3Targeted Mutagenesis of NAC Transcription Factor Gene,OsN AC041,Leading to Salt Sensitivity in Rice显示文摘Salinity is a major abiotic stress factor that seriously affects plant growth. Many genes are involved in the response to salt stress with various metabolism pathways. A number of plant transcription factor family genes have been found to be involved in the salt stress response, and NAM, ATAF and CUC(NAC) transcription factors are thought to act as active regulators during abiotic stress, especially salt stress. In this study, we detected a rice NAC transcription factor coding gene, OsNAC041, and confirmed that it influenced the germination of seeds under salt stress and salt tolerance of plants. OsNAC041 was primarily expressed in the leaves and located in the nucleus. Furthermore, the CRISPR/Cas9 method was used to obtain a targeted osnac041 mutant, of which the plant height was higher than that of the wild-type, showing increased salt sensitivity. Moreover, RNA-seq analysis revealed a number of differentially expressed genes(DEGs) involved in several important signaling pathways in the osnac041 mutant. Subsequently, Kyoto Encyclopedia of Genes and Genomes annotation also revealed differential expression of DEGs associated with mitogen-activated protein kinase signaling, peroxisome, eukaryotictype ABC transporters, photosynthesis and plant hormones, which are involved in stress-related signaling pathways. Overall, our study suggested that OsNAC041 was involved in the salt stress response in rice. 4Kindlin-2 expression in adult tissues correlates with their embryonic origins显示文摘Kindlin-2 functions in the maintenance of homeostasis and in human diseases.This study investigated the interrelationship between Kindlin-2 expression in tissues and the corresponding germ layers from which these tissues originated.Kindlin-2 expression was examined in normal adult human organs and human cancer tissues by immunohistochemical analyses.Analysis of Kindlin-2 mRNA levels in adult human organs in the Oncomine dataset revealed Kindlin-2 is highly expressed in mesoderm-derived organs.However,Kindlin-2 was negative or weakly expressed in endoderm/ectoderm-derived organs.Interestingly,the abnormal expression of Kindlin-2 was observed in a variety of human cancers.In agreement with its expression profile in humans,Kindlin-2 was also highly expressed in mesoderm-derived organs in mouse embryos with the exception of strong Kindlin-2 expression in ectoderm-derived spinal cord and ganglia,tissues that are highly mobile during embryonic development.Importantly,we demonstrated the expression level of Kindlin-2 in adult organs correlated with their embryonic dermal origins and deregulation of Kindlin-2 in tissues is associated with tumor progression.This finding will help us understand the dual role of Kindlin-2 in the regulation of tumor progression and embryonic development.ZHAN Jun YANG Mei CHI XiaoChun ZHANG Jing PEI XueLian REN CaiXia GUO YongQing LIU Wei ZHANG HongQuan 2014Science China(Life Sciences)2014,57,7:8

6Dendritic BiVO4 decorated with MnOx co-catalyst as an efficient hierarchical catalyst for photocatalytic ozonation显示文摘An appropriate co-catalyst can significantly promote the photocatalytic efficacy, but this has been seldom studied in the visible-light photocatalysis combined with ozone, namely photocatalytic ozonation. In this work, a dendritic bismuth vanadium tetraoxide (BiVO4) material composited with highly dispersed MnOv nanoparticles was synthesized, and its catalytic activity is 86.6% higher than bare BiVO4 in a visible light and ozone combined process. Catalytic ozonation experiments, ultraviolet- visible (UV-Vis) diffuse reflectance spectra and photoluminescence spectra jointly indicate that MnOA plays a triple role in this process. MnOv strengthens the light adsorption and promotes the charge separation on the composite material, and it also shows good activity in catalytic ozonation. The key reactive species in this process is OH, and various pathways for its generation in this process is proposed. 11Kruppel-like factor 10 protects against acute viral myocarditis by negatively regulating cardiac MCP-1 expression显示文摘Viral myocarditis(VMC)is a cardiac disease associated with myocardial inflammation and injury induced by virus infection.Cardiomyocytes have recently been regarded as key players in eliciting and modulating inflammation within the myocardium.Kruppel-like factor 10(KLF10)is a crucial regulator of various pathological processes and plays different roles in a variety of diseases.However,its role in VMC induced by coxsackievirus B3(CVB3)infection remains unknown.In this study,we report that cardiac KLF10 confers enhanced protection against viral myocarditis.We found that KLF10 expression was downregulated upon CVB3 infection.KLF10 deficiency enhanced cardiac viral replication and aggravated VMC progress.Bone marrow chimera experiments indicated that KLF10 expression in nonhematopoietic cells was involved in the pathogenesis of VMC.We further identified MCP-1 as a novel target of KLF10 in cardiomyocytes,and KLF10 cooperated with histone deacetylase 1(HDAC1)to negatively regulate MCP-1 expression by binding its promoter,leading to activation of MCP-1 transcription and recruitment of Ly6C^(high) monocytes/macrophages into the myocardium.This novel mechanism of MCP-1 regulation by KLF10 might provide new insights into the pathogenesis of VMC and a potential therapeutic target for VMC.Jie Yang Hongkai Zhang Xuelian Wang Jing Guo Lin Wei Yahui Song Yuan Luo YinXia Zhao Malayannan Subramaniam Thomas C.Spelsberg Lie Wang Wei Xu Min Li 2021Cellular & Molecular Immunology2021,18,9:3
12The hist one varia nt SI_H2A.Z regulates carote noid biosynthesis and gene expression during tomato fruit ripening显示文摘The conserved histone variant H2A.Z is essential for transcriptional regulation;defense responses;and various biological processes in plants,such as growth,development,and flowering.However,little is known about how H2A.Z affects the developmental process and ripening of tomato fruits.Here,we utilized the CRISPR/Cas9 gene-editing system to generate a sl_hta9 sl_hta11 double-mutant,designated sl_h2az,and found that these two mutations led to a signi fi cant reduction in the fresh weight of tomato fruits.Subsequent messenger RNA(mRNA)-seq results showed that dysfunction of SI_H2A.Z has profound effects on the reprogramming of genome-wide gene expression at different developmental stages of tomato fruits,indicating a ripening-dependent correlation between SI_H2A.Z and gene expression regulation in tomato fruits.In addition,the expression of three genes,SIPSY1,SlPDS,and SlVDE,encoding the key enzymes in the biosynthesis pathway of carotenoids,was signi fi cantly upregulated in the later ripening stages,which was consistent with the increased contents of carotenoids in sl_h2a.z double-mutant fruits.Overall,our study reveals a role of SI_H2A.Z in the regulation of carotenoids and provides a resource for the study of SI_H2A.Z-dependent gene expression regulation.Hence,our results provide a link between epigenetic regulation via histone variants and fruit development,suggesting a conceptual framework to understand how histone variants regulate tomato fruit quality.Xuedong Yang Xuelian Zhang Youxin Yang Hui Zhang Weimin Zhu Wen-Feng Nie 2021Horticulture Research2021,8,1:2

