4Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 6Silibinin ameliorates hepatic lipid accumulation and oxidative stress in mice with non-alcoholic steatohepatitis by regulating CFLAR-JNK pathway显示文摘Non-alcoholic steatohepatitis(NASH) is a chronic metabolic syndrome and the CFLAR-JNK pathway can reverse the process of NASH. Although silibinin is used for the treatment of NASH in clinical, its effect on CFLAR-JNK pathway in NASH remains unclear. This study aimed to investigate the effect of silibinin on CFLAR-JNK pathway in NASH models both in vivo and in vitro. The in vivo study was performed using male C57 BL/6 mice fed with methionine– choline-deficient diet and simultaneously treated with silibinin for 6 weeks. The in vitro study was performed by using mouse NCTC-1469 cells which were respectively pretreated with oleic acid plus palmitic acid, and adenovirus-down Cflar for 24 h,then treated with silibinin for 24 h. After the drug treatment, the key indicators involved in CFLAR-JNK pathway including hepatic injury, lipid metabolism and oxidative stress were determined. Silibininsignificantly activated CFLAR and inhibited the phosphorylation of JNK, up-regulated the mRNA expression of Pparα, Fabp5, Cpt1α, Acox, Scd-1, Gpat and Mttp, reduced the activities of serum ALT and AST and the contents of hepatic TG, TC and MDA, increased the expression of NRF2 and the activities of CAT, GSH-Px and HO-1, and decreased the activities and expression of CYP2 E1 and CYP4 A in vivo.These effects were confirmed by the in vitro experiments. 7Measuring Conflict Functions in Generalized Power Space显示文摘One of the most important open issues is that the classical conflict coefficient in D-S evidence theory (DST) cannot correctly determine the conflict degree between two pieces of evidence. This drawback greatly limits the use of DST in real application systems. Early researches mainly focused on the improvement of Dempster’s rule of combination (DRC). However, the current research shows it is very important to define new conflict coefficients to determine the conflict degree between two or more pieces of evidence. The evidential sources of information are considered in this work and the definition of a conflict measure function (CMF) is proposed for selecting some useful CMFs in the next fusion work when sources are available at each instant. Firstly, the definition and theorems of CMF are put forward. Secondly, some typical CMFs are extended and then new CMFs are put forward. While most ingested foreign bodies usually pass through the gastrointestinal tract uneventfully, sharp foreign bodies such as toothpicks infrequently cause intestinal perforation and may even result in death. 11Scalable synthesis of nanoporous silicon microparticles for highly cyclable lithium-ion batteries显示文摘Nanoporous silicon is a promising anode material for high energy density batteries due to its high cycling stability and high tap density compared to other nanostructured anode materials.However,the high cost of synthesis and low yield of nanoporous silicon limit its practical application.Here,we develop a scalable,low-cost top-down process of controlled oxidation of Mg2Si in the air,followed by HCl removal of MgO to generate nanoporous silicon without the use of HF.By controlling the synthesis conditions,the oxygen content,grain size and yield of the porous silicon are simultaneously optimized from commercial standpoints.In situ environmental transmission electron microscopy reveals the reaction mechanism;the Mg2Si microparticle reacts with O2 to form MgO and Si,while preventing SiO2 formation.Owing to the low oxygen content and microscale secondary structure,the nanoporous silicon delivers a higher initial reversible capacity and initial Coulombic efficiency compared to commercial Si nanoparticles(3,033 mAh/g vs.2,418 mAh/g,84.3%vs.73.1%).Synthesis is highly scalable,and a yield of 90.4%is achieved for the porous Si nanostructure with the capability to make an excess of 10 g per batch.Our synthetic nanoporous silicon is promising for practical applications in next generation lithium-ion batteries.Jiangyan Wang William Huang Yong Seok Kim You Kyeong Jeong Sang Cheol Kim Jeffrey Heo Hiang Kwee Lee Bofei Liu Jaehou Nah Yi Cui 2020Nano Research2020,13,6:9
