的全球及半球尺度的地面气温异常趋势值最接近观测值,表明它们具有较好的预估能力。由于这四个模式在地面气温异常的多年代际趋势上具有较好的模拟及预估性能,故选择它们来预估2006—2099年的地面气温异常在全球及半球尺度上的变化特征。结果显示在RCP4.5(RCP8.5)情景下,所选四个模式集合平均的全球、北半球及南半球年平均地面气温异常趋势值分别为0.17(0.29)、0.22(0.36)及0.11(0.23)℃·decade^(–1),其趋势值明显小于未经过模式筛选的CMIP5模式集合的结果。Nan Xing Jianping Li Lanning Wang 2017Engineering2017,3,1:4
18Insight into the interaction between Ni-rich LiNi0.8Co0.1Mn0.1O2 cathode and BF4--introducing electrolyte at 4.5V high voltage显示文摘Owing to the high specific capacity and high voltage,Ni-rich(LiNi0.8Co0.1Mn0.1O2,LNCM811)cathode has been considered as one of the most promising candidate cathode materials for next generation lithium ion batteries,whereas severe capacity fading greatly hinders its practical application.Notably,the compatibility of Ni-rich materials with LiBF4-containing electrolyte has not yet been realized.Herein,1 M LiPF6-based electrolyte with introducing 2 M LiBF4 is proposed to dramatically improve the cyclic stability of high voltage LNCM811/Li half-cell.Addition of high concentrated LiBF4 improves the moisture stability of electrolyte,which hinders the generation of harmful by-product HF,resulting in improved interfacial stability of LNCM811.Lithium plating/stripping reaction of Li/Li symmetric cell confirms that the enhanced cyclic stability is ascribed to the improved interfacial stability of LNCM811 instead of lithium electrode.Morphology and composition characterization results reveal that LiBF4 participates in the CEI film-forming reaction,resulting in suppressed oxidation of electrolyte and interfacial structural destruction of LNCM811.Guangyuan Lan Hebing Zhou Lidan Xing Jiawei Chen Zifei Li Rude Guo Yanxia Che Weishan Li 2019Journal of Energy Chemistry2019,28,12:3