14Dietary calcium requirements of broilers fed a conventional corn-soybean meal diet from 1 to 21 days of age显示文摘Background:The current calcium(Ca)recommendation for broilers is primarily based on studies conducted more than 30 years ago with birds of markedly different productive potentials from those which exist today.And the response indicators in these studies are mainly growth performance and bone ash percentage.Therefore,the present study was carried out to investigate the effect of dietary Ca level on growth performance,serum parameters,bone characteristics and Ca metabolism-related gene expressions,so as to estimate dietary Ca requirements of broilers fed a conventional corn-soybean meal diet from 1 to 21 days of age.Methods:A total of 4201-day-old Arbor Acres male broilers were randomly assigned to 1 of 7 treatments with 6 replicates(10 birds per cage)and fed the corn-soybean meal diets containing 0.60%,0.70%,0.80%,0.90%,1.00%,1.10%or 1.20%Ca for 21 days.Each diet contained the constant non-phytate phosphorus content of about 0.39%.Results:The average daily gain decreased linearly(P<0.001)as dietary Ca level increased.The serum and tibia alkaline phosphatase(ALP)activities,tibia bone mineral density(BMD),middle toe BMD,tibia ash percentage,tibia breaking strength,and tibia ALP protein expression level were affected(P<0.05)by dietary Ca level,and showed significant quadratic responses(P<0.02)to dietary Ca levels.The estimates of dietary Ca requirements were 0.80 to 1.00%based on the best fitted broken-line or quadratic models(P<0.03)of the above serum and bone parameters,respectively.Conclusions:The results from the present study indicate that the Ca requirements would be about 0.60%to obtain the best growth rate,and 1.00%to meet all of the Ca metabolisms and bone development of broilers fed a conventional corn-soybean meal diet from 1 to 21 days of age.Shiping Bai Yunfeng Yang Xuelian Ma Xiudong Liao Runlian Wang Liyang Zhang Sufen Li Xugang Luo Lin Lu 2022Journal of Animal Science and Biotechnology2022,13,4:2

19Multiscale structures of Zr-based binary metallic glasses and the correlation with glass forming ability显示文摘Thermal behaviors and structures of three Zr-based binary glass formers, Zr_(50)Cu_(50), Zr_(64)Cu_(36) and Zr_(64)Ni_(36), were investigated and compared using differential scanning calorimetry(DSC), transmission electron microscopy(TEM), high energy X-ray diffraction(XRD) and small angle X-ray scattering(SAXS). The high energy XRD results show that the bulk glass former Zr_(50)Cu_(50) has a denser atomic packing efficiency and reduced mediumrange order than those of marginal glass formers Zr_(64)Cu_(36) and Zr_(64)Ni_(36). Based on TEM observations for the samples after heat treatment at 10 K above their crystallization onset temperatures, the number density of crystals for Zr_(50)Cu_(50) was estimated to be 10^(23)-10^(24) m^(-3), which was four-orders higher than that in Zr_(64)Cu_(36)and Zr_(64)Ni_(36) metallic glasses. SAXS results indicate that Zr_(50)Cu_(50) has higher degree of nanoscale inhomogeneities than those in Zr_(64)Cu_(36) and Zr_(64)Ni_(36) at as-cast state.
