1Ablation of gut microbiota alleviates obesity-induced hepatic steatosis and glucose intolerance by modulating bile acid metabolism in hamsters显示文摘Since metabolic process differs between humans and mice, studies were performed in hamsters, which are generally considered to be a more appropriate animal model for studies of obesityrelated metabolic disorders. The modulation of gut microbiota, bile acids and the farnesoid X receptor(FXR) axis is correlated with obesity-induced insulin resistance and hepatic steatosis in mice. However,the interactions among the gut microbiota, bile acids and FXR in metabolic disorders remained largely unexplored in hamsters. In the current study, hamsters fed a 60% high-fat diet(HFD) were administeredvehicle or an antibiotic cocktail by gavage twice a week for four weeks. Antibiotic treatment alleviated HFD-induced glucose intolerance, hepatic steatosis and inflammation accompanied with decreased hepatic lipogenesis and elevated thermogenesis in subcutaneous white adipose tissue(sWAT). In the livers of antibiotic-treated hamsters, cytochrome P450 family 7 subfamily B member 1(CYP7 B1) in the alternative bile acid synthesis pathway was upregulated, contributing to a more hydrophilic bile acid profile with increased tauro-β-muricholic acid(TβMCA). The intestinal FXR signaling was suppressed but remained unchanged in the liver. 4Hierarchically electrospraying a PLGA@chitosan sphere-in-sphere composite microsphere for multi-drug-controlled release显示文摘Sequential administration and controlled release of different drugs are of vital importance for regulating cellular behaviors and tissue regeneration,which usually demands appropriate carriers like microspheres(MS)to control drugs releases.Electrospray has been proven an effective technique to prepare MS with uniform particle size and high drug-loading rate.In this study,we applied electrospray to simply and hierarchically fabricate sphere-in-sphere composite microspheres,with smaller poly(lactic-co-glycolic acid)MS(~8-10 lm in diameter)embedded in a larger chitosan MS(~250-300 lm in diameter).The scanning electron microscopy images revealed highly uniform MS that can be accurately controlled by adjusting the nozzle diameter or voltage.Two kinds of model drugs,bovine serum albumin and chlorhexidine acetate,were encapsulated in the microspheres.The fluorescence-labeled rhodamine-fluoresceine isothiocyanate(Rho-FITC)and ultraviolet(UV)spectrophotometry results suggested that loaded drugs got excellent distribution in microspheres,as well as sustained,slow release in vitro.In addition,far-UV circular dichroism and matrixassisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF-MS)results indicated original secondary structure and molecular weight of drugs after electrospraying.Generally speaking,our research proposed a modified hierarchically electrospraying technique to prepare sphere-in-sphere composite MS with two different drugs loaded,which could be applied in sequential,multi-modality therapy.Zhu Liu Weilong Ye Jingchuan Zheng Qindong Wang Guowu Ma Huiying Liu Xiumei Wang 2020Regenerative Biomaterials2020,7,4:5
5Estimating the quality of stripe in structured light 3D measurement显示文摘Affected by noise, light blocking, color and shape of object, the quality of captured stripes in structured light three dimensional(3 D) measurement system is degenerated. As the quality of captured stripes is one of the key factors for measurement accuracy, some large error data is introduced into the measurement results which can only be recognized artificially with prior knowledge of the object to be measured. In this paper, a method is proposed to estimate the quality of stripe image. In the method, two parameters, skewness coefficient of stripe gray distribution and the noise level, are used to estimate the quality of stripe. The simulation results show that the bigger the skewness coefficient is, the bigger the error of stripe locating results is. Meanwhile, the smaller the noise level is, the smaller the error of stripe locating results is. The method has been used to estimate the experimental image, and the same conclusion can be obtained. 