12Oral microbiota in human systematic diseases显示文摘Oral bacteria directly affect the disease status of dental caries and periodontal diseases. The dynamic oral microbiota cooperates with the host to reflect the information and status of immunity and metabolism through two-way communication along the oral cavity and the systemic organs. The oral cavity is one of the most important interaction windows between the human body and the environment.The microenvironment at different sites in the oral cavity has different microbial compositions and is regulated by complex signaling,hosts, and external environmental factors. These processes may affect or reflect human health because certain health states seem to be related to the composition of oral bacteria, and the destruction of the microbial community is related to systemic diseases. In this review, we discussed emerging and exciting evidence of complex and important connections between the oral microbes and multiple human systemic diseases, and the possible contribution of the oral microorganisms to systemic diseases. This review aims to enhance the interest to oral microbes on the whole human body, and also improve clinician’s understanding of the role of oral microbes in systemic diseases. 13Assessment of prospective hazards resulting from the 2017 earthquake at the world heritage site Jiuzhaigou Valley, Sichuan, China显示文摘On August 8, 2017, a Ms = 7.0 magnitude earthquake occurred in the Jiuzhaigou Valley, in Sichuan Province, China(N: 33.20°, E: 103.82°). Jiuzhaigou Valley is an area recognized and listed as a world heritage site by UNESCO in 1992. Data analysis and field survey were conducted on the landslide, collapse, and debris flow gully, to assess the coseismic geological hazards generated by the earthquake using an unmanned aerial vehicle(UAV), remote-sensing imaging, laser range finders, geological radars, and cameras. The results highlighted the occurrence of 13 landslides, 70 collapses, and 25 potential debris flow gullies following the earthquake. The hazards were classified on the basis of their size and the potential property loss attributable to them. Consequently, 14 large-scale hazards, 30 medium-sized hazards, and 64 small hazards accounting for 13%, 28%, and 59% of the total hazards, respectively, were identified. Based on the variation tendency of the geological hazards that ensued in areas affected by the Kanto earthquake(Japan), Chi-chi earthquake(Taiwan China), and Wenchuan earthquake(Sichuan China), the study predicts that, depending on the rain intensity cycle, the duration of geological hazard activities in the Jiuzhaigou Valley may last over ten years and will gradually decrease for the following five to ten yearsbefore returning to pre-earthquake levels. 14Spatial distribution of heavy metals(Cu, Pb, Zn, and Cd)in sediments of a coastal wetlands in eastern Fujian, China显示文摘We investigated the spatial distribution(horizontal and vertical concentrations) of copper(Cu),lead(Pb), zinc(Zn), and cadmium(Cd) in five wetland types(mudflat, aquaculture wetland, water area, farmland wetland and mangrove) from three areas(Ningde, Fuding,and Xiapu), China. Cu concentrations in five wetland types descended in the order: farm wetland, mudflat, aquaculture,water area and mangrove. Pb concentrations decreased in the order: aquaculture, mangrove, farm wetland, mudflat,and water area. Zn content decreased in the order: farm wetland, water area, aquaculture, mudflat and mangrove,and Cd content decreased as follows: mangrove, aquaculture, water area, mudflat, and farm wetland. Comparison of the concentrations of the same heavy metals in different areas showed that the highest Cu(63.75 mg kg^-1) and Zn(152.32 mg kg^-1) concentrations occurred in Ningde coastal wetlands; Pb(110.58 mg kg^-1) and Cd(2.81 mg kg^-1) contents were highest in Fuding wetlands,and the average contents of all heavy metals were very low in Xiapu wetlands. Examination of the vertical distribution showed that the Cu content was high in all mudflat layers;Pb and Cd concentrations were highest in aquaculture and mangrove wetlands, respectively, and Zn content was highest in farm wetlands. The spatial distribution of Cu and Zn contents for different areas decreased as follows:Ningde [ Fuding [ Xiapu, for Pb and Cd were most concentrated in Fuding coastal wetlands. 15Screening of Serum Biomarkers for Distinguishing between Latent and Active Tuberculosis Using Proteome Microarray显示文摘Objective To identify potential serum biomarkers for distinguishing between latent tuberculosis infection(LTBI) and active tuberculosis(TB). Methods A proteome microarray containing 4,262 antigens was used for screening serum biomarkers of 40 serum samples from patients with LTBI and active TB at the systems level. The interaction network and functional classification of differentially expressed antigens were analyzed using STRING 10.0 and the TB database, respectively. Enzyme-linked immunosorbent assays(ELISA) were used to validate candidate antigens further using 279 samples. The diagnostic performances of candidate antigens were evaluated by receiver operating characteristic curve(ROC) analysis. Both antigen combination and logistic regression analysis were used to improve diagnostic ability. Results Microarray results showed that levels of 152 Mycobacterium tuberculosis(Mtb)-antigenspecific IgG were significantly higher in active TB patients than in LTBI patients(P < 0.05), and these differentially expressed antigens showed stronger associations with each other and were involved in various biological processes. Eleven candidate antigens were further validated using ELISA and showed consistent results in microarray analysis. ROC analysis showed that antigens Rv2031 c, Rv1408, and Rv2421 c had higher areas under the curve(AUCs) of 0.8520, 0.8152, and 0.7970, respectively. In addition, both antigen combination and logistic regression analysis improved the diagnostic ability. 