| 6 | microRNA-18a Promotes Cell Migration and Invasion Through Inhibiting Dicer l Expression in Hepatocellular Carcinoma In Vitro显示文摘Objective To investigate the effects of microRNA-18a(miR-18a) on migration and invasion of hepatocellular carcinoma(HCC) cells,and its possible mechanism associated with Dicer l. Methods HepG 2 and HepG 2.2.15 cells were transfected with miR-18 a inhibitor using Lipofectamine. Cell invasion was evaluated by transwell invasion assay,and cell migration was detected by transwell migration and wound-healing assays. Moreover,luciferase reporter assay was used to identify whether Dicer expression was regulated by miR-18 a. Real-time RT-PCR and western blot were performed to analyze Dicer 1 expression. In addition,a functional restoration assay was performed to investigate whether miR-18 a promotes HCC cell migration and invasion by directly targeting Dicer 1. Results miR-18 a inhibitor can suppress the migration and invasion of HCC cells. Furthermore,suppression of Dicer l expression by small interfering RNA essentially abolished the inhibition of cell migration and invasion induced by miR-18 a inhibitor,restorating these activities to levels similar to the parental HCC cells. Interestingly,suppression of miR-18 a in HCC cells resulted in enhanced expression of Dicer l. In addition,the results of a luciferase assay demonstrated targeted regulation of Dicer l by miR-18 a. During the formation of glass network structure, Pb2 tprefers to break the Sie Oe Si bond and subsequently bond to F for charge compensation, and Cd2 tprefers to break the SieO eA l bond and is surrounded by O2. Pb2 tand F gather together and form the fluoride nanocrystals,while Cd2 tremains in oxide matrix after thermal treatment. | 15 | Copper-cysteamine nanoparticle-mediated microwave dynamic therapy improves cancer treatment with induction of ferroptosis显示文摘Photodynamic Therapy(PDT)holds a great promise for cancer patients,however,due to the hypoxic characteristics of most solid tumors and the limited penetration depth of light in tissues,the extensive clinical application of PDT is limited.Herein,we report microwave induced copper-cysteamine(Cu-Cy)nanoparticles-based PDT as a promising cancer treatment to overcome cancer resistance in combination with ferroptosis.The treatment efficiency of Cu-Cy-mediated microwave dynamic therapy(MWDT)tested on HCT15 colorectal cancer(CRC)cells via cell titer-blue cell viability assay and live/dead assay reveal that Cu-Cy upon MW irradiation can effectively destroy HCT15 CRC cells with average IC-50 values of 20μg/mL.The cytotoxicity of Cu-Cy to tumor cells after MW stimulation can be alleviated by ferroptosis inhibitor.Furthermore,Cu-Cy mediated MWDT could deplete glutathione peroxide 4(GPX4)and enhance lipid peroxides(LPO)and malondialdehyde(MDA).Our findings demonstrate that MW-activated Cu-Cy killed CRC cells by inducing ferroptosis.The superior in vivo antitumor efficacy of the Cu-Cy was corroborated by a HCT15 tumor-bearing mice model.Immunohistochemical experiments showed that the GPX4 expression level in Cu-Cy+MW group was significantly lower than that in other groups.Overall,these findings demonstrate that Cu-Cy nanoparticles have a safe and promising clinical application prospect in MWDT for deep-seated tumors and effectively inhibit tumor cell proliferation by inducing ferroptosis,which provides a potential solution for cancer resistance. | Hui Zhou Zhongtao Liu Zijian Zhang Nil Kanatha Pandey Eric Amador William Nguyen Lalit Chudal Li Xiong Wei Chen Yu Wen | 2023 | Bioactive Materials2023,,6: | 1 |

| 20 | Precipitate characteristics and their effects on the mechanical properties of as-extruded Mg-Gd-Li-Y-Zn alloy显示文摘In this work,a high-performance Mg-8.4 Gd-4.4 Li-3.5 Y-1.4 Zn(wt%)alloy was successfully prepared by low-temperature hot extrusion.The studied alloy has a TYS of 295±1 MPa and an EL of 2.5%±0.2%at room temperature,159±8 MPa and 15.9%±1.4%at 150℃,and 143±4 MPa and 19.8%±1.8%at200℃.The high performance at tempe ratures up to 200°is attributed to the dispersion strengthening of three Mg3 RE phase variants,the solid solution strengthening of alloying elements and the bimodal structure consisting of fine DRXed grains with random texture and coarse un-DRXed grains with basal texture.A novel island shapedβ_(1 R)phase(FCC,a=0.78 nm)with a size of 30 nm-100 nm is observed in the studied alloy.Theβ_(1 R)phase precipitates on the{1100}_(α)prismatic planes.The strain-induced,dislocationassisted dynamic precipitation during low-temperature hot extrusion is responsible for the formation of theβ_(1 R)phase.The growth of theβ_(1 R)phase is owing to the aid of the formation and transformation of zig-zag structures.The basal slip of dislocations is the dominate deformation mechanism of the studied alloy in the early stage of tensile deformation at room temperature,while non-basal slip of and dislocations at 200℃.Due to the strong interaction betweenβ_(1 R)precipitates and dislocations,the densely distributedβ_(1 R)precipitates significantly improve the mechanical properties via the Orowan strengthening both at room temperature and elevated temperatures. | Zijian Yu Xi Xu Adil Mansoor Baotian Du Kang Shi Ke Liu Shubo Li Wenbo Du | 2021 | Journal of Materials Science & Technology2021,,29: | 1 |