11Microbiome-metabolome analysis reveals cervical lesion alterations显示文摘Cervical cancer(CC)continues to be one of the most common cancers among females worldwide.It takes a few years or even decades for CC to arise in a minority of women with cervical precancers.An increasing corpus of studies today indicates that local microecology and carcinogenesis are intimately related.To investigate the changes in cericovaginal microecology with the development of cervical cancer,we performed 16S rDNA sequencing and metabolomic analysis in cericovaginal fluid from 10 LSIL patients,10 HSIL patients,10 CC patients and 10 healthy controls to reveal the differential flora and metabolites during cervical carcinogenesis.Carcinogenesis is associated with alterations in microbiome diversity,individual taxa,and functions with notable changes in Lactobacillus,Prevotella and Aquabacterium,as well as in cervicovaginal metabolites that correlate with cervicovaginal microbial patterns.Increased bacterial diversity and a decline in the relative abundance of Lactobacillus,the dominant species in the cericovaginal flora,are observed when cervical lesions advance.According to KEGG pathway enrichment analysis,lipids and organic acids change as cervical cancer progresses,and the phenylalanine,tyrosine,and tryptophan biosynthesis pathway is essential for the development of cervical cancer.Our results reveal that microbic and metabolomic profiling is capable of distinguishing CC from precancer and highlights potential biomarkers for the early detection of cervical dysplasia.These differential microorganisms and metabolites are expected to become a potential tool to assist in the diagnosis of cervical cancer.Hanjie Xu Lou Liu Feng Xu Min Liu Yuexiao Song Jiale Chen Huiying Zhan Ye Zhang Dexiang Xu Yu Chen Mudan Lu Daozhen Chen 2022Acta Biochimica et Biophysica Sinica2022,54,10:0
12Fructose-1,6-bisphosphatase 1 dephosphorylates IκBα and suppresses colorectal tumorigenesis显示文摘Emerging evidence demonstrates that some metabolic enzymes that phosphorylate soluble metabolites can also phosphorylate a variety of protein substrates as protein kinases to regulate cell cycle,apoptosis and many other fundamental cellular processes.However,whether a metabolic enzyme dephosphorylates protein as a protein phosphatase remains unknown.Here we reveal the gluconeogenic enzyme fructose 1,6-biphosphatase 1(FBP1)that catalyzes the hydrolysis of fructose 1,6-bisphosphate(F-1,6-BP)to fructose 6-phosphate(F-6-P)as a protein phosphatase by performing a high-throughput screening of metabolic phosphatases with molecular docking followed by molecular dynamics(MD)simulations.Moreover,we identify IκBαas the substrate of FBP1-mediated dephosphorylation by performing phosphoproteomic analysis.Mechanistically,FBP1 directly interacts with and dephosphorylates the serine(S)32/36 of IκBαupon TNFαstimulation,thereby inhibiting NF-κB activation.MD simulations indicate that the catalytic mechanism of FBP1-mediated IκBαdephosphorylation is similar to F-1,6-BP dephosphorylation,except for higher energetic barriers for IκBαdephosphorylation.Functionally,FBP1-dependent NF-κB inactivation suppresses colorectal tumorigenesis by sensitizing tumor cells to inflammatory stresses and preventing the mobilization of myeloid-derived suppressor cells.Our finding reveals a previously unrecognized role of FBP1 as a protein phosphatase and establishes the critical role of FBP1-mediated IκBαdephosphorylation in colorectal tumorigenesis.Wencheng Zhu Huiying Chu Yajuan Zhang Tianhang Luo Hua Yu Hongwen Zhu Ye Liu Hong Gao Yun Zhao Quanlin Li Xiongjun Wang Guohui Li Weiwei Yang 2023Cell Research2023,33,3:0
13Two-dimensional SnS2 nanosheets on Prussian blue template for high performance sodium ion batteries显示文摘Three-dimensional Prussian blue (PB) nanostructures was obtained via a one-step hydrothermal method. Subsequently, two-dimensional tin disulfide (SnS2) nanosheets were grown onto PB through a facile hydrothermal synthesis. The as prepared SnS2/PB is further employed as the anode of sodium ion batteries (SIBs). S11S2/PB nanoarchitecture delivers a specific capacity of 725.7 mAh·g-1 at 50 mA·g-1. When put through more than 200 cycles, it achieved a stable cycling capacity of 400 m Ah·g^-1 at 200 m A·g^i. The stable Na^+ storage properties of SnS2/PB was attributed to the synergistic effect among the conductive PB carbon, used as the template in this work. These results obtained potentially paves the way for the development of excellent electrochemical performance with stable performance of SIBs.Glenn J.