16The Optimal Cross-section Design of the “Trapezoid-V” Shaped Drainage Canal of Viscous Debris Flow显示文摘Debris flow drainage canal is one of the most widely used engineering measures to prevent and manage debris flow hazards.The shape and the sizes of the cross-section are important parameters when design debris flow drainage canal.Therefore,how to design the appropriate shape and sizes of the cross-section so that the drainage canal can have the optimal drainage capacity is very important and few researched at home and abroad.This study was conducted to analyze the hydraulic condition of a Trapezoid-V shaped drainage canal and optimize its cross-section.By assuming characteristic sizes of the cross-section,the paper deduced the configuration parameter of the cross-section of a Trapezoid-V shaped debris flow drainage canal.By theory analysis,it indicates that the optimal configuration parameter is only related to the side slope coefficient and the bottom transverse slope coefficient.For this study,the Heishui Gully,a first-order tributary of the lower Jinsha River,was used as an example to design the optimal cross-section of the drainage canal of debris flow.YOU Yong 2011Journal of Mountain Science2011,8,1:8

18Factors associated with significant liver fibrosis assessed using transient elastography in general population显示文摘AIM: To investigate the prevalence of significant liver fibrosis assessed using transient elastography(TE) and its predictors in asymptomatic general population.METHODS: A total of 159 subjects without chronic viral hepatitis who underwent comprehensive medical health check-up between January 2012 and July 2012 were prospectively recruited. Significant liver fibrosis was defined as liver stiffness value > 7.0 k Pa.RESULTS: The mean age and body mass index(BMI) of the study population(men 54.7%) was 56.0 years and 24.3 kg/m2. Among the study subjects, 11(6.9%) showed significant liver fibrosis. On univariate analysis, BMI, alanine aminotransferase(ALT), homeostasis model assessment of insulin resistance, carotid intimal media thickness(IMT), number of calcified plaques on carotid ultrasound, and visceral fat area on computed tomography were significantly higher in subjects with significant liver fibrosis than in those without(all P < 0.05). However, on multivariate analysis, BMI [odds ratio(OR) =1.487; P = 0.045], ALT(OR = 1.078; P = 0.014), carotid IMT(OR = 3.244; P = 0.027), and the number of calcified carotid plaques(OR = 1.787; P = 0.031) were independent predictors of significant liver fibrosis. CONCLUSION: The prevalence of significant liver fibrosis assessed using TE was 6.9% in apparently healthy subjects. 20Release of VEGF and BMP9 from injectable alginate based composite hydrogel for treatment of myocardial infarction显示文摘Myocardial infarction(MI)is one of cardiovascular diseases that pose a serious threat to human health.The pathophysiology of MI is complex and contains several sequential phases including blockage of a coronary artery,necrosis of myocardial cells,inflammation,and myocardial fibrosis.Aiming at the treatment of different stages of MI,in this work,an injectable alginate based composite hydrogel is developed to load vascular endothelial active factor(VEGF)and silk fibroin(SF)microspheres containing bone morphogenetic protein 9(BMP9)for releasing VEGF and BMP9 to realize their respective functions.The results of in vitro experiments indicate a rapid initial release of VEGF during the first few days and a relatively slow and sustained release of BMP9 for days,facilitating the formation of blood vessels in the early stage and inhibiting myocardial fibrosis in the long-term stage,respectively.Intramyocardial injection of such composite hydrogel into the infarct border zone of mice MI model via multiple points promotes angiogenesis and reduces the infarction size.Taken together,these results indicate that the dual-release of VEGF and BMP9 from the composite hydrogel results in a collaborative effect on the treatment of MI and improvement of heart function,showing a promising potential for cardiac clinical application.Yong Wu Tianqi Chang Weiqian Chen Xiaoyu Wang Jingjing Li Yueqiu Chen You Yu Zhenya Shen Qian Yu Yanxia Zhang 2021Bioactive Materials2021,6,2:7